Targeting The Bcl2 Family: Advances And Challenges In Bh3 Mimetic-Based Therapies, Nabanita Mukherjee, James Sheetz, Yiqun Shellman
Student Papers, Posters & Projects
The BCL2 family of proteins plays a pivotal role in regulating apoptosis and cellular homeostasis, making them critical therapeutic targets in cancer and other diseases characterized by pathological cell survival. BH3 mimetics, small molecules that selectively inhibit anti-apoptotic BCL2 family members, have achieved significant clinical success, particularly in hematologic malignancies. However, several challenges remain, including resistance mechanisms, toxicity (such as MCL1 inhibitor-associated cardiotoxicity), and the intricate balance between apoptotic and non-apoptotic functions. This review provides a comprehensive overview of BCL2 family biology, the development and clinical application and outcomes of BH3 mimetics, and the emerging resistance mechanism known as double-bolt …

All Decisions
The Appellate Term affirmed the Civil Court's dismissal of the tenant's complaint for breach of lease; breach of warranty of habitability; and retaliation. The court properly applied res judicata; finding these claims arose from the same transactions as those previously settled or litigated in a prior landlord and tenant proceeding; including an allegation of harassment that should have been raised in Housing Court. Additionally; the tenant's affirmation opposing the landlord's motion was inadmissible for failing to include the mandatory language required by CPLR 2106.

All Decisions
In a nonpayment proceeding where the tenant interposed a rent overcharge defense, the court granted the tenant's motion for discovery. The court found the apartment's rent history unreliable due to missing registrations and a significant rent increase, warranting discovery beyond the statutory lookback period. The landlord's cross-motion for outstanding and pendente lite use and occupancy was denied without prejudice, as the tenant raised colorable defenses of rent overcharge and warranty of habitability under RPAPL § 745(2)(a)(iv)-(v).

Cowles Foundation Discussion Papers
In many real recommender systems, novel items are added frequently over time. The importance of sufficiently presenting novel actions has widely been acknowledged for improving long-term user engagement. A recent work builds on Off-Policy Learning (OPL), which trains a policy from only logged data, however, the existing methods can be unsafe in the presence of novel actions. Our goal is to develop a framework to enforce exploration of novel actions with a guarantee for safety. To this end, we first develop Safe Off-Policy Policy Gradient (Safe OPG), which is a model-free safe OPL method based on a high confidence off-policy …

Predictors Of Lower-Limb Arterial Occlusion Pressure Across Commonly Used Cuff Widths, Isaac J. Wedig, Isaac M. Lennox, Erich J. Petushek, John J. Durocher, John Mcdaniel, Steven J. Elmer
Michigan Tech Publications
We compared predictors of lower-limb arterial occlusion pressure (AOP) across commonly used blood flow restriction (BFR) cuff widths (11, 13, 18 cm) and developed prediction equations to estimate AOP for each cuff. Participants (n = 116) underwent measurements of thigh circumference (TC), systolic (SBP) and diastolic (DBP) blood pressure, and AOP was assessed using Doppler ultrasound in a seated position. Multiple linear regression models with commonality analysis and mixed-effects models were used to identify and compare predictors of AOP between each cuff. LASSO regression with bootstrap resampling was used to develop and internally validate prediction equations. TC, SBP, DBP, age, …

B cells express many protein ligands, yet their regulatory functions are incompletely understood. We profiled ligand expression across murine B sublineage cells, including those activated by defined receptor signals, and assessed their regulatory capacities and specificities through in silico analysis of ligand-receptor interactions. Consequently, we identified a B cell subset that expressed cytokine interleukin-27 (IL-27) and chemokine CXCL10. Through the IL-27–IL-27 receptor interaction, these IL-27/CXCL10-producing B cells targeted CD40-activated B cells in vitro and, upon induction by immunization and viral infection, optimized antibody responses and antiviral immunity in vivo. Also present in breast cancer tumors and retained there through CXCL10-CXCR3 …
Representation, Dynamics And Optimization Of Physical Structures, Fan Feng
Representation, Dynamics And Optimization Of Physical Structures, Fan Feng
Dartmouth College Ph.D Dissertations
Computational tools play a vital role in structural analysis and design, enabling engineers and researchers to optimize performance, reduce material usage, and explore novel configurations across solids and fluids. Capturing, analyzing, and optimizing these structures while balancing efficiency, stability, and adaptability remains a central challenge. This thesis proposes three complementary methods that advance representation, simulation, and optimization. We present an impulse (gauge) fluid formulation that utilizes the impulse gauge variable to reduce numerical diffusion and better preserve coherent flow features such as vorticity at practical resolutions. We introduce a differentiable Voronoi representation for cellular solids that provides smooth implicit boundaries …

The aim of this study was to understand the experiences of rural, first-generation college (RFGC) students in relation to dominant culturally normative expectations about postsecondary educational and workforce trajectories. The study adopted patterns of engagement with a master narrative as both a conceptual framework and unit of analysis, examining both biography and cultural ecologies as well as highlighting agency in the lives and choices of study participants. Evidence is drawn from 4 in-depth narrative interviews with each of 14 RFGC students, conducted both at school and students’ homes, from rural and small-town communities. Results show that a master narrative of …

Stressing The Stress Tests, Natasha Sarin, Til Schuermann
Journal of Financial Crises
In the spring of 2023, Silicon Valley Bank, Signature Bank, and First Republic Bank failed, owing to predictable rate hikes by the Federal Reserve. It’s clear from these recent events that the current US stress-testing system is ill-suited to mitigate risk associated with the banking system, especially when looking at smaller regional banks that aren’t subjugated to annual stress testing. Stress testing as a regulatory tool is also exposed to significant legal risks following recent Supreme Court jurisprudence that draws into question the viability of the status quo. Today, regulators and other interested parties must change tacks to craft a …

The Tax Trench Deepens, Tracey M. Roberts
The Tax Trench Deepens, Tracey M. Roberts
Florida Tax Review
Congress, long divided, has been in gridlock for decades. The result has been regulatory ossification. Existing statutes lack the flexibility to manage the country’s many technological, social, environmental, sectoral and market challenges. However, without a Senate supermajority vote to override the filibuster, new legislation fails. While the budget reconciliation process has provided one of the few pathways around the filibuster, that process is limited to budgetary outlays and revenue measures. Consequently, a “tax trench” has been worn from Congress repeatedly pushing major regulatory and public benefits legislation through the process as tax provisions. Now, the tax trench is set to …
Tax Regulations In A Loper Bright Light, Blaine G. Saito
Tax Regulations In A Loper Bright Light, Blaine G. Saito
Florida Tax Review
The Supreme Court’s decision in Loper Bright Enterprises. v. Raimondo overturned Chevron deference. In doing so, Loper Bright loosed chaos into many areas of administrative law, including tax. Indeed, the majority opinion in Loper Bright undid Chevron but left open the question of what the actual standard of review is, namely, whether it is de novo or Skidmore applies.
Tax too will suffer some of the chaos, especially as courts work out the matters. But both the positive citation in the opinion of Skidmore along with some of the language supporting long-standing regulations that are contemporaneously promulgated with the statute’s …
Have Your Cake And Eat It Too, Using Apportionment To Preserve Congress's Tax Power, Donald B. Tobin
Have Your Cake And Eat It Too, Using Apportionment To Preserve Congress's Tax Power, Donald B. Tobin
Florida Tax Review
The Constitution provides Congress with extremely broad taxing power, and the Supreme Court has recognized that this power is “virtually without limitation.” As budget deficits and income inequality in the United States increase, policymakers are examining options to tax wealth, either through a wealth tax or through taxes on untaxed capital gains. Opponents of these measures have turned to the apportionment requirement in the Constitution and argued that these taxes are not “income taxes” and instead are “direct taxes” that must be apportioned among the states. They further argue that since these taxes cannot be apportioned fairly, any wealth tax …

The Decline Of Chevron, The Rise Of Loper Bright And The Shifting Sands Of Tax Law, Doron Narotzki, Tamir Shanan
Florida Tax Review
The repeal of the Chevron deference doctrine has triggered what some may argue is a seismic shift in how tax law may be interpreted and applied, which may have far-reaching implications for taxpayers, the Internal Revenue Service and the federal government. This Article examines the evolving landscape of U.S. federal tax law in the wake of the Loper Bright decision, specifically focusing on the tax and economic consequences for key tax areas such as the General Anti-Abuse Rule and the complicated foreign tax credit and transfer pricing regimes, as well as the rules that characterize financial instruments or equity investments …

Moore Questions, Some Answers: Fixing The Personal Tax System Despite Constitutional Constraints, David Gamage, John R. Brooks, Edward J. Mccaffery
Florida Tax Review
Moore v. United States was expected to rule on the constitutional necessity of the tax-law realization requirement originating from Eisner v. Macomber, a potential impediment to progressive tax reform efforts aimed at shutting down the planning techniques of Buy Borrow Die. The various opinions in Moore, however, provided no definitive answer to this core question, instead leaving many more questions. Amid the lingering uncertainty, we argue that various responses to the problem of wealthy Americans’ not needing to pay any taxes remain possible after Moore. An incremental, “mix-and-match” approach to progressive tax reform may best suit …

Containing The Blast Radius: Can Congress Save The Code From Realization?, Reuven Avi-Yonah, Elise Hocking, Muskan Sharma
Florida Tax Review
The Supreme Court’s decision in Moore v. United States avoided enshrining realization as a constitutional requirement for taxing income. But four Justices were in favor of doing so, and the majority opinion explicitly avoided deciding the issue. Thus, it is quite plausible to imagine that a future case would garner five votes in favor of constitutionalizing realization. This Article addresses the question whether Congress can do anything preemptively to contain the “blast radius” of such an explosive decision and limit the ensuing “fiscal calamity.”
Charles And Kathleen Moore And The Coming Tax Armageddon, Calvin H. Johnson
Charles And Kathleen Moore And The Coming Tax Armageddon, Calvin H. Johnson
Florida Tax Review
The Supreme Court decided last term, against predictions, that U.S. shareholders could be taxed on the undistributed earnings of their foreign corporation. The Court expressly did not decide the “Armageddon question,” whether a wealth tax or mark-to-market tax would be constitutional.
In the coming Armageddon, the Court needs to reverse the error in Pollock v. Farmers’ Loan & Trust Co., which said that the Constitution “prevent[s] an attack upon accumulated property by mere force of numbers.” That is exactly wrong. Apportionment of direct tax in fact was written to reach the wealth of the states using the labor of …

Florida Tax Review
In the 2023 Term, the Supreme Court decided three cases that are likely to have significant implications for the federal tax system. Moore v. United States presages a resuscitated constitutional realization requirement for the income tax—a requirement long thought moribund, if not dead, by most commentators and policymakers. Loper Bright Enterprises v. Raimondo jettisoned the Chevron doctrine’s deferential standard of review of agency rulemaking, returning the standard to some perhaps modified version of its more searching pre-Chevron status. And Corner Post, Inc. v. Board of Governors of the Federal Reserve System held that the statute of limitations for most …
