The tallgrass prairie of North America has changed drastically since colonial settlement, with up to 99% of this region converted for agriculture and other land-uses. Concurrent with grassland conversion, grassland birds have experienced the most extreme, consistent, and widespread population declines of any avian guild. Agricultural lands in the U.S. Midwest were able to provide adequate habitat for several bird species until the 1950’s; however, altered and intensified management practices have degraded much of remaining suitable habitat and undermined ecosystem functions. Pneumonia is a major factor affecting populations of free-ranging bighorn sheep (Ovis canadensis) across western North America. Pneumonia can occur in large-scale epizootics, during which greater than half of the population typically dies. After these epizootics, surviving ewes continue to conceive and bear lambs. However, lamb recruitment may remain low due to periodic or annual pneumonia outbreaks causing high lamb mortality rates, sometimes greater than 90%. Our study focused on the Rapid City, South Dakota bighorn sheep (BHS) herd that has recorded pneumonia-induced population decline since 2009. Bighorn sheep (Ovis canadensis) and domestic sheep (Ovis aries) are members of the same genus and thus share multiple pathogens that can be spread between them. One specific respiratory pathogen of concern is Mycoplasma ovipneumoniae (M. ovipneumoniae), which has been linked to pneumonia epizootics in bighorn sheep that are often characterized by all age die offs upon initial exposure followed by years of low lamb recruitment. Domestic sheep have been identified as one carrier of M. ovipneumoniae with transmission between sheep species occurring when there is close contact on the landscape. The subterranean estuary (STE) is a subsurface ecosystem where fresh groundwater mixes with intruding, recirculated seawater at the coastline. Despite being present globally, STEs and their potential impacts as hotspots for biogeochemical processing, or as a source of nutrients to coastal systems, are not well understood. STEs have been recognized as highly reactive zones for biogeochemical reactions, which influence the fate and transport of nutrients, organic matter, and trace metals discharged to the coastal ocean. Biogeochemical processing of nitrogen (N) in STEs influences N in submarine groundwater discharge (SGD) including its availability for use by primary producers and cascading eutrophication. …

No-till is increasingly promoted as a vital agricultural practice to address the challenge of improving soil quality while maintaining the ability to produce food, feed, fuel, and fiber. Precision agriculture offers techniques and tools that can be used to help accommodate the growing population. One specific precision agriculture tool is remote sensing data, which can be used to image fields as an effort to better predict or understand the crops. Under this program all U.S. pork producers and importers pay $0.40 for every $100 worth of pigs is sold (Pork Checkoff). Phosphorus (P) pollution has become a concern among multiple scientific organizations as it leads to eutrophication, an algal bloom that depletes lacustrine and marine ecosystems of native species. Multiple strategies can be implemented to reduce phosphorus loss from agriculture fields, which is often implicated as a cause of eutrophication. Soil phosphorus chemistry results in phosphate fertilizers absorbing to clay minerals over time. Soil phosphorus is lost from agricultural fields primarily through wind and water erosion. The entrainment map, developed by Dr.Diekman and Dr.Bose, is claimed to be a 1-dimensional map that produces a better prediction for phase-locking than methods than the phase response curve for circadian models. Changes in winter conditions, such as increased temperatures and decreased ice coverage, have been observed worldwide. The responses of many lake fish populations to changing winters are projected to be inadequate to counter the speed and magnitude of climate change. Such environmental changes have been hypothesized to explain the low recruitment observed in freshwater whitefishes (Salmonidae Coregoninae). My research focused on measuring the impact changing winter conditions may have on coregonine reproductive phenology and developmental and morphological traits to better predict changes in coregonine populations as a result of climate change.

Grasslands historically covered 46 million km2 of Earth’s surface representing nearly 42% of the living vegetation. Encroachment of woody species onto grasslands and savannas is a widely researched global phenomenon, with eastern redcedar (ERC) (Juniperus virginiana L.) trees being the most prominent woody encroacher in the Northern Great Plains (NGP) due to the lack of prescribed fire and planted ERC escaping from shelterbelts. One of the remarkable steps in loline alkaloid biosynthesis is the installation of an ether bridge between two unactivated C atoms in 1-exo-acetamidopyrrolizidine (AcAP). LolO, a 2-oxoglutarate-dependent nonheme Fe oxygenase, catalyzes both the hydroxylation of AcAP and the resulting alcohol's cycloetherification to give N-acetylnornoline (NANL). The mechanism of hydroxylation is well understood, but the mechanism of the oxacyclization is not. I synthesized difluorinated analogs of AcAP in an attempt to further understand the mechanism of the unusual cycloetherification step.

In nature, DNA exists primarily in a highly compacted form. The compaction of DNA in vivo is mediated by cationic proteins; histone in somatic nuclei and arginine-rich peptides called protamines in sperm chromatin. The packaging in the sperm nucleus is significantly higher than somatic nuclei resulting in a final volume roughly 1/20th that of a somatic nucleus. This tight packaging results in a near crystalline packaging of the DNA helices. Autophagy is an essential recycling process that occurs within eukaryotic cells, however, the individual functions of the current thirty-two known autophagic proteins are not yet entirely understood. At this time, it is known that the autophagic protein Atg7 works upstream of both Atg3 and Atg10. Atg3 is affected by Atg7 and allows for the attachment of Atg8 and the lipid PE which is part of the autophagosome membrane. The Atg10 pathway is also affected by Atg7 but attaches Atg12 to Atg5 which upregulates the function of Atg3 and ultimately Atg8 lipidation.
