In this thesis, the successful fabrication of 3D-printed microfluidic devices will be discussed. Fabrication is performed with a low-cost commercially available stereolithographic 3D printer utilizing a custom PEGDA resin formulation tailored for low non-specific protein adsorption based on my colleagues' work [Rogers et al., Anal. Chem. 83, 6418 (2011)]. Horizontal microfluidic channels with designed rectangular cross sectional dimensions as small as 300 um wide and 150 um tall are printed with 100% yield, as are cylindrical vertical microfluidic channels with 300 um designed (334 um actual) diameters. The purpose of this project is to quantify the effects of increasing spinal ligament fidelity on the mechanics of the human lumbar spine using finite element analysis (FEA). In support of this goal, a material characterization study was completed to provide anisotropic, nonlinear material parameters for the human anterior longitudinal ligament. (ALL). Cadaveric samples of the human ALL were tested using a punch test technique. Multi- axial force-deformation data were gathered and fit to a commonly used transversely isotropic material model using an FEA system identification routine. Heavy reliance on GPS is preventing unmanned air systems (UAS) from being fully inte- grated for many of their numerous applications. In the absence of GPS, GPS-reliant UAS have difficulty estimating vehicle states resulting in vehicle failures. Additionally, naively using erro- neous measurements when GPS is available can result in significant state inaccuracies. We present a simultaneous localization and mapping (SLAM) solution to GPS-degraded navigation that al- lows vehicle state estimation and control independent of global information. In this thesis, frame synchronization for iNET formatted SOQPSK-TG communications is considered. Frame synchronization for M-ary linear modulations (MQAM, MPSK, etc.) are known in the literature using pilot detection methods, but are based on a signal model that does not apply to SOQPSK-TG. Maximum likelihood frame synchronizers are derived for an SOQPSK-TG system following assumptions found in the literature. The analysis shows that a reinterpretation of known detectors operating on the samples of the received waveform and locally stored samples of the pilot is the optimum approach for this case. Three-dimensional two-phase unsteady CFD simulations were run on three and four-blade inducers for the purpose of analyzing differences in cavitation stability at design and off-design flow rates. At design flow rates, there were very small differences between the breakdown curves for the three and four-bladed inducers. However, at lower cavitation numbers, the three-bladed inducer exhibited up to three times the rotor forces than the four-bladed inducer. When moving to off-design flow rates, both inducers experienced multiple modes of cavitation instabilities including rotating cavitation, alternate-blade cavitation, and cavitation surge. This work investigates the application of active noise control (ANC) to speech. ANC has had success reducing tonal noise. In this work, that success was extended to noise that is not completely tonal but has some tonal elements such as speech. Limitations such as causality were established on the active control of human speech. An optimal configuration for control actuators was developed for a sphere using a genetic algorithm. This paper presents a strategy for optimal estimation of parameters for towed cable systems using moving horizon estimation (MHE). The main contributions of the work include a novel formulation of MHE using a dead-band that explicitly rejects measurement noise, real-time implementation results, and the investigation of time-varying stochastic disturbances as well as unknown yet constant disturbances. Further analysis is conducted on the observability and sensitivity of key parameters to determine which parameters can be estimated by the proposed approach using real-time streaming data from experiments. With the increasing demand to integrate unmanned aircraft systems (UAS) into the National Airspace System (NAS), new procedures and technologies are necessary to ensure safe airspace operations and minimize the impact of UAS on current airspace users. Currently, small UAS face limitations on their use in civil airspace because they lack the ability to detect and avoid other aircraft. This article presents a framework that consists of an Automatic Dependent Surveillance-Broadcast (ADS-B)-based sensor, track estimator, conflict/collision detection, and resolution that mitigates collision risk. A review of current variable-stiffness actuators reveals a need for more simple, cost effective, and lightweight designs that can be easily incorporated into a variety of human-interactive robot platforms. This thesis considers the potential use of compliant mechanisms to improve the performance of variable-stiffness actuators. The advantages and disadvantages of various concepts using compliant mechanisms are outlined, along with ideas for further exploration. A novel nanosized liposomal carrier was prepared consisting of perfluorocarbon emulsion droplet(s) encapsulated inside a liposome, which is called an eLiposome. This 200-nm construct is useful for acoustically activated drug delivery because ultrasound causes the emulsion droplet to expand from liquid to gas, which expansion ruptures the eLiposome bilayer and releases the internal contents. This study focuses on the parameters that influence control of the release of calcein (a model drug), including ultrasound amplitude, insonation time, and perfluorocarbon vapor pressure as a function of temperature and composition. In this thesis, the Recursive-Random Sample Consensus (R-RANSAC) multiple target tracking (MTT) algorithm is further developed and applied to video taken from static platforms. Development of R-RANSAC is primarily focused in three areas: data association, the ability to track maneuvering objects, and track management. The probabilistic data association (PDA) filter performs very well in the R-RANSAC framework and adds minimal computation cost over less sophisticated methods. The interacting multiple models (IMM) filter as well as higher-order linear models are incorporated into R-RANSAC to improve tracking of highly maneuverable targets. This thesis describes two research efforts to improve two different aspects of hydrologic data collection and management. First described is a recipe for the design, development, and deployment of a low-cost environmental data logging and transmission system for environmental sensors and its connection to an open source data-sharing network. Modeling Stokes Flow Using Hierarchical Structure-Preserving B-Splines, Kendrick Monroe Shepherd

A new spline space, the hierarchical structure-preserving B-spline space, is introduced and implemented in the analysis of Stokes flow. The space, when properly constrained, is shown to be stable and to have at least optimal convergence rates in the velocity field and suboptimal convergence rates in the pressure field. However, results show that superoptimal convergence can often be expected in the pressure field, likely due to error reduction in the velocity field. Like other hierarchical spline spaces, these splines are shown to greatly increase accuracy and to drastically lower computation times for analyses on domains whose solution fields have singularities …

However, field data to characterize emergent vegetation at many reservoirs may not be available or may be limited to small, isolated areas. Both of these have potential for application in the design of deployableand foldable arrays of spatial mechanisms.Origami is considered a compliant mechanisms because it achieves its motion through thedeflection of paper creases. Compliant mechanisms generally do not allow for continuous rotation;however, the compliant kaleidocycle represents an exception to this generality. The research in this dissertation is divided between three different approaches for predicting the shear strength of reinforcement masonry shear walls. Each approach provides increasing accuracy and precision in predicting the shear strength of masonry walls. The three approaches were developed or validated using data from 353 wall tests that have been conducted over the past half century. The data were collected, scrutinized, and synthesized using principles of meta-analysis. Predictions made with current Masonry Standards Joint Committee (MSJC) shear strength equation are unconservative and show a higher degree of variation for partially-grouted walls.
