The foodborne pathogen Campylobacter jejuni is one of the leading causes of human gastroenteritis. This bacterium is not a robust organism compared with many other foodborne pathogens and requires special conditions in the laboratory for its growth. In nature, however, this organism is able to survive in very diverse and hostile environments and produce disease in humans. The different mechanisms by which C. jejuni survives stressful conditions in the environment remain unclear. Reproductive effort varies as organisms make reproductive decisions based on available energy. Eighty Brahman-influenced cows (5.6 BCS) were assigned to graze common bermudagrass (CB) or toxic tall fescue (E+) during a 60-d breeding season. Body condition (BC) was assessed at d 0, 30, and 60 of breeding season. Cows were managed to achieve marginal (4.2 BCS) or good (6.3 BCS) BC during last trimester. Heifer birth and weaning (WW; 7 to 8 mo) weights were recorded. The Angus sired heifers were weighed and randomly assigned to E+ or non-toxic (Novel) tall fescue at 9 to 10 mo of age for 190d. Three studies were conducted to evaluate how the broiler has changed over the years due to intense genetic selection. Four different broiler genotypes, one unselected since the 1950's (HER) representing the old meat-type bird and three current strains (Ross 308, Ross 708, and a test product (TY)) were studied. Differences in growth, morphometry, and response in performance and processing yield to different nutrient levels were studied.

Bioenergetic mechanisms responsible for ATP production are essential in carrying out maintenance and cell-specific functions. In this study, hepatocytes (liver cells) were used to test both endogenous and exogenous stress on cellular respiration. The secondary lipid peroxide, 4-hydroxynonenal (HNE), was used because it alters bioenergetics by increasing mitochondrial proton leak that attenuates mitochondrial radical production and, therefore, endogenous oxidative stress. Three experiments were conducted to evaluate the responses of broiler performance to Lysine (Lys) and Methionine (Met) during the starter, grower, and finisher feeding phases. All the experimental diets were formulated based on the Ideal Protein Concept.

Two experiments were conducted to characterize polymorphisms (SNP) in the bovine FSHβ gene promoter region to examine breed differences in SNP, and to determine effects of SNP on reproduction in beef cattle. Experiment 1 - DNA samples were collected from 5 Angus (ANG), 13 Balancer (BAL), & 16 Brahman influenced (BI) bulls. Polymorphisms were identified by sequencing of 3 sequential PCR products from the promoter region revealed 17 SNP & 4 insertion/deletions (INDEL). The present study investigated the distribution of the vasotocin subtype four receptor (VT4R) in brain and pituitary gland of the chicken, Gallus gallus. The anterior pituitary cell types associated with the VT4R were also determined. Two polyclonal antibodies were raised in rabbit against a cocktail of peptides, 15 amino acids from the amino terminal region and 17 amino acids from the carboxy terminal region of VT4R receptor. The antibody was validated utilizing the Western blot and immunocytochemistry. The Louisiana Waterthrush, Parkesia motacilla, is a migratory wood-warbler and breeding season resident of the eastern United States. Males defend breeding territories that extend linearly along clear, fast-flowing, gravel-bottomed, forest streams. Defense includes two song types, primary and extended song. As riverine specialists, birds rely upon aquatic invertebrates as prey and riparian habitat features for nesting. They use a unique foraging maneuver, leaf-pulling, that involves picking up or pulling a leaf from water, and turning it over to search for prey. Sperm mobility has been shown to be a major determinant of male fertility in broiler breeders. A whole genome SNP scan identified chromosomal regions (QTLs) that control sperm mobility. I examined spatial and temporal consumption dynamics using an energy intake model and a bioenergetics model of rainbow trout, Oncorhynchus mykiss, and brown trout, Salmo trutta, within three catch-and-release (C-R) areas in Bull Shoals and Norfork tailwaters to determine whether trout populations were limited by food supply. Campylobacter spp. has been identified as one of the leading causative agents of food borne diarrheal illness. Epidemiological evidence has shown that poultry is the main source for human infection. Poultry are asymptomatic carriers of Campylobacter within their gastrointestinal tract, with colonization levels reaching 106-108 cfu/g cecal content. Surveys of domestic poultry flocks have estimated that approximately 90% of flocks are positive for Campylobacter colonization. Risk assessment studies have determined that by reducing levels of Campylobacter colonization during rearing, incidences of human infection will be significantly reduced. The Smyth line (SL) of chickens is a well-established animal model for the study of spontaneous autoimmune vitiligo, displaying all the characteristics of human vitiligo. In humans, phenolic derivative 4-tertiary butyl phenol (4-TBP) has been shown to trigger vitiligo in susceptible individuals. In vitiligo-prone SL-chickens, live turkey herpesvirus (HVT) infection at hatch was identified as the most effective trigger of SL-vitiligo expression. In 1935, the National Poultry Improvement Plan (NPIP) was created to control Salmonella gallinarum and Salmonella pullorum . These two pathogens were devastating economically for poultry producers. Through cooperative efforts using vaccination and strict biosecurity, these two pathogens were eradicated from the United States. Currently, the NPIP program is targeting two other poultry pathogens, Salmonella enterica serovar Enteritidis and Mycoplasma . In the broiler industry it targets 2 specific Mycoplasma species (synoviae, gallisepticum). Two groups of bulls were utilized to evaluate if single nucleotide polymorphisms (SNP) in the Bos taurus OPN gene (GenBank accession # AY878328.1) promoter region were related to bull semen quality variables as evaluated by computer assisted semen analysis (CASA). The studies that comprise this thesis were performed in an attempt to identify and eliminate stressful conditions that may lead to infertility in the male bovine. The first project was performed to determine if ergot alkaloids directly affect bovine sperm motility. Percentage of motile spermatozoa was affected (P = 0.015) by a three-way interactions between time, concentration, and alkaloid. Ergotamine and dihydroergotamine decreased (P = 0.01) sperm motility in a concentration and time dependant manner and ergonovine had a minimal effect on overall sperm motility. The number of static spermatozoa also was affected (P < 0.01) by a three way interaction and increased as ET and DHET concentrations increased. Percentages of progressively motile and rapidly motile spermatozoa decreased (P < 0.01) in a two way interaction between alkaloid and concentration. Overall, sperm motility was decreased by ET and DHET. Furthermore, the qualities of motility as represented by progressive, rapid, path velocity (VAP), progressive velocity (VSL), track speed (VCL), beat frequency (BCF), lateral amplitude (ALH), straightness (STR), elongated, and area were decreased by those alkaloids. These data verify that ergot alkaloids commonly found in toxic tall fescue are detrimental to bovine spermatozoa. In the second study two antioxidants (alpha-tocopherol and ascorbic acid) were added to bovine sperm culture media and cryopreservation extender. The antioxidant capabilities of these two vitamins could help reduce free radical production and help preserve sperm viability. There was an interactive effect between ascorbic acid concentration and storage method on the bovine sperm motility characteristics: motile, progressive, rapid, track speed (VCL), and straightness (P < 0.05). A bull by ascorbic acid interaction (P < 0.05) was observed for motile, progressive, rapid, path velocity (VAP), progressive velocity (VSL), and VCL characteristics. Alpha tocopherol had no affects on sperm motility characteristics. Lipid peroxidation was affected (P < 0.01) by bull and ascorbic acid. Malondialdehyde concentrations for ascorbic acid treatments (0, 5, 10, 20 mM) were 3.25, 4.2, 2.96, and 2.15 ìM respectively. Results from the second study indicate that the addition of ascorbic acid may reduce sperm motility, but help prevent lipid peroxidation.

Lactate dehydrogenase (LDH) catalyzes the conversion of the pyruvate to lactate (forward) or lactate to pyruvate (reverse) in the last step of glycolysis. Objectives were to document the effects of LDH haplotypes and its SNPs, found in the promoter and coding sequence site, and body condition on beef cow production. A series of experiments were conducted to determine the effects of on-arrival vs. delayed respiratory vaccination and exposure to persistently infected bovine viral diarrhea virus challenge on health, gain performance, and physiological and immunological measurements in newly received stocker cattle. Two experiments evaluated timing (d 0 vs. 14) of respiratory, clostridial, or both vaccinations in newly received stocker calves during stress-induced immunosuppression. This research was undertaken to identify an antimicrobial for use during chilling in beef processing, including periods of temperature abuse, as well as to evaluate membrane filtration as means to remove essential oils from bacteria to prevent further antibacterial action. Cold pressed terpeneless Valencia orange oil was examined in combination with various temperatures (37°C, 10°C, and 4°C) to determine its antimicrobial activity against various strains of bacteria.
