The endoplasmic reticulum (ER) plays an indispensable role in cellular processes, including maintenance of calcium homeostasis, and protein folding, synthesized and processing. Disruptions in these processes leading to ER stress and the accumulation of misfolded proteins can instigate the unfolded protein response (UPR), culminating in either restoration of balanced proteostasis or apoptosis. A key player in this intricate balance is CLCC1, an ER-resident chloride channel, whose essential role extends to retinal development, regulation of ER stress, and UPR. The importance of CLCC1 is further underscored by its interaction with proteins localized to mitochondria-associated endoplasmic reticulum membranes (MAMs), where it participates …

Imaging glucose metabolism with [18F]fluorodeoxyglucose positron emission tomography has transformed the diagnostic and treatment algorithms of numerous malignancies in clinical practice. The cancer phenotype, though, extends beyond dysregulation of this single pathway. Reprogramming of other pathways of metabolism, as well as altered perfusion and hypoxia, also typifies malignancy. These features provide other opportunities for imaging that have been developed and advanced into humans. In this review, we discuss imaging metabolism, perfusion, and hypoxia in cancer, focusing on the underlying biology to provide context. We conclude by highlighting the ability to image multiple facets of biology to better characterize cancer and …

Heart disease involves irreversible myocardial injury that leads to high morbidity and mortality rates. Numerous cell-based cardiac in vitro models have been proposed as complementary approaches to non-clinical animal research. However, most of these approaches struggle to accurately replicate adult human heart conditions, such as myocardial infarction and ventricular remodeling pathology. The intricate interplay between various cell types within the adult heart, including cardiomyocytes, fibroblasts, and endothelial cells, contributes to the complexity of most heart diseases. Consequently, the mechanisms behind heart disease induction cannot be attributed to a single-cell type. Thus, the use of multi-cellular models becomes essential for creating …

We performed genome-wide association studies of breast cancer including 18,034 cases and 22,104 controls of African ancestry. Genetic variants at 12 loci were associated with breast cancer risk (P < 5 × 10-8), including associations of a low-frequency missense variant rs61751053 in ARHGEF38 with overall breast cancer (odds ratio (OR) = 1.48) and a common variant rs76664032 at chromosome 2q14.2 with triple-negative breast cancer (TNBC) (OR = 1.30). Approximately 15.4% of cases with TNBC carried six risk alleles in three genome-wide association study-identified TNBC risk variants, with an OR of 4.21 (95% confidence interval = 2.66-7.03) compared with those carrying fewer than two risk alleles. A polygenic risk score (PRS) showed an area under the receiver operating characteristic curve of 0.60 for the prediction of breast cancer risk, which outperformed PRS derived using data from females of European ancestry. Our study markedly increases the population diversity in genetic studies for breast cancer and demonstrates the utility of PRS for risk prediction in females of African ancestry.

Repair of large bone defects caused by severe trauma, non-union fractures, or tumor resection remains challenging because of limited regenerative ability. Typically, these defects heal through mixed routines, including intramembranous ossification (IMO) and endochondral ossification (ECO), with ECO considered more efficient. Current strategies to promote large bone healing via ECO are unstable and require high-dose growth factors or complex cell therapy that cause side effects and raise expense while providing only limited benefit. Herein, we report a bio-integrated scaffold capable of initiating an early hypoxia microenvironment with controllable release of low-dose recombinant bone morphogenetic protein-2 (rhBMP-2), aiming to induce ECO-dominated …

The purpose of this study is to examine the dosimetric difference between the step-and-shot intensity modulated radiation therapy (IMRT) plans on Elekta Unity and our clinical volumetric modulated arc therapy (VMAT) plans on Varian TrueBeam for HN SBRT.

METHOD: Fourteen patients treated on TrueBeam sTx with VMAT treatment plans were …

Parent’S Perspectives On The Impact Of Recurrent Otitis Media On Speech And Language Development, Andrea Rose Wilson May 2024

Parent’S Perspectives On The Impact Of Recurrent Otitis Media On Speech And Language Development, Andrea Rose Wilson

Theses and Dissertations

The purpose of this study is to investigate the parent’s perspective on whether otitis media delayed their child’s speech and language development. It also focuses on intervention options and the parent’s relationship with their medical providers regarding intervention options for otitis media. Research has shown the importance of early intervention; however, when it comes to recurrent otitis media, many physicians require patients to express symptoms for extended periods before surgical solutions are considered (Granath, 2021). An electronic survey was distributed to parents and caregivers on various parenting Facebook pages. The study was open for one month before the research was …

“Can I Have It Non-Personalised?” An Empirical Investigation Of Consumer Willingness To Share Data For Personalized Services And Ads, Monika Leszczynska, Daria Baltag May 2024

“Can I Have It Non-Personalised?” An Empirical Investigation Of Consumer Willingness To Share Data For Personalized Services And Ads, Monika Leszczynska, Daria Baltag

Faculty Scholarship

European regulators, courts, and scholars are currently debating the legality of data processing for personalization purposes. Should businesses require separate consent for processing user data for personalized advertising, especially when offering free services reliant on such ads for revenue? Or is general consent for the contract enough, given personalized advertising’s role in fulfilling contractual obligations? This study investigates whether these legal distinctions reflect differences in people’s willingness to share data with businesses for personalization. Are consumers less willing to share their data for personalized advertising than for personalized services that they clearly contracted for? Does that change if the service …

Over 50% of patients with systemic LCH are not cured with front-line therapies and data to guide salvage options are limited. We describe 58 patients with LCH who were treated with clofarabine. Clofarabine monotherapy was active against LCH in this cohort including heavily pretreated patients with systemic objective response rate of 92.6%, higher in children (93.8%) than adults (83.3%). BRAFV600E+ variant allele frequency in peripheral blood correlated with positive clinical responses. Prospective multi-center trials are warranted to determine optimal dosing, long-term efficacy, late toxicities, relative cost and patient reported outcomes of clofarabine compared to alternative LCH salvage therapy strategies.

Background: Obesity or overweight raises the risk of developing 13 types of cancer, representing 40% of all cancers diagnosed in the United States annually. Given the ongoing debate surrounding the impact of metabolically healthy obesity (MHO) on cardiovascular outcomes, it is crucial to comprehend the incidence of Major Adverse Cardiovascular and Cerebrovascular Events (MACCEs) and the influence of MHO on these outcomes in cancer patients. Methods: Data of hospitalized cancer patients with and without obesity were analyzed from the National Inpatient Sample 2016-2020. Metabolically healthy patients were identified by excluding diabetes, hypertension, and hyperlipidemia using Elixhauser comorbidity software, v.2022.1. After …

Broadway Theatre And The Covid-19 Pandemic: How Pre-Shutdown Longevity Impacted Production Success, Emmeline A. Lignowski May 2024

Broadway Theatre And The Covid-19 Pandemic: How Pre-Shutdown Longevity Impacted Production Success, Emmeline A. Lignowski

College Honors Program

On March 12, 2020 New York’s Broadway theaters halted performances due to the COVID-19 pandemic. After a year and a half of closure, Broadway finally reopened in August 2021, however, the market was drastically different from its pre-shutdown state. In the midst of a global pandemic, new productions faced increased challenges when trying to establish themselves on the Broadway stage, and even old productions struggled to recoup losses. In this paper, multiple regression analysis and a Cox proportional hazards model were used to test whether a production’s pre-shutdown longevity impacted its success after Broadway reopened. It was found that productions …

Supporting The Creative Side Of Creative Coding: Helping Students Wield Code In The Pursuit Of Their Own Dreams, Gretchen K. Larsen May 2024

Supporting The Creative Side Of Creative Coding: Helping Students Wield Code In The Pursuit Of Their Own Dreams, Gretchen K. Larsen

Dissertations and Doctoral Documents, University of Nebraska-Lincoln, 2023–

Creative coding has become more common in art and new media schools, however research describing how teachers navigate the unique challenges of teaching code as a creative medium is limited. This paper presents the details of a creative coding course designed to support the ‘creative’ side of creative coding. As a result of this research, I present a framework for supporting the ‘creative’ side of creative coding, which connects to arts-based education theories, modified for the creative code context. This paper asks—what helps students use creative coding as an artistic medium in the pursuit of their own questions and meaning? …
