Developing A Protocol For Non-Cellular Transplantation Of Donor Mitochondria Into Recipient Cells, Andrew Parton
Developing A Protocol For Non-Cellular Transplantation Of Donor Mitochondria Into Recipient Cells, Andrew Parton
Honors Theses
Tumor innervation has emerged as a critical feature of cancer progression, with increased nerve density correlating with enhanced aggressiveness, metastatic dissemination, and poor clinical outcome. However, the functional contribution of neurons to tumor biology remains incompletely defined. We recently identified the intercellular transfer of mitochondria from neurons to cancer cells as a mechanism that promotes tumor progression. We therefore hypothesized that disruption of this transfer could attenuate tumor aggressivity. To directly investigate this process, we developed an approach to isolate mitochondria from donor cells and transplant them into recipient cancer cells independently of canonical cell-cell interactions. Transferred mitochondria were tracked …

Adventitial Macrophage Accumulation Impairs Perivascular Nerve Function In Mesenteric Arteries With Inflammatory Bowel Disease, Elizabeth A. Grunz, Benjamin W. Jones, Olubodun Michael Lateef, Sidharth Sen, Katie Wilkinson, Trupti Joshi, Erika M. Boerman
Biomedical Sciences
Introduction: Inflammatory bowel disease involves aberrant immune responses and is associated with both cardiovascular disease risk and altered intestinal blood flow. However, little is known about how inflammatory bowel disease affects regulation of perivascular nerves that mediate blood flow. Previous work found perivascular nerve function is impaired in mesenteric arteries with Inflammatory bowel disease. The purpose of this study was to determine the mechanism of impaired perivascular nerve function.
Methods: RNA sequencing was performed on mesenteric arteries from IL10−/− mice treated with H. hepaticus to induce disease (inflammatory bowel disease) or left non-gavaged (Control). For all other studies, Control and …
Fingernail Enhancement Microbiomes Study, Kendall E. Bare
Fingernail Enhancement Microbiomes Study, Kendall E. Bare
Undergraduate Honors Theses
Policies in healthcare regarding fingernail enhancements have not been supported by current science. This study examined bacterial colony growth on gel nail enhancements, regular nail polish, and bare natural nails to evaluate which condition harbors the least bacterial microbiota. The hypothesis proposed that gel enhancements would demonstrate the lowest bacterial counts compared to regular polish and unpolished nails. This investigation was conducted in the context of a local healthcare system dress code policy permitting only regular nail polish in clinical settings. Six participants volunteered and underwent nail swabbing in a microbiology laboratory using sterile cotton applicators and a neutral solution. …
Preclinical Animal Modeling To Investigate Redox-Mediated Molecular Dysfunction In Traumatic Brain Injury, Brandon Z. Mcdonald
Preclinical Animal Modeling To Investigate Redox-Mediated Molecular Dysfunction In Traumatic Brain Injury, Brandon Z. Mcdonald
Department of Agricultural and Biological Systems Engineering: Dissertations, Theses, and Student Research
Impacts associated with traumatic brain injury (TBI) induce a multitude of neurological morbidities mediated by a spread of biochemical dysfunction. Redox stress is a significant contributor to TBI pathophysiology and several redox stress markers have substantial evidence to support their role in diagnosing preclinical and clinical TBI. However, there is a critical need to verify their translational utility through correlations with behavioral, imaging, and molecular modalities. In chapter two, we examined the therapeutic efficacy of an antioxidant treatment (NPC3) using a controlled cortical impact (CCI) mouse model of TBI. NPC3 alleviated glial expression and reactivity while influencing the expression of …

Human Nutrition and Hospitality Management Undergraduate Honors Theses
Preventive strategies targeting modifiable lifestyle factors related to food, nutrition, and health are needed as chronic health conditions continue to rise. Nutrition literacy refers to having the knowledge and skills needed to obtain, understand, and apply nutrition information to make informed dietary decisions. Without adequate nutrition education and food security, vulnerable populations are at risk of malnutrition and poor dietary behaviors. Educational interventions that increase food literacy can help improve dietary choices and food selection skills.
The purpose of this creative project was to develop a nutrition education toolkit for Extension agents to implement among rural populations with low health …
The Impact Of Glucose Exposure On Organic Cation Transporters (Oct) Function In A Corneal Epithelial Cell Line, David A. Garza
The Impact Of Glucose Exposure On Organic Cation Transporters (Oct) Function In A Corneal Epithelial Cell Line, David A. Garza
Theses & Dissertations
Organic cation transporters (OCT) facilitate the movement of neurochemicals and medications, contribute to the removal of cellular waste, and facilitate neuronal communication. Three subtypes of these high-capacity, low-affinity transporters (OCT1, OCT2, and OCT3) are differently expressed in peripheral tissues such as the liver, kidneys, as well as the central nervous system. OCTs are also expressed in ocular tissues. Chronic hyperglycemia is a complication commonly associated with diabetes that can lead to ocular diseases such as diabetic keratopathy, which affects the cornea. In this project, we expose immortalized human corneal epithelial cells (HCE-S) to various glucose concentrations to study their impact …

Validation Of A Real-Time Pcr Laboratory Developed Test For The Detection Of Pneumocystis Jirovecii In Bronchoalveolar Lavage Samples, Caitlyn Moon
Honors Scholar Theses
Pneumocystis jirovecii is an opportunistic, host-obligate fungal pathogen responsible for causing P. jirovecii pneumonia (PJP) in humans. PJP is treated with trimethoprim-sulfamethoxazole (TMP-SMX), a strong broad-spectrum antibiotic that carries risk of renal and hepatotoxicity. Therefore, rapid diagnosis or exclusion of PJP is crucial to ensure PJP-positive patients receive effective treatment and PJP-negative patients avoid unnecessary antibiotics. Unlike most microorganisms, P. jirovecii cannot be cultured, presenting complications for the typical methods of diagnosing fungal pathogens. Historically, the gold standard for diagnosis was direct microscopic examination with stains such as methenamine silver and calcofluor white, but this diagnostic technique requires highly skilled …
The Development Of A Crispr-Based Calibrated Functional Assay For Classifying Pathogenicity Of Msh2 And Msh6 Variants In Lynch Syndrome, Olivia N. Amodeo
The Development Of A Crispr-Based Calibrated Functional Assay For Classifying Pathogenicity Of Msh2 And Msh6 Variants In Lynch Syndrome, Olivia N. Amodeo
Honors Scholar Theses
Lynch syndrome is a hereditary disease caused by the inheritance of a mismatch repair gene variant. Individuals with this condition are predisposed to cancer development, most commonly colorectal cancer. Current guidelines for variant classification are based on numerous evidence categories, including functional evidence. However, many novel clinical variants are not well characterized, and evidence is difficult to obtain if functional assays are not calibrated.
To address this, our lab created a calibrated functional assay that calculates an odds of pathogenicity score for MSH2 and MSH6 gene variants that can be used as evidence for classifying variants of uncertain significance. This …
Impact Of Inhibiting Mitochondrial Calcium Uptake On Calcium Homeostasis And Postmortem Metabolism In Porcine Muscle, Fatema Rahaman Mishu
Impact Of Inhibiting Mitochondrial Calcium Uptake On Calcium Homeostasis And Postmortem Metabolism In Porcine Muscle, Fatema Rahaman Mishu
All Graduate Theses and Dissertations, Fall 2023 to Present
After harvesting the animal, pork muscle continues to undergo changes as it turns from muscle into meat. During this early period, the muscle uses up energy and produces acid, which lowers pH and can affect important quality traits such as color and how well the meat holds water. A key driver of these early changes is calcium, a natural mineral inside muscle cells that helps control contraction and energy use. In this study, we tested whether mitochondria (the cell’s “powerhouse”) help control calcium after harvest and, in turn, influence early postmortem changes in pork. We used a research compound (DS16570511) …
Mechanisms Of Synergy Between Nanosecond Pulsed Electric Field (Nspef) And Cold Atmospheric Pressure Plasma Discharge (Cap) In Pancreatic Cancer: The Role Of Ros-Mediated Oxidative Stress And Apoptosis, Zobia Minhas
Biomedical Sciences Theses & Dissertations
Pancreatic ductal adenocarcinoma is a highly lethal malignancy characterized by poor prognosis and resistance to conventional therapies. Novel treatment strategies are needed to overcome this resistance. Non-thermal physical modalities, including nanosecond pulsed electric field (nsPEF) and cold atmospheric pressure plasma discharge (CAP), have emerged as potential anticancer approaches; however, the mechanisms underlying their combined effects remain unclear.
This study evaluated whether nsPEF enhances CAP-induced cytotoxicity and investigated the associated molecular mechanisms using the murine Pan02 pancreatic cancer cell line. Cell viability was assessed by WST-1 metabolic activity assays and electric cell–substrate impedance sensing (ECIS). Intracellular reactive oxygen species (ROS) were …

Pattern Dynamics And Stochasticity Of Brain Rhythms And Spike Trains In A Tauopathy Mouse Model Of Alzheimer’S Disease, Clarissa M. Hoffman
Pattern Dynamics And Stochasticity Of Brain Rhythms And Spike Trains In A Tauopathy Mouse Model Of Alzheimer’S Disease, Clarissa M. Hoffman
Dissertations and Theses (Open Access)
Systems neuroscience posits that every aspect of perceived physical reality, every aspect of animal and human behavior, and every cognitive phenomenon emerges from patterns of neuronal activity. While most researchers embrace this idea, there are major difficulties in describing and analyzing these complex neuronal dynamics—spike flows produced by cells ensembles, synchronized extracellular field oscillations, and other patterns—which limits our understanding of how the activity of individual neurons and the whole-animal cognition and behavior might be connected. In particular, we lack the approaches and even the semantics for connecting the individual cell outputs and the integrated results of their activity. Current …
Platelet Gαz Regulates Ovarian Tumor Growth And Mediates The Effects Of Chronic Stress, María G. Torres Carrizalez
Platelet Gαz Regulates Ovarian Tumor Growth And Mediates The Effects Of Chronic Stress, María G. Torres Carrizalez
Dissertations and Theses (Open Access)
Ovarian cancer is the most aggressive gynecologic cancer, with 12,450 estimated deaths in 2026. Elevated platelet counts are commonly observed in patients with ovarian cancer and are associated with enhanced tumor growth and poor prognosis. Patients with ovarian cancer are frequently exposed to elevated levels of stress, which has been correlated with accelerated disease progression and increased mortality. Interestingly, the platelet Gαz protein is activated by epinephrine through the α2A-adrenergic receptor, suggesting a potential mechanistic link between stress signaling and platelet-driven tumor progression. To investigate this, I evaluated the role of platelet Gαz in ovarian cancer using control (Gzfl/fl …
Abnormal Trafficking And Processing Of Multiple Matrix Metalloproteinases Drive Cartilage Defects In Congenital Disorders Of Glycosylation, Chia-Lun Wu
All Dissertations
Congenital Disorders of Glycosylation (CDG) are rare metabolic diseases caused by defects in glycosylation. Despite identification of over 200 CDG types, the mechanisms linking glycosylation defects to diverse clinical phenotypes remain unclear. This dissertation uses zebrafish models of PMM2-CDG and STT3-CDG to redefine CDG pathogenesis, shifting from a simple glycan deficiency model to one involving disrupted cellular spatial organization.
We identify a protease-dependent pathway underlying craniofacial cartilage defects. Specifically, defective proteolytic processing of N-cadherin, a key adhesion molecule in chondrogenesis, is a central driver of pathology. We further uncover an unconventional trafficking mechanism in which ER stress and altered secretory …
Alternative Abl1 Isoform Expression In Oncogenic Translocations: Implications For Cancer Progression, Jessica Teague
Alternative Abl1 Isoform Expression In Oncogenic Translocations: Implications For Cancer Progression, Jessica Teague
Biological Sciences
Abstract (also on poster): Previous studies in our lab show that an alternate isoform of Bruton’s tyrosine kinase, BTK-C, is required for the survival of several epithelial-derived cancer cells. BTK-C is transcribed from an alternative promoter and includes a 34-amino-acid N- terminal extension with a palmitoylation sequence that alters its subcellular localization. Nine other human kinases have similar alternative N-termini (ANT), often producing palmitoylated or non-palmitoylated isoforms from different transcriptional start sites. Among these, ABL1 is a known oncogenic kinase and key drug target. Translocation mutations at the 5′ end of ABL1 are a major cause of adult leukemias. This …

Purpose: Radiation therapy (RT) has emerged as an effective local therapy option for patients with unresectable hepatocellular carcinoma (HCC) and is associated with improved overall survival (OS). However, some patients still have poor outcomes after RT, and biomarkers are needed to stratify prognostic groups. High circulating growth hormone (GH) levels promote HCC proliferation and survival and are associated with aggressive disease. In this study, we evaluated whether pretreatment GH levels are associated with OS and progression-free survival (PFS) in patients who underwent RT for unresectable HCC.
Methods and materials: Patients undergoing RT for HCC during 2017-2023 were identified from a …

From Invisible Injury To Measurable Impact: Exploring Associations Between Peripheral Biomarkers Of Neuroinflammation And Mental Health Outcomes After Mtbi, Lauren Leggett Smith
Psychology Theses & Dissertations
Mild traumatic brain injury (mTBI), often referred to as a “silent epidemic,” accounts for the majority of traumatic brain injuries worldwide and is increasingly linked to persistent psychiatric outcomes. Despite its prevalence, mTBI remains understudied due to diagnostic challenges, underreporting, and misperceptions of its clinical significance. The present study examined whether peripheral biomarkers of neuroinflammation and astroglial injury—tumor necrosis factor-alpha (TNF-α), interleukin-6 (IL-6), and glial fibrillary acidic protein (GFAP)— mediate associations between mTBI history and psychiatric symptom severity, including depression, anxiety, and posttraumatic stress disorder (PTSD). Participants were 138 traumaexposed adults recruited from an urban university and surrounding community, with …
Understanding The Role Of Toxs As A Bile Sensing Component Of The Toxrs Virulence Regulatory System In Vibrio Spp., Minje Kim
Dartmouth College Ph.D Dissertations
Pathogenic Vibrio bacteria, such as V. cholerae and V. parahaemolyticus, colonize in the human small intestine to cause severe gastrointestinal disease. During the infection, these bacteria encounter bile salts, antimicrobial cholesterol metabolites secreted into the intestine. Pathogenic Vibrio species have evolved to utilize bile salts as signals to regulate virulence. The signaling depends in part on ToxRS, a conserved co-component transmembrane transcription regulator. ToxRS consists of the transcription factor ToxR and its membrane-tethered binding partner ToxS. ToxS is required for ToxR stability and full transcriptional activity. Although bile salts are known to influence ToxRS-dependent virulence gene expression, the molecular …
Molecular Mechanisms Of Pfas Induced Protein Dysfunction: Implications For Neurodegenerative Risk, Daisy Lee Wilson
Molecular Mechanisms Of Pfas Induced Protein Dysfunction: Implications For Neurodegenerative Risk, Daisy Lee Wilson
Open Access Theses & Dissertations
Per- and polyfluoroalkyl substances (PFAS) are persistent environmental contaminants with widespread human exposure and well-established links to adverse health outcomes. However, despite increasing epidemiological and experimental evidence, the molecular mechanisms underlying PFAS-induced neurotoxicity remain incompletely defined. This dissertation investigates PFAS toxicity across biological scales, integrating protein biophysics with neuronal cell models to elucidate pathways relevant to neurodegeneration. At the molecular level, we demonstrate that PFAS disrupts the structure and function of globular proteins, including the impairment of α-lactalbumin's calcium-binding capacity and of β-lactoglobulin's retinol binding, through concentration-dependent conformational destabilization. By applying these findings to a neuronal model, we show that …

Prediabetes Prediction Before Disease Onset Using Multimodal Health Data: A Machine Learning Approach, Luisa Veronica Gracia Mazuca
Open Access Theses & Dissertations
Prediabetes is a critical health condition that increases the risk of developing type 2 diabetes. The hemoglobin A1c (HbA1c) test diagnoses patients with prediabetes, but the disease has already caused metabolic alterations. Early detection is essential for timely interventions, and machine learning models offer a promising approach to identify prediabetic individuals through the analysis of biomarkers such as cytokines. We compared four classifiers-logistic regression, decision tree, random forest, and k-nearest neighbors - using cytokines (TNF-α, MCP-1, IL-1β, IL-6, IFN-γ), age, BMI, and waist-to-hip ratio (WHR). Models were evaluated using stratified 5-fold cross-validation and ROC-AUC. K-Nearest Neighbors (k-NN) achieved the highest …
Determining Zap-70 Signaling Pathways Mediated By Ccr7 In Tcr T Cell Activation, Vivian Torres
Determining Zap-70 Signaling Pathways Mediated By Ccr7 In Tcr T Cell Activation, Vivian Torres
Open Access Theses & Dissertations
Activation of T lymphocytes is the main event that occurs in the adaptive immune response after engagement of the T cell receptor (TCR) with the peptide of the major histocompatibility complex (pMHC) molecules that are demonstrated by antigen presenting cells [1]. A key step in this process is the recruitment and activation of the zeta chain associated protein of 70 kDa (ZAP-70). ZAP-70 couples TCR engagement to downstream intracellular signaling pathways through phosphorylation of tyrosine residues within intracellular tyrosine activation motifs (ITAMs) [2]. Although there have been extensive studies on the signaling response through TCR activation, the regulatory influence of …

Phase-change nanodroplets (PCNDs) continue to generate significant research interest due to their potential to extravasate into tissue, to be targeted for molecular imaging and drug delivery, and to undergo an induced phase-change to "activated" microbubbles (MBs) for ultrasound (US) imaging. To accurately quantify molecular markers, however, one assumes a consistent proportion of PCNDs in a region of interest (ROI) are stably activated and imaged. Herein we present a framework for developing a diagnostic sequence that is optimized for PCND activation uniformity, contrast, and acquisition time. To develop this framework, activation was examined at three scales of increasing complexity: single, adjacent, …

Investigating Arginase Activity In The Murine Macrophage Raw 264.7 Cell Line, Abigail G. Johnson
Undergraduate Theses and Capstone Projects
Macrophages are a type of white blood cell that have important functional roles in the innate immune system, the body’s first line of defense against invading pathogens. Macrophages possess two phenotypic forms, M1 and M2, which are functionally and metabolically different and respond to different stimuli. M1 macrophages respond to bacterial products, such as lipopolysaccharide (LPS), by producing pro-inflammatory mediators. Furthermore, M2 macrophages respond to 8 Bromo cAMP (8Br) by producing tissue repair and wound healing properties. Second messenger signaling through cAMP plays an important role in the resolution of inflammation and inflammatory mediators. Metabolism plays a key role in …

Mechanistic Insights Into Enterolactone-Mediated Glut4-Dependent Glucose Uptake, Merritt Smith
Senior Honors Theses
Type 2 diabetes mellitus (T2D) is one of the leading causes of morbidity and mortality in the United States, with approximately 25% of healthcare expenditures directed toward its management and associated metabolic complications. In response to the urgent need for novel therapeutic strategies, recent research has focused on bioactive compounds derived from dietary sources. In light of that, we identified the plant lignan matairesinol (MA) and its gut-derived metabolite, enterolactone (ENL), as agents capable of enhancing glucose uptake in 3T3-L1 differentiated adipocytes. These results suggest that enterolactone may represent a promising dietary supplement for improving glycemic control in patients with …
