Chulalongkorn University Theses and Dissertations (Chula ETD)
Carbapenems are regarded as a last-resort option for treating a wide range of Gram-positive and Gram-negative bacterial infections. Unfortunately, the prevalence of carbapenem-resistant genes has been on the rise among patients. Some factors contribute to this trend, including prolonged hospital stays, prior antibiotic usage, inappropriate or insufficient antibiotic treatment, and contamination through wounds or feces. The emergence of carbapenem-resistant (CR) genes gained significant attention, particularly after 2005. These CR genes are frequently carried on mobile genetic elements such as plasmids or transposons, enabling their transmission to other bacteria. Klebsiella pneumoniae carbapenemase (KPC) and New delhi metallo-β-lactamase (NDM) stand out as …

Chulalongkorn University Theses and Dissertations (Chula ETD)
Heat shock proteins (HSPs) are host chaperones that can be used by viruses to facilitate various steps of viral infections. In this study, we found that Litopenaeus vannamei heat shock protein 90 (LvHSP90) is a viral-responsive protein, due to a significant transcriptional and translational upregulation following white spot syndrome virus (WSSV) infection. Suppression of LvHSP90 significantly reduced WSSV infection which was corresponded with significantly decreased expressions of viral genes, including the immediate-early genes and viral DNA polymerase. Conversely, injecting shrimp with WSSV that had been co-incubated with a recombinant LvHSP90 (rLvHSP90) promoted WSSV infection as evidenced by an increased cumulative …

Role Of Grb2 Intracellular Localization In Progesterone Receptor Positive Breast Cancer, Nattamolphan Wittayavimol
Role Of Grb2 Intracellular Localization In Progesterone Receptor Positive Breast Cancer, Nattamolphan Wittayavimol
Chulalongkorn University Theses and Dissertations (Chula ETD)
Progesterone receptor (PR) extranuclear signaling mediates mainly through PR polyproline domain (PPD) interaction with SH3 domain of cytoplasmic signaling molecules. PR-PPD interfered with Grb2-SOS binding through SH3 interaction resulting in decreased cell proliferation and correlates with a good prognosis in PR positive breast cancer. Grb2 is a key adaptor molecule and expressed in both nucleus and cytoplasm. We hypothesized that change in Grb2 localization alters PR transcription resulting in better clinical outcomes in PR positive breast cancer. BT-474 breast cancer cells expressing inducible Grb2-SV402x (nuclear Grb2, nuGrb2) or Grb2-MYR (membrane Grb2, memGrb2) were constructed. MemGrb2-BT-474 showed higher levels of PR …

Viral Rna M6a Methylome Profilingusing Nanopore Direct Rna Sequencing, Thada Sukprasong
Chulalongkorn University Theses and Dissertations (Chula ETD)
N6-methyladenosine (m6A) modification is the most common RNA modification in eukaryotic mRNAs and viruses, playing a pivotal role in gene regulation. Currently, nanopore sequencing has emerged as a technique capable of identifying m6A positions by observing differences in ionic currents between wild-type and METTL3-knockout samples. This method sequences native RNA strands directly, bypassing reverse transcription. The xPore program compares the ionic currents from both samples to determine m6A positions and their modification rates. However, obtaining METTL3-knockout RNAs is not always achievable. To address this limitation, m6Anet predicts m6A positions and provides the probability of m6A sites using only wild-type RNA. …
Metabolomic Analysis Of Lungs From Breast Cancer Metastatic Mice Models Treated With Thai Traditional Medical Formulary, Kotchanipa Rukruam
Metabolomic Analysis Of Lungs From Breast Cancer Metastatic Mice Models Treated With Thai Traditional Medical Formulary, Kotchanipa Rukruam
Chulalongkorn University Theses and Dissertations (Chula ETD)
Triple-negative breast cancer (TNBC) is a highly invasive subtype, leading to lung metastasis in 40% of patients with TNBC metastasis. Lung metastases significantly reduce median survival compared to other breast cancer types. Hence, there are efforts to investigate TNBC treatments, including modern and alternative therapies. The Thai traditional medical formulary (TTMF) is broadly used as an alternative treatment for Thai cancer patients. However, there are no studies on metabolic changes after treatment in advanced TNBC. Additionally, obtaining metabolites from biological samples is challenging due to the lack of a universal protocol for metabolite extraction. Consequently, this thesis aimed to first …
Genome Characterization And Significant Gene Annotation Of Mycobacteria Based On Data From High-Throughput Dna Sequencing Technology, Ajala Prommi
Chulalongkorn University Theses and Dissertations (Chula ETD)
Mycobacteria encompass a variety of species, including pathogenic strains such as Mycobacterium tuberculosis complex (MTBC) and nontuberculous mycobacteria (NTM). These organisms are responsible for significant health challenges globally and in Thailand, particularly due to tuberculosis (TB) and its drug-resistant forms. This dissertation aims to characterize the whole genome of various Mycobacterium species and annotate significant genes utilizing high-throughput DNA sequencing technology. Key objectives include investigating the mutations in Mycobacterium tuberculosis responsible for resistance to primary anti-TB drugs, specifically isoniazid, and analyzing the resistome of Mycobacterium abscessus, a rapidly growing NTM, isolated from patients in Thailand. The study also seeks to …

Identification And Characterization Of Genes Involved In Antioxidant Traits In Local Thai Rice (Oryza Sativa L.), Putut Rakhmad Purnama
Chulalongkorn University Theses and Dissertations (Chula ETD)
Developing rice (Oryza sativa L) cultivars with high antioxidant activities have become increasingly important since they have nutritional advantages for human health. Hence, the researchers must precisely elucidate the genetic bases controlling antioxidant-related traits in rice grains. In this study, we aimed to identify the genes involved in antioxidant traits in local Thai rice. The first strategy was conducting a genome-wide association analysis (GWAS) using 233 accessions of local Thai rice. A total of 119,541 single nucleotide polymorphic (SNP) markers was used as genotype data. In contrast, the five antioxidant assays, ABTS, DPPH, FRAP, TFC, and TFC, were used as …

Effects Of Salinity Changes On Osmotic Properties And Oxidative Stresses In Razor Clam Solen Sp. In Don Hoi Lot Tidal Flat, Sappasith Dechkittithum
Chulalongkorn University Theses and Dissertations (Chula ETD)
Estuaries are important coastal ecosystems that serve as transition zones between freshwater and seawater. The Don Hoi Lot tidal flat, located in the upper Gulf of Thailand, is a crucial estuarine ecosystem in the country. However, this area faces increasing environmental challenges from both human activities and natural changes. The razor clam, Solen regularis, is a notable species in this region. This study examined how salinity variations impact the osmotic properties and antioxidant enzyme activities in five tissues (foot, mantle, adductor muscle, gill, and digestive gland) of the razor clam S. regularis collected from Don Hoi Lot. Razor clams were …
