Center for Brain, Biology, and Behavior: Faculty Publications

Background

Conduct disorder (CD) involves a group of behavioral and emotional problems that usually begins during childhood or adolescence. Structural brain alterations have been observed in CD, including the amygdala, insula, ventrolateral and medial prefrontal cortex, anterior cingulate cortex, and fusiform gyrus. The current study developed a multivariate generalized linear model (GLM) to differentiate adolescents with CD from typically developing (TD) adolescents in terms of grey matter volume (GMV).

Methods

The whole‐brain structural MRI data were collected from 96 adolescents with CD (mean age = 16.188 ± 1.259 years; mean IQ = 104.292 ± 8.107 ; 63 males) and 90 …

Climate change has adversely affected maize productivity. Thereby, a holistic understanding of metabolic crosstalk among its organs is important to address this issue. Thus, we reconstructed the first multi-organ maize metabolicmodel, iZMA6517, and contextualized itwith heat and cold stress transcriptomics data using expression distributed reaction flux measurement (EXTREAM) algorithm. Furthermore, implementing metabolic bottleneck analysis on contextualized models revealed differences between these stresses. While both stresses had reducing power bottlenecks, heat stress had additional energy generation bottlenecks. We also performed thermodynamic driving force analysis, revealing thermodynamics-reducing power-energy generation axis dictating the nature of temperature stress responses. Thus, a temperaturetolerant maize ideotype can …

This paper proposes an algorithm for the optimal installation of inverter-based resources (IBR) composed of wind energy conversion systems, photovoltaic systems, and battery energy storage systems in distribution systems using genetic algorithm (GA) and the cuckoo search (CS) as optimization techniques. The OpenDSS software is used to calculate the power flow in the distribution system with different penetration levels of IBRs. It is used a standard load shape of the IEEE 123 bus system programmed in OpenDSS and irradiance, temperature, and wind speed curves from Brazil. The proposed algorithm, using a genetic algorithm and cuckoo search, was able to define …

Staphylococcus pseudintermedius is historically understood as a prevalent commensal and pathogen of dogs, though modern clinical diagnostics reveal an expanded host-range that includes humans. It remains unclear whether differentiation across S. pseudintermedius populations is driven primarily by niche-type or host-species. We sequenced 501 diagnostic and commensal isolates from a hospital, veterinary diagnostic laboratory, and within households in the American Midwest, and performed a comparative genomics investigation contrasting human diagnostic, animal diagnostic, human colonizing, pet colonizing, and household-surface S. pseudintermedius isolates. Though indistinguishable by core and accessory gene architecture, diagnostic isolates harbor more encoded and phenotypic resistance, whereas colonizing and surface …

Introducing biomolecules into cells is an important process in the field of gene therapy and in developing cures for debilitating illnesses such as HIV and cancer. However, the currently used methods for cellular transfection are slow, inefficient and expensive, causing them to be restrictive. A solution to this problem would be the carbon nanotube (CNT) array devices developed by our research group, which have been shown to be highly efficient at transporting biomolecules into cells. In the current manufacturing process, a sacrificial template with vertically aligned nanopores is used to deposit carbon in, the template then is etched away to …

Prostate cancer (PCa) brings huge public health burden in men. A growing number of conventional observational studies report associations of multiple circulating proteins with PCa risk. However, the existing findings may be subject to incoherent biases of conventional epidemiologic studies. To better characterize their associations, herein, we evaluated associations of genetically predicted concentrations of plasma proteins with PCa risk. We developed comprehensive genetic prediction models for protein levels in plasma. After testing 1308 proteins in 79 194 cases and 61 112 controls of European ancestry included in the consortia of BPC3, CAPS, CRUK, PEGASUS, and PRACTICAL, 24 proteins showed significant …

Oral cancer pain remains a significant public health concern. Despite the development of improved treatments, pain continues to be a debilitating clinical feature of the disease, leading to reduced oral mobility and diminished quality of life. Opioids are the gold standard treatment for moderate-to-severe oral cancer pain; however, chronic opioid administration leads to hyperalgesia, tolerance, and dependence. The aim of this review is to present accumulating evidence that epidermal growth factor receptor (EGFR) signaling, often dysregulated in cancer, is also an emerging signaling pathway critically involved in pain and opioid tolerance. We presented preclinical and clinical data to demonstrate how …

Identifying circulating proteins associated with cognitive function may point to biomarkers and molecular process of cognitive impairment. Few studies have investigated the association between circulating proteins and cognitive function. We identify 246 protein measures quantified by the SomaScan assay as associated with cognitive function (p < 4.9E-5, n up to 7289). Of these, 45 were replicated using SomaScan data, and three were replicated using Olink data at Bonferroni-corrected significance. Enrichment analysis linked the proteins associated with general cognitive function to cell signaling pathways and synapse architecture. Mendelian randomization analysis implicated higher levels of NECTIN2, a protein mediating viral entry into neuronal cells, with higher Alzheimer's disease (AD) risk (p = 2.5E-26). Levels of 14 other protein measures were implicated as consequences of AD susceptibility (p < 2.0E-4). Proteins implicated as causes or consequences of AD susceptibility may provide new insight into the potential relationship between immunity and AD susceptibility as well as potential therapeutic targets.

Wills Eye Hospital Papers

Several nomenclature and grading systems have been proposed for conjunctival melanocytic intraepithelial lesions (C-MIL). The fourth "WHO Classification of Eye Tumors" (WHO-EYE04) proposed a C-MIL classification, capturing the progression of noninvasive neoplastic melanocytes from low- to high-grade lesions, onto melanoma in situ (MIS), and then to invasive melanoma. This proposal was revised to the WHO-EYE05 C-MIL system, which simplified the high-grade C-MIL, whereby MIS was subsumed into high-grade C-MIL. Our aim was to validate the WHO-EYE05 C-MIL system using digitized images of C-MIL, stained with hematoxylin and eosin and immunohistochemistry. However, C-MIL cases were retrieved from 3 supraregional ocular pathology …

Cardeza Foundation for Hematologic Research

Two of the most prevalent neurodegenerative disorders (NDDs), Alzheimer's disease (AD) and Parkinson's disease (PD), present significant challenges to healthcare systems worldwide. While the etiologies of AD and PD differ, both diseases share commonalities in synaptic dysfunction, thereby focusing attention on the role of neurotransmitters. The possible functions that platelets may play in neurodegenerative illnesses including PD and AD are becoming more acknowledged. In AD, platelets have been investigated for their ability to generate amyloid-ß (Aß) peptides, contributing to the formation of neurotoxic plaques. Moreover, platelets are considered biomarkers for early AD diagnosis. In PD, platelets have been studied for …

The What And How Of Essential Thematic Analysis, Elih Sutisna Yanto

The Qualitative Report

Gareth Terry and Nikki Hayfield's book, Essentials of Thematic Analysis, introduces readers to reflexive thematic analysis, a method for analyzing interview and focus group transcripts, qualitative survey responses, and other qualitative data. This method is based on the understanding that we all exist in a context from which we can see and speak. In this way, researchers produce knowledge that represents situated truths and allow them to understand others’ perspectives on a given topic. The book shows how to construct a “positioned reality of the situation” from qualitative data. According to the authors, this method is not a methodology …

Consanguineous marriage, defined as marriage between close relatives, is a prevalent practice in many traditional societies. It is commonly perceived to secure property rights, maintain social cohesion, and fortify kinship structures. However, consanguineous marriages are also associated with an increased risk of inbreeding and consequent chromosomal disorders. Pakistan has one of the highest rates of consanguineous marriage worldwide, with an estimated 60% of all marriages occurring between close relatives. This study focuses on the persistence of consanguinity in Kabirwala, Pakistan, a town in the Punjab province. It investigates the trend of consanguinity and relative affinity according to generations in Kabirwala. …

Integrated Periphyton Biofilters In Marine Recirculating Aquaculture Systems, Adam N. Bell Nov 2023

Integrated Periphyton Biofilters In Marine Recirculating Aquaculture Systems, Adam N. Bell

USF Tampa Graduate Theses and Dissertations

Marine aquaculture is a growing industry that has the potential to reduce eutrophication, overfishing, and loss of biodiversity. Recirculating aquaculture systems (RAS) are a type of aquaculture system that decreases discharge, raises biological control, and conserves water; however, RAS are challenged due to high energy cost, low biodiversity, water quality control, and waste nutrients. Periphyton biofilters are a new technology that allows for feed production, water quality control, microbial diversity, and resource recovery in marine aquaculture systems. The overarching question that this dissertation investigates is, how can periphyton biofilters be integrated into RAS to address the challenges of modern sustainable …

Brain Metastasis-Associated Fibroblasts Secrete Fucosylated Pvr/Cd155 That Induces Breast Cancer Invasion, Emma Adhikari

USF Tampa Graduate Theses and Dissertations

Breast cancer (BC) continues to be the most diagnosed cancer and the second leading cause of cancer deaths in women worldwide. High mortality, in most cancers, is due to metastasis; BC is the most common source of one of the deadliest forms of metastasis–brain metastasis (BM) in female. Patients with triple negative breast cancer (TNBC) exhibit the highest incidence of BM and poorest survival outcomes upon diagnosis with BM. The prognosis of breast cancer brain metastasis (BCBM) remains poor because the blood–brain barrier (BBB) prevents the uptake of drug substances, including chemotherapeutic agents, targeted agents, and toxins, from the brain. …

Procedures For Assessment Of The Marine Carbon System And Evaluation Of Access And Ocean Acidification Literacy In The Deaf Community, Loraine Martell-Bonet

USF Tampa Graduate Theses and Dissertations

One major goal of this dissertation addresses the need for improved laboratory procedures to measure chemical changes in seawater caused by the uptake of atmospheric CO2, especially in coastal environments. Due to the proximity of anthropogenic stressors in the coastal zone, the biogeochemical complexity of the CO2 system is more dynamic than the open ocean. My principal research objectives were addressed by developing methods for (1) improving the accuracy of pH measurements over a wide range of salinities and (2) investigating the acid-base properties of natural organics in the coastal ocean.

The accuracy of pH electrode calibrations is affected when …

The spinning disk technology has previously been utilized to isolate bacterial components from blood in hours instead of days. We hypothesized that this platform could be applied as an alternative approach to isolating plasma RNA from a whole blood sample. We consequently tested the efficacy of the spinning disk technology to extract plasma from whole blood upstream of RNA isolation and analysis. To do so, we collected plasma using either the spinning disk or the typical two-spin centrifuge method. We found that the spinning disk method results in significantly more hemolysis during collection than the conventional two-spin centrifuge method. However, …

Introduction: Chronic pain and posttraumatic stress disorder (PTSD) are prevalent comorbid conditions, particularly in Veterans; however, there are few integrated treatments for chronic pain and PTSD. Instead, interventions are typically implemented separately and may involve addictive opioids. Although there are highly effective, non-pharmacological treatments for PTSD, they are plagued by high dropout, which may be exacerbated by comorbid pain, as these PTSD treatments typically require increased activity. Importantly, a noninvasive pain treatment, tDCS (transcranial direct current stimulation) shows indications of effectiveness and may be integrated with psychological treatments, even when delivered via telehealth. This study examines the feasibility and initial …

Simple Summary

Chromosomal translocations involving the mixed lineage leukemia (MLL) gene generate potent fusion oncogenes and cause acute myeloid leukemia or lymphocytic leukemia, which account for ~75% infant and 5–10% child/adult acute leukemia cases with a poor prognosis (5-year survival rates < 45%). Protein–protein interactions between the most frequent MLL fusion partner proteins AF9/ENL and AF4 or histone methyltransferase DOT1L are critical to malignant gene expression and are therefore a potential drug target for cancer. Compound screening followed by medicinal chemistry studies identified several novel small-molecule inhibitors showing strong inhibition of these protein–protein interactions, significant suppression of characteristic gene expression, and robust cellular anticancer activities with negligible cytotoxicity. These compounds are useful chemical probes for biological studies of these protein–protein interactions, as well as pharmacological leads for further drug development against MLL-rearranged and other leukemias.

Abstract

Chromosomal translocations involving the mixed lineage leukemia (MLL) gene cause 5–10% acute leukemias with poor clinical outcomes. Protein–protein interactions (PPI) between the most frequent MLL fusion partner proteins AF9/ENL and AF4 or histone methyltransferase DOT1L are drug targets for MLL-rearranged (MLL-r) leukemia. Several benzothiophene-carboxamide compounds were identified as novel inhibitors of these PPIs with IC50 values as …

The γ-aminobutyric acid-mediated (GABAergic) system participates in many aspects of organismal physiology and disease, including proteostasis, neuronal dysfunction, and life-span extension. Many of these phenotypes are also regulated by reactive oxygen species (ROS), but the redox mechanisms linking the GABAergic system to these phenotypes are not well defined. Here, we report that GABAergic redox signaling cell nonautonomously activates many stress response pathways in

Eudaimonic Happiness, Temperament, And Political Ideology, Anthony Walsh

Journal of Ideology

This article looks at eudaimonic happiness and political ideology seen in terms of liberalism and conservativism. It appears that these two ideologies are driven by different temperaments that have been broadly viewed as Platonic and Aristotelian. It is often said that Aristotle is the master of those who know as Plato is the master of those who dream. Temperament is composed of a number of sub-traits that are highly heritable which, separately and together, move us to perceive the world in certain ways and thus help to forge worldviews that are emotionally attractive to us. Liberalism/conservativism is also consistently found …
