It is common practice in the livestock animal production industries to feed high levels of protein to maximize the yield of each animal in production. However, diets high in protein can cause decreased reproductive success within these animals. The causes behind this decrease are not fully understood. Increased dietary protein leads to increased levels of both ammonia and urea in the blood of the animal. The dam vaginal microbiota is the first major microbial inoculating community within the neonate. The composition of the dam vaginal microbiota has implications in calf commensal microbiota development. Alterations of the dam microbial community prior to parturition could alter inoculating communities and immune responses in both the dam and calf. Thus, authors aimed to elucidate the microbial community composition of the bovine dam vaginal and calf nasal microbiota post-partum after utilizing betadine lavages (BL). Nutrient restriction is a relatively common production insult to pregnancy in cattle. Recently, melatonin supplementation has been investigated as a possible therapeutic to rescue the negative effects of nutrient restriction. Neurotransmitters have been implicated as having negative programming effects in mouse and human pregnancies, manifested as metabolic and neurologic disorders. The role of neurotransmitters in fetal development has only begun to be understood in mice and humans. Embryos resulting from assisted reproductive technologies, such as in vitro fertilization (IVF) or intracytoplasmic sperm injection (ICSI), develop with lower efficiencies than embryos resulting from their in vivo counterparts. The reasons behind the developmental discrepancies remain largely unknown. Because the egg is the primary determinant of embryo developmental success, it is reasonable to consider inherent egg quality as a possible cause. The hypothesis for this project is that there are distinct mRNA transcript patterns, or molecular “fingerprints,” that distinguish high- versus low-quality eggs developing within the same environment. Major subpopulations of gamma delta T cells within ruminant and pigs are defined by expression of WC1, a hybrid pattern recognition receptor/co-receptor to the T cell receptor (TCR). It is known that when WC1 is knocked down cells fail to respond. Showing that WC1 plays an active role in the stimulation of bovine gamma delta T cells. Here we explored the spatio-temporal dynamics of WC1 and TCR interaction using imaging flow cytometry and stochastic optical reconstruction microscopy. Oocyte vitrification, while beneficial for research and species conservation, has limited success due to cryoinjury to the meiotic spindle. This study aimed to evaluate treatments to improve meiotic spindle recovery after vitrification by treating bovine oocytes with either a microtubule stabilizing agent, or a microtubule recovery agent. In the first two experiments, Taxol and epothilone B were used to treat bovine oocytes before vitrification. Both compounds have microtubule stabilizing properties and are known antimitotic compounds commonly used in cancer treatment. Experiment 1 tested the hypothesis that ad libitum consumption of a 1:1 blend (MIX) of sodium selenite (ISe) and SELPLEX vs solely ISe as selenium (Se) supplements (27 ppm) in a vitamin-mineral mix (V-M) by steers subjected to summer-long grazing of toxic endophyte-infected tall fescue (TE) would ameliorate the negative effects of TE by positively affecting the expression pattern of prolactin (PRL) and NF-kB pathway genes by circulating leukocytes (WBC). This study assessed the effects both DTT and progesterone have on bovine spermatozoa to induce decondensation or the acrosome reaction in order to facilitate male pronuclear formation after ICSI. Sperm prepared by swim-up in a 5 mM concentration of DTT displayed time dependent morphological changes resulting in decondensation over a 6-hour period. An estimated 90% of treated sperm displayed early changes in morphology after the second hour of incubation. Beef cattle consuming endophyte infected tall fescue typically exhibit reduced performance in terms of both decreased dry matter intake (DMI) and growth rates. It has been suggested that lower concentrations of circulating IGF-1 (an important stimulator of the mTOR pathway and ultimately protein synthesis), as observed with consumption of ergot alkaloids, contribute to reduced growth rates. Intraovarian growth factors play a vital role in influencing the fate of ovarian follicles. They affect proliferation versus apoptosis of granulosa cells (GCs), and can influence whether small antral follicles continue their growth or undergo atresia. Transforming Growth Factor-alpha (TGFα), an oocyte-derived growth factor, is thought to regulate granulosa cell function, yet has been largely overshadowed by current interest in TGF-beta superfamily members, such as bone morphogenetic proteins (BMPs) and anti-Mullerian hormone (AMH). Overall, the goals of this thesis were to improve oocyte and embryo culture with the antioxidant polydatin and to optimize slow freezing procedures. Fescue toxicosis is problematic for ruminant livestock, causing weight loss and low productivity when fed endophyte-infected forages. Complete underlying mechanisms of toxicosis are unknown therefore; the objective of the study was to determine if ruminally dosed ergot alkaloids impact rumen motility. Cannulated steers were pair-fed a forage diet and ruminally dosed with endophyte-free (E-) or endophyte-infected (E+) tall fescue seed. An 8-h period of rumen motility collection began 4-h after feeding by monitoring pressure change via a wireless telemetry and transducer system. In vitro production and transfer of embryos has become a common practice within the dairy industry to efficiently breed superior animals and meet the consumption demand of the growing population. Cyropreservation is necessary for the application of commercialized embryo transfer, however, in vitro-produced embryos show morphological and physiological defects which negatively impact their ability to withstand cryopreservation in comparison to their in vivo counterparts. MicroRNAs (miRNAs) are a family of small non-coding RNAs (sRNA) that play a key role in regulating gene expression by binding to their complementary mRNA and altering translation. It has been reported that this down-regulation of specific areas of the genome plays a role in male fertility. Piwi-interacting RNAs (piRNAs) are another class of germline-specific non-coding RNAs that form silencing complexes to maintain germline genome integrity. Three studies were conducted to improve and incorporate reproductive management practices into beef cow-calf production. First study evaluated serial use of Estrotect estrous detection patches as a simple, cost-effective reproductive management tool to identify cyclic animals before breeding, distinguish between females conceiving to AI versus natural service, and determine seasonal pregnancy rate after bull removal. Also determined, was effectiveness of altered timing of GnRH treatment (1 d ± CIDR removal) in a modified 14-d CIDR-Select Synch protocol. When evaluated over a 4-wk period, estrous detection patches correctly (P < 0.01) identified 79% of cyclic and 86% of non-cyclic heifers. Patches were 96 and 98% accurate in identifying heifers and cows pregnant by AI, respectively, and were 76 and 87% accurate in identifying pregnant heifers and cows at the end of the breeding season (P < 0.01). Treatment with GnRH at CIDR removal reduced labor costs and animal handling without compromising estrous response (both ~63.0%) and AI pregnancy rates (~76 and 77%; P > 0.1). A study investigated the use of Anti-Mullerian hormone (AMH) and/or follicle counts as a predictor of subsequent superovulatory response and embryo production in 79 beef cows. Before initiation of superovulation, ultrasonography was used to scan the ovaries of each donor cow to record the number of 3 to 5 mm follicles present, and a blood sample was collected for measure of serum AMH. At the time of embryo collection, the ovaries of donor cows were palpated to estimate the number of corpora lutea (CL) present on each ovary. The main objective of this research was to examine the lipolytic changes in triglyceride and phospholipid as well as the incidence of germinal vesicle breakdown during IVM of heat-stressed oocytes compared to non-stressed oocytes. To this end, cumulus-oocyte complexes were matured for 0, 2, 4, 6 or 24 hIVM at 38.5 or 41.0°C (first 12 h only, then transferred to 38.5°C). Triglyceride and phospholipid levels decreased by 2 hIVM (P = 0.0009 and P = 0.0005, respectively) but remained fairly constant to 24 hIVM; lipid decline was not affected by maturation temperature. Endophyte-infected tall fescue produces many ergot alkaloids, which have been shown to be vasoconstrictive in various vessel types of bovine. On the other hand, substantial evidence has been reported on the vasodilative effects of formononetin and biochanin A in different vessel types in humans and rats. So, a study was conducted using mesenteric vasculature collected from heifers shortly after slaughter. In vitro maturation (IVM) is a reproductive technique critical to in vitro embryo production (IVP) in commercial livestock industries, research, and human infertility treatment. Currently, IVP has low efficiency due to an inadequate IVM system in which premature meiotic resumption results in low oocyte viability. This project was funded by the United States of Department of Agriculture (USDA), which funds research aimed at improving production and animal health. The aim of this study was to advance knowledge of maternal immune tolerance to the fetus and mechanisms bovine non-classical MHC class I proteins employ to interact with immune cells and render them inert towards the fetus.

Mastitis is an economically distressing disease in the dairy industry. Bacterial pathogens enter the gland and encounter cell types that release immune mediators including interleukin (IL)-8. IL-8 has two membrane bound receptors: CXCR1 and CXCR2. CXCR1 and CXCR2 are expressed on neutrophils and other cell types in other species but their expression is unknown in the bovine mammary gland. To test this, mammary tissue samples from six Holstein dairy cows were subject to dual immunofluorescence with bovine specific CXCR1 and CXCR2 antibodies and cell type markers.
