Alzheimer's disease (AD) is the most common form of dementia, a disease that gradually destroys brain cells and leads to progressive decline in mental function. The presence of high densities of neuritic plaques composed of Abeta in the cerebral cortices is a criterion for the post-mortem diagnosis of AD. The view that Abeta deposition drives the pathogenesis of AD (amyloid hypothesis) has received support from a wide range of molecular, genetic, and animal studies. This hypothesis has been the focus of therapeutic intervention leading to the development of anti-Abeta immunotherapy as a potential treatment. There is a great deal of …

Revisiting the American Gothic via Julia Kristeva's theory of "the abject" demonstrates how Gothic strategies expose the historical contradictions of race in works by Mark Twain, Charles W. Chesnutt, and Toni Morrison. As theorized by Kristeva in Powers of Horror, the archaic process in which the subject attempts to constitute itself as homogeneous by casting off or "abjecting" all that cannot be assimilated to the self-same necessarily opens the way to repeated returns of the abject(ed) and the "horror" it provokes. Because the Gothic enacts the return of the abject, it was itself abjected from the literary canon until recently. …

This study establishes a link between research on organizational source credibility and the effects of public information campaigns. Research has established that source credibility is one factor audiences evaluate when responding to messages and that credible information sources enhance message acceptance, while untrustworthy sources can interfere with desired message effects. Although source credibility studies have typically focused on the person delivering a message, recent studies indicate that audience perceptions of the organization sponsoring a message has a direct effect on message acceptance as well. The Reproducing Kernel Element Method (RKEM) implementation of the Fleck-Hutchinson phenomenological strain gradient theory in 1D, 2D and 3D is implemented in this research. Fleck-Hutchinson theory fits within the framework of Touplin- Mindlin theories and deals with first order strain gradients and associated work conjugate higher-order stress. Theories at the intrinsic or material length scales find applications in size dependent phenomena. In elasticity, length scale enters the constitutive equation through the elastic strain energy function which depends on both strain as well as the gradient of rotation and stress. This treatise presents a transcendental argument for corporate social responsibility. The argument is that corporate social responsibility, or CSR, is best understood as a collective moral practice that is a precondition for sustainable business. There are a number of theories and definitions of CSR in the contemporary business literature. These theories include considerations of economic, legal, social, and environmental notions of what a corporation ought to take responsibility for based on either motives or concerns of accountability for corporate acts. Organizational researchers focus on group level variables such as organizational climate and organizational structure. The purpose of the current meso-level study is to examine the effects of the structure of an academic department on faculty members' job performance, job satisfaction, and prevalence of counterproductive work behavior (CWB), or harmful behaviors while at work. The sample consisted of 1135 full time faculty members working in 229 academic departments throughout the United States and Canada. Results of the study suggested that faculty members working in a more organically structured department report higher levels of job satisfaction. Additionally, productive faculty members working in …

The Dona and Roberts Bay watershed comprises one of the five major watersheds in Sarasota County, Florida. It is connected to the Gulf of Mexico via Venice Inlet. Like many estuaries in the Florida, significant modifications have been made to the drainage basins, principally to the main tributaries. The system has undergone several anthropogenic modifications to enhance water management efforts. Creeks that comprise fresh water input into the system have been dammed in order to inhibit the upstream flow of salt water during the dry season. Flavonoids are a long and well known class of natural products. Their potential health benefits can be attributed to their antioxidant activity, and modulation of cell signaling pathways. Green tea one of the most widely consumed beverages, consist of flavonoids such as catechins and tannins. Epigallocatechin gallate (EGCG) the major catechin of green tea exhibits multiple health benefits due to its antioxidant nature. Research on organizational citizenship behaviors (OCB) has focused on the positive aspects of the construct, neglecting the possibility that individuals who engage in OCB may suffer negative consequences. Thus, the present study expands the literature by examining the possibility that OCB is also related to negative individual-level factors, such as work-to-family conflict (WIF). In a replication and extension of Bolino and Turnley's (2005) research, the present study offers an in-depth analysis of the relationship between OCB and WIF, examining two potential mediators (work time and role overload) and two potential moderators (gender and perceptions of OCB as discretionary). Two highly polar sol-gel coatings were developed for capillary microextraction (CME). One of the coatings contained cyanopropyl-polydimethylsiloxane (CN-PDMS) and the other low molecular weight polyethylene glycol. These highly polar coatings were immobilized via sol-gel chemistry allowing for direct chemical bonding to the inner surface of fused silica capillaries. These sol-gel coated microextraction capillaries were employed in CME for solvent-free microextraction and preconcentration of trace analytes (polar, moderately polar, and nonpolar) from aqueous matrices. CN-PDMS and short chain PEG extraction phases exhibit both polar and polarizable characteristics. Therefore, both sol-gel CN-PDMS and short chain sol-gel PEG coatings were able to extract …

This thesis improves a previous kinematic analysis and develops the elastic portion of the analysis of a curved compliant beam. This analysis is used to develop a Pseudo-Rigid-Body Model for the curved compliant beam. The Pseudo-Rigid-Body Model consist of kinematic and elastic parameters which can be used to simplify the computation of the large deflections of the beam as it undergoes spherical motion. The kinematic parameters that are developed are the characteristic radius, Gamma*length, the parametric angle coefficient, c_theta, and the kinematic parametrization limit, Capital_theta_max(Gamma). When Henry James sat down to write his "amusette" as he called The Turn of the Screw (1898), he created various ambiguities in the text as a means of confusing and surprising his readers or, in other words, catching them off guard. The purpose of the present study is to examine the theme of literature in Rayuela (Julio Cortázar, 1963) and Tres tristes tigres (Guillermo Cabrera Infante, 1967), taking into account the importance of this theme within the socio-historical and intellectual context of 1960's Latin America, an era characterized not only by the triumph of the Cuban Revolution in the political field, but also the height of poststructuralist literary theory, which arrives in Latin America via Europe. The MDM2 homolog MDMX is an important regulator of p53 during mouse embryonic development. DNA damage promotes MDMX phosphorylation, nuclear translocation, and degradation by MDM2. Here we show that MDMX copurifies with 14-3-3, and DNA damage stimulates MDMX binding to 14-3-3. Chk2-mediated phosphorylation of MDMX on S367 is important for stimulating 14-3-3 binding, MDMX nuclear import by a cryptic NLS, and degradation by MDM2. Mutation of MDMX S367 inhibits ubiquitination and degradation by MDM2, and prevents MDMX nuclear import. Expression of 14-3-3 stimulates the degradation of phosphorylated MDMX. Chk2 and 14-3-3 cooperatively stimulate MDMX ubiquitination and overcome the inhibition of …

The principal objective of this paper is to analyze Sir Gawain's efforts to balance the conflicting requirements of the Code of Chivalry with the basic needs of human nature to develop insights into the Gawain's character. Using an amalgamated definition of chivalry as the standard, I will examine Gawain's attempts to achieve his goal of being the perfect chivalric knight, determine the nature of his obstacles, and analyze the development of his character. In trying to live up to perfection, Gawain discovers that he is not perfect.

The purpose of this study was to identify and examine the role of dispositional characteristics in effective mentoring relationships. A learning and development framework was incorporated to examine the relationships of protégé and mentor characteristics, mentoring provided, and developmental mentoring outcomes. First, relationships between individual characteristics and mentoring provided were examined. Second, relationships between individual characteristics and partner developmental outcomes were examined. Third, mentoring provided was examined as a mediator of individual characteristics and partner developmental outcomes. The final sample consisted of 93 protégé-mentor pairs. The effects of the toxic dinoflagellate, karenia brevis (Wilson clone), on larval survival and development of the northern quahog (=hard clam, Mercenaria mercenaria), eastern oyster (Crassostrea virginica) and bay scallop (Argopecten irradians) were studied in the laboratory. The effects of K. brevis on feeding activities of juveniles from these species plus the green mussel (Perna viridis) were also examined. Finally, adult bay scallops were exposed to K. brevis for two weeks to investigate possible cytotoxic effects. Survival of 3-day-old larvae was generally > 85% for all shellfish species at Karenia brevis densities of 100 cells . ml-1 or less, and not …

This thesis is a broad survey of the uses of ambiguity in academic discourse. I note the uses of ambiguity in literary criticism, linking ambiguity with epistemic relativism. Then I pose the question, is the notion of "reframing" in psychotherapy analogous to the concept of "spin" in propaganda, advertising and public relations? In a consideration of theories that posit the social construction of reality, I examine articles by Judith Butler and Ian Hacking, noting the ambiguous reception of performativity and nominalism, respectively, within academia. Goethe's Faust and Nabokov's Humbert both are erudite, middle-aged European scholars who, experiencing a convergence of academic and existential ennui, set eyes upon a young girl and instantly are consumed with lust. In both works the girls' widowed mothers die as a result of the protagonists' lustful intentions; a cross-country flight ensues; the once-respected scholars are wanted for murder; and Gretchen and Lolita each suffer from their sexual and emotional objectification. But the connections between Goethe's play and Nabokov's novel extend far beyond plot points, or even their decidedly different receptions in early 19th century Germany versus mid-20th century America. …

Adolescence is a period of development that is associated with increased risk taking behaviors and experimenting with drugs of abuse, including alcohol and nicotine. Early onset of use of these agents may be associated with long-term changes in behavior and enhanced sensitivity to the subsequent effects of alcohol in adulthood. We have designed and examined a compliant knee mechanism that may offer solutions to problems that exist for infants and toddlers who are just learning to walk. Pediatric prosthetic knees on the market today are not well designed for infants and toddlers for various reasons. Children at this age need a prosthetic that is light in weight, durable, and stable during stance. Of the eleven knees on the market for children, all but three are polycentric or four-bar knees, meaning they have multiple points of movement. This empirical research study investigated the phenomenon of ancient language teacher cognition in the technology-rich contexts of two secondary teachers of Latin. Although one teacher was an inexperienced teacher working in a traditional, face-to-face classroom in an independent school, the other teacher was an experienced teacher working at a public virtual school. Features of teacher cognition, such as pre-active planning and inter-active decision-making were examined from a communities-of-practice theoretical perspective (Wenger, 1998). In this dissertation, I explore the question of how women alcoholics achieve sobriety. Using narrative inquiry, I focus on the identity transformation that must occur in order to maintain sobriety and how a drinking self is deconstructed and reconstructed as a sober self. Today, alcoholism is still viewed as stigmatizing in our society and in all Western cultures. The stigma of alcoholism makes it difficult for alcoholics to communicate their experiences to people who have not had similar experiences. Analysis of extremes may aid in developing predictive models in hydro-climatology by giving enhanced understanding of processes driving the extremes and perhaps delineate possible anthropogenic or natural causes. Speaker or voice recognition is the task of automatically recognizing people from their speech signals. This technique makes it possible to use uttered speech to verify the speaker's identity and control access to secured services. Surveillance, counter-terrorism and homeland security department can collect voice data from telephone conversation without having to access to any other biometric dataset. Protein-protein interactions are central to most biological processes. Although in the field of drug discovery there is a great interest in targeting protein-protein interactions, the discovery and development of small-molecules, which effect these interactions has been challenging. The purpose of this project is to determine if in situ click chemistry is a practical approach towards testing whether Bcl-XL is capable of assembling it's own inhibitory compounds. Abbott laboratories developed compound ABT-737, which binds with high affinity (Ki < 1 nM) to the binding sites of Bcl-XL.36.
