The tallgrass prairie was once one of the most diverse grasslands on the planet, however over 95% has been converted to agriculture in the past 150 years. The remaining prairie is still host to a wide variety of plants, insects, mammals, and soil-dwelling microorganisms, creating an incredibly complex ecosystem. The first aim of this study is to compare three approaches for the measurement of nematode diversity within a 10-mile-long protected prairie habitat corridor near Lincoln, Nebraska. Corn smut is caused by the fungus Ustilago maydis. Smut rarely occurs in Utah and is not considered economically important for grain production. For home gardeners and small farms, serious smut infections on sweet corn can cause ears to be inedible. U. maydis spreads from plant to plant by wind-borne or water-splashed spores onto silks or through existing wounds. Therefore, keeping corn plants insect- and disease-free reduces the number of entry points for infections. This fact sheet reviews the disease cycle, identification, and management.

Human APOBEC3 (apolipoprotein B mRNA-editing catalytic polypeptide-like 3) enzymes are capable of inhibiting a wide range of endogenous and exogenous viruses using deaminase and deaminase-independent mechanisms. These enzymes are essential components of our innate immune system, as evidenced by (a) their strong positive selection and expansion in primates, (b) the evolution of viral counter-defense mechanisms, such as proteasomal degradation mediated by HIV Vif, and (c) hypermutation and inactivation of a large number of integrated HIV-1 proviruses. Roseau cane (Phragmites australis) is a wild plant with an extensive root system that is useful in areas vulnerable to soil erosion. In Louisiana, this plant plays an important role protecting the coastal infrastructure of the lower Mississippi River delta (MRD). The legume Oxytropis sericea hosts a fungal endophyte, Alternaria oxytropis, which produces secondary metabolites (SM), including the toxin swainsonine. Polyketide synthase (PKS) and non-ribosomal peptide synthase (NRPS) enzymes are associated with biosynthesis of fungal SM. To better understand the origins of the SM, an unannotated genome of A. oxytropis was assessed for protein sequences similar to known PKS and NRPS enzymes of fungi. Contigs exhibiting identity with known genes were analyzed at nucleotide and protein levels using available databases. Spinach (Spinacia oleracea) is a self-pollinated, dioecious winter crop. Prevalent challenges to the production of spinach include disease pressure imposed by downy mildew, which is caused by Peronospora effusa (=P. farinosa f. sp. spinaciae [Pfs]). A total of 19 new races of P. effusa have emerged, imposing serious challenges to the disease management in spinach production. Accordingly, this study was designed to explore the genetic components for establishing the basis of durable disease resistance development against the downy mildew pathogen (P. effusa 13) in spinach, through the use of various genome engineering approaches.

Rice is the staple food for more than half the world’s population, with nearly 500 million metric tons of rice consumed globally every year. Any risk of loss due to disease, waste, or spoilage is therefore noteworthy. This dissertation explores the passage of rice from paddy to pantry to plate, with special attention given to disease and storage life. The most important disease of rice is blast, caused by the ascomycete Magnaporthe oryzae, which can cause losses of ~30% every year. In the Philippines, phytoplasma disease symptoms of little leaf and witches’ broom have been observed in sponge gourd (Luffa sp.), bitter gourd (Momordica charantia) and bamboo (Bambusa vulgaris). However, the cause of the disease remained unidentified. Molecular analysis was conducted to detect the presence and identify the phytoplasma associated with the diseased plants. Nested PCR has detected the presence of phytoplasmas in sponge gourd, bitter gourd and bamboo showing symptoms of little leaf and witches’ broom. The genetic variability of Fusarium oxysporum f. sp. vasinfectum isolates from Gossypium barbadense L. seedlings were assessed using pathogenicity, mycelial compatibility, and RAPD analyses. The results showed that the disease incidence percentages and compatibility percentages considerably varied, extending from 33.2 to 86.6 % and 6 to 60 %, respectively in North Egypt and from 33.3 to 66.6 % and 0.0 to 20% in South Egypt. It also showed that the isolates ISO-8 and ISO-19 had the lowest disease incidence (33.2%) as compared with isolate ISO-5 (86.6%). Structurally and chemically diverse fatty acids, referred to as unusual or specialized fatty acids, are found in the seed oils of diverse plant species. Positive-strand (+)RNA viruses take advantage of the host cells by subverting a long list of host protein factors and transport vesicles and cellular organelles to build membranous viral replication organelles (VROs) that support robust RNA replication. Sphingolipids have roles as membrane structural components and as bioactive molecules in plants. In Physcomitrella patens, 4-hydroxysphinganine (phytosphingosine, t18:0) is the predominant sphingolipid long-chain base (LCB). To assess the functional significance of t18:0, CRISPR-Cas9 mutagenesis was used to generate mutant lines lacking the sole SPHINGOID BASE HYDROXYLASE (SBH) gene encoding the hydroxylase responsible for converting sphinganine (d18:0) to t18:0. Total sphingolipid content in sbh protonemata was 2.4-fold higher than in wild-type. Modest changes in glycosyl inositolphosphorylceramide (GIPC) glycosylation patterns occurred. Sphingolipidomic analyses of mutants lacking t18:0 indicated modest alterations in acyl-chain pairing with d18:0 in GIPCs and ceramides, but dramatic …

Community association populations are composed of phenotypically and genetically diverse accessions. Once these populations are genotyped, the resulting marker data can be reused by different groups investigating the genetic basis of different traits. Because the same genotypes are observed and scored for a wide range of traits in different environments, these populations represent a unique resource to investigate pleiotropy. The introduction of the Chinese chestnut blight in 1904 decimated native American chestnut (Castanea dentata (Marshall) Borkh.) populations. In this study we aim to: 1) document the location of individual chestnuts in our vicinity; 2) document healthy versus blight infected individuals; 3) characterize putative differences in the soil microbiome between infected and non-infected individuals, as well as between chestnuts and hardwood neighbors. With this information we hope to provide new insights into mechanisms that may enhance blight and fungal resistance in American chestnuts through the understanding of the interactions between the trees and their surrounding soil microbiome.

Understanding the genetic basis of complex traits is a fundamental goal of evolutionary genetics. Yet, the genetics controlling complex traits in many important species such as hemp (Cannabis sativa) remain poorly investigated. Because hemp’s change in legal status with the 2014 and 2018 U.S. Federal Farm Bills, interest in the genetics controlling its numerous agriculturally important traits has steadily increased. Potato blackleg and soft rot (PBSR), which can be caused by Dickeya spp. and Pectobacterium spp., is a serious problem worldwide. The recent outbreak of PBSR in the Northeastern USA, caused primarily by D. dianthicola, has resulted in significant economic losses since 2015. This seedborne disease is highly associated with and therefore spread by seed tuber distribution. In the early 2000s and in 2012-2013, viral disease devastated strawberry production in California and Florida respectively; causing multimillion dollar losses. Myxomycetes and dictyostelids are Amoebozoans that are cosmopolitan inhabitants of a variety of habitats, particularly forest environments. Both groups reproduce using spores which are primarily dispersed via wind in myxomycetes but this characteristic poses a problem for dictyostelids. The spores of dictyostelids are incased in a mucilaginous matrix that makes wind ineffective except in exceptional cases. It has been suggested that animals such as birds may play an important yet understudied role in the dispersal of these organisms. Cercospora spp. and Corynespora spp. are two common foliar fungal pathogens in Arkansas amongst other soybean producing areas. Two primary diseases caused by Cercospora spp. are Cercospora Leaf Blight (CLB, caused mainly by C. kikuchii) and Frogeye Leaf Spot (C. sojina). Both diseases affect foliage, and when lesions collapse, leaves may fall prematurely resulting in yield loss. In the specific case of CLB, this is a disease on the rise since 2000, and also causes seed infection reducing seed quality. The Sandhills of Nebraska is a complex ecosystem, covering 50,000 km2 in central and western Nebraska and predominantly of virgin grassland. Grasslands are the most widespread vegetation in the U.S. and once dominated regions are currently cultivated croplands, so it stands to reason that some of the current plant pathogens of cultivated crops originated from grasslands, particularly soilborne plant pathogens.
