School of Veterinary and Biomedical Sciences: Faculty Publications

A small number (<2%) of mammalian retinal ganglion cells express the photopigment melanopsin and are intrinsically photosensitive (ipRGCs). Light depolarizes ipRGCs and increases intracellular calcium levels ( [Ca2+]i ) but the signaling cascades underlying these responses have yet to be elucidated. To facilitate physiological studies on these rare photoreceptors, highly enriched ipRGC cultures from neonatal rats were generated using anti-melanopsin-mediated plate adhesion (immunopanning). This novel approach enabled experiments on isolated ipRGCs, eliminating the potential confounding influence of rod/cone-driven input. Light induced a rise in [Ca2+]i (monitored using fura-2 imaging) in the immunopanned ipRGCs and the source of this Ca2+ signal was investigated. The Ca2+ responses were inhibited by 2-aminoethoxydiphenyl borate, SKF-96365 (1–2-(4-methoxyphenyl)-2-[3-(4-methoxyphenyl)propoxy]ethyl-1H-imidazole), flufenamic acid, …

Enhancement Of Efficacy Of Prrsv Vaccines By Altering The Glycosylation Pattern Of Viral Glycoproteins, Asit K. Pattnaik Oct 2007

Enhancement Of Efficacy Of Prrsv Vaccines By Altering The Glycosylation Pattern Of Viral Glycoproteins, Asit K. Pattnaik

School of Veterinary and Biomedical Sciences: Faculty Publications

PRRSV, the causative agent of PRRS is of major economic significance to the pork industry in the USA and around the world. Current commercial vaccine does not provide adequate protection against PRRSV outbreaks. Therefore, there is an urgent need for development of more efficacious vaccine to combat PRRS. Our previous studies have suggested that (i) induction of neutralizing antibody response is an important correlate of evaluating the efficacy of a vaccine; (ii) neutralizing antibodies can be enhanced by hypoglycosylation of the major surface glycoprotein (GP5). While it is known that GP5 plays a prominent role in neutralizing antibody induction, it …

Veterinary Science and Medicine Collection

This study examined stimulus–response properties of somatosensory receptors on the head of rainbow trout, Oncorhynchus mykiss, using extracellular recording from single cells in the trigeminal ganglion. Of 121 receptors recorded from 39 fish, 17 were polymodal nociceptors, 22 were mechanothermal nociceptors, 18 were mechanochemical receptors, 33 were fast adapting mechanical receptors and 31 were slowly adapting mechanical receptors. Mechanical thresholds were higher in polymodal nociceptors than in either slowly adapting or fast adapting mechanical receptors, whereas thermal thresholds of mechanothermal nociceptors were higher than those of polymodal nociceptors. Polymodal nociceptors and mechanochemical receptors gave similar responses to topical applications of …

Low Molecular Weight Protein Tyrosine Phosphatase (Lmw-Ptp) And Its Possible Physiological Functions Of Redox Signaling In The Eye Lens, Kuiyi Xing, Ashraf Raza, Stefan Lofgren, M. Rohan Fernando, Ye-Shih Ho, Marjorie F. Lou Aug 2007

Low Molecular Weight Protein Tyrosine Phosphatase (Lmw-Ptp) And Its Possible Physiological Functions Of Redox Signaling In The Eye Lens, Kuiyi Xing, Ashraf Raza, Stefan Lofgren, M. Rohan Fernando, Ye-Shih Ho, Marjorie F. Lou

School of Veterinary and Biomedical Sciences: Faculty Publications

Low molecular weight protein tyrosine phosphatase (LMW-PTP) was cloned from human lens epithelial B3 cells (HLE B3) and the recombinant enzyme was purified to homogeneity. The pure enzyme reacted positively with anti-LMW-PTP antibody, displayed tyrosine-specific phosphatase activity and was extremely sensitive to H2O2. The inactivated LMW-PTP could be regenerated by thioltransferase (TTase)/GSH system as demonstrated by both activity assay and by mass spectrometry (MS). The MS study also showed that an intramolecular disulfide bond was formed between C13 and C18 at the active site, and was reduced by the TTase/GSH system. The putative role of LMW-PTP in regulating platelet derived …

A Comparison Of Pyronin Y-Methyl Green Stain And Methylene Blue Stain For Somatic Cell Count In Sheep Milk, Emily Mirek, Stacey O’Donnell

Senior Honors Projects

Somatic cell count is a key method used to evaluate the quality of milk in today’s growing dairy sheep industry. Somatic cells are body cells, primarily the white blood cells, found in a milk sample. If an infection such as mastitis is present, the number of somatic cells in the milk increases (Gonzalo, et al. 1992). Producers routinely perform somatic cell counts on cow and goat milk. For dairy cattle in the United States, the somatic cell count can not exceed 750,000 cells per milliliter, for commercial milk. For dairy goats the accepted limit is 1,000,000 cells per mL of …

The Human/Animal Interface: Emergence And Resurgence Of Zoonotic Infectious Diseases, Michael Greger

Transgenesis Collection

Emerging infectious diseases, most of which are considered zoonotic in origin, continue to exact a significant toll on society. The origins of major human infectious diseases are reviewed and the factors underlying disease emergence explored. Anthropogenic changes, largely in land use and agriculture, are implicated in the apparent increased frequency of emergence and reemergence of zoonoses in recent decades. Special emphasis is placed on the pathogen with likely the greatest zoonotic potential, influenzavirus A.

Turkish Journal of Veterinary & Animal Sciences

The aim of this study was sex determination in Nymphicus hollandicus without giving it any harm and obtaining accurate results by DNA analysis. CHD genes are preserved within avian Z and W sex chromosomes. The intron regions of the CHDW and CHDZ genes vary between male (ZZ) and female (ZW) individuals. The method used in this study was based on this difference. DNA was extracted from feathers instead of blood. The intron regions of CHDW and CHDZ genes were amplified by sex specific primers (P2 and P8). PCR products were screened by agarose gel electrophoresis. Individuals showing double (ZW) and …

Effect Of Feeding Supplemental Tallow On The Performance Of Lactating Nili-Ravi Buffaloes, Haq Nawaz, Muhammad Yaqoob, Muhammad Abdullah Jan 2007

Effect Of Feeding Supplemental Tallow On The Performance Of Lactating Nili-Ravi Buffaloes, Haq Nawaz, Muhammad Yaqoob, Muhammad Abdullah

Turkish Journal of Veterinary & Animal Sciences

In order to determine the feeding value of tallow for lactating Nili-Ravi buffaloes, 4 buffaloes were fed 0%, 2%, 4%, or 6% tallow in a 4 X 4 Latin square experiment. The intakes of dry matter (DM), organic matter (OM), crude protein (CP), acid detergent fiber (ADF), and neutral detergent fiber (NDF) decreased (P < 0.01), but the intakes of ether extract (EE) (P < 0.01) and digestible energy (DE) (P < 0.05) increased with increasing levels of tallow in the diets. Intake of net energy for lactation (NE_L) did not differ (P < 0.01) with varying levels of supplemental tallow. Daily milk yield increased (P < 0.01) from 11.00 to 13.2 kg/day. Production of 4% fat corrected milk (FCM), solid corrected milk (SCM), and energy corrected milk (ECM) increased quadratically (P < 0.05) with the increasing level of tallow in the diets. Milk fat content and total milk fat increased linearly (P < 0.01) and quadratically (P < 0.05), respectively, as the level of tallow in the diet increased. There was no difference in protein and lactose content due to feeding tallow. Total solid content increased quadratically (P < 0.05) from 16.45% to 17.67% but solid not fat (SNF) percentages did not vary with varying levels of tallow. The gross energy (GE) of the milk increased from 1096 to 1160 Kcal/kg (P < 0.01) with increasing levels of tallow in the diets. Energetic efficiency of milk production improved in a quadratic (P < 0.05) manner with 2%, 4%, and 6% tallow, with the highest being 47.6% with 4% supplemental tallow. The proportion of C_{8:0} to C_{16:1} fatty acids (FA) decreased from 53.66% to 35.52%, whereas the concentration of C_{18:0} to C_{20:0} increased (44.93% to 62.84%) in the milk fat of buffaloes fed diets containing different levels of tallow. Gains in body weight (BW) were higher (P < 0.05) in buffaloes fed diets containing supplemental tallow as compared to those fed the control diet. This study suggests that tallow up to 6% of diet DM is a suitable fat supplement as an economical energy source for lactating buffaloes.

Investigation Of Some Biochemical Parameters And The Antioxidant System In Calves With Dermatophytosis, Mahmut Karapehli̇van, Erdoğan Uzlu, Necati̇ Kaya, Orhan Kankavi̇, Kerem Ural, Mehmet Çi̇ti̇l

Turkish Journal of Veterinary & Animal Sciences

The study included 10 clinically healthy calves (group 1) and 10 clinical cases of dermatophytosis (group 2). The influence of dermatophytosis on some serum biochemical profiles and the antioxidant system was investigated. Serum malondialdehyde (MDA), reduced glutathione (GSH), uric acid, nitric oxide (NO), total sialic acid (TSA), lipid-bound sialic acid (LBSA), total protein (TP), and albumin levels were measured in groups 1 and 2. Calves in group 2 had significantly higher (P < 0.001) MDA, NO, TSA, and LBSA levels, and a higher P < 0.01 TSA:TP ratio; however, GSH and uric acid levels were significantly lower (P < 0.001). There was no significant difference between the groups in total protein and albumin. These findings suggest a relationship between dermatophytosis, the antioxidant system, and lipid oxidation.
