A Persistent Homology Framework For Scrna-Seq: Assessing Clustering Robustness And Quantifying Preprocessing And Integration Effects On Topological Features., Jonah Daneshmand

As single-cell RNA sequencing (scRNA-seq) data expands, robust methods for integrating diverse datasets are critical. This dissertation applies Persistent Homology (PH), a technique from Topological Data Analysis (TDA), to a collection of scRNA-seq datasets spanning eight tissue types to quantify how data integration affects topological features and biological interpretability. We assessed global topological structure using Betti curves, Euler characteristics, and persistence landscapes across raw, normalized, and integrated data representations. Our analysis revealed a performance inversion: while conventional methods excelled on unintegrated data, high-granularity topological methods, particularly those sensitive to global data structure, became superior after integration. Introduction. E-cigarettes (e-cigs) have been shown to be pro-arrhythmic in both human and male rodent studies; however, little is known about how e-cig use impacts patients with a predisposed heightened risk for arrhythmias, including pregnant women. Both pregnancy and e-cig use have independently been shown to increase heart rate, diminish heart rate variability, and increase ventricular premature beats, which is suspected to be due to sympathetic dominance. Importantly, β-blockers have been shown to reduce sympathetic activity, reduce heart rate, increase HRV, and reduce arrhythmias. Thus, the use of β-blockers may help prevent adverse health effects driven by sympathetic dominance from …

Crystallization behavior of three edible fats: interesterified soybean oil (IESBO), interesterified palm olein (IEPO), and fully hydrogenated palm kernel oil (FHPKO) were explored to understand how this process is affected by different factors. The study focused on how the chemical composition of these fats affects their behavior like how much time they require to be crystallized at a certain temperature. By using different scientific methods, how quickly each fat began to crystallize and how much energy it took to start the process was measured. Friction stir welding (FSW) is a manufacturing process used to join materials through intense heat and pressure. To better understand and optimize this FSW process, mathematical models incorporating non-Newtonian Navier-stokes equations for large plastic deformation and heat equation for the heat transfer are used to simulate the FSW process. However, solving these coupled and nonlinear equations with high accuracy requires significant computational power and time. This thesis has applied model order reduction, more precisely the Proper Orthogonal Decomposition (POD) and Discrete Empirical Interpolation Method (DEIM), to efficiently solve the FSW system in low-dimensional space. One such disease is chronic wasting disease (CWD), a fatal illness affecting deer and related species in North America and other regions. CWD is caused by prions, misfolded proteins that can remain infectious for years after shedding by infected hosts.

Female breast cancer (BC) remains the most commonly diagnosed non-skin cancer and the second leading cause of cancer death among U.S. women. Despite advances in detection and treatment, substantial inequities in BC outcomes persist, driven in part by structural forces such as residential segregation. This dissertation examined the relationship between residential segregation, BC outcomes, and key contextual factors in Louisville, Kentucky. Area-level SDOH factors that coexist to form “high-risk” clusters are more prevalent in rural than urban areas. The goal of this work is to better understand these interactions by further characterizing an effector from Microbotryum lychnidis-dioicae and heterologously expressing this and other effectors in heterologous species to see if the range of these effectors can play a role in pathogenicity when expressed in distantly and closely related fungal species. Background: Falls are the leading cause of reported injuries within the hospital setting. Injuries from falls occur in about20-45% of the time and is the leading cause of injury and injury death among the adult population. Inpatient falls are associated with an additional $14,0000 per occurrence and a prolonged hospitalization by 6.3 days. Increased falls rates affect hospitals rating, accreditation and reimbursements. This hospital is experiencing inpatient falls that exceed national average and corporate goals.

This qualitative study explored the life experiences of Black alumni and their participation in intentional, multicultural programming of cultural facilities to create a sense of counterspace for Black students on historically White college campuses. The study captured the stories of 12 Black alumni who graduated from seven historically White institutions (HWIs), between the years of 2010 and 2016, across four different states. The 12 study participants completed undergraduate and terminal degrees during the years 2009 - 2016 and identified as Black, African, or bi-racial. In today’s sociopolitical climate, restrictive reproductive laws continue to disproportionately target Black women, reinforcing historic patterns of racialized and gendered oppression. This thesis argues that Black women will recommence their usage of traditional herbal healing as both a survival strategy and an act of reclaiming bodily autonomy in defiance of hostile legal, financial, and medical systems. The Gullah Geechee community serves as a blueprint for this transition, offering cultural continuity, intergenerational knowledge, and a model for community-based healthcare and resistance. In 2012, Washington’s landmark McCleary decision confirmed and expanded previous interpretations of the state constitution’s “paramount duty” clause, affirming a vital positive right to education and a judicial willingness to enforce it against the other branches. Notwithstanding a tense six-year period of judicial supervision after the ruling, ultimately the state legislature enacted broad education funding reforms and the appropriations and revenues to implement the additional resources for schools. Given competition among stakeholders for public funding for state programs, in the post-McCleary period, other litigants have asked courts to find similar positive constitutional rights to state services and funding. Commentators have forecasted the demise of high-priced commercial casebooks for two decades. Yet little has changed. This Symposium Essay explores the headwinds facing free and low-cost books in the law school casebook market. It suggests that the biggest problem is “casebook selection inertia.” Given the centrality of the assigned casebook to a typical law school course, professors face strong incentives to make a safe choice from among the leading casebooks—typically those published by the handful of established academic presses who set the highest prices. In 2016, the Russian government purchased political advertisements from U.S. social media companies like Facebook to sway the U.S. presidential election. These purchases violated the foreign national prohibition (“the Prohibition”), which Congress codified as part of the Federal Election Campaign Act Amendments of 1974 (FECA). This Note explains why Congress should expand the Prohibition to capture the recipients of independent expenditures that violate the Prohibition and fine people who run online platforms that receive independent expenditures or contributions made by foreign nationals in violation of the Prohibition.
