Implantable drug delivery devices have many benefits over traditional drug administration techniques and have attracted a lot of attention in recent years. By delivering the medication directly to the tissue, they enable the use of larger localized concentrations, enhancing the efficacy of the treatment. Passive-release drug delivery systems, one of the various ways to provide medication, are great inventions. However, they cannot dispense the medication on demand since they are nonprogrammable. This has cause a growing number of patients to develop disabilities over time due to spinal cord injuries and stroke among other neurological ailments. The field of neurotechnology research has a bright and promising future as more devices are created. However, there are still many gaps in the field as the potential for neuromodulation grows. Devices such as the Micro-Leads StimZ system and the Ripple Grapevine/Summit system help bridge that gap by allowing for a broader variety of closed-loop neuromodulation experiments to be implemented thanks to their portability and versatility. The Tissue Engineering Lab at California Polytechnic State University San Luis Obispo focuses on creating tissue-engineered Blood Vessel Mimics (BVMs) designed for the preclinical testing of neurovascular devices. These BVMs are composed of silicone models, representing anatomically accurate neurovasculatures, that are sodded with vascular cell types and then cultivated in bioreactors (which maintain physiologic conditions). These silicone models are currently sourced externally from industry partners, so the primary goal of this thesis was to develop the means and methods for the Tissue Engineering Lab to manufacture silicone models in-house.

Breast cancer is a global problem, and it is inevitable to detect cancerous cells at early stages. In recent years, microwave imaging technology has been widely applied in biomedical applications for its non-ionizing radiation. In this study, we design a micro-strip patch antenna with an inverted T-type notch in the partial ground to detect tumor cells inside the human breast. The size of the current antenna is small enough (18 mm × 21 mm × 1.6 mm) to distribute around the breast phantom. Breast cancer has become the most prominent cancer worldwide in women. Annual mammograms are encouraged for women of high risk to increase early detection allowing for lumpectomies rather than mastectomies to occur. Prior to a lumpectomy, a biopsy must be taken to determine if the tissue is cancerous, and a breast cancer biopsy marker (BBM) is left in the region of possible cancerous tissue. In a Type 1 Diabetic, Insulin can be administered in a pump system. There are two types of insulin that must be given: basal and bolus. Basal insulin is a long-acting form of insulin that works in the background while fasting, while Bolus insulin is rapid/short acting given in response to food to immediately begin working to lower blood sugar.

Degenerative joint disease, or osteoarthritis, is a common occurrence in the knee joint. This can often result in joint pain, decrease in range of motion, and ultimately disability. One way to counteract osteoarthritis is the incorporation of a total knee arthroplasty (TKA). TKAs replace the damaged bone and soft tissue surrounding the knee with metal and polyethylene components. Ideally this will improve the joint’s performance and reduce pain. However, there is still a number of TKA patients who remain dissatisfied. In order to investigate this, in depth research must be done on the design and performance of TKAs.

Many patients struggle with disabilities that hinder their ability to walk. This project aimed to create a leg assembly capable of variable gravity so that it could be combined with a Robotic Walking Training Device, and lead to better rehabilitation options for patients. This was accomplished by deriving equations of joint torque, creating circuit diagrams for Arduino systems, modeling leg assemblies in CAD, and finally combining it to create a working small-scale prototype. Transoral robotic surgery (TORS) is a minimally invasive approach in treating head and neck cancers and has demonstrated improved surgical outcomes with reduced morbidity when compared to traditional open surgery. However, the lack of haptic feedback makes it more difficult to assess the tumor extent and locate critical structures (e.g., artery and nerves). In standard-of-care TORS, preoperative imaging is used as guidance; however, it becomes inaccurate intraoperatively: the patient’s neck is hyper-extended, surgical instruments are introduced, and soft tissues deform significantly. Total ankle replacement (TAR) implants are an effective option to restore the range of motion of the ankle joint for arthritic patients. An effective tool for analyzing these implants’ mechanical performance and longevity in-silico is finite element analysis (FEA). ABAQUS FEA was used to statically analyze the von Mises stress and contact pressure on the articulating surface of the bearing component in two newly installed fixed-bearing total ankle replacement implants (the Wright Medical INBONE II and the Exactech Vantage). Breast Cancer Related Lymphedema (BCRL) is a common co-morbidity in cancer survivors following neoadjuvant therapies such as chemotherapy, radiation, and/or surgery. It is brought about by the disruption in the lymphatic system (think lymph node biopsy) that leads to a buildup of lymphatic fluid in the arm. This dissertation aimed to advance knowledge of how subglottal stenosis impacts voice production physiology. An in-house fluid-structure-acoustic interaction approach based on the hydrodynamic/acoustic splitting technique was employed. This technique was rigorously verified for simulating phonation by matching the acoustic behavior to a compressible flow solver for phonation-relevant geometries. Simulations of an idealized 2D vocal tract model demonstrated the effects of supraglottal acoustic resonance on vocal fold kinematics and glottal flow waveform. Results showed that the acoustic coupling between higher harmonics and formats generated pressure oscillations, modifying vocal fold dynamics and glottal flow rate.

A Calibrated Cuff Plethysmography device was built, tested for verification, and used to experiment on human subjects to measure the cardiovascular response of consuming a beet supplement, specifically looking at arterial compliance and pressure-area curves. Each subject was tested four times. A baseline was measured under normal conditions and after five-minute hyperemia conditions. 10 subjects were given 6 ounces of water mixed with either purple Kool-Aid (control), a SuperBeets supplement, or a SuperBeets Sport supplement and after 45 minutes, measurements were taken undergoing normal and hyperemia conditions once more. Reactive chemistries for protein chemical modification play an instrumental role in chemical biology, proteomics, and therapeutics. Depending on the application, the selectivity of these modifications can range from precise modification of an amino acid sequence by genetic manipulation of protein expression machinery to a stochastic modification of lysine residues on the protein surface. Approximately 19% of breast cancer patients undergoing breast conserving surgery (BCS) must return for a re-excision surgery due to positive margin reported by pathology. This problem is mainly associated with lack of intraoperative cancer margin assessment tools. Our previous work demonstrated that lipid-weighted photoacoustic imaging (PAI) can be used intraoperatively to successfully differentiate between cancerous and healthy tissue with high sensitivity. As the prevalence of cardiovascular diseases continues to exponentially grow in populations across the globe, the necessity of determining underlying factors, effective methods of diagnoses, and universally available preventive measures also grows. Early detection of endothelial dysfunction, a proven precursor of cardiovascular diseases, can be extremely impactful in encouraging preventative measures and early intervention before medical conditions become chronic.
