Here, we develop and apply a semi-quantitative method for the high-confidence identification of pseudouridylated sites on mammalian mRNAs via direct long-read nanopore sequencing. A comparative analysis of a modification-free transcriptome reveals that the depth of coverage and specific k-mer sequences are critical parameters for accurate basecalling. By adjusting these parameters for high-confidence U-to-C basecalling errors, we identify many known sites of pseudouridylation and uncover previously unreported uridine-modified sites, many of which fall in k-mers that are known targets of pseudouridine synthases. Cholesterol synthesis is both energy- and oxygen-intensive, yet relatively little is known of the regulatory effects of hypoxia on pathway enzymes. We previously showed that the rate-limiting and first oxygen-dependent enzyme of the committed cholesterol synthesis pathway, squalene monooxygenase (SM), can undergo partial proteasomal degradation that renders it constitutively active. Here, we show hypoxia is a physiological trigger for this truncation, which occurs through a two-part mechanism: (1) increased targeting of SM to the proteasome via stabilization of the E3 ubiquitin ligase MARCHF6 and (2) accumulation of the SM substrate, squalene, which impedes the complete degradation of SM and liberates …

BACKGROUND: We have previously shown coxsackievirus B (CVB) to be a potent inducer of congenital heart disease (CHD) in mice. The clinical relevance of these findings in humans and the roles of other viruses in the pathogenesis of CHD remain unknown.

METHODS: We obtained plasma samples, collected at all trimesters, from 89 subjects (104 pregnancies), 73 healthy controls (88 pregnancies), and 16 with CHD-affected birth (16 pregnancies), from the Perinatal Family Tissue Bank (PFTB). The second one is to examine the translation of this term into English. This essay reviews Matthew F. Delmont’s new book, Half American: The Epic Story of African Americans Fighting World War II at Home and Abroad (2022). The book enriches the ongoing scholarship related to critical race theory and the effects of executive action on the lived experience of Black Americans. Delmont presents a well-woven narrative of the experience of Black American soldiers during World War II. Pieced together from letters, court documents, and articles published during the war, this book sheds light on accounts previously buried beneath a shield of trauma, frustration, and disbelief.

Background: A large proportion of women in the northern region of Nigeria do not utilise antenatal care (ANC) services. As a result, the region has the worst maternal and child health indicators. This study aims to identify the socio-demographic determinants of the non-utilisation of ANC services by pregnant women to provide evidence for policymakers to base decisions towards addressing the problem.

Methods: Data from the 2018 Nigeria Demographic and Health Survey was used with a sample of 14,421 women with a pregnancy history. Descriptive, bivariate, and hierarchical regression analyses were applied to the data using STATA software version 15.

Background: Reporting of adverse events following immunization (AEFI) is not the sole responsibility of health workers providing routine immunization services, but also of health workers providing clinical treatments and working in other health units. This study aimed to assess health workers’ level of knowledge as well as reporting attitudes and practices on AEFI in Ondo State, Nigeria.

Background: Lifestyle intervention is effective in preventing type 2 diabetes mellitus (T2DM). The efficacy of intervention components across different sociocultural settings is relevant for real world implementation. Thus, this study aimed to evaluate the effectiveness of a culturally tailored diabetes prevention program (CTDPP) for at-risk Muslim people in Thailand.

Method: The CTDPP was developed and culturally adapted based on formative research. Many take Kantian ethics to founder when it comes to our duties to animals. In this paper, I advocate a novel approach to this problem. The paper is divided into three sections. In the first, I canvass various passages from Kant in order to set up the problem. In the second, I introduce a novel approach to this problem. In the third, I defend my approach from various objections. By way of preview: I advocate rejecting the premise that nonhuman animals are nonrational.

Recent experiments in bonded PMMA layers have shown dramatic changes in dynamic crack growth characteristics depending on the interface location and toughness. In this paper we present a peridynamic (PD) analysis of this phenomenon and determine three elements that are essential in a model reproducing the observed fracture behavior: (1) softening near the crack tip to account for changes in PMMA due to heat-generation induced by the high strain rates reached around the crack tip in dynamic fracture; (2) independent extension (mode I) and shear (mode II) modes of fracture; (3) a two-parameter fracture model, which matches both strength and …

Predicting the reliability of board-level solder joints is a challenging process for the designer because the fatigue life of solder is influenced by a large variety of design parameters and many nonlinear, coupled phenomena. Machine learning has shown promise as a way of predicting the fatigue life of board-level solder joints. Recharge Assessment Of The Memphis Aquifer Beneath An Urban Watershed Following Stream Restoration, Jarod Przybylski

Stream restoration was conducted by the West Tennessee River Basin Authority to restore flow and correct erosional problems in the Sandy Creek watershed, an urban site in western Tennessee. In this study, post-restoration water balance data of one year are compared pre-restoration water balance data of three years to understand the short-term effects of stream restoration on the recharge rate at the Sandy Creek watershed. Pre-restoration recharge rates for the three water budget years (Oct.-Sept.) were estimated as 0.529 m/m2 for 2017-2018, 0.682 m/m2 for 2018-2019, and 0.406 m/m2 for 2020-2021. Psoriasis is a chronic autoimmune inflammatory skin disorder characterized by epidermal hyperplasia and aberrant immune response. In addition to aberrant cytokine production, psoriasis is associated with activation of the Akt/mTOR pathway. mTOR/S6K1 regulates T-lymphocyte activation and migration, keratinocytes proliferation and is upregulated in psoriatic lesions. Several drugs that target Th1/Th17 cytokines or their receptors have been approved for treating psoriasis in humans with variable results necessitating improved therapies. Fisetin, a natural dietary polyphenol with anti-oxidant and anti-proliferative properties, covalently binds mTOR/S6K1. Through a structured decision-making framework, this study aims to better understand the key factors influencing transportation adaptation planning in practice. Qualitative, semi-structured, in-depth interviews with various stakeholders were conducted to identify the main concerns, challenges, objectives, tradeoffs, and evaluation variables in transportation adaptation planning. Stakeholders were identified through preliminary interviews with transportation planning experts from the metropolitan planning organization using typical case and snowball sampling methods. Key aspects related to the major concerns, objectives, priorities, adaptation plan evaluations, implementation challenges, and potential conflicts and tradeoffs are identified. Tired of the power that mega platforms wield over the web, a growing chorus of internet users has hailed the arrival of blockchain technology, believing it can be used to build a new internet. Called “Web 3.0” by some, the new internet would allow users to exchange goods and services—digital currencies, cloud computing power, data storage—without needing a central intermediary to validate transactions. Instead, users would transact through decentralized platforms that use consensus-based mechanisms to verify users’ exchanges.
