This study presents the development of a collagen-based bioink for 3D printing tendon-to-bone tissue grafts, specifically designed for rotator cuff repair applications. Three different bioink formulations were developed: (i) 5.1% (w/v) collagen I, (ii) 5% (w/v) collagen I and collagen II, and (iii) 4.5% (w/v) collagen I combined with β-TCP. The bioink containing collagen I and collagen II was prepared by mixing the two components in a 1:1 ratio, while the collagen I plus β-TCP formulation was made by incorporating 20 wt % β-TCP into the collagen I solution. Graph-based Retrieval-Augmented Generation (GraphRAG) enhances large language models (LLMs) by grounding their reasoning in structured knowledge graphs, making them more reliable for multi-hop reasoning and factual QA. A central mechanism in Think-on-Graph systems such as ToG[14] and FastToG[7] is community detection, which groups locally related nodes into compact subgraphs so that LLMs can reason over focused, information-rich neighborhoods instead of traversing the entire graph. The Deep Underground Neutrino Experiment (DUNE) aims to advance understanding of neutrino properties using large liquid Argon time projection chambers (LArTPCs). The ProtoDUNE-II Horizontal Drift (HD) detector, constructed at CERN in Switzerland, serves as 1:20 scale prototype to validate design and operational parameters for DUNE’s Far Detectors. Accurate prediction of cryostat thermal behavior is critical to predicting liquid Argon purity distributions and detector performance, motivating the development of a validated computational fluid dynamics (CFD) model. The primary objective of this thesis is to accurately estimate the capacity of distributed energy resources (DERs) using the spatiotemporally downscaled local solar irradiance for their efficient integration into transmission-level grid operations. Accurate estimation of DER capacity is crucial for their enhanced integration into real-time electricity markets. Different methods of spatiotemporally downscaling the solar irradiance are presented in this thesis, followed by their integration into the 12 house distribution network – a low voltage residential distribution system. This thesis presents recommendations for optimizing hypersonic scramjets via a numerical study focusing on fuel type and cavity flameholder design and the impacts they have on supersonic combustion. The research is divided into two main investigations aimed at improving scramjet engine efficiency using two distinct geometrical configurations, a square cavity engine and an axisymmetric cavity engine. This paper investigates the derivation and performance of new stiffness coefficients. The estimated coefficients aimed to improve the geometric stiffness matrix representation and their application in elastic second-order analysis for steel frames. The newly developed (C1-C4) coefficients incorporate non-linear effects and reduce computational efforts to efficiently enhance the accuracy of second-order analysis. These coefficients are particularly beneficial for braced structures where they allow more refined analysis using fewer elements per member, especially as the load applied to the frame approaches the critical buckling load. Identifying adaptive mutations in genetic data is challenging due to the low frequency of occurrence of such events, and because signatures of selection are intertwined with the footprints of various other evolutionary forces that shape our genomes. Even when a larger region appears to be under selection, genomic sites that are linked to adaptive mutations have similar statistical signals, and thus can obfuscate the identification of the actual adaptive mutation. Flash floods are the costliest natural disaster in Appalachia. The goal of this study is to better understand flood response in headwater streams and the role of these systems in the generation of downstream floods. We applied a process-based hydrologic model using data from the Robinson Forest environmental monitoring network to estimate the contribution of headwaters to downstream floods. We then used the model to understand the effects of landscape disturbance in headwaters on downstream floods. This research introduces an unstable slope management program (USMP) for Tennessee based on federal slope management standards, along with improved methods for landslide monitoring with unmanned aerial systems (UAS) lidar and photogrammetry. In mountainous regions, monitoring slope hazards is a critical function of transportation management. A mobile field assessment form created with Survey123 was used to collect 22 unstable slope ratings in eastern Tennessee. Location points were appended with photographs, notes, and site information. Landslide scores ranged from 325 (Fair) to 1005 (Poor). UAS monitoring of a slow-moving soil landslide along I-40 near Rockwood, TN, produced high-resolution lidar and photogrammetry …

Architected materials with intricate three-dimensional (3D) geometries offer unique properties and functionalities but face challenges in manufacturing costs and complexity. This research explores the integration of simple 2D Greek Key-inspired lattices into fractal designs to achieve exceptional mechanical performance. Using 3D printing and kerfing, these fractal structures demonstrate enhanced flexibility and energy absorption capabilities. Through three-point bending and in-plane tensile tests, the 2D fractal lattices exhibited a 600-fold reduction in bending force and a tensile stretch capacity of up to 520%, significantly outperforming brittle parent materials. In this thesis, a novel control strategy for an inverter-driven linear compressor system is proposed in which the overall electrical losses of a coupled inverter-linear motor system are reduced through the use of a modified square wave inverter modulation scheme to minimize inverter switching losses at the expense of a modest increase in conduction losses. Gymnasts are at a high risk of Achilles tendon (AT) injuries due to intense loads during takeoffs and landings. To better understand better the relationship between impact loading and its effects on tendon adaptation and athletic performance, this study uses Inertial Measurement Units (IMUs) to track AT loading across a collegiate gymnastics season. This data were compared against physiological and athletic performance metrics, obtained through force plates and ultrasound imaging, to explore how mechanical load influences reported pain and athletic performance. The Next Round Brewing project was a project that sought to use treated wastewater to make potable drinking water using peracetic acid (PAA) and the advanced oxidation process and break the stigma around reclaimed water. In experiments, PAA was an efficient biocide which reduced the level of pathogens to within EPA standards. When PAA was coupled with the advanced oxidation process all organic matter was also reduced to within acceptable levels, and no biocide was left over in the final product, meaning PAA properly dissociated into harmless byproducts. Currently, cryovials on dry ice or liquid nitrogen vapor transportation vessels are primary ways utilized to transport mammalian cells from one location to another. Due to the safety concerns of dry ice and liquid nitrogen, associated costly shipping charges, and limitations of proper packaging and labeling of materials, these conventional shipping techniques are unacceptable. It is urgent to develop a safe and environmentally friendly protocol to preserve and ship the biological samples from one place to another.
