Modern coal preparation facilities incorporate a wide array of solid-solid and solid-liquid separation processes for rejecting mineral matter to meet market specifications. The coarse mineral matter is typically placed into engineered refuse piles whereas the fine refuse is either stored in impoundments or co-disposed with the coarse refuse. The discharge water from the refuse material represents an environmental concern due to the potential release of trace elements, and the subsequent elevation of total dissolved solids and conductivity. The models currently used to examine the behavior transportation systems are usually modespecific. That is, they focus on a single mode (i.e. railways, highways, or waterways). The lack of integration limits the usefulness of models to analyze the intermodal movement of freight. This project developed a GIS-based model of the three primary surface modes as well as intermodal connections. The resulting Integrated Freight Network Model (IFNM) accommodates highly detailed about shipping costs, transfer costs, traffic volumes (including non-freight auto traffic), and network interconnectivity properties. For decades, attempts have been made at increasing the efficiency of blasting to reduce costs and increase production. The purpose of this study was to develop and evaluate the flow rate, spray pattern, and droplet spectra characteristics of an actively controlled variable-orifice nozzle at constant carrier pressures. A commercially available variable-orifice nozzle (VariTarget) was modified to allow for direct electromechanical control of the metering stem. The modified system was tested at five carrier pressures ranging from 138 to 414 kPa and five metering stem (and thus orifice) positions. The metering stem position range was chosen because it provided a linear response in flow rate at each carrier pressure. Flow rate testing indicated a turndown ratio of 2.4:1 at …

Chemo-radiation therapy is a principal modality for the treatment of head and neck cancers, and its efficacy depends on the interaction of tumor oxygen with free radicals. In this study, we adopted a novel hybrid diffuse optical instrument combining a commercial frequency-domain tissue oximeter (Imagent) and a custom-made diffuse correlation spectroscopy (DCS) flowmeter, which allowed for simultaneous measurements of tumor blood flow and blood oxygenation. Using this hybrid instrument we continually measured tumor hemodynamic responses to chemo-radiation therapy over the treatment period of 7 weeks. Hypercholesterolemia is a risk factor for osteoporosis but the underlying mechanism is unknown. Previous evidence suggests that osteoporosis results from an impaired regulation of osteoblasts by fluid pressure fluctuations in the bone matrix. Recently, our laboratory showed that enhanced cholesterol in the cell membrane, due to hypercholesterolemia, alters leukocyte mechanosensitivity. We predict a similar link between osteoblasts and hypercholesterolemia leading to osteoporosis. Specifically, we hypothesize that extracellular cholesterol modifies the osteoblast sensitivity to pressure. MC3T3-E1 cells were exposed to hydrodynamic pressures regimes (mean=40mmHg, amplitude=0-20mmHg, frequency=1Hz) for 1-12 hours. Orthostatic intolerance (OI), i.e., the inability to maintain stable arterial pressure during upright posture, is a major problem for astronauts after spaceflight. Therefore, one important goal of spaceflight-related research is the development of countermeasures to prevent post flight OI. Given the rarity and expense of spaceflight, countermeasure development requires ground-based simulations of partial gravity to induce appropriate orthostatic effects on the human body, and to test the efficacy of potential countermeasures.

Hollow fiber membranes are widely used in blood oxygenators to remove carbon dioxide and add oxygen during cardiopulmonary bypass operations. These devices are now widely used off-label by physicians to perform extracorporeal blood oxygenation for patients with lung failure. Unfortunately, the hollow fiber membranes used in these devices fail prematurely due to blood plasma leakage and gas emboli formation.

This project formed ultrathin (~100nm) polymer coatings on polymer hollow fiber membranes. The coatings were intended to “block” existing pores on the exterior surfaces while permitting high gas fluxes. This coating is synthesized using surface imitated control radical polymerization.

Because of the rising interest in the use of corn biomass as a feedstock for biofuel production, the effects of corn stover removal on soil erosion were explored. The following full-factorial study compared fungal activity on lignocellulosic biomass that was inoculated with three different amounts of fungus, and grown using three different airflow rates. These treatments were compared to a control which consisted of biomass that was not inoculated but was exposed to the same growth conditions in the environmental chamber. The objectives of the following experiment were to determine the inoculum density and airflow rate required to optimize Phanerochaete chrysosporium lignin degradation. Additionally, this study quantifies the saccharification yield from the pretreated switchgrass.

Weathered sandstone materials have seen increased use in reclamation due to the wide-spread adoption of the Forest Reclamation Approach (FRA) in Appalachia. Runoff from these newly FRA reclaimed sites has the potential to adversely impact aquatic environments without fine sediment retention. To reduce the size and capital investment of settling ponds, flocculant utilization was investigated. Preliminary jar tests were conducted using composite weathered mine spoil samples acquired from a surface coal mine in eastern Kentucky. The purpose of this study was to investigate methods of processing lignocellulosic material so that its monosacharides can be more easily accessed for fermentation, the lack of which is hindering the economics and widescale adoption of lignocellulosic biorefining. Fabrication of effective algae cultivation systems adjacent to coal-fired power plants to fixate waste CO2 would represent a sizable step towards achieving a carbon neutral energy cycle. However, emission gas would elevate the algal cultivation system temperature and decreases its pH without expensive preprocessing. Increased temperature and acidity constitutes a profound stress on the algae. Surface mining is a commonly used method for extracting coal in the Appalachian Coalfields of the U.S. This mining practice produces excess spoil or overburden, which is often placed in adjacent valleys resulting in the creation of valley fills. These valley fills bury headwater streams, which in turn can negatively impact downstream ecosystems. Despite the industry focus on chemical synthesis, plants remain a source of potent and complex bioactive metabolites. Many of these have evolved as defensive compounds targeted on key proteins in the CNS of herbivorous insects, for example the insect dopamine transporter (DAT). This thesis recounts the study of contour blasting practices in the Appalachia coal region. Contour blasting practices vary widely and problems are often encountered. Several different sites were visited and contour blasting practices at each were studied. Based on the information gathered, a comprehensive plan was developed for blasting operations to follow and then was tested and compared to an example of blasting practices without use of the plan.
