Vcp: A Gatekeeper For Intracellular Proteopathic Seeding, Jiang Zhu May 2022

Vcp: A Gatekeeper For Intracellular Proteopathic Seeding, Jiang Zhu

Arts & Sciences Graduate Student Theses and Dissertations

Protein inclusions such as β-amyloid, tau, α-synuclein, and TDP-43 are considered the pathologic hallmarks of many neurodegenerative diseases. These proteins are prone to misfold, aggregate, and template new aggregates. Accumulating evidence suggests that those proteins in their high-molecular-weight forms can serve as a "seed", spread through an interconnected brain network, and induce new inclusions. Therefore, it is essential to understand the mechanism of proteopathic seeding. In this dissertation, we performed a whole genomic CRISPR-Cas9 KO screening to identify gene modifiers of αS seeding. Within the screening, we found several hits of endolysosomal function and trafficking, including VCP. VCP is a …

Three Essays On Local Government: Responsiveness, Transparency, And Discriminatory Ordinances, Bryant James Moy May 2022

Three Essays On Local Government: Responsiveness, Transparency, And Discriminatory Ordinances, Bryant James Moy

Arts & Sciences Graduate Student Theses and Dissertations

This dissertation explores democratic governance at the local level: responsiveness, transparency, and the emergence of discriminatory policies. First, in Chapter 1, I consider whether citizens in overlapping governing units are represented by their elected officials. In other words, given a patchwork system of overlapping local institutions, can residents direct public policy? To answer this question, I implement a framework that takes into account multiple overlapping governing institutions: cities, counties, school districts, and special districts. In doing so, I create a novel measure of local ideological preferences that varies dynamically. I find that both cross-sectional and dynamic responsiveness exists at the …

Cripping Utopia: Revolutionary Psychocorporealities In Cuba And The Diaspora, Charlotte Emily Merrigan May 2022

Cripping Utopia: Revolutionary Psychocorporealities In Cuba And The Diaspora, Charlotte Emily Merrigan

Arts & Sciences Graduate Student Theses and Dissertations

As a utopian, anti-imperialist project, Cuba’s 1959 Revolution consolidated idealized performances of revolutionary citizenship, in the process regulating ideal forms of embodiment and/or cognition and narrowing the parameters of what good health and productive social reform meant. “Cripping Utopia” therefore challenges the putative able-bodiedness and able-mindedness of the archetypal revolutionary citizen. Throughout the project, I translate and adapt concepts from the historically Anglo-centric field of Critical Disability Studies to the context of the Cuban revolutionary process, from 1959 to the early 2000s. Unlike capitalist or neoliberal societies that make disability into a reason for exclusion or a source of profit, …

The Resilience Of Agro-Pastoral Communities In High Altitude Central Tibet: Zooarchaeological And Stable Isotope Investigations At Bangga, 3000–2200 Bp, Zhengwei Zhang May 2022

The Resilience Of Agro-Pastoral Communities In High Altitude Central Tibet: Zooarchaeological And Stable Isotope Investigations At Bangga, 3000–2200 Bp, Zhengwei Zhang

Arts & Sciences Graduate Student Theses and Dissertations

Our understanding of the development of high-altitude adaptations worldwide has been enriched by recent anthropological research. Study of life on the Tibetan Plateau—dubbed “the Roof of the World”—has highlighted specific challenges of seasonal availability of food and extreme weather systems. Recent archaeological studies have revealed that pastoralism has a long history of resilience on the extreme high altitude Tibetan Plateau. However, specific risks to early flocks on the Tibetan Plateau and ways that herders managed their animals to mitigate these have not been well explored. Dated to 3000–2200 Cal BP, the archaeological settlement of Bangga represents one of the earliest …

Low-Temperature Plasticity Of Rock-Forming Minerals, Michael Keith Sly May 2022

Low-Temperature Plasticity Of Rock-Forming Minerals, Michael Keith Sly

Arts & Sciences Graduate Student Theses and Dissertations

The rheology of the Earth’s crust and mantle is in part governed by the physical and mechanical properties of feldspar, quartz, calcite, and olivine. Over the past several decades a significant number of experimental studies have been performed on these minerals. Conventional experiments are generally performed at elevated temperatures because much of the viscoplastic deformation of these minerals is accommodated by high-temperature mechanisms. There is, however, evidence of viscoplastic deformation at lower temperatures (T ≤ 0.3Tm) in the uppermost mantle and throughout the crust. Micromechanical experimental techniques, including nanoindentation and micropillar deformation, can be used to achieve plastic deformation at …

Contribution To Data Science: Time Series, Uncertainty Quantification And Applications, Dhrubajyoti Ghosh May 2022

Contribution To Data Science: Time Series, Uncertainty Quantification And Applications, Dhrubajyoti Ghosh

Arts & Sciences Graduate Student Theses and Dissertations

Time series analysis is an essential tool in modern world statistical analysis, with a myriad of real data problems having temporal components that need to be studied to gain a better understanding of the temporal dependence structure in the data. For example, in the stock market, it is of significant importance to identify the ups and downs of the stock prices, for which time series analysis is crucial. Most of the existing literature on time series deals with linear time series, or with Gaussianity assumption. However, there are multiple instances where the time series shows nonlinear trends, or when the …

Application Of Bismuth Oxychloride As A Colorimetric Uv Sensor Material, Kyle James Troche May 2022

Application Of Bismuth Oxychloride As A Colorimetric Uv Sensor Material, Kyle James Troche

Nanoscience and Microsystems ETDs

Sunlight contains about 9% UV radiation with roughly one-third of it penetrating the atmosphere and reaching the Earth’s surface. UV radiation from the sun is classified into three different types: UVA (315- 499 nm), UVB (280- 314nm), and UVC (100- 279 nm). Prolonged exposure to artificial UV radiation or direct sunlight can still induce many adverse effects such as sunburn, weakening of the immune system, and skin cancer. A wide variety of photoresistor, photodiode and colorimetric UV sensors are currently being researched to help monitor UV radiation. Our research is focused on developing a colorimetric UV sensor that is inherently …

Constructing Roles And Discourse: Presidential Speech And European Union Accession In The Former Yugoslavia, 2000-2021, John Horner

Dissertations

Yugoslavia’s dissolution in the 1990s resulted in seven distinct nation-states vying for functional institutions, ethno-nationalistic coalescence, and external validation. To this end the European Union (EU) offered a pathway to nation-state building and membership via democratization, economic liberalization, and legal and civil improvements. However, to date only Slovenia (2004) and Croatia (2013) are EU member-states. Montenegro, North Macedonia, and Serbia are candidate countries. Bosnia and Herzegovina and Kosovo are potential candidates. What are the incentives for Western Balkan countries to reform and join the EU? Were motivations driven by rational, economic benefits of membership, or was the impetus identity-oriented to …

Assembly Of The Peripheral Arm Subunits Of Escherichia Coli Complex I And Analysis Of Clinical Mutations, Hind Alkhaldi May 2022

Assembly Of The Peripheral Arm Subunits Of Escherichia Coli Complex I And Analysis Of Clinical Mutations, Hind Alkhaldi

Biological Sciences Theses and Dissertations

Respiratory Complex I from E. coli is a proto-type of the mitochondrial enzyme, consisting of a 6-subunit peripheral arm (B-CD-E-F-G-I) and a 7-subunit membrane arm. When subunits E-F-G (N-module), were expressed alone they formed an active complex as determined by co-immunoprecipitation and native gel electrophoresis. When co-expressed with subunits B and CD, only a complex of E-F-G was found. When these five subunits were co-expressed with subunit I and two membrane subunits, A and H, a complex of B-CD-E-F-G-I was membrane-bound, constituting the N- and Q-modules. Assembly of Complex I was also followed by splitting the genes between two plasmids, …

Activation Of P53-Regulated Pro-Survival Signals And Hypoxia-Independent Mitochondrial Targeting Of Tigar By E6 Oncoproteins Of High-Risk Hpvs, Lacin Yapindi

Biological Sciences Theses and Dissertations

The high-risk human papillomaviruses are small, non-enveloped double-stranded DNA viruses that infect the basal epithelial keratinocytes. The majority of HPV infections are self-limiting and asymptomatic; however, in 2-4% of all infected individuals, persistent infections with high-risk HPVs lead to malignancies, including head-and-neck cancers and anogenital carcinomas, particularly cervical cancers. The HPV life cycle is strongly tied to the normal differentiation process of the epithelial tissue. Upon cell entry, the viral genome is maintained as episomal DNA within the nuclei of HPV-infected basal keratinocytes and amplified to a low copy number as the cells divide. As a part of the self-renewing …

Steps Toward The Total Synthesis Of The Chrysosporazine Family, Katie Kaczynski May 2022

Steps Toward The Total Synthesis Of The Chrysosporazine Family, Katie Kaczynski

Electronic Theses and Dissertations

Cancer is one of the leading causes of death worldwide, with 19.3 million new cancer cases and 10 million deaths in 2020 alone. In the United States, cancer was the second leading cause of death after heart disease. One of the reasons that cancer is so frequently terminal is that cancerous cells frequently develop multidrug resistance (MDR) to frontline chemotherapeutical treatments, resulting in cancers cannot be treated and are inevitably fatal. This makes the discovery of ways to combat MDR so important in the treatment of cancer. The chrysosporazines are a family of recently discovered (2019) natural products that have …

The Catholic Paradox Of Villette, Kevin R. Bie May 2022

The Catholic Paradox Of Villette, Kevin R. Bie

Senior Honors Projects, 2020-current

Villette, published in 1853, was Charlotte Brontë’s last novel. Brontë explores both narrative and religious complexities through her narrator, Lucy Snowe. Orphaned Lucy Snowe embarks on a new life in a predominantly Catholic country where her Protestant identity is challenged. Catholicism is presented as a temptation for Lucy. Brontë reveals Lucy’s story through her notable fictional autobiography structure, but Lucy Snowe complicates the relationship between narrator and reader. Lucy explicitly capitalizes on the structure of fictional autobiography, critiquing her narration and fostering a personal relationship with the reader.

This thesis analyzes the Catholic paradox in Charlotte Brontë’s Villette by …

From Patrons To Landlords: The Transformation Of Class Relations In Zanzibar Through Wakf Reform, Isabel Spafford May 2022

From Patrons To Landlords: The Transformation Of Class Relations In Zanzibar Through Wakf Reform, Isabel Spafford

Honors Theses

This study examines the role of wakf reforms in reshaping class relationships in Zanzibar during the British protectorate. Prior to the establishment of the British protectorate in Zanzibar, wakf dedications maintained patron-client relationships between the landowning class and poor clients that were established during the time of slavery but continued after abolition. I argue that wakf dedications were essential to continuing these relationships, and therefore British wakf reforms were necessary to achieve British colonial goals of dissolving patron-client relationships and establishing a capitalist system based on wage labor and ground rent. I analyze the relationship of the British colonial class, …
