Objective: The structure of CLPS1 (polysaccharide fraction 1 of Codonopsis lanceolata) was determined to provide a basis for the study of polysaccharide activity of C. lanceolata and the development of natural plant polysaccharides. Methods: After extraction, separation, and purification, the separation component CLPS1 was determined by gas chromatography, infrared spectroscopy, nuclear magnetic resonance, periodate oxidation, Smith degradation, and methylation analysis to determine the structure of CLPS1. Results: The specific rotation of CLPS1 is +44°, the content of the total polysaccharide is 97.6%, the uronic acid of CLPS1 is 13.18%, and the molecular weight of CLPS1 is 91.7. The monosaccharide …

Objective: In order to explore the extraction method and antioxidant activity of crude polysaccharide from edible fungus resources of reed. Methods: The characteristic Agrocybe praecox of reed edible fungus of Dongting Lake District was used as raw material, the extraction rate of polysaccharide was used as the index, and the extraction was carried out by ultrasonic-assisted hot water alcohol precipitation method. Objective: The study aimed to optimize the preparation process and investigate the quality of coix bran dietary fiber. Methods: The coix bran was used as raw material; the effects of solid-liquid ratio, the amount of amylase and alcalase on the extraction rate were analyzed by usingorthogonal experiments. The chemical composition and physicochemical properties of the dietary fiber obtained under the optimal preparation conditions was analyzed. Results: The optimal extraction parameters of dietary fiber were as follows: solid-liquid ratio was 1∶10 (g/mL), the amount of amylase and alcalase was 100 U/g, respectively, and the extraction rate was up to 84.39%. In view of the limitations of the traditional processing technology of Pidan, this review summarizes the innovative processing technologies of Pidan, such as gradient temperature control, ultrasonic assisted, vacuum decompression, pulsating pressure, etc., and describes the changes of water, pH value, protein, free amino acids, texture, mineral elements and micro-morphology during the picking process of Pidan. Moreover, the development directions of Pidan products are prospected.

The composition, structure and functional properties of Agaricus blazei polysaccharide were summarized. The extraction techniques of polysaccharides and the features of different extraction methods were enumerated and compared. The problems in the research of A. blazei was discussed, such as unsystematic basic research, immature technological conditions, imperfect equipment and facilities, and low degree of specificity in seeds. In the end,the research direction of A. blazei polysaccharide was prospected.

Online professional development became a necessity during the pandemic for educators, including Instructional Technology Coaches, to continue to gain knowledge to improve instruction. In this study, Instructional Technology Coaches participated in an online professional development series of synchronous and asynchronous sessions called Pathways. Two theoretical frameworks were used to support the research: Adult Learning Theory with an emphasis on the Theory of Computer-based Instruction for Adults, and TPACK. The seven characteristics of effective professional development created by Darling-Hammond et al. (2017) provided a foundation on which design elements to include in Pathways. 1. Pulsed subsidy events create ephemeral fluxes of hyper-abundant resources that can shape annual patterns of consumption and growth for recipient consumers. However, environmental conditions strongly affect local resource availability for much of the year, and can heavily impact consumer foraging and growth patterns prior to pulsed subsidy events. Thus, a consumer's capacity to exploit pulse subsidy resources may be influenced by antecedent environmental conditions, but this has rarely been shown in nature and is unknown in aquatic ecosystems. 2. Indoor air quality (IAQ) is adversely affected by both indoor and outdoor environments, and rental homes occupied by low-income families suffer the most. Poor indoor air can be detrimental to the health of occupants, especially for susceptible populations like those with respiratory diseases such as asthma. Although IAQ monitoring is necessary, it is a challenging, costly, and time-intensive task. Moreover, the protocol for assessing IAQ in residential settings varies, and additional research is needed to establish a standard operating procedure that incorporates new monitoring technologies, such as low-cost air sensors. The coqui frog (Eleutherodactylus coqui) was introduced to the island of Hawai'i in the 1980s, and has spread across much of the island. There is concern they will invade higher elevation areas where negative impacts on native species are expected. It is not known if coqui change behavior and baseline physiology in ways that allow them to invade higher elevations. We investigated where coqui are found across the island and whether that includes recent invasion into higher elevations. Ribosome biogenesis is an energetically costly process, and tight regulation is required for stoichiometric balance between components. This requires coordination of RNA polymerases I, II, and III. Lack of nutrients or the presence of stress leads to downregulation of ribosome biogenesis, a process for which mechanistic target of rapamycin complex I (mTORC1) is key. mTORC1 activity is communicated by means of specific transcription factors, and in yeast, which is a primary model system in which transcriptional coordination has been delineated, transcription factors involved in regulation of ribosomal protein genes include Fhl1p and its cofactors, Ifh1p and Crf1p. Resident female wood ducks (Aix sponsa) in the southeastern United States are managed through nest box programs, with the goal of enhancing local hunting opportunities. Historically, hooded mergansers (Lophodytes cucullatus) were the only other cavity-nesting waterfowl species in the southeastern United States known to use these nest boxes. However, the recent northward range expansion of black-bellied whistling-ducks (Dendrocygna autumnalis) has extended into the range of wood ducks and mergansers. All three species are brood parasites, potentially using one another as hosts for their parasitic eggs and impacting their reproductive output. My research evaluated the …

UNLABELLED: Lipid droplets (LD) are dynamic organelles that serve as hubs of cellular metabolic processes. Emerging evidence shows that LDs also play a critical role in maintaining redox homeostasis and can mitigate lipid oxidative stress. In multiple cancers, including prostate cancer, LD accumulation is associated with cancer aggressiveness, therapy resistance, and poor clinical outcome. Prostate cancer arises as an androgen receptor (AR)-driven disease. Among its myriad roles, AR mediates the biosynthesis of LDs, induces autophagy, and modulates cellular oxidative stress in a tightly regulated cycle that promotes cell proliferation. In previous work, my research group and I reported on the synthesis of Anaephenes A and B and confirmed their biological activities. We also found that these compounds were active against drug resistant bacteria such as methicillin-resistant Staphylococcus aureus (MRSA). This finding lead to a series of structure activity relationship studies (SAR) to develop analogs with improved biological activities. Near the start of my graduate studies, my team and I reported an SAR where we generated eighteen analogs. At the heart of the DNA/ALVAC/gp120/alum vaccine’s efficacy in the absence of neutralizing antibodies is a delicate balance of pro- and anti-inflammatory immune responses that effectively decreases the risk of SIVmac251 acquisition in macaques. Vaccine efficacy is linked to antibodies recognizing the V2 helical conformation, DC-10 tolerogenic dendritic cells eliciting the clearance of apoptotic cells via efferocytosis, and CCR5 downregulation on vaccine-induced gut homing CD4+ cells. RAS activation is also linked to vaccine efficacy, which prompted the testing of IGF-1, a potent inducer of RAS activation with vaccination. Rheumatic illnesses are chronic diseases that cause the immune system to attack the bodies joints, muscles, bones, and organs (Mayo Clinic Health System, 2022). In many cases, rheumatic illnesses can cause severe disability and end organ damage leading to mortality if left untreated. Many rheumatological patients are on biologic disease modifying anti-rheumatic drugs (bDMARDs) and this has significantly improved the outcome for patients with rheumatic diseases (Humphreys et al., 2016). Objective: This study aimed to explore the composition and the functional bioactivity of tea polyphenols from cocoa tea. Methods: Chemical components of crude green tea polyphenol extracts from cocoa tea were analyzed and identified using high resolution mass spectrometry. Additionally, their potential functional effects were predicted by network pharmacological analysis. Results: The in vitro hyperglycemic potential of crude green tea polyphenol from cocoa tea was preliminarily confirmed based on their strong inhibition on α-glucosidase and α-amylase. Suicidal ideation disproportionally affects the LGBTQ+ community. Community connectedness and mindfulness can be protective against suicidal ideation, whereas emotion regulation difficulty is associated with increased suicidal ideation. Community connectedness, emotion regulation difficulty and mindfulness have demonstrated relationships to each other in the LGBTQ+ community and while they are each independently associated with suicidal ideation, their combined impact on suicidal ideation has not yet been examined. Introduction—Approximately 1% of the world’s population is impacted by epilepsy, a chronic neurological disorder characterized by seizures. One-third of epileptic patients are resistant to AEDs, or have medically refractory epilepsy (MRE). One non-invasive treatment that exists for MRE includes the ketogenic diet, a high-fat, low-carbohydrate diet. Despite the KD’s success in seizure attenuation, it has a few risks and its mechanisms remain poorly understood. The KD has been shown to improve metabolism and mitochondrial function in epileptic phenotypes. Potassium channels have implications in epileptic conditions as they have dual roles as metabolic sensors and control neuronal excitation.

Genomic abnormalities are strongly associated with cancer and infertility. In this study, we develop a simple and efficient method - multiple genetic abnormality sequencing (MGA-Seq) - to simultaneously detect structural variation, copy number variation, single-nucleotide polymorphism, homogeneously staining regions, and extrachromosomal DNA (ecDNA) from a single tube. MGA-Seq directly sequences proximity-ligated genomic fragments, yielding a dataset with concurrent genome three-dimensional and whole-genome sequencing information, enabling approximate localization of genomic structural variations and facilitating breakpoint identification. Additionally, by utilizing MGA-Seq, we map focal amplification and oncogene coamplification, thus facilitating the exploration of ecDNA's transcriptional regulatory function.

Glioblastoma multiforme (GBM) is the most deadly brain tumor, effective treatment options for which still remain elusive. The current treatment procedure of maximal resection followed by chemotherapy has proved to be grossly insufficient to prevent disease progression and death. Despite best efforts, the maximum survival post-diagnosis is a mere 1.5 years. Therefore, there is a huge unmet clinical need to find effective therapeutic procedures to prevent the pathogenesis and relapse of GBM. Small-molecule inhibitors of signaling pathways are an attractive option to prevent various types of tumors. However, no effective small-molecule inhibitors have been successful against GBM in clinical trials. …

Selenium is an essential trace element in living organisms, and is present in selenoenzymes with antioxidant activity, like glutathione peroxidase (GPx) and thioredoxin reductase (TrxR). The search for small selenium-containing molecules that mimic selenoenzymes is a strong field of research in organic and medicinal chemistry. In this review, we review the synthesis and bioassays of new and known organoselenium compounds with antioxidant activity, covering the last five years. A detailed description of the synthetic procedures and the performed in vitro and in vivo bioassays is presented, highlighting the most active compounds in each series.

Coffee drinks are sugar-sweetened beverages made from coffee powder, sugar, and water through heating. High consumption of coffee drinks is associated with sugar content, which can increase the risk of obesity. The aim of this study was to determine the differences in coffee drink consumption levels based on individual characteristics and environmental factors among non-health undergraduate students at the University of Indonesia in 2023. Ultra-processed food/UPF is a product that undergoes a series of industrial techniques and processes, also has low nutritional value. Overconsumption of UPF can increase the risk of overweight and obesity. The purpose of this study is to determine the differences in the proportion of UPF consumption based on individual characteristics, environmental factors, and lifestyle factors among non-health undergraduate students in Universitas Indonesia in 2023.
