Triple negative breast cancer (TNBC) is an aggressive and heterogeneous molecular subtype of breast cancer that has limited available treatments. The organometallic cationic ruthenium derivative Ru-IM was found to be highly water soluble, was differentially toxic for breast cancer cells and tumors, and was involved in the pI3K/AKT/mTOR pathway. However, a full understanding of genetic and epigenetic factors that can be part of regulatory mechanisms of Ru-IM is yet to be determined. Dragonflies and damselflies (Odonata Fabricius, 1793) are among the most widely recognized and charismatic insect groups, whose significance have permeated throughout artwork, culture, and religion. They are highly mobile predators, currently consisting of ~ 6,500 species, and possessing some of the complex visual color systems, visual acuity, and flight patterns on Earth. The vertebrate retina is a complex tissue made up of seven major cell subtypes which during development must differentiate and localize into a highly organized structure. These neuronal cells do not regenerate if damaged or diseased, and the progressive deterioration of cone photoreceptor cells is the leading cause of age related blindness. Currently there are no cures for this disease, but one promising therapeutic avenue is cell transplantation research. If we can further elucidate the gene regulatory networks that define cone cell development, this knowledge could be applied to the generation of healthy cone cells in clinically relevant models.

Psychopathy is a complex personality disorder characterized by distinct emotional and interpersonal deficits, and affects approximately 25% of the prison population and 4.5% of nonincarcerated populations. The manifestation of psychopathy can be understood through two distinct variants, each demonstrating unique etiological pathways and clinical presentations. Primary psychopathy is characterized by core interpersonal and affective deficits, including shallow emotions and manipulativeness, while demonstrating reduced autonomic responsivity and lower anxiety levels. Secondary psychopathy presents with higher levels of impulsivity, anxiety, and emotional dysregulation, showing stronger associations with various psychopathologies. Despite the severity, 98% of drugs have failed as treatments for AD. Alzheimer’s Disease (AD) is a complex neurodegenerative disease that has limited treatment options and no cure. AD is characterized by robust pathology including extracellular amyloid beta plaques, intracellular neurofibrillary tangles, neuronal and synaptic loss, neuroinflammation, and brain atrophy. AD presents with cognitive decline and memory loss but only years after neuropathology has started to accumulate. Osteosarcoma is a rare but aggressive bone malignancy most commonly affecting adolescents and young adults (AYA). With 5-year survival rate stagnating over the last four decades despite the advancements in multi-modal therapy, drug repurposing offers a rapid path to new osteosarcoma treatments. Recent studies suggest that Riluzole, a drug approved for amyotrophic lateral sclerosis (ALS), may be repurposed for osteosarcoma treatment due to its ability to slow tumor progression and induce apoptosis. The Bone Morphogenetic Proteins (BMPs) are secreted peptide ligands of the Transforming Growth Factor beta (TGF-β) family, initially identified for their roles in development and differentiation across animal species. They are now increasingly recognized for their roles in physiology and infectious disease. In the nematode Caenorhabditis elegans, the BMP ligand DBL-1 controls fat metabolism and immune response, in addition to its roles in body size regulation and development. DBL-1 regulates classical aspects of innate immunity, including the induction of anti-microbial peptides. Inhibition of Return (IOR) and Attentional Blink (AB) are two distinctly defined phenomena that display some intriguingly similar properties. They both describe suppression effects following a visual stimulus that has captured attention, both may be preceded by an enhancement, and both share partially overlapping effect time windows which can be manipulated by task difficulty. Anorexia nervosa (AN) is a severe psychiatric disorder with the highest mortality rate among mental health conditions, yet effective biological treatments remain limited. The activity-based anorexia (ABA) paradigm, which combines food restriction with wheel access to model the hyperactivity and self-starvation cycle characteristic of AN, shows considerable individual variability in vulnerability. The striatum is a central node in action selection and associative learning, integrating cortical, thalamic, and neuromodulatory input. Among its principal modulators, dopamine (DA) and acetylcholine (ACh) are known for their temporally coordinated yet mechanistically distinct influences on striatal output. However, the upstream signals that regulate their dynamic interplay, particularly within cholinergic interneurons (CINs)—the primary source of striatal ACh—remain incompletely characterized. The Southwestern US and northern Mexico continue to serve as ideal regions to elucidate the suite of ecological and evolutionary processes influencing lineage diversification. The complex geological history of arid North America, coupled with paleoclimate change during the Pleistocene and diverse ecoregions makes a multipronged approach to hypothesis testing necessary. We combine thousands of loci from a genotyping-by-sequencing (GBS) approach along with mitochondrial DNA (mtDNA) sequences from the Greater Earless Lizard (Cophosaurus texanus) to examine diversity and test for cryptic population structure. This project investigated the accumulation of DNA on duct tape rolls stored in cars. Specifically, we analyzed samples recovered from three areas (the inside, outside, and flat sides) of unused duct tape stored under the driver's seat and trunk of cars for seven days. Twelve volunteers were recruited, resulting in 64 DNA samples from 12 cars. We evaluated DNA quantities and STR genotyping outcomes after amplification with the GlobalFiler™ kit. On average, the DNA yields from under the driver's seat were higher than from the trunk, but the difference was not significant. The inside of the tape roll yielded significantly …

Fast-growing plant species, like Lantana camara and Acanthus pubescens, could threaten biodiversity in Kibale National Park, Uganda. I surveyed plants and wildlife in areas where these plants were frequently encountered compared to those areas where they are infrequent. I found mixed results based on plant traits and the animals considered. My findings support management efforts on these two species to prevent forest degradation.

Monozygotic (MZ) twins possess identical DNA sequences, posing a problem for forensic scientists, as they are incapable of distinguishing MZ twins from each other using traditional DNA identification methods. However, the epigenomes of MZ twins will differ and might be used for their forensic discrimination. Here, we explored the variability of an assay previously developed by our laboratory that showed high discrimination potential for MZ twins using buccal swab DNA, cg18562578 MethylQuant. Monozygotic (MZ) twins have identical DNA, making traditional profiling methods insufficient in their discrimination. However, MZ twins can be differentiated by their levels of DNA methylation. Our research group previously developed a methodology to detect cg18562578 DNA methylation levels in MZ twins based on a two- step PCR approach and a cg18562578 MethylQuant Assay. That study generated data for only 64% of the samples it investigated, and showed a discrimination power of 77% to discriminate between MZ twins. Organic acids, as natural metabolites, play crucial roles in human metabolism and health. Tumor Necrosis Factor (TNF), a pivotal mediator in immune regulation and inflammation, is a key therapeutic target. We evaluated ten organic acids as TNF modulators using in silico molecular docking, followed by detailed ADMET (Absorption, Distribution, Metabolism, Excretion, and Toxicity) profiling and molecular dynamics (MD) simulations for three lead candidates: gallic, aconitic, and crocetin acids. This collaborative class zine assignment uses a shared PowerPoint format to deepen student learning while building community. Students contribute visual reflections (graphics, diagrams, quotes, or doodles) weekly or periodically throughout the semester. Faculty provide reflection prompts about key takeaways and unclear concepts (sample prompts included). The resource includes both sample LMS instructions and a ready-to-use PowerPoint zine template that works across all disciplines. By combining metacognitive reflection with creative expression, students consolidate learning by finding new connections and applications. The collaborative format creates community connections and joy while students learn from peers' diverse perspectives.

With lower detection limits for forensic DNA analysis, it is important to have a better understanding of background DNA levels unrelated to the crime to facilitate accurate data interpretation. Duct tape is used in crimes, e.g. to bind victims and seal drug packaging, and may be tested for DNA. To determine DNA background levels, twelve volunteers were recruited to donate a used roll of duct tape from their home. Str Analysis Performance Comparison For Two Capillary Electrophoresis Instruments: The Seqstudio And The 3500 Genetic Analyzers, Yana Leichyk

This study compares the performance of two capillary electrophoresis instruments—the SeqStudio™ Genetic Analyzer and the 3500™ Genetic Analyzer—for forensic short tandem repeat (STR) analysis. Using the GlobalFiler STR kit and certified reference material (SRM 2391d), five key performance metrics were evaluated: sizing precision, signal reproducibility, detection sensitivity, mixture resolution, and overload tolerance. STR profiles were generated from single-source and mixture samples with DNA inputs ranging from 500 pg to 15.6 pg. Both instruments met the < 0.15 bp sizing precision requirement for forensic casework. Signal reproducibility tests showed that the SeqStudio often produced higher average RFU values, but with greater variability, while the 3500 demonstrated more stable signal performance across days and samples. Mixture detection was effective on both platforms up to 1:10 ratios. Dropout of minor alleles began to appear at 1:20 ratios, especially in green and yellow dye channels, on both instruments. Sensitivity studies confirmed full profile recovery down to 62.5 pg, with increasing allelic and locus dropout at lower inputs. Enhanced injection voltage (1.5 kV) did not improve low-template performance. In overload testing (1–5 ng input), the 3500 maintained a consistent signal output, while the SeqStudio exhibited signal suppression at 5 ng, likely due to internal normalization. The SeqStudio exhibited better spectral overlap compensation, with fewer occurrences of spectral bleed-through artifacts. These findings support the use of both platforms in forensic casework, while emphasizing the importance of platform-specific calibration and analytical threshold settings for optimal performance.

Recognizing that libraries serve multiple constituencies and subject areas, this chapter documents and advocates for development of transdisciplinary arts-based research (ABR) and culture-related projects linked to environmental challenges. Libraries contribute collections and spaces, as well as the research of library and information scientists. Libraries are currently among invisible contributors to sustainability planning and services. The chapter will link this invisibility to the value of what visual arts refer to as negative space elements in subjects ranging from traditional ecological knowledge to environmental science. The ability to differentiate between threat and safety cues is essential for adaptive behavior, yet this process is disrupted in psychiatric disorders, such as post-traumatic stress disorder (PTSD), where safety cues fail to downregulate fear expression. To better understand the neurobiological mechanisms of safety learning, I investigate how safety conditioning vs contextual fear conditioning modify behavioral and cellular mechanisms in the infralimbic (IL) region of the medial prefrontal cortex (mPFC) at several time points after learning. Through this work, I identify a novel mechanism whereby safety learning promotes learning dependent neuro-glial interactions that promote conditions for plasticity in the mPFC. …

Parasites, and the infectious diseases they carry, are thought to have significantly impacted our evolutionary trajectory for millennia. Exploring how parasites are acquired and transmitted by their hosts in wild, natural populations is integral to understanding how similar processes may have shaped host-parasite co-evolutionary relationships over time. However, parasitological studies in humans and in non-human primates have produced inconsistent results, highlighting the dynamic, nuanced, and unique relationships individual parasites have with their hosts. Translation initiation in eukaryotes is a highly regulated process essential for accurate protein synthesis. It is a dynamic process that involves a complex interplay between messenger RNAs (mRNAs), ribosomal subunits, and a host of initiation factors, ensuring precise start codon selection and the subsequent assembly of the translation machinery.
