Seagrasses are important coastal resources facing numerous stressors, and losses have been documented from local to global assessments. Under the broad theme of habitat loss and fragmentation, a study of historical change in total area and landscape configuration of seagrasses in the Mississippi and Chandeleur Sounds was conducted. Mapping data was collated from a multitude of previous projects from 1940 to 2011.

Comparisons of seagrass area among various studies that used different mapping methods can result in overestimation of area change and misleading conclusions of change over time. The vegetated seagrass area (VSA) data were generalized to a common resolution …

Humpback whales (Megaptera novaeangliae) belonging to the “breeding G-stock” annually migrate from the Antarctic Peninsula and southern Chile to the southeastern Pacific to reproduce. Associations between mother-calf pairs and escorts were examined in the Gulf of Tribugá, northern Colombian Pacific, using photo-identification and behavioral/spatial sampling. Research hypotheses included: 1. The association between cows and escorts is short-lived, consistent with a male reproductive strategy, 2. Thousands of Lepidoptera species build shelters as caterpillars using plant material and their own silk. Although these caterpillars and their shelters are recognized as playing important ecological roles, the structural diversity of shelters and the costs and benefits of different shelters to their builders are still poorly understood. In this dissertation, I use natural history observations, observational and manipulative field projects, and molecular and phylogenetic tools to investigate these questions for a diverse and abundant shelter-building caterpillar community within the dry forest of Palo Verde National Park, Costa Rica. The gray triggerfish (Balistes capriscus) and the queen triggerfish (Balistes vetula) are two exploited reef fish distributed in tropical and temperate shelf waters of the Atlantic Ocean and the Mediterranean Sea. Both species are highly sedentary as adults but disperse pelagic larvae for extended periods of time potentially allowing connectivity across long distances under the action of oceanic currents. In this work population structure, phylogeography, and migration patterns were examined in the two species and contrasted with predictions of larval transport based on surface circulation data. The problem was to find out why African American and Hispanic students were showing very little interest in science. While the scientific community is in consensus that coastal systems are threatened by climate change, few climate change studies test the effects of more than one variable directly related to climate change. The dissolved oxygen (DO) levels of the ocean are currently subject to both global warming and eutrophication; 94% of all hypoxia zones are expected to experience >2°C increase by 2035. This dissertation aims to examine how a model organism responds to simultaneous thermal and DO stress involving four levels of DO (100%, 70%, 50%, and 20%) saturation and three temperatures (15°C, 20°C, and 25°C).

The mitochondria are responsible for the bulk of energy production in eukaryotes. They possess their own genome that works in conjunction with the nuclear genome to accomplish the extraordinarily important task of energy conversion. When species hybridize there will be a mismatch in evolutionary histories between these two genomes. The deleterious interactions of these genomes have been studied in great detail (i.e. hybrid breakdown). While most works have focused on the species-rich Eastern Highlands region, there has been a recent shift towards plethodontid assemblages within the Coastal Plain. The Pearson and Rosenberg (P-R) conceptual model of macrobenthic succession was used to assess the impact of hypoxia (dissolved oxygen [DO] ≤ 2 mg/L) on the macrobenthic community on the continental shelf of northern Gulf of Mexico for the first time. The model uses a stress-response relationship between environmental parameters and the macrobenthic community to determine the ecological condition of the benthic habitat. The ecological significance of dissolved oxygen in a benthic habitat is well understood. In addition, the annual recurrence of bottom-water hypoxia on the Louisiana/Texas shelf during summer months is well documented.

A challenge faced by organisms with complex life cycles is how environmental factors experienced at an earlier life stage affect the fitness at that stage or are carried over to subsequent life stages. I used container-inhabiting mosquitoes: Aedes albopictus, Aedes aegypti, Aedes triseriatus and Culex quinquefasciatus to study the interactions and performance of life history stages under specific environmental factors. A recent taxonomic revision of the Neotropical catfish genus Rhamdia (Pisces: Siluriformes: Heptapteridae) reduced a number of described species to synonymy, especially under a broadly circumscribed R. quelen. Evidence is presented here from DNA sequence data, external and internal morphology, and morphometrics that argues for the recognition of R. guatemalensis in Central and northern South America and R. saijaensis and R. cinerascens in the Pacific drainages of Colombia and Ecuador, respectively. The recent and drastic decline in tortoise numbers has resulted in a multi-faceted approach to conserve this species. I used a population genetic approach to determine the population structure, genetic diversity and barriers to gene flow at a broad, regional and local scale. Estuaries are dynamic regions in which there can be significant modification of the riverine flux of trace elements to the open ocean due to various geochemical, physical, and biological processes. Additionally, estuaries are often subject to anthropogenic inputs of trace elements. The first portion of this study investigated the source, behavior, and sediment interaction of anthropogenic stable cesium (Cs) in St. Louis Bay (SLB), MS. Selected trace elements (TEs), dissolved organic carbon, and nutrients were studied in Louisiana Shelf waters including the Mississippi (MR) and Atchafalaya (AR) River plumes during periods of high, intermediate, and low river discharges. Seasonal variations in TEs were observed at low salinity, reflecting seasonal changes in the river water endmembers. Shelf surface water dissolved Mo, Cs, U, Ni, and Cu showed conservative behavior with minor scattering in some high salinity waters. Vibrio parahaemolyticus (Vp) is a gram-negative bacterium found naturally in marine and estuarine environments. Vp is found in oysters including those which are later consumed by the public. Sub-populations of potentially virulent Vp contain specific virulence factors and are relevant human pathogens capable of causing gastroenteritis, wound infection, and death. The tdh and trh genes, both encoding hemolysins, have been correlated with the majority of clinical Vp isolates but have not been shown to be the definitive virulence factors.

Effects Of Hypoxia And 4-Tert-Octylphenol On Gene Expression Profiles Of The Sheepshead Minnow (Cyprinodon Variegatus), Arthur Alan Karels

Hypoxia occurs in estuaries of northern Gulf of Mexico and world-wide, with increasing frequency/severity via eutrophication and anthropogenic influences. Hypoxia inducible factors (HIFs) form transcriptional complex and bind DNA at hypoxia responsive elements (HREs) in promoter regions of genes needed for systemic and cellular adaptation of fish to low dissolved oxygen (hypoxia, DO <2.0 mg/ml). Hypoxia-induced activation of HIF-αs can lead to a cascade of downstream activation, such as erythropoietin (EPO). Return to normal DO levels (normoxia), prolyl hydroxylases (PHDs) are activated to degrade HIF-αs back to baseline. Fish are affected by environmental estrogen mimics, like 4-tert-octylphenol (4tOP), binding estrogen receptor alpha (ERα) at estrogen responsive elements (EREs) and activating genes vitellogenin (VTG). Previous research showed overlap or crosstalk between these two mechanistic pathways. Hypoxia triggers unknown factors regulating ERE-mediated ERα signaling pathway, and stressor combinations could increase/decrease hypoxic or endocrine pathway. Research examined molecular/physiological effects of hypoxia (acute and chronic, moderate and severe) and 4tOP (~60μg/L)on adult male and/or female sheepshead minnow (Cyprinodon variegatus). Three genes identified, cloned, and sequenced (HIF-1α, HIF-2α, and PHD3), plus previously identified genes EPO and VTG, were examined in liver/testes exposed to hypoxia and/or 4tOP for cellular/physiological changes. Endpoints examined included mRNA expression from real-time PCR of HIF-1α, HIF-2α, PHD3, EPO, and VTG using cDNA from total RNA extracts, and microarray analyses of genes expressed during the transition from hypoxia back to normoxia. Phylogenetic analyses confirmed isolation of two HIF-α isofoms (HIF-1α and HIF-2α) and the PHD3 isoform. Significant up-regulation of PHD3 occurred within 10 hrs of chronic hypoxia, and persisted when severe (1.5 mg/L) and declined when moderate (~2.5mg/L). Significant up-regulation of HIF-1α and EPO occurred within 30 minutes to 2 hours of onset of acute severe and very severe (~1.08mg/L) hypoxia. Hypoxia acted similar to an estrogen mimic, with huge up-regulation of VTG gene expression in males, and increased VTG levels (additive effect) when hypoxia was combined with 4tOP. Microarray analyses showed 125 genes with significant transcriptional change, with up- or down-regulation from transitions of: (1) hypoxia (72 hrs) to normoxia (74 hrs) and (2) hypoxia+4tOP (72 hrs) to normoxia+4tOP (74 hrs).

Movement ecology is a component of nearly all aspects of animal behavior and an animal’s decision to move is likely influenced by a complex combination of exogenous and endogenous factors. Therefore, an examination of the causes and consequences of organismal movement provides a conceptual framework for understanding complex behavioral strategies. My dissertation research is focused on the movement ecology of an intercontinental migratory songbird during spring migration. I adopted experimental approaches to study the factors influencing how a songbird migrant, red-eyed vireos (Vireo olivaceus), makes decisions in unfamiliar landscapes from the initiation of spring stopover.

The course of the ancestral Tennessee River has been debated in both the geological and biological literature for over 100 years. Several of the proposed courses for the ancestral Tennessee place its course across the state of Mississippi. Geochemical analysis of sedimentary rocks in the Pascagoula River basin supports these hypotheses suggesting that the rocks in the Pascagoula basin were derived from the Highland Rim of Tennessee and northern Alabama, while geochemical analysis of rocks from the Pearl River basin point to deposition from a mixture of sources including the ancestral Mississippi River and perhaps the ancestral Tennessee. The role of species conservation is becoming increasingly important due to the mounting pressures from humans on habitats and populations of organisms. This is particularly evident in riverine ecosystems throughout the world where the human demand for freshwater resources is increasing, and consequently, the number of imperiled aquatic organisms is also growing. The seven chapters of this dissertation primarily focus on the Yellow-blotched Sawback, Graptemys flavimaculata, an endangered riverine turtle that is endemic to the Pascagoula River system of southeast Mississippi, USA. This study was the first to examine five significant enteric viruses in human fecal material, sewage, and oysters to show a genetic relationship between human enteric viruses and different sample matrices. Fecal samples were collected from an area hospital and examined for norovirus genotype I (NoV GI), norovirus genotype II (NoV Gil), hepatitis A virus (HA V), adenovirus (ADV), and enteroviruses. Researchers have suggested that there is an increasing apathy in the study of natural history both in academic settings and in the scientific community (Schmidly, 2005). Natural history is the cornerstone of ecological literacy. However, most studies of environmental knowledge do not directly address knowledge of local natural history. Instead, they concern knowledge of human environmental issues, environmental concepts, or broad ecological knowledge. Ecoliteracy established the study of natural history as fundamental to environmental knowledge and seeks to determine levels of knowledge of local environments and factors associated with that knowledge (Pilgrim et al., 2007).
