It is common practice in the livestock animal production industries to feed high levels of protein to maximize the yield of each animal in production. However, diets high in protein can cause decreased reproductive success within these animals. The causes behind this decrease are not fully understood. Increased dietary protein leads to increased levels of both ammonia and urea in the blood of the animal. These increased metabolite levels may be a possible explanation for the decline in reproductive success. Fibroblast growth factor 21 (FGF21) has been identified in multiple mammalian species as a molecular marker of energy metabolism while also providing negative feedback to the gonads. However, the role of FGF21 in regulating the energetic and reproductive physiology of beef heifers and cows has yet to be characterized. Herein, we investigated the temporal concentrations of FGF21 in female beef cattle from the prepubertal period to early lactation. Circulating concentrations of FGF21, non-esterified fatty acids, plasma urea nitrogen, glucose, and progesterone were assessed. The dam vaginal microbiota is the first major microbial inoculating community within the neonate. The composition of the dam vaginal microbiota has implications in calf commensal microbiota development. Alterations of the dam microbial community prior to parturition could alter inoculating communities and immune responses in both the dam and calf. Thus, authors aimed to elucidate the microbial community composition of the bovine dam vaginal and calf nasal microbiota post-partum after utilizing betadine lavages (BL). Water is an essential nutrient and is required for growth, metabolism, lactation, and reproduction. Without water, livestock production would not be possible. Despite its importance, water requirements in livestock have not received much attention from the research community since the 1950s. The most recent edition of the Nutrient Requirements of Beef Cattle (8th edition) relies on literature from the 1950s or earlier for defining water requirements in beef cattle. Nutrient restriction is a relatively common production insult to pregnancy in cattle. Recently, melatonin supplementation has been investigated as a possible therapeutic to rescue the negative effects of nutrient restriction. Neurotransmitters have been implicated as having negative programming effects in mouse and human pregnancies, manifested as metabolic and neurologic disorders. The role of neurotransmitters in fetal development has only begun to be understood in mice and humans. Neurotransmitters have not been analyzed in cattle, much less within the context of a compromised pregnancy. Embryos resulting from assisted reproductive technologies, such as in vitro fertilization (IVF) or intracytoplasmic sperm injection (ICSI), develop with lower efficiencies than embryos resulting from their in vivo counterparts. The reasons behind the developmental discrepancies remain largely unknown. Because the egg is the primary determinant of embryo developmental success, it is reasonable to consider inherent egg quality as a possible cause. The hypothesis for this project is that there are distinct mRNA transcript patterns, or molecular “fingerprints,” that distinguish high- versus low-quality eggs developing within the same environment. Major subpopulations of gamma delta T cells within ruminant and pigs are defined by expression of WC1, a hybrid pattern recognition receptor/co-receptor to the T cell receptor (TCR). It is known that when WC1 is knocked down cells fail to respond. Showing that WC1 plays an active role in the stimulation of bovine gamma delta T cells. Here we explored the spatio-temporal dynamics of WC1 and TCR interaction using imaging flow cytometry and stochastic optical reconstruction microscopy. Alternatives to routine antibiotic treatment of dairy cattle during the dry period before their next calving are of interest. This was a preliminary study of whether intramammary infusion of casein hydrolysate, administered alone or combined with standard dry treatment, accelerated the rate of mammary involution early in the dry period. Four treatments were studied in a split udder design. One udder half was assigned a treatment, and the contralateral half was administered dry cow treatment + internal teat sealant as a control. Microarray analysis using Affymetrix Bovine GeneChip 1.0 ST Array to determine RNA expression analysis was performed on somatic granulosa cells from two different groups of cows classified based on androstenedione concentration within the follicular fluid (Control vs High A4) of estrogen-active dominant follicles. The normalized linear microarray data was deposited to the NCBI GEO repository (GSE97017 - RNA Expression Data from Bovine Ovarian Granulosa Cells from High or Low Androgen-Content Follicles). Dairy calf immune development leaves calves susceptible to illness during the pre-weaned stage of life. Previous studies have shown that the use of probiotics during this time may result in increased growth rates and decreased incidence of illness, particularly diarrhea and scours. We hypothesized that by providing probiotics during the time from birth until weaning, calves on study would experience similar growth and health benefits. Neonatal calves (N=32) were provided milk replacer three times daily, with half of the calves receiving Probios® supplements mixed in with their first morning feeding. Oocyte vitrification, while beneficial for research and species conservation, has limited success due to cryoinjury to the meiotic spindle. This study aimed to evaluate treatments to improve meiotic spindle recovery after vitrification by treating bovine oocytes with either a microtubule stabilizing agent, or a microtubule recovery agent. In the first two experiments, Taxol and epothilone B were used to treat bovine oocytes before vitrification. Both compounds have microtubule stabilizing properties and are known antimitotic compounds commonly used in cancer treatment. Bison are an icon of the American West and an ecologically, commercially, and culturally important species. Despite numbering in the hundreds of thousands today, conservation concerns remain for the species, including the impact on genetic diversity of a severe bottleneck around the turn of the 20th century and genetic introgression from domestic cattle. Pooling individual samples prior to DNA extraction can mitigate the cost of DNA extraction and genotyping; however, these methods need to accurately generate equal representation of individuals within pools. The objective of this study was to determine accuracy of pool construction of blood samples based on white blood cell counts compared to two common DNA quantification methods. This research was carried out to determine the seroprevalence of Bovine Viral Diarrhoea Virus (BVDV), Bovine Herpes Virus 1 (BoHV-1), and Bovine Herpes Virus 4 (BoHV-4) in cattle (n = 188) in Ağrı province of Eastern Anatolia Region, Turkey. The presence of BVDV, BoHV-1, and BoHV-4 antibodies in sera samples were determined by an ELISA test kit. Experiment 1 tested the hypothesis that ad libitum consumption of a 1:1 blend (MIX) of sodium selenite (ISe) and SELPLEX vs solely ISe as selenium (Se) supplements (27 ppm) in a vitamin-mineral mix (V-M) by steers subjected to summer-long grazing of toxic endophyte-infected tall fescue (TE) would ameliorate the negative effects of TE by positively affecting the expression pattern of prolactin (PRL) and NF-kB pathway genes by circulating leukocytes (WBC). This study assessed the effects both DTT and progesterone have on bovine spermatozoa to induce decondensation or the acrosome reaction in order to facilitate male pronuclear formation after ICSI. Sperm prepared by swim-up in a 5 mM concentration of DTT displayed time dependent morphological changes resulting in decondensation over a 6-hour period. An estimated 90% of treated sperm displayed early changes in morphology after the second hour of incubation. Sperm displaying partial decondensation of the nucleus was estimated as 42% by the 3rd hour and increased to 62% by the 5th hour of incubation. Fully decondensed sperm or …

In vitro produced bovine embryos are critical to the cattle industry. However, these embryos have altered morphology, epigenetics, and metabolism when compared to their in vivo counterparts. The aim of this thesis was to alter maturation and culture media to improve the developmental competence of in vitro bovine embryos. Approximately 90% of beef cattle on feed in the United States receive at least one anabolic implant, which results in increased growth, efficiency, and economic return to producers. However, the complete molecular mechanism through which anabolic implants function to improve skeletal muscle growth remains unknown. Beef cattle consuming endophyte infected tall fescue typically exhibit reduced performance in terms of both decreased dry matter intake (DMI) and growth rates. It has been suggested that lower concentrations of circulating IGF-1 (an important stimulator of the mTOR pathway and ultimately protein synthesis), as observed with consumption of ergot alkaloids, contribute to reduced growth rates. Cows were treated with follicle-stimulating hormone (FSH) in a single dose and then subjected to ovum pick-up (OPU) for evaluation of oocyte recovery and in vitro fertilization (IVF). Cows underwent OPU followed by application of FSH (200 mg) on day 2 and new OPU on day 3. This methodology was repeated 3 times sequentially. In FSH-free the cows (control), OPU was performed at intervals of one week. The number of oocytes recovered was higher in the control cows (22.2 ± 1.5) compared to the treated cows (16.9 ± 1.4; P < 0.05). Yes-associated protein 1 (YAP1) is a major component of the Hippo signaling pathway. Although the exact extracellular signals that control the Hippo pathway are currently unknown, increasing evidence supports a critical role for the Hippo pathway in embryonic development, regulation of organ size, and carcinogenesis. Granulosa cells (GCs) within the ovarian follicle proliferate and produce steroids and growth factors, which facilitate the growth of follicle and maturation of the oocyte.We hypothesize that YAP1 plays a role in proliferation and estrogen secretion of GCs. Intraovarian growth factors play a vital role in influencing the fate of ovarian follicles. They affect proliferation versus apoptosis of granulosa cells (GCs), and can influence whether small antral follicles continue their growth or undergo atresia. Transforming Growth Factor-alpha (TGFα), an oocyte-derived growth factor, is thought to regulate granulosa cell function, yet has been largely overshadowed by current interest in TGF-beta superfamily members, such as bone morphogenetic proteins (BMPs) and anti-Mullerian hormone (AMH). Bovine in vitro produced embryos suffer from poor developmental competence and altered metabolism which hinders their cryotolerance. Overall, the goals of this thesis were to improve oocyte and embryo culture with the antioxidant polydatin and to optimize slow freezing procedures. This thesis was designed as three experiments, and in each experiment, oocytes were aspirated from abattoir ovaries, matured for 23h, fertilized with semen from 1 of 3 bulls, and cultured in synthetic oviductal (SOF) based medium (SCF1) in 38.5 °C in 5% O2, 5% CO2 and 90% N2. Ergot alkaloids, in their active isomeric form, affect animal health and performance, and adsorbents are used to mitigate toxicities by reducing bioavailability. Adsorbents with high specificity (molecularly imprinted polymers: MIP) adsorb ergot alkaloids in vitro, but require evaluation for biological implications. Using ex vivo myography, synthetic polymers were evaluated for effects on the bioactivity of ergotamine tartrate (ETA). Polymers were first evaluated using isotherms. Lateral saphenous veins were collected from 17 steers for four independent studies: dose response of ETA, adsorbent dose response, validation of pre-myograph incubation conditions and MIP/ non-molecularly imprinted polymer (NIP) comparison.
