This thesis covers a two part project: the production methods to create a double collagen binding domain molecule with a growth factor for wound healing applications and the development of a new in-house production method for isolating C1q from bovine blood. The wound healing molecule was created using transformation, sonication, and purification before being tested via electrophoresis SDS page and Western blots to confirm the molecule’s presence. The purpose of this honors thesis is to create an experiment for the CHEG Lab I course. This report explains the motivation for creating this heat convection experiment, the results of performing the experiment, and provides recommendations for future work on this experiment. Multiple experiments were performed to assess materials and parameters to be investigated. It was determined that a 0.5” plate has smaller percent error and accommodates for the desired timeframe for a Lab I experiment compared to the first plate used (1.5” thickness). When cancer metastasizes from a primary tumor site to secondary site through the bloodstream or lymph, the cancer becomes more difficult to treat. For this reason, it is vital to study what indicates the metastatic potential of a tumor. Current research has shown that cell lines with high metastatic potential display increased levels of metabolic adaptability over their nonmetastatic counterparts after undergoing hypoxic conditions. Women, people of color, and those from low-income families have less access to entrepreneurial knowledge due to the education gap. The worldwide prevalence of diabetes mellitus is rapidly increasing with about 9.3% of the adult population living with the disease. People with diabetes have trouble regulating their blood glucose levels which typically leads to hyperglycemia. Under normal physiological conditions, erythrocytes can undergo deformations in response to shear stress when passing through capillaries with a smaller diameter. Poorly managed hyperglycemia can lead to the glycosylation of erythrocyte membrane proteins and hemoglobin. Greater than 70% of strokes are of ischemic etiology and involve the occlusion of key arteries in the cardiovascular system [2]. Tissue plasminogen activator (IV-tPA) is the current gold standard for thrombolytic approaches [3]; however, this therapeutic is only effective between 3 and 4.5 hours from the patient’s last know well [4]. Slit-like pore membranes are preferred over conventional membranes with cylindrical pores since slit-like pores have two length scales that can be manipulated to better control cut off and flux through the membrane. Water electrolysis has been proposed as a renewable source of hydrogen, a possible replacement to harmful fossil fuels. This process can be broken down into two half reactions, the hydrogen evolution reaction and the oxygen evolution reaction. Because the oxygen evolution reaction has slow kinetics and a high overpotential to overcome, a catalyst is needed to speed up the kinetics and ensure faster and more efficient production of molecular hydrogen. Since it was first isolated and characterized in 2004, graphene has shown the potential for a technological revolution. This is due to its amazing physical properties such as high electrical conductivity, high thermal conductivity, and extreme flexibility. Freestanding graphene membranes naturally possesses an intrinsic rippled structure, and these ripples are in constant random motion even room temperatures. Occasionally, the ripples undergo spontaneous buckling (change of curvature from concave to convex and vice versa) and the potential energy associated with this is a double well potential. This dissertation work presents two novel converter topologies (a three-level ANPC inverter utilizing hybrid Si/SiC switches and an Asymmetric Alternate Arm Converter (AAAC) topology) that are suitable for high efficiency and high-power density energy conversion systems. The operation principle, modulation, and control strategy of these newly introduced converter topologies are presented in detail supported by simulation and experimental results. A thorough design optimization of these converter topologies (Si/SiC current rating ratio optimization and gate control strategies for the three-level ANPC inverter topology and component sizing for the asymmetric alternate arm converter topology) are also presented.

Polymeric membrane fouling is a long-standing challenge for water filtration. Metal/metal oxide nanoparticle functionalization of the membrane surface can impart anti-fouling properties through the reactivity of the metal species and the generation of radical species. Copper oxide nanoparticles (CuO NPs) are effective at reducing organic fouling when used in conjunction with hydrogen peroxide, but leaching of copper ions from the membrane has been observed, which can hinder the longevity of the CuO NP activity at the membrane surface. Chemotherapy has proven effective to reduce tumor burden for CRC patients, but there are several disadvantages associated with the use of mainstay maximtolerated dose (MTD) chemotherapeutic strategies. Harmful Algal Blooms (HABs) are becoming a global concern due to their increasing distribution, frequency, intensity, and the occurrence of toxins. While it is known that eutrophication influences algal blooms, there is less known about what triggers these HABs to produce toxins, especially microcystin. In this study, we conducted 21 community bioassays at Lake Fayetteville, a hypereutrophic reservoir in Fayetteville, Arkansas, from April-November 2021 to examine how the addition of phosphorous and nitrogen influence cyanobacteria concentrations, microcystin concentrations, and microcystin toxin production. This work establishes a method for assessing on-target precision due to CRISPR-Cas9 gene editing, especially within the context of exon skipping therapy for Duchenne Muscular Dystrophy. The proposed method utilizes an Oxford nanopore long-read sequencing approach to sequence amplified regions of DNA that have been edited using CRISPR-Cas9. NIH3T3 and C2C12 cell lines were treated with a dual-guide CRISPR-Cas9 system, that targets and deletes exon 23 from the DMD gene in mouse samples. Deletion PCR revealed deletion of exon 23 in both DNA and cDNA samples. Additionally, sequencing using Oxford Nanopore revealed targeted exon 23 deletion as the most prevalent …

In electric vehicles and airliners, the motors and power electronics are integrated into a compact space, leading to unprecedently high power density. ARKSAT-2 is a cube satellite developed by the University of Arkansas for its second CubeSat mission. There are two objectives of the ARKSAT-2 mission. The first objective of this mission is to test a novel cold gas thruster propulsion system using water-propylene propellant. This propulsion system will be used for attitude control of the satellite.
