Scalable Assembly Of Nanoparticles Onto Templated Substrates, John Juchnowski
Undergraduate Research Posters 2016
Anisotropic nanoparticles, such as carbon nanotubes and noble metal nanorods, have excellent electrical, mechanical, and thermal properties. This work examined techniques for the nanopatterning of silicon substrates for the subsequent deposition of anisotropic nanoparticles in order to exploit their properties in macroscopic applications. Argon-ion-sputter induced rippling of Si (100) as well as a microparticle mask were examined as methods for templating a silicon substrate. For the ion-sputter

Glycerolipid Analysis Of Adaptation To Saline Changes In The Culture Conditions Of Algae, Scenedesmus Dimorphus, By Gc-Ms, Tyler Fitzgerald, Satya Girish Chandra Avula, Chandana Mannem
Undergraduate Research Posters 2016
Since fossil fuels are decreasing over time an alternative energy source will be soon required. The algae, Scenedesmus dimorphus, grows in freshwater and is known for its fast growth of glycerolipid content which is used for biodiesel production. After the algae is grown in optimal conditions, the released fatty acids and glycerolipids are transformed into fatty acid methyl esters (FAMEs) which are used as biodiesel. The FAMEs were quantitatively determined by gas chromatography mass spectrometry (GC-MS) to determine the total glycerolipid content in the different algae samples. The samples that were analyzed include freshwater controls and saline adapted samples. Analysis …

Active Stiffening Of F-Actin Network Dominated By Structural Transition Of Actin Filaments Into Bundles, Shengmao Lin, Xinwei Han, Gary C.P. Tsui, David Hui, Linxia Gu
Department of Mechanical and Materials Engineering: Faculty Publications
Molecular motor regulated active contractile force is key for cells sensing and responding to their mechanical environment, which leads to characteristic structures and functions of cells. The F-actin network demonstrates a two-order of magnitude increase in its modulus due to contractility; however, the mechanism for this active stiffening remains unclear. Two widely acknowledged hypotheses are that active stiffening of F-actin network is caused by (1) the nonlinear force-extension behavior of cross-linkers, and (2) the loading mode being switched from bending to stretching dominated regime. Direct evidence supporting either theory is lacking. Here we examined these hypotheses and showed that a …

Cyber Center Publications
Cells alter their gene expression in response to exposure to various environmental changes. Epigenetic mechanisms such as DNA methylation are believed to regulate the alterations in gene expression patterns. In vitro and in vivo studies have documented changes in cellular proliferation, cytoskeletal remodeling, signal transduction, bone mineralization and immune deficiency under the influence of microgravity conditions experienced in space. However microgravity induced changes in the epigenome have not been well characterized. In this study we have used Next-generation Sequencing (NGS) to profile ground-based “simulated” microgravity induced changes on DNA methylation (5-methylcytosine or 5mC), hydroxymethylation (5-hydroxymethylcytosine or 5hmC), and simultaneous gene …

Effect Of Twisted Fiber Anisotropy In Cardiac Tissue On Ablation With Pulsed Electric Fields, Fei Xie, Christian W. Zemlin
Effect Of Twisted Fiber Anisotropy In Cardiac Tissue On Ablation With Pulsed Electric Fields, Fei Xie, Christian W. Zemlin
Bioelectrics Publications
Background: Ablation of cardiac tissue with pulsed electric fields is a promising alternative to current thermal ablation methods, and it critically depends on the electric field distribution in the heart.
Methods: We developed a model that incorporates the twisted anisotropy of cardiac tissue and computed the electric field distribution in the tissue. We also performed experiments in rabbit ventricles to validate our model. We find that the model agrees well with the experimentally determined ablation volume if we assume that all tissue that is exposed to a field greater than 3 kV/cm is ablated. In our numerical analysis, we considered …

Analysis Of Ca²⁺ Mobilization Of Nanosecond Pulses In Cells, Iurii Semenov
Bioelectrics Publications
Exposure to nanosecond electric pulses (nsEPs) evokes transient rise of cytosolic free Ca²⁺ concentration ([Ca²⁺]i) in the cells. We quantified the external Ca²⁺ uptake, intracellular Ca²⁺ release and uncover interplay of these processes for CHO-K1 cells upon exposure to single 60 ns pulse in the range of amplitudes from 0 to 30 kV/cm. NsEPs are distinguished by the ability to electroporate intracellular membranes. We studied how duration of nsEPs is related to efficiency of intracellular membranes electroporation comparing ability of single 300-, 60- and 10 ns pulses to recruit Ca²⁺ from endoplasmic reticulum (ER) in respect to the uptake of …

The goal of this study was to investigate the role of MLC phosphatase (MLCP) in a LPS model of acute lung injury (ALI). We demonstrate that ectopic expression of a constitutively-active (C/A) MLCP regulatory subunit (MYPT1) attenuates the ability of LPS to increase endothelial (EC) permeability. Down-regulation of MYPT1 exacerbates LPS-induced expression of ICAM1 suggesting an anti-inflammatory role of MLCP. To determine whether MLCP contributes to LPS-induced ALI in vivo, we utilized a nanoparticle DNA delivery method to specifically target lung EC. Expression of a C/A MYPT1 reduced LPS-induced lung inflammation and vascular permeability. Further, increased expression of the …

A dielectrically loaded wideband rod antenna has been studied as a pulse delivery system to subcutaneous tissues. Simulation results applying 100-ps electrical pulse show that it allows us to generate a critical electric field for biological effects, such as brain stimulation, in the range of several centimeters. In order to reach the critical electric field for biological effects, which is similar to 20 kV/cm, at a depth of 2 cm, the input voltage needs to be 175 kV. The electric field spot size in the brain at this position is similar to 1 cm(2). Experimental studies in free space with …

Investigation Of Enzymatically Synthesized Glycogen As A Novel Nanodendrimer For Therapeutic Delivery, Sarah Ann Engelberth
Legacy Theses & Dissertations (2009 - 2024)
The field of medicinal chemistry is ever expanding, designing and discovering new therapeutic strategies. Oftentimes, it is challenging for these therapeutics to undergo clinical translation due to ineffective administration or unwanted toxicity in vivo. As such, drug delivery vehicles are designed to overcome these hurdles, allowing for delivery to the site of action by improving biodistribution, protecting therapeutic cargo, and decreasing toxicity. The work presented here aims to investigate a naturally-derived carbohydrate nanodendrimer, enzymatically synthesized glycogen (ESG) for drug delivery. This nontoxic, highly-branched, glucose-based structure has interior void volumes to allow for cargo encapsulation as well as a large density …

Toward Magnetic Resonance Only Treatment Planning: Distortion Mitigation And Image-Guided Radiation Therapy Validation, Ryan Glen Price
Wayne State University Dissertations
While MR-only treatment planning has shown promise, there are still several well-known challenges that are currently limiting widespread clinical implementation.
Firstly, MR images are affected by both patient-induced and system-level geometric distortions that can significantly degrade treatment planning accuracy. . In addition, the availability of comprehensive distortion analysis software is currently limited. Also while many groups have been working toward a synthetic CT solution, further study is needed on the implementation of synCTs as the reference datasets for linac-based image-guided radiation therapy (IGRT) to help determine their robustness in an MR-only workflow.

Surface-Initiated Polymerizations For The Rapid Sorting Of Rare Cancer Cells, Jacob L. Lilly
Theses and Dissertations--Chemical and Materials Engineering
Cancer metastasis directly accounts for an estimated 90% of all cancer related deaths and is correlated with the presence of malignant cells in systemic circulation. This observed relationship has prompted efforts to develop a fluid biopsy, with the goal of detecting these rare cells in patient peripheral blood as surrogate markers for metastatic disease as a partial replacement or supplement to tissue biopsies. Numerous platforms have been designed, yet these have generally failed to support a reliable fluid biopsy due to poor performance parameters such as low throughput, low purity of enriched antigen positive cells, and insufficiently low detection thresholds …

Biventricular Finite Element Modeling And Quantification Of 3d Langragian Strains And Torsion Using Dense Mri, Zhanqiu Liu
Theses and Dissertations--Mechanical and Aerospace Engineering
Statistical data suggests that increased use of evidence-based medical therapies has largely contributed to the decrease in American death rate caused by heart disease. And my studies are about two applications of magnetic resonance imaging (MRI) as a non-invasive approach in evidence-based health care research. In my first study, the achievement of a pulmonary valve replacement surgery was assessed on a patient with tetralogy of Fallot (TOF). In order to evaluate the remodeling of right ventricle, two biventricular finite element models were built up for pre-surgical images and post-surgical images. In my second study, 3D Lagrangian strains and torsion in …
Development And Application Of Gadolinium Free Cardiac Magnetic Resonance Fibrosis Imaging For Multiscale Study Of Heart Failure In Patients With End Stage Renal Disease, Tori A. Stromp
Theses and Dissertations--Physiology
Cardiac magnetic resonance (CMR) is a powerful tool to noninvasively image ventricular fibrosis. Late gadolinium enhancement (LGE) CMR identifies focal and, with T1 mapping, diffuse fibrosis. Despite prevalent cardiac fibrosis and heart failure, patients with end stage renal disease (ESRD) are excluded from LGE. Absence of a suitable diagnostic has limited the understanding of heart failure and obstructed development of therapies in the setting of ESRD. A quantitative, gadolinium free fibrosis detection method could overcome this critical barrier, propelling the advancement of diagnostic, monitoring, and therapy options. This project describes the development of a gadolinium free CMR technique and application …

Études primaires
Expertise is a key factor modulating the risk of low back disorders (LBD). Through years of practice in the workplace, the typical expert acquires high level specific skills and maintains a clean record of work-related injuries. Ergonomic observations of manual materials handling (MMH) tasks show that expert techniques differ from those of novices, leading to the idea that expert techniques are safer. Biomechanical studies of MMH tasks performed by experts/novices report mixed results for kinematic/kinetic variables, evoking potential internal effect of expertise. In the context of series of box transfers simulated by actual workers, detailed internal loads predicted by a …

How the human brain plans, executes, and monitors continuous and fluent speech has remained largely elusive. For example, previous research has defined the cortical locations most important for different aspects of speech function, but has not yet yielded a definition of the temporal progression of involvement of those locations as speech progresses either overtly or covertly. In this paper, we uncovered the spatio-temporal evolution of neuronal population-level activity related to continuous overt speech, and identified those locations that shared activity characteristics across overt and covert speech. Specifically, we asked subjects to repeat continuous sentences aloud or silently while we recorded …

A New Technique To Improve The Operation Of Prosthetic Limbs During Muscle Fatigue, H. Albunashee, G. Rasool, K. Iqbal, G. White
Journal of the Arkansas Academy of Science
Prosthetic limbs hold a promise to renew the quality of life for the amputee. Neural commands are decoded via a classifier to generate control signals for the prosthetic devices. In the literature, many challenges and limitations have been identified that affect the prosthesis operation. One such drawback is muscle fatigue which degrades the surface electromyogram (sEMG) signals, and consequently, the performance of the deployed classification algorithm declines from 90% to 50% of average accuracy. We used a new technique using the Linear Discrimination Analysis (LDA) algorithm and the muscle synergy-based task discrimination (MSD) algorithm to improve the classification accuracy. In …

Mechanical & Aerospace Engineering Faculty Publications
In light of the need of tissue palpation for Robotics-assisted Minimally Invasive Surgery (RMIS), this paper presents a microfluidic-based tactile sensor for palpating mice tissues for tumor localization. The core of the sensor is a 3x3 sensing-plate/transducer array built into a single polydimethylsiloxane (PDMS) microstructure, with a transducer spacing of 3.75mmx1.5mm. Mounted on a robot, the sensor is pressed against a tissue region with a pre-defined indentation depth pattern, and consequently the stiffness distribution across the tissue region translates to the deflection distribution of the sensing-plate array and is captured by the transducer array underneath as resistance changes. Thus, the …

Hydroband: The Continuous Hydration Monitoring System, Karina E. Atkinson, Catherine P. Benedict
Williams Honors College, Honors Research Projects
Dehydration is defined as a mild, moderate, or extreme loss of water in the body. It can result in fever, muscle cramps, headaches, dizziness, or heat stroke in athletes. Ways to determine hydration levels include analysis of the urine, blood, or sweat, body weight changes, skin elasticity tests, and increases in core temperature. Patent search, research journals, and outside studies in the health and fitness market showed that a convenient and continuous hydration monitoring system was not available. Changes in core body temperature proved to be the quickest way to measure hydration levels. The HydroBand system is geared towards athletes …

Recognizing Pain Using Novel Simulation Technology, Justin C. Grace
Honors Undergraduate Theses
Effective pain management and time to treatment is essential in patient care. Despite scientific evidence supporting the need to treat pain and an emphasis on addressing pain as a priority, pain management continues to be an unresolved issue. As a member of the health care team, nurses are integral to optimal pain management. Currently, nursing schools have limited innovative or alternative methods for teaching pain assessment and management. Simulation in nursing education provides a unique opportunity to expose students to realistic patient situations and allow them to learn and make mistakes without causing harm. However, modern low- and high-fidelity simulation …
