Consolidated Chamber Design And Protocol For Olfactory Conditioning Assay With Drosophila Melanogaster, Sasha Bronovitskiy, Andres Castillo, Michael Yan, Fang Ju Lin

The olfactory conditioning assay is widely used in Alzheimer’s disease research to quantify learning and memory in Drosophila melanogaster. The assay tests ability to recall an aversive conditioned stimulus of scent paired with electrical shock when presented a choice between shock-associated and unrelated scents. The T-maze, a commonly used apparatus for olfactory conditioning assays, employs an elevator mechanism to transfer live flies from the shock-delivering training chamber to the scent selection point. This elevator mechanism is known to cause fly casualty. T-mazes are not commercially available and often difficult to reproduce. The aim of this thesis is to investigate the effect of physical activity and the administration of s-equol on rates of neurogenesis in HIV-1 transgenic rats. HIV-1 is a neurodegenerative disease that leads to the wide range of cognitive impairment described as HAND. Neurogenesis is a newly discovered process that involves the birth of new neurons during our adult life within the dentate gyrus of the hippocampus. Therefore, inducing neurogenesis may be a proper therapeutic that can mitigate some of the cognitive destruction seen as a result of HIV-1 infection. The role of the two hemispheres in processing metaphoric language is controversial. While some studies have reported a special role of the right hemisphere (RH) in processing metaphors, others indicate no difference in laterality relative to literal language. Some studies have found a role of the RH for novel/unfamiliar metaphors, but not conventional/familiar metaphors. It is not clear, however, whether the role of the RH is specific to metaphor novelty, or whether it reflects processing, reinterpretation or reanalysis of novel/unfamiliar language in general. Most previous studies investigating the neural correlates of reading have presented text using serial visual presentation (SVP), which may not fully reflect the underlying processes of natural reading. In the present study, eye movements and BOLD data were collected while subjects either read normal paragraphs naturally or moved their eyes through “paragraphs” of pseudo-text (pronounceable pseudowords or consonant letter strings) in two pseudo-reading conditions. Eye movement data established that subjects were reading and scanning the stimuli normally.
