Measuring Ligand Exchange On The Surface Of Gold Nanoparticles: A Study Of Competitive Binding Of Ligands Utilizing Fluorescent Resonance Energy Transfer, Daniel D'Unger
All Theses
This study was conducted to track ligand exchange on the surface of metallic nanoparticles in order obtain a better understanding of binding strength of various functional groups. This study develops groundwork for further understanding ligand exchange as nanoparticles transition from engineered to natural systems. A problem exists in knowing the fate of these particles if they are released into natural systems, by accident or with intent. To gain a better understanding of what may occur we look at the binding strength of different ligands. This is done to gain knowledge of the possibility of displacement of ligands occurring in the …

Biomass And Lipid Production From Heterotrophic And Mixotrophic Fed-Batch Cultivations Of Microalgae Chlorella Protothecoides Using Glycerol, Shwetha Sivakaminathan
All Theses
Chlorella protothecoides is a microalga that can grow both photo-autotrophically and/or heterotrophically under different culture and environmental conditions. In this study both the heterotrophic growth and mixotrophic growth have been conducted. The heterotrophic experiments were conducted completely in the dark while the mixotrophic experiments had the dark cycles with periodic light exposure. The aim of the study was to independently understand the effect of each mode on biomass and lipid yields.
For the heterotrophic experiments, glycerol was used as an external organic carbon source while yeast extract was used as the nitrogen source. The carbon and nitrogen source were added …
Functionalized Nanoparticles As Removable Coatings For Reverse Osmosis Membranes, Megan Smith
Functionalized Nanoparticles As Removable Coatings For Reverse Osmosis Membranes, Megan Smith
All Theses
Reverse osmosis (RO) desalination is increasingly used to produce potable water throughout the world. Despite their promising abilities, membrane filtration processes are limited by fouling. Fouling is a broad term for organics, inorganics, colloids and organisms that interact physically, chemically, or biologically with the membrane surface, resulting in reduced flux and shortened membrane lifespan. There is potential to create a snakeskin-like barrier between the membrane and foulants by electrostatically binding a coating material to the membrane that can be released by pH manipulation, removing foulants in the process. The feasibility of using functionalized nanoparticles as removable adsorptive coatings on RO …
Vibration And Acoustic Performance Of In-Plane Honeycomb Sandwich Panels, Xiao Gong
Vibration And Acoustic Performance Of In-Plane Honeycomb Sandwich Panels, Xiao Gong
All Theses
Sandwich panel structures constructed with cellular honeycomb cores allow for control of acoustic performance due to their ability to optimize effective orthotropic material properties with changes in cell geometry. By modification of topology and geometric parameters of a unit cell, desirable effective properties can be obtained and used to design lightweight structures with reduced vibration and increased sound transmission loss properties. Thus investigating the relation between the geometric configuration of the honeycomb core and vibration and acoustic behavior is important to optimize design of sandwich panels.
In this work, a finite element model is developed in MATLAB to evaluate the …
Oral Delivery Of Peptide Drugs For Mitigation Of Crohn's Disease, Paul Blichmann
Oral Delivery Of Peptide Drugs For Mitigation Of Crohn's Disease, Paul Blichmann
All Theses
Protein drugs are typically administered intravenously, but this practice has clear disadvantages such as widespread circulation and swift clearance from the body. Orally delivered protein drugs are not yet available but potentially offer improved distribution, retention, and activity by use of protective matrices and cell-specific targeting. Much work has been done on delayed release formulations for the upper intestine, but there has not been overwhelming evidence of protection of peptides from gastric conditions. In many instances the large intestine may be a better release target due to lower proteolytic activity. Afflictions of the colon such as Inflammatory Bowel Disease and …
Microstructural And Mechanical Characterization Of Soy-Based Polyethylene Fibers, Ozgun Ozdemir
Microstructural And Mechanical Characterization Of Soy-Based Polyethylene Fibers, Ozgun Ozdemir
All Theses
Bio-composite fibers have attracted increasing attention from environmentally-conscious consumers because of their low cost and biodegradability. Soy protein isolates and other purified forms have been proposed as bio-based substitutes for pure synthetic polymers for use in textiles. However, fibers from pure soy proteins do not possess adequate properties for use as textile fibers. Therefore, this research aimed to investigate composite fibers consisting of soy and polyethylene for their microstructure and mechanical properties. Changes in properties with increasing filler content and long-term properties of fibers were systematically examined.
First, the microstructure of the fibers was investigated by optical analysis. Six different …
Relating Burst Pressure To Seal Peel Strength In Pouches, William Bernal
Relating Burst Pressure To Seal Peel Strength In Pouches, William Bernal
All Theses
This work was aimed at studying the relationship between burst pressure and seal peel strength. Past researchers developed a force balance equation based on the analysis of a force diagram to relate burst pressure to seal strength. The theoretical formula S=P(D/2) (S=Seal Strength, P=Burst Pressure, D=Restraining Plate Gap) has been studied by past researchers with contrasting results.
The theoretical formula was tested by varying dwell time to produce seals of varying strength. Pouches were burst tested at each of these dwell times to obtain burst pressures, burst peeling times, and burst locations. Corresponding peel tests were performed with crosshead speeds …
Deformation And Adhesion Of Thin Metallic Films On Flexible Substrates, Julia Reid
Deformation And Adhesion Of Thin Metallic Films On Flexible Substrates, Julia Reid
All Theses
Flexible electronics are conductive circuits, components and lines integrated onto elastic substrates. These systems often consist of metallic thin films deposited onto thin sheets of glass, metals stainless steel or polymeric materials such as polyethylene or polystyrene. These systems are currently being studied for a wide variety of aeronautic and astronautical applications including: electronic skins, solar sails, and antennae structures. For applications include only moderately elevated temperatures (near 200 ¡C) and low strains (insert range), engineers have proposed incorporation metallic lines onto polymeric substrates. Like traditional microelectronic systems with films on hard substrates, the reliability is controlled by the film-substrate …
The Formulation Of A Bone Targeted Drug Delivery System Of Poly(Glycolic Acid)-Poly(Ethylene Glycol) Coated Hydroxyapatite Nanoparticles For The Delivery Of Statins, Erika Jelen
All Theses
Bone is a form of mineralized connective tissue that provides strength and rigidity to the skeleton. The two primary components within bone tissue are an organic extracellular matrix, containing type I collagen, and an inorganic mineral component composed mainly of calcium phosphate hydroxyapatite crystals. Over time the microarchitecture of bone can break down due to a variety of different factors, mainly the onset of osteoporosis in post-menopausal women, Paget's disease, and the experience of a loss of gravity during space flight. Currently there are about ten million people in the United States alone suffering from osteoporosis.
The prevention of further …
Design Of Advanced Fouling-Resistant And Self-Cleaning Membranes For Treatment Of Oily And Impaired Waters, Daniel Wandera
Design Of Advanced Fouling-Resistant And Self-Cleaning Membranes For Treatment Of Oily And Impaired Waters, Daniel Wandera
All Dissertations
The overall goal of my Doctor of Philosophy (PhD) research was to design and develop advanced anti-fouling and self-cleaning membranes for treating impaired waters. Initial work focused on the development of membranes to treat produced water, which is oily wastewater that is co-produced during oil and gas exploration. Economical, environmentally sustainable treatment of the large volumes of produced water is a grand challenge for oil and gas companies. While membranes offer many advantages over more conventional treatment methods, membrane-based treatment processes for oily waters often fail due to membrane fouling. Therefore, the primary objective of my doctoral research was to …

Overactive bladder (OAB) is a bladder disorder that is characterized by bladder storage symptoms of urgency with or without urge incontinence, frequency, nocturia, and, as of 2003, affected approximately 16.5% of adults in the United States with an annual treatment cost of over $65 billion. While therapies are available to mitigate the symptoms of OAB, there are no treatments for the cause of OAB, due to the lack of understanding of the etiology of the disorder. Recent research has provided evidence that the bladder urothelium is not just a passive barrier, but is also sensitive to various chemical and mechanical …

Prediction Of The Wear & Evolution Of Cutting Tools In A Carbide / Ti-6al-4v Machining Tribosystem By Volumetric Tool Wear Characterization & Modeling, Mathew Kuttolamadom
All Dissertations
The objective of this research work is to create a comprehensive microstructural wear mechanism-based predictive model of tool wear in the tungsten carbide / Ti-6Al-4V machining tribosystem, and to develop a new topology characterization method for worn cutting tools in order to validate the model predictions. This is accomplished by blending first principle wear mechanism models using a weighting scheme derived from scanning electron microscopy (SEM) imaging and energy dispersive x-ray spectroscopy (EDS) analysis of tools worn under different operational conditions. In addition, the topology of worn tools is characterized through scanning by white light interferometry (WLI), and then application …
Prognostic And Health-Management Oriented Fuel Cell Modeling And On-Line Supervisory System Development, Xian Zhang
Prognostic And Health-Management Oriented Fuel Cell Modeling And On-Line Supervisory System Development, Xian Zhang
All Dissertations
Of the fuel cells being studied, the proton exchange membrane fuel cell (PEMFC) is viewed as the most promising for transportation. Yet until today, the commercialization of the PEMFC has not been widespread in spite of its large expectation. Poor long term performances or durability, and high production and maintenance costs are the main reasons. For the final commercialization of fuel cells in the transportation field, durability issues must be addressed, while costs should be further brought down. At the same time, health-monitoring and prognosis techniques are of great significance in terms of scheduling condition-based maintenance (CBM) to minimize repair …
Laboratory Characterization And Theoretical Analysis Of Contaminant Mass Transfer, Xiaoling Liu
Laboratory Characterization And Theoretical Analysis Of Contaminant Mass Transfer, Xiaoling Liu
All Dissertations
Chlorinated volatile organic compounds (CVOCs) in fractured, low permeability material can be a long-term source of groundwater contaminants and they are among the most difficult remediation challenges. Thermal methods are a possible remediation option, where heat is used to boil water and the volatile compounds are stripped out by partitioning into the resulting water vapor. Theoretical analysis indicates that only a small fraction of pore water removal may lead to a significant geometric reduction in the dissolved concentration [(Udell(, 1998]. Previous modeling studies [(Falta and Murdoch(, 2011; (Pruess(, 1983] suggest that a lower permeability limit exists for effective mass transfer …
On Simulation And Modeling Of Turbulent Non-Premixed Reacting Shear Flames At Low And High Pressure, Justin Foster
On Simulation And Modeling Of Turbulent Non-Premixed Reacting Shear Flames At Low And High Pressure, Justin Foster
All Dissertations
A database of simulations of fundamental turbulent H2/O2 and H2/Air shear layer flames has been developed using the direct numerical simulation technique in which all length and time scales of a flow are fully resolved without resort to turbulence modeling. The formulation includes the fully-compressible form of the governing equations, a pressure dependent detailed chemical kinetics scheme (9 species, 19 steps), a cubic real gas equation of state, realistic property models, and generalized heat and mass diffusion models. Simulations were conducted for a temporally developing shear layer geometry and cover a wide range of initial pressures (1 atm < P < 125 atm) and flame Reynolds numbers (850 < Re < 4500). These supercomputing simulations are the largest yet performed for high pressure conditions to the author's knowledge. Computational requirements for the largest simulations included mesh sizes of ~3/4 billion grid points and ~2.2 million CPU-hrs conducted on nearly 4,000 CPU cores on Clemson's Palmetto cluster.
The database …
Propagation Of Uncertainty In Light-Frame Wood Buildings, Seyed Masood Hassanzadehshirazi
Propagation Of Uncertainty In Light-Frame Wood Buildings, Seyed Masood Hassanzadehshirazi
All Dissertations
Light-frame wood construction is the most common form of construction for residential and low-rise commercial buildings (e.g. hotels and motels) in North America. In these buildings, shearwalls are the primary systems resisting lateral loads induced by earthquakes and winds. Past earthquake events have revealed that the structural performance of nominally identical light-frame wood buildings varied significantly. Although much research has been conducted investigating the uncertainty in the performance of wood shearwalls and buildings under earthquake loading, the focus of many of these studies was on the uncertainty due to the earthquake motions, referred as earthquake-to-earthquake uncertainty, the influences of variability …
Model Of A Mechanical Clock Escapement, John Wagner, David Moline, Eugene Vold
Model Of A Mechanical Clock Escapement, John Wagner, David Moline, Eugene Vold
Publications
The mechanical tower clock originated in Europe during the 14th century to sound hourly bells and later display hands on a dial. An important innovation was the escapement mechanism, which converts stored energy into oscillatory motion for fixed time intervals through the pendulum swing. Previous work has modeled the escapement mechanism in terms of inelastic and elastic collisions. We derive and experimentally verify a theoretical model in terms of impulsive differential equations for the Graham escapement mechanism in a Seth Thomas tower clock. The model offers insight into the clock’s mechanical behavior and the functionality of the deadbeat escapement mechanism.
Deviations In Standard Aggregate Gradation And Its Affects On The Properties Of Portland Cement Concrete, Marcus Balitsaris
Deviations In Standard Aggregate Gradation And Its Affects On The Properties Of Portland Cement Concrete, Marcus Balitsaris
All Theses
Aggregate gradation is the particle size distribution of both the stone and sand present in the concrete matrix. It is an aggregate property that has been heavily researched for over a century but the effects of which on concrete properties is still somewhat misunderstood. Past research has revealed that aggregate gradation dictates the proportion of aggregate to cement paste in concrete, and can play a major variable that determines the overall durability of the construction material. Increasingly, aggregate in South Carolina are failing to meet the standard aggregate gradation for portland cement concrete, however, the effects of such failed aggregate …
Resource Allocation Framework: Validation Of Numerical Models Of Complex Engineering Systems Against Physical Experiments, Joshua Hegenderfer
Resource Allocation Framework: Validation Of Numerical Models Of Complex Engineering Systems Against Physical Experiments, Joshua Hegenderfer
All Dissertations
An increasing reliance on complex numerical simulations for high consequence decision making is the motivation for experiment-based validation and uncertainty quantification to assess, and when needed, to improve the predictive capabilities of numerical models. Uncertainties and biases in model predictions can be reduced by taking two distinct actions: (i) increasing the number of experiments in the model calibration process, and/or (ii) improving the physics sophistication of the numerical model. Therefore, decision makers must select between further code development and experimentation while allocating the finite amount of available resources. This dissertation presents a novel framework to assist in this selection between …

Assembly Time Modeling Through Connective Complexity Metrics, James L. Mathieson, Bradley A. Wallace, Joshua D. Summers
Assembly Time Modeling Through Connective Complexity Metrics, James L. Mathieson, Bradley A. Wallace, Joshua D. Summers
All CEDAR Publications
This paper presents an approach for the development of surrogate models predicting the assembly time of a system based on complexity metrics of the physical system architecture when detailed geometric information is unavailable. A convention for modelling physical architecture is presented, followed by a sample of 10 analysed systems used for training and three systems used for validation. These systems are evaluated on complexity metrics developed from graph theoretic measures. An example model is developed based on a series of regressions of trends observed within the sample data. This is validated against the systems that are not used to develop …
Evaluation Of An Enrichment Culture That Reductively Dechlorinates Γ-Hexachlorocyclohexane, Christina Anderson
Evaluation Of An Enrichment Culture That Reductively Dechlorinates Γ-Hexachlorocyclohexane, Christina Anderson
All Theses
In a previous study using microcosms from a site contaminated with γ- hexachlorocyclohexane (γ-HCH), rapid anaerobic biodegradation of γ-HCH to benzene and chlorobenzene was observed. Enrichment cultures were developed and subsequently transferred to an anaerobic sulfate-free HEPES buffered medium without loss of γ-HCH dechlorination, thus suggesting γ-HCH undergoes anaerobic biodegradation via chlororespiration. The overall objective of this thesis was to further characterize the enrichment culture.
To do so, one specific objective was to determine the effect of vancomycin on γ-HCH in an enrichment culture grown in a sulfate-free HEPES buffered medium using hydrogen as the electron donor. This objective would …

Piercing The Cloud And Marking The Onion: Removing The Anonymity Of The Tor Network, James Ashworth
Piercing The Cloud And Marking The Onion: Removing The Anonymity Of The Tor Network, James Ashworth
All Theses
In this work, we extend previous work done on removing anonymity from The Onion Routing network (Tor). We explore previous techniques for removing Tor's anonymity developed on a private Tor network, and attempt to reproduce these results on the global public network.
We find that the previous work done on the private network is unable to be carried over to the public network. This is mainly due to the level of jitter on the public network overwhelming our earlier method for compromising Tor's anonymity. We develop a new method for compromising Tor's anonymity by using a clustering algorithm to analyze …
Surface Characterization Of Nephila Clavipes Dragline Silk, Benoit Faugas
Surface Characterization Of Nephila Clavipes Dragline Silk, Benoit Faugas
All Theses
To design synthetic fibers for extreme applications, many research groups are trying to identify how the structure of natural fibers (such as spider silk) leads to desirable properties such as high tensile strength.
Spider silk is a biological fiber having mechanical properties that exceed those of the best man made fibers in terms of high tensile strength and large extensibility, this combination providing the silk with a large work of rupture. While the bulk structure, composition and properties have been intensively studied elsewhere, this study focuses on elucidating the Nephila clavipes spider dragline silk surface structure and composition, as well …

A New Metaheuristic Algorithm Motivated By The Rooting Phenomena In Plants, Behzad Nemati
All Theses
In this thesis, we present a new metaheuristic algorithm for analyzing challenging optimization problems. Initial testing reveals the potential for the algorithm, which is based on the rooting phenomena found in plants in nature, to quickly produce effective solutions to challenging combinatorial optimization problems in the class NP. We present computational results describing the proposed metaheuristic's efficacy for an important class of scheduling problems and discuss areas of future work and application.
Finite Element Modeling And Design Of Honeycomb Sandwich Panels For Acoustic Performance, David Griese
Finite Element Modeling And Design Of Honeycomb Sandwich Panels For Acoustic Performance, David Griese
All Theses
Honeycomb cellular metamaterial structures offer many distinct advantages over homogenous materials because their effective material properties depend on both their constituent material properties and their geometric cell configuration. From this, a wide range of targeted effective material properties can be achieved thus supporting forward design by tailoring the honeycomb cellular materials and properties for specific applications. One area that has not been fully explored is the set of acoustic properties of honeycomb materials and how these can offer increased design flexibility when targeting acoustic performance. Understanding these relations, the designer can effectively tune designs to perform better in specific acoustic …

Application Of Microbial Fuel Cells In A Forested Wetland, Jianing Dai
All Theses
ABSTRACT
Electricity can be generated from forest residues, especially the organic matter from litter fall, in freshwater forested wetland environments with a microbial fuel cell (MFC) system. Electricity generation efficiency was examined for cypress, tupelo, and pine litter extracts, using dual-chamber reactors. The maximum power density generated with cypress extracts was on the level of 320 mW/m2, higher than tupelo (230 mW/m2), and pine (210 mW/m2). The efficacy of cypress extracts was also higher than tupelo and
pine, demonstrating that organic matter from cypress litter is a preferred substrate for electricity generation with MFCs. Meanwhile, low aromaticity of the substrate …
