The Apollo Command Module had a tendency to flip over upon impact with the ocean surface after returning from space (9/19 times). In an effort to better characterize the inversion process for future water landing vehicles, experimental results for a simplified buoyant cylindrical puck impacting the water surface are presented. This study focuses on the dependence of inversion upon vertical velocity, horizontal velocity, and the pitch angle of the puck relative to the free surface. High-speed images reveal an asymmetric cavity that forms upon water impact. The asymmetric cavity then collapses, applying a moment, which can be sufficient to invert …

The focus of this work is the integration of alkali vapor atomic vapor cells into common silicon wafer microfabrication processes. Such integrated platforms enable the study of quantum coherence effects such as electromagnetically induced transparency, which can in turn be used to demonstrate slow light. Slow and stopped light devices have applications in the optical communications and quantum computing fields. This project uses hollow core anti-resonant reflecting optical waveguides (ARROWs) to build such slow light devices. An explanation of light-matter interactions and the physics of slow light is first provided, as well as a detailed overview of the fabrication process. …

My research broadly covers various important aspects of microfluidic devices and biosensors. Specifically, this dissertation reports: (1) a new and effective room temperature method of bonding polydimethylsiloxane (PDMS) microfluidics to substrates such as silicon and glass, (2) a new microfluidic pump concept and implementation specifically designed to repeatedly drive a small sample volume (<1 µL) very rapidly (~500 µL/min) through a sensor-containing flow channel to significantly decrease sensor response time through advection-driven rather than diffusion-driven mass transport, (3) use of a new microfluidic material based on polyethylene glycol diacrylate (PEGDA) to implement impedance-based dynamic nanochannel sensors for protein sensing, and (4) an investigation of galvanoluminescence and how to avoid it for conditions important to fluorescence-based dielectrophoresis (DEP) microfluidic biosensors. Over the last decade, the Nordin research group has developed a lab-on-a-chip (LOC) biosensor based on silicon photonic microcantilever arrays integrated with polydimethylsiloxane (PDMS) microfluidics for protein biomarker detection. Integration requires reliable bonding at room temperature with adequate bond strength between the PDMS element and microcantilever sensor substrate. The requirement for a room temperature process is particularly critical because microcantilevers must be individually functionalized with antibody-based receptor molecules prior to bonding and cannot withstand significant heating after functionalization. I developed a new room temperature bonding method using PDMS curing agent as an intermediate adhesive layer. Two curing agents (Sylgard 184 and 182) were compared, as well as an alternate UV curable adhesive (NOA 75). The bond strength of Sylgard 184 was found to be stronger than Sylgard 182 under the same curing conditions. Overnight room temperature curing with Sylgard 184 yields an average burst pressure of 433 kPa, which is more than adequate for many PDMS sensor devices. In contrast, UV curable epoxy required a 12 hour bake at 50 °C to achieve maximum bond strength, which resulted in a burst pressure of only 124 kPa. In many biosensing scenarios it is desirable to use a small sample volume (<1 µL) to detect small analyte concentrations in as short a time as possible. I report a new microfluidic pump to address this need, which we call a reflow pump. It is designed to rapidly pump a small sample volume back and forth in a flow channel. Ultimately, the flow channel would contain functionalized sensor surfaces. The rapid flow permits use of advection-driven mass transport to the sensor surfaces to dramatically reduce sensor response times compared to diffusion-based mass transport. Normally such rapid flow would have the effect of decreasing the fraction of analyte molecules in the volume that would see the sensor surfaces. By configuring the pump to reflow fluid back and forth in the flow channel, the analyte molecules in the small sample volume are used efficiently in that they have many opportunities to make it to the sensor surfaces. I describe a 3-layer PDMS reflow pump that pumps 300 nL of fluid at 500 µL/min for 15 psi actuation pressure, and demonstrate a new two-layer configuration that significantly simplifies pump fabrication. Impedance-based nanochannel sensors operate on the basis of capturing target molecules in nanochannels such that impedance through the nanochannels is increased. While simple in concept, the response time can be quite long (8~12 hours) because the achievable flow rate through a nanochannel is very limited. An approach to dramatically increase the flow rate is to form nanochannels only during impedance measurements, and otherwise have an array of nanotrenches on the surface of a conventional microfluidic flow channel where they are exposed to normal microfluidic flow rates. I have implemented such a dynamic nanochannel approach with a recently-developed microfluidic material based polyethylene glycol diacrylate (PEGDA). I present the design, fabrication, and testing of PEGDA dynamic nanochannel array sensors, and demonstrate an 11.2 % increase in nanochannel impedance when exposed to 7.2 µM bovine serum albumin (BSA) in phosphate buffered saline (PBS). Recently, LOC biosensors for cancer cell detection have been demonstrated based on a combination of dielectrophoresis (DEP) and fluorescence detection. For fluorescence detection it is critical to minimize other sources of light in the system. However, reported devices use a non-noble metal electrode, indium tin oxide (ITO), to take advantage of its optical transparency. Unfortunately, use of non-noble metal electrodes can result in galvanoluminescence (GL) in which the AC voltage applied to the electrodes to achieve DEP causes light emission, which can potentially confound the fluorescence measurement. I designed and fabricated two types of devices to examine and identify conditions that lead to GL. Based on my observations, I have developed a method to avoid GL that involves measuring the impedance spectrum of a DEP device and choosing an operating frequency in the resistive portion of the spectrum. I also measure the emission spectrum of twelve salt solutions, all of which exhibited broadband GL. Finally, I show that in addition to Au, Cr and Ni do not exhibit GL, are therefore potentially attractive as low cost DEP electrode materials.

A case study has been conducted on the reduction of n-methyl-2-pyrrilodone (NMP) solvent waste in the manufacture of polyimide and polybenzoxazole precursors. The evaluation includes the environmental and economic assessment of solvent recovery and substitution strategies. A two-step distillation process proved effective in recovering 95% of the NMP at a purity of 99.97% from the aqueous solvent waste stream. Yearly operating costs were reduced by 83% and total life cycle emissions were reduced by 44%, due to reduction in virgin NMP use and hazardous waste disposal. With capital acquisition, the recovery option would still result in a net present value …

To accommodate this large volume of people, ventricular assist devices (VADs) are necessary to provide mechanical circulatory support. Current VADs exhibit issues such as thrombosis and hemolysis caused by large local pressure drops and turbulent flow within the pump. Chronic obstructive pulmonary disease (COPD) including emphysema is a devastating condition, increasing in prevalence in the US and worldwide. There remains no cure for COPD, rather only symptomatic treatments. Due to unique challenges of the lung, translation of therapies for acute lung injury to target chronic lung diseases like COPD has not been successful. The goal of this research was to develop a system of injectable hydrogels to deliver stem cells to musculoskeletal defects, thereby allowing cells to remain at the treatment site and secrete soluble factors that will facilitate tissue regeneration. First, production parameters for encapsulating cells in microbeads were determined. This involved investigating the effects of osmolytes on alginate microbead properties, and the effects of alginate microbead cell density, alginate microbead density, and effects of osteogenic media on microencapsulated cells. Left ventricular assist devices (LVADs) are an increasingly utilized therapy for end-stage heart failure. Thrombosis within the graft from the pump to the aorta has been documented, but is poorly researched. This study examines the effect of graft geometry, as measured by radius of curvature, bend angle, and diameter, on thrombogenic flow patterns within the graft for a range of flow conditions. It also examines the effect blood properties, including viscosity and density, on these flow patterns. The results indicated that radius of curvature had a powerful effect on thrombogenic flow patterns. Flowrate and bend angle were also influential. The …

Ge1-xSnx alloys are among a small class of benign semiconductors with composition tunable bandgaps in the near-infrared spectrum. As the amount of Sn is increased the band energy decreases and a transition from indirect to direct band structure occurs. Hence, they are prime candidates for fabrication of Si-compatible electronic and photonic devices, field effect transistors, and novel charge storage device applications. Success has been achieved with the growth of Ge1-xSnx thin film alloys with Sn compositions up to 34%. The impact of false information injection is investigated for linear dynamic systems with multiple sensors. First, it is assumed that the system is unaware of the existence of false information and the adversary is trying to maximize the negative effect of the false information on Kalman filter's estimation performance under a power constraint. Cell proliferation and contact inhibition play a major role in maintaining epithelial cell homeostasis. A hallmark of epithelial cells is strong cell-cell junctions. These junctions include E-Cadherin, a type of adherens junction that is critical for both barrier function and contact inhibition. Prior experiments by other groups have shown that adherens junctions are subject to mechanical tension. Externally applied forces (e.g. stretch) results in changes in E-Cadherin forces that coordinate proliferation. My current work tests the hypothesis that E-Cadherin forces mediate the spatial regulation of cell proliferation even in the absence of externally applied forces.

High-temperature, high-pressure (HTHP) conditions are exemplified in ultra-deep petroleum reservoirs and can be exhibited within diesel engines. Accurate pure component hydrocarbon data is essential in understanding the overall behavior of petroleum and diesel fuel at these conditions. The present study focuses on the HTHP properties of toluene since this hydrocarbon is frequently used to increase the octane rating of gasoline and toluene occurs naturally in crude oil. This is a report on the study of the drying of nanoporous polymer foam material fabricated by photolithogtaphic methods. Three drying methods were employed, which were air drying, supercritical drying and freeze drying. After fabrication and drying, physical properties of the polymer foams were measured. These measurements included density of the material, Young’s modulus, surface area, and the shape of the skeletal particles. The measurements determined the effect of the polymer concentration and the effect of drying methods. It was determined that polymer concentration had a much larger effect on the properties of the materials than the drying method.

The need for automation, optimality and efficiency has made modern day control and monitoring systems extremely complex and data abundant. However, the complexity of the systems and the abundance of raw data has reduced the understandability and interpretability of data which results in a reduced state awareness of the system. Within NASA's Air Traffic Management Technology Demonstration # 1 (ATD-1), Interval Management (IM) is a flight deck tool that enables pilots to achieve or maintain a precise in-trail spacing behind a target aircraft. Previous research has shown that violations of aircraft spacing requirements can occur between an IM aircraft and its surrounding non-IM aircraft when it is following a target on a separate route. The goal of this study is to quantify the transient performance of microfabricated superhydrophobic surfaces when used in underwater applications. A mathematical framework is developed and used to predict the stability, longevity, and drag reduction benefits of submerged superhydrophobic surfaces with two- or three-dimensional micro-textures. In addition, a novel design is proposed to improve the drag-reduction benefits of lubricant-infused surfaces, by placing a layer of trapped air underneath the lubricant layer. Friction stir welding (FSW) is a solid-state joining process that utilizes a non-consumable, rotating tool to plunge into abutting faces and transverse along the weld path. Material nearby the tool is softened by frictional heat generated via local friction and plastic deformation. Severe plastic deformation of the interface along with a state of hydrostatic pressure results in creation of a joint by metallurgical bonding. Arterial tissue failure leads to a number of potentially life-threatening clinical conditions, such as atherosclerotic plaque rupture and aortic dissection which develop unpredictably and rapidly in vivo. Thus, a full understanding of the two conditions will provide a solid basis for medical advances in the intervention and prevention of the occurrence of this life-threatening event. Specifically, the NF-κB signaling pathway plays a central role. In the US alone, 20% of citizens will be over the age of 65 by the year 2030, and the largest challenge facing this growing demographic is not a new disease but a simple motion - the fall. These events are the premier cause of fatal and nonfatal injuries. In fact, a fall is so common that every 17 seconds an older adult is treated for fall-related injuries, and every 30 minutes, an older adult dies from fall complications.
