Marine ectothermic fishes – fishes that do not regulate internal body temperature independent from the water temperature – are some of the most vulnerable organisms to rapid and alarming increases in environmental temperature occurring due to climate change mediated-ocean warming. For tropical fish species that live near their thermal maxima, this warming could be of grave consequence to individuals, populations, and whole marine ecosystems. Certain key life stages such as embryos and reproducing adults may be particularly vulnerable to warming given the excess energetic costs of these life stages, which may disallow compensation for extreme changes in temperature. Stressors have a negative impact on reproductive efficiency in humans and other animals, which has an economic cost due to infertility treatments for humans and reduced income for food producers. We wished to determine the molecular pathways by which metabolic disturbances and low-level inflammation impact the hypothalamic pituitary gonadal (HPG) axis using an in vitro model. To this end, we used the GT1-7 cell line, an immortalized line expressing the Kiss1R receptor that responds to kisspeptin stimulation with the release of GnRH to assess the impact of stressors on the GnRH-releasing cells. Female mimicry in the red-sided garter snake, Thamnophis sirtalis parietalis, is described in several well-studied evolutionary advantages, but an aspect that remains elusive is how reproductive traits are linked to the 3-fold higher circulating androgens in males with this phenotype. In this study, we implanted male garter snakes (n=15 per group) with either a blank implant (SHAM; control) or a T implant, the latter simulating the female mimic phenotype. Following simulated low-temperature dormancy, males were scored for courtship intensity over three days of behavioral trials with females housed in the same facility. In this body of research, reproductive and adrenal hormones and behavior were used to evaluate individual and group responses to physiological, social, and environmental changes in zoo-housed Asian elephants, considering factors of sex, age, and life stage. Animals experience physiological, social, and environmental changes as part of their natural history and individual life experience. Measures of both positive and negative states are needed to assess the impact of these changes at the individual and group level. While much effort has been put into understanding vitellogenesis in insects and other organisms, much less is known of this process in ticks. There are several steps that facilitate yolk formation in developing oocytes of which the vitellogenin receptor (VgR) is a key component. The tick VgR binds vitellogenin (Vg) circulating in the hemolymph to initiate receptor-mediated endocytosis and its transformation into vitellin (Vn). The conversion of Vg into Vn, the final form of the yolk protein, occurs inside oocytes of the female tick ovary. Vn is critical to tick embryos since it serves as the nutritional source for their …

Neurokinin B (NKB) and kisspeptin are critical for fertility and potent stimulators of gonadotropin-releasing hormone (GnRH) and luteinizing hormone (LH) secretion in sheep. We previously demonstrated that administration of NKB receptor (NK3R) agonist, senktide, within the retrochiasmatic area (RCh) resulted in surge-like release of LH and increased expression of kisspeptin neurons in the arcuate nucleus (ARC) of female lambs. We concluded that NKB-responsive neurons in the RCh stimulate GnRH secretion by inducing kisspeptin release from neurons in the ARC.

Quagga mussels (Dreissena rostriformis bugensis) originate from brackish waters of the Ponto-Caspian area and are nuisance invasive species in North American and European freshwaters. Their invasion has caused major economic and ecological damages in the Laurentian Great Lakes. Through selective filter feeding, quagga mussels have promoted the growth of cyanobacteria. Harmful cyanobacteria blooms have the potential to produce toxins, which can be toxic to humans and wildlife. Although quagga mussels can increase the prevalence of cyanobacteria blooms, it is unknown how they are physiologically affected by cyanobacteria and understanding these dynamics can be useful in the management of this invasive …

The goal of this study was to examine potential adaptation within two generations of Gulf killifish exposed to polycyclic aromatic hydrocarbons (PAH) while also investigating the use of mucus as an effective and non-lethal alternative to determine concentrations of 11-ketotestosterone (11-KT) and vitellogenin (VTG) of F. grandis. Multiple biomarkers were used to assess adaptation within these fish, such as lysozyme (enzyme critical in innate immunity), and 11-ketotestosterone and vitellogenin (endocrine hormones often affected by PAHs).

All vertebrates respond to stressful situations through the release of hormones called glucocorticoids (CORT). These hormones alter processes within the body to prioritize long term survival over immediate reproduction. It is hypothesized that this is helps individuals survive until conditions become more favorable for successful reproduction. Historically, stress was hypothesized to primarily to pull organisms out of reproductive life history stages when unfavorable conditions made offspring survival slim (otherwise known as the “CORT-tradeoff hypothesis”). EMS has been determined to influence the intrafollicular environment of mare ovaries. In humans, insulin resistance has been linked to polycystic ovaries as part of Polycystic Ovarian Syndrome (PCOS). In this dissertation I examined the relationship between individual energetic status, hormone production, and life history trait expression in field-active Timber Rattlesnakes, Crotalus horridus. In chapter one I reviewed what is known regarding these relationships in snakes and defined major research goals. In chapter two I described the seasonal profile of testosterone (T) and corticosterone (CORT) in relation to the breeding season and to individual energetic status in males. Results showed that the seasonal pattern of T production in C. horridus was different than other pit viper species with similar mating patterns. Testosterone was elevated in the months leading up …

Shark fisheries are a multimillion dollar industry in the United States and have significant contributions both recreationally and commercially. In order to maintain this industry, fisheries must be properly managed. An understanding of life history and reproduction is crucial in order to adequately manage shark fisheries. The finetooth shark, Carcharhinus isodon, is a member of the small coastal shark (SCS) fishery complex. The objective of this thesis was to evaluate the influence of steroids and gonadotropins on reproduction in beef cattle. In experiment 1, beef heifers were used to determine the influence of growth-promoting implants on growth, reproductive development, estrous behavior, and pregnancy rate. Heifers were assigned to 1 of 4 implant treatment groups: control (CON); trenbolone acetate (TBA); trenbolone acetate plus estradiol (TBA+E2) or zeranol (ZER). Heifers were implanted once, A.I. and exposed to bull during this experiment. Body weight, BCS, HH, RTS, estrous behavior and pregnancy data were collected throughout this experiment. Reports have linked energy balance along with adipocyte derived leptin action to improved fertility. Recent evidence indicates that leptin hormone is present in breast milk and leptin receptors are well expressed in mammary epithelial cells. The hypothesis that insufficiency of leptin restraint in the hypothalamus may underlie infertility in rodents and the failure of lactating breast to express adequate amount of milk was tested. Female Sprague-Dawley rats were injected leptin through intracerebroventricular cannulation (ICVC) of the third ventricle. Female rats were mated with stud males and observed throughout gestation.
