"This thesis investigates the preparation, characterization, and transfection efficiency of various ionizable lipid nanoparticles (LNPs) formulated for the delivery of mRNA encoding enhanced green fluorescent protein (EGFP) in RAW 264.7 macrophages and MDA-MB-231 breast cancer cell lines. The ionizable cationic lipid studied are ALC-035, C12-200, C14-4, PPZ-A10, DLin-MC3-DMA. The study addresses the need for effective gene delivery systems to provide a safe and versatile platform that protects and transports nucleic acids into target cells. The National Football League (NFL) is a leading professional American football organization experiencing rapid growth in popularity. However, the high-contact nature of the sport frequently results in player injuries, with head trauma and concussions being major concerns. This project aims to mitigate these risks by developing an advanced helmet liner designed to reduce the impact of direct collisions and torsional shear forces. This thesis presents the design and implementation of a smart irrigation system that combines Internet of Things hardware with a Long Short-Term Memory (LSTM) neural network for predictive soil moisture management. The goal is an affordable and reliable solution that uses real-time sensor data and environmental data to schedule irrigation before the substrate moisture drops below its target range. Power quality disturbances (PQDs) are among the primary challenges facing modern electrical systems, as they degrade the performance and lifespan of connected equipment. This thesis investigates the relationship between the rate at which voltage waveform data are sampled, the reliability of these measurements, and the ability of deep neural networks to classify PQDs accurately. A one-dimensional convolutional neural network (CNN) was trained and evaluated across multiple sampling rates and signal-to-noise ratios to quantify how information loss in the temporal and spectral domains affects classification reliability. Most literature has focused on the effect of shrinkage-reducing admixture (SRA) in shrinkage mitigation resistance of concrete. This study aims to examine the impact of SRA on the efficiency of macro synthetic fibers (MSF) in enhancing the flexural post-cracking behavior of fiber-reinforced super workable concrete (FR-SWC). A comparative analysis of the influence of fiber combinations on the flexural post-cracking behavior of beams is also included. Results showed that a higher dosage of SRA, particularly at 5 % by mass of binder, had a noticeable negative impact on flexural post-cracking performance of beams while exhibiting positive effect on workability and shrinkage …

Hybrid poplars are widely recognized for their effectiveness in remediating subsurface aromatic hydrocarbon contaminants, including benzene, toluene, ethylbenzene, and xylene isomers (BTEX). While BTEX compounds are frequently found in the transpiration streams of poplars at contaminated sites, the microbial dynamics within these trees, particularly in response to hydrocarbon exposure, remain underexplored. This study utilized high-throughput amplicon sequencing to investigate the trunk microbiome in hybrid poplars at a field-scale toluene phytoremediation site. Roadbed thaw settlement is a unique challenge for the durability of low-volume roads (LVRs) in permafrost regions. Air convection embankment (ACE) is an effective technique that acts as a semi-heat-transfer system to control temperature variation and reduce the thaw depth of subsoil. This project aimed to develop a laboratory-scale solution that replicates produced water conditions (CO2 + brine) found in crude pipelines, with the added requirement of promoting the growth of sulfate-reducing bacteria (SRB). A previously successful study was used as a reference, which employed a hybrid salt solution containing essential nutrients and environmental factors. The proposed solution included 10 g sodium chloride, 1 g sodium pyruvate, 0.2 g ammonium chloride, and 0.8 g sodium sulfate per liter of deionized water. Traditionally, traffic incident management (TIM) programs coordinate the deployment of emergency resources to immediate incident requests without accommodating the interdependencies on incident evolutions in the environment. However, ignoring these inherent interdependencies while making current deployment decisions is shortsighted, and the resulting naive deployment strategy can significantly worsen the overall incident delay impact on the network. To characterize the theoretical behavior, we analyzed the worst-case-timing communication between 4 nodes using Arduino Minima. The project focused on understanding the message transmission and reception via CAN Bus, with 4 Arduino R4 Minima and a Level Voltage Converter to shift from 3.3V to 5V. After trials and errors, we successfully measured the worst-case timing for sending and receiving messages across the 4 Arduino R4 Minima nodes. These findings provided valuable insights into the effectiveness of CAN communication.

This thesis investigates the autogenic sediment transport regime in a low-gradient stream system in the inner bluegrass region of central Kentucky, USA The seasonality of sediment transport seasonality was not well addressed in prior research for baseflow and low flow periods, to our knowledge, due to the assumption that low flows do not account for considerable fluid transport capacity to move sediment. The hypothesis is that during the autogenic regime the sediment transport in the low-gradient system is being driven by geophysical, biogeochemical, and biological processes. This paper identifies barriers, challenges, and requirements to the widespread adoption of Design for Deconstruction and Reuse (DfDR) of wood and engineered wood products in the United States (U.S.) to increase circularity in the built environment. A series of interviews with industry representatives were performed to summarize the state-of-the-art related to DfDR and gain insight into the perspectives and attitudes of the primary stakeholders; the results of these interviews were compared to the current academic literature. These higher-order processes facilitate the achievement of goal-directed behavior and enable both adaptive decision-making and emotional regulation. This paper explores implementing and evaluating a Bluetooth Low Energy (BLE)-based indoor localization system using Received Signal Strength Indicator (RSSI) and Angle of Arrival (AoA) data via machine learning. A survey of localization technologies (RFID, GPS, ZigBee, and BLE) provides context on capabilities and limitations in indoor positioning. IQ data and phase-based angle estimation show how BLE 5.1’s direction-finding features enable sub-meter accuracy. "This research investigates the bond behavior of magnesium potassium phosphate cement (MKPC) coated mild steel deformed bars in conventional concrete and concrete repair materials. For typical steel reinforcement in concrete, a thin “passive layer” develops around the reinforcement, which protects the reinforcement from corrosion. The concrete carbonates over time, reducing the protection of the passive layer and allowing chlorides, which corrode the reinforcement, to penetrate at an increasing rate. In some cases, to combat this corrosion, a protective layer is added to the reinforcement prior to installation. Common protective layers include epoxy coatings or zinc galvanization. This study explores the integration of spectral mixtures of fetal heart rate (FHR) and uterine contraction (UC) signals to enhance the prediction of fetal acidosis, utilizing the CTU-CHB dataset. Several classification models were trained using two distinct oversampling techniques and inputs, demonstrating that models incorporating spectral mixtures significantly outperform those using raw signals. These models, particularly when combined with convolutional neural networks (CNNs) and attention mechanisms, achieved a notable F1-score of 0.98, with the highest model achieving an area under the Receiver Operating Characteristic (ROC) curve of 0.95.
