Examining The Role Of Microglia In Diabetic Retinopathy: Phenotypes, Mechanisms, And Therapies, Charles W. Pfeifer

Retinal microglia activation and function is a pivotal component of ocular disease pathology, but their involvement in diabetic retinopathy, a complex neurovascular complication of diabetes, remains unclear. The purpose of this thesis work is to thoroughly examine features of microglia activation in DR, identify mechanistic underpinnings of their contributions to neuroinflammatory features of disease pathogenesis, and assess immunotherapeutic targeting of microglia as a potential approach to treating early-stage DR. Here, we show that chronic hyperglycemia elicits morphological, ultrastructural, and transcriptional changes in retinal microglia during early-stage DR while other elements of the retinal neurovascular unit remain unperturbed. The fungal pathogen Candida albicans is linked to chronic brain diseases such as Alzheimer's disease (AD), but the molecular basis of brain anti-Candida immunity remains unknown. We show that C. albicans enters the mouse brain from the blood and induces two neuroimmune sensing mechanisms involving secreted aspartic proteinases (Saps) and candidalysin. Saps disrupt tight junction proteins of the blood-brain barrier (BBB) to permit fungal brain invasion. Saps also hydrolyze amyloid precursor protein (APP) into amyloid β (Aβ)-like peptides that bind to Toll-like receptor 4 (TLR4) and promote fungal killing in vitro while candidalysin engages the integrin CD11b (Mac-1) on microglia. …

Post-COVID conditions (PCCs) are common and have significant morbidity. Risk factors for PCC include advancing age, female sex, obesity, and diabetes mellitus. Little is known about treatment, inflammation, and PCC. Among 882 individuals with confirmed SARS-CoV-2 infection participating in a randomized trial of COVID-19 convalescent plasma (CCP) vs control plasma with available biospecimens and symptom data, the association between early CCP treatment, cytokine levels, and PCC was evaluated. Cytokine and chemokine levels were assessed at baseline, day 14, and day 90 using a multiplexed sandwich immunoassay (Meso Scale Discovery). Objectives: Central sensitivity syndrome disorders such as fibromyalgia, provoke continued debate, highlighting diagnostic and therapeutic uncertainty. The Hyland model provides a way of understanding and treating the medically unexplained symptoms of central sensitivity syndromes using complexity theory and principles of adaption in network systems. The body reprogramming is a multi-modal intervention based on the Hyland model designed for patients living with medically unexplained symptoms. This preliminary, naturalistic and single-arm service evaluation set out to evaluate outcome after attending a body reprogramming course in patients living with fibromyalgia or central sensitivity syndrome. We have previously identified TopBP1 (topoisomerase IIβ-binding protein 1) as a promising target for cancer therapy, given its role in the convergence of Rb, PI(3)K/Akt, and p53 pathways. Based on this, we conducted a large-scale molecular docking screening to identify a small-molecule inhibitor that specifically targets the BRCT7/8 domains of TopBP1, which we have named 5D4. Our studies show that 5D4 inhibits TopBP1 interactions with E2F1, mutant p53, and Cancerous Inhibitor of Protein Phosphatase 2A. This leads to the activation of E2F1-mediated apoptosis and the inhibition of mutant p53 gain of function. In addition, 5D4 disrupts the interaction of TopBP1 …

The consequences of returning infectious pathogen test results identified incidentally in research studies have not been well-studied. Concerns include identification of an important health issue for individuals, accuracy of research test results, public health impact, potential emotional distress for participants, and need for IRB permissions. Blood RNA-sequencing analysis for non-human RNA in 3984 participants from the COPDGene study identified 228 participants with evidence suggestive for hepatitis C virus (HCV) infection. We hypothesized that incidentally discovered HCV results could be effectively returned to COPDGene participants with attention to the identified concerns. The efficacy of therapeutic T-cells is limited by a lack of positive signals and excess inhibitory signaling in tumor microenvironments. We previously showed that a constitutively active IL7 receptor (C7R) enhanced the persistence, expansion, and anti-tumor activity of T-cells expressing chimeric antigen receptors (CARs), and C7R-modified GD2.CAR T-cells are currently undergoing clinical trials. To determine if the C7R could also enhance the activity of T-cells recognizing tumors via their native T-cell receptors (TCRs), we evaluated its effects in Epstein–Barr virus (EBV)-specific T-cells (EBVSTs) that have produced clinical benefits in patients with EBV-associated malignancies. Metastatic colorectal cancer (mCRC) with mutated BRAF exhibits distinct biological and molecular features that set it apart from other subtypes of CRC. Current standard treatment for these tumors involves a combination of chemotherapy (CT) and VEGF inhibitors. Recently, targeted therapy against BRAF and immunotherapy (IT) for cases with microsatellite instability (MSI) have been integrated into clinical practice. While targeted therapy has shown promising results, resistance to treatment eventually develops in a significant portion of responsive patients. This article aims to review the available literature on mechanisms of resistance to BRAF inhibitors (BRAFis) and potential therapeutic strategies to overcome them.

Glioblastomas (GBM) are highly invasive, malignant primary brain tumors. The overall prognosis is poor, and management of GBMs remains a formidable challenge, necessitating novel therapeutic strategies such as dendritic cell vaccinations (DCVs). While many early clinical trials demonstrate an induction of an antitumoral immune response, outcomes are mixed and dependent on numerous factors that vary between trials. This essay examines the structural choices Virginia Woolf makes in Orlando to mirror her prose with the relative models of time the protagonist experiences throughout the story. Comparing the leading theories in particle physics at the time Woolf wrote to her work shows that she was aware of these developments and incorporated this knowledge into her writing. Specifically, the relativity of time is used to mirror the relative nature of Orlando’s identity, as they change from man to woman throughout the course of the novel.

In the Portland Metro Area, the signal crayfish (Pacifastacus leniusculus) is a keystone species. Signal crayfish consume inaccessible plant matter and detritus, feed various game fish, engineer riverbeds, and are even harvested commercially for human consumption. Recognizing signal crayfishes’ multifaceted ecological and social significance, the purpose of this study was to assess the habitat factors that impact signal crayfish health and behavior in the Tryon Creek Watershed. This article studies Espejo fiel de vidas (Life’s True Mirror, 1720) of Daniel Israel López Laguna as an example of Baroque Spanish poetry that reworks the book of Psalms for an audience that like the author were conversos, or New Christians, that is, Spanish and Portuguese Catholics of Jewish origin and their descendants. Likewise, it studies the geographic and spiritual journey of one such Iberian New Christian through the Hispano-Portuguese Jewish diaspora who became a practicing Jew upon settling in Jamaica. While the Western novel is typically associated with cowboys and overarching masculine accomplishments of “settling,” the representation of women’s work beyond narratives of being “tamed” and the settling-down of motherhood has been overlooked by writers and ecocritics alike. College students are at high risk for experiencing sexual violence. Living without parents, lack of understanding of consent, new relationships, high stress, and experimenting with drugs and alcohol are some factors that increase the risk of sexual violence among college students. Pacific University has existing prevention programs aimed to decrease the incidence of sexual violence, but existing workshops only target first year students and athletes. How does the use of antibiotics in animal husbandry impact antibiotic resistance genes (ARGs) and integron prevalence? This article examines the prevalence of nine ARGs (OXA-2, tetB, tetG, blaCMY-2, ereB, NDM, ermB, sul1, and sul2) and two class one integron sequences (intI1 and CS) at an organic goat dairy farm. A Gendered Historical Discourse Of The Naxalbari Movement, Pritha Sarkar

This paper analyzes the Indian English novel The Lowland (2012) by Jhumpa Lahiri and examines its representation of the Naxalbari movement’s (1965-1975) gendered history to locate women, their roles, their marginalized position, and the growth of their individual independent identities. The Naxalbari movement is the first major peasant protest within 20 years of Indian independence. Though the first actions of the movement were in a village of North Bengal, this paper mainly concentrates on the movement’s activities after it was urbanized and joined by the middle class. It, therefore, tries to locate the position of middle-class women within the movement. …

This auto-ethnographic essay draws upon Foucault’s Archaeology of Knowledge to discuss the condition of Indian women in the Humanities in academia today. While acknowledging the encouragingly gender-inclusive projections in India’s National Education Policy vision statement from 2020, I argue for more probing engagement with the concrete reality of being a woman teacher and researcher in the increasingly competitive and corporatized milieu of higher education. My methodology has been a close reading of the NEP’s vision statement to analyze recurrences of terms and concepts as pointers to its discursive field. With the advent of social media, the media environment has become more participatory for its users, making it possible for older adults to produce content for social media and be agential in online spaces. This article observes a group of older women known as Activist Grannies (Aktivistimummot in Finnish) and 60+ Finnish women bloggers who identify as “grannies” to discover what kind of agency social media potentially enables for older women. Triumphant Or Trapped Pakistani Women? A Feminist Critique Of Mueenuddin’S “Nawabdin Electrician” And Haq’S Song “Chamkeeli”, Amna Khan

In patriarchal societies, women are traditionally subjugated and suppressed in one way or another. Men are privileged and kept at the center. They speak, express, and dream while benefiting from the autonomy provided to them by the phallogocentric system. By contrast, women are marginalized. Patriarchal writers define women as weak, fragile, helpless, docile, submissive, and emotional. However, this paper reveals that in Daniyal Mueenuddin’s “Nawabdin Electrician” and Abrar-ul-Haq’s song “Chamkeeli,” regardless of a change in times and “gender performativity,” Pakistani male writers continue to stigmatize women. Historically speaking, women have been associated with madness, be it Medea from Ancient Greece, the medieval trials of the witches of Salem, or so called “hysterical” women in the Victorian era. Even in 21st-century literature, arts, and media, the madness of women is widely discussed and often romanticized. Some women authors employed the madwoman trope to show the effects of patriarchal oppression on women. Other studies have associated women’s madness in literature with subversion. This work presents a general framework for developing a multi-parameter 1-D chaotic system for uniform and robust chaotic operation across the parameter space. This is important for diverse practical applications where parameter disturbance may cause degradation or even complete disappearance of chaotic properties. The wide uninterrupted chaotic range and improved chaotic properties are demonstrated with the aid of stability analysis, bifurcation diagram, Lyapunov exponent (LE), Kolmogorov entropy, Shannon entropy, and correlation coefficient. We also demonstrate the proposed system’s amenability to cascading for further performance improvement. In the past twenty years, multiple studies have shown the relationship between childhood adversity and later negative health consequences. Yet the extent to which the public is aware of this relationship is unclear. We surveyed Black and White college students about their knowledge of the long-term effects of Adverse Childhood Experiences (ACEs). Students read vignettes comparing children exposed and unexposed to ACEs, and predicted their mental, physical, and social health as adults. Participants were aware of the effect of ACEs on later mental and social health, but not as aware of the risks on physical health. Black and White students …
