We present a detailed, xeno-free protocol for the rapid differentiation of human induced pluripotent stem cells (hiPSCs) into microglia using the well-characterized KOLF2.1J reference line. This system employs doxycycline-inducible expression of six transcription factors (6-TF), stably integrated into the CLYBL safe harbor locus, to drive uniform microglial differentiation within two weeks. Adapted from Dräger et al. (1), our protocol includes key optimizations for KOLF2.1J, including culture on Laminin-521 to support xeno-free conditions. The resulting i-Microglia exhibit hallmark features of mature microglia, including expression of P2RY12, loss of the pluripotency marker SSEA4, phagocytic activity, and upregulation of immune markers (e.g., CD80, …

Ectopic pregnancy remains a significant contributor to pregnancy-related morbidity and mortality, yet little is known about whether its occurrence follows discernible temporal patters that could inform prediction and prevention efforts. This dissertation aimed to explore the temporal characteristics of ectopic pregnancy through three interrelated projects: development of a new theoretical framework, a scoping review of the literature on ectopic pregnancy temporal patterns, and an empirical study using electronic health records data from a ten-year period at a single academic medical center. Per- and polyfluoroalkyl substances (PFAS) are synthetic compounds that have been detected in water sources of urban and rural environments with evidence of adverse human health effects from exposure. Humans are primarily exposed through ingestion of contaminated water and food. PFAS exposure has been linked to negative multi-system health outcomes (liver, kidneys, cardiovascular, reproductive, and immune). PFAS occur as persistent mixtures in environmental media with decades long consequences to water quality. While national agencies have launched large-scale PFAS water quality monitoring studies, limited data exists for contamination of rural water in Nebraska, especially private wells. A critical area of cancer research, symptom management, has evolved from focusing on individual symptoms to recognizing the complexity of co-occurring symptoms, commonly referred to as symptom clusters. However, most studies on pediatric cancer symptom clusters have focused primarily on the treatment phase or long-term survivorship, overlooking the early post-treatment period. We present a method to identify and categorize gravitational wave candidate triggers identified by matched filtering gravitational wave searches (pipelines) caused by transient noise (glitches) in gravitational wave detectors using support vector machine (SVM) classifiers. Our approach involves training SVM models on pipeline triggers which occur outside periods of excess noise to distinguish between triggers caused by random noise and those induced by glitches. This method is applied independently to the triggers produced by the GstLAL search pipeline on data from the laser interferometer gravitational-wave observatory Hanford and Livingston observatories during the second half of the O3 observing run. Summary In this Doctor of Nursing Practice (DNP) project, I evaluated whether the project site organization’s implementation of leadership rounding reduced patient falls within a medical-surgical unit. Despite established fall prevention strategies, the unit continued to experience ongoing incidents, prompting the implementation of structured leadership rounding. The practice-focused question assessed changes in fall rates over a 3-month postintervention period compared to the preceding 3 months. A retrospective chart review was conducted from December 2023 to June 2024, excluding March 2024 due to partial implementation. I used descriptive statistics to analyze trends. Gram-negative bacteria produce chaperone-usher pathway (CUP) pili, which are extracellular adhesive protein fibers critical for host-pathogen interactions. An outer-membrane usher protein, together with a periplasmic chaperone, assembles thousands of pilin subunits into a pilus fiber. Each pilus is tipped by an adhesin that mediates host and tissue tropisms by binding to receptors with stereochemical specificity. Type 1 pili expressed by uropathogenic Escherichia coli (UPEC) are essential virulence factors in urinary tract infections (UTIs) and facilitate attachment to mannosylated glycoproteins on bladder epithelial cells. Background/aim: Childhood-onset arthritis may result from various rheumatic diseases. This study aimed to systematically evaluate their distribution and characteristics in a large pediatric cohort.
Materials and methods: We retrospectively analyzed all pediatric patients who presented with arthritis to the pediatric rheumatology clinic between January 2000 and January 2023.
Results: A total of 1713 pediatric patients who presented with arthritis were included in this study (median age at diagnosis: 6.8 years; F/M = 1.2). Most of the patients (n = 859, 50.1%) had juvenile idiopathic arthritis (JIA) (especially oligoarticular JIA). Background/aimː This study aims to investigate the prevalence of malignancy in patients with inflammatory bowel disease (IBD) followed up in a tertiary reference center.
Materials and methodsː IBD patients with at least 6 months of follow-up from the gastroenterology clinic between 2000 and 2022 wereevaluated retrospectively in a tertiary center. Patient information was obtained from the patient registration system.
Resultsː There were 697 patients in the study, 320 (45.9%) of these were female. The mean age of the patients at IBD diagnosis was 33.4± 13.1 years. The mean follow-up time was 93.1 ± 64.8 (median 84, IQR (25–75) (36–144)) months. …

Background/aim: Gynecological malignancies, including those affecting the uterus, cervix, vagina, vulva, and adnexa, pose significant physical and psychosocial burdens. Early detection and effective management of these malignancies are critical for improving outcomes. This study aims to evaluate metabolic uptake patterns in gynecological organs using fluorodeoxyglucose positron emission tomography (FDG-PET) and to analyze their malignancy potential in women with nongynecological cancers.

Background/aim: Microplastics (MPs) are a growing concern due to their pervasive environmental presence and potential environmental and human exposure impacts. Given evidence of systemic MP presence in human tissues and fluids, and the ear canal's environmental exposure, this novel study aimed to identify these particles in human cerumen. Cerumen was collected from the proximal ear canal to minimize potential airborne contamination.
