In the era of targeted therapy, there has been increasing concern about the development of oncology drugs based on the "more is better" paradigm, developed decades ago for chemotherapy. Recently, the US Food and Drug Administration (FDA) initiated Project Optimus to reform the dose optimization and dose selection paradigm in oncology drug development. To accommodate this paradigm shifting, we propose a dose-ranging approach to optimizing dose (DROID) for oncology trials with targeted drugs. DROID leverages the well-established dose-ranging study framework, which has been routinely used to develop non-oncology drugs for decades, and bridges it with established oncology dose-finding designs to …

The early events that lead to the inflammatory and immune-modulatory effects of radiation therapy (RT) in the tumor microenvironment (TME) after its DNA damage response activating the innate DNA-sensing pathways are largely unknown. Neutrophilic infiltration into the TME in response to RT is an early innate inflammatory response that occurs within 24-48 h. Using two different syngeneic murine tumor models (RM-9 and MC-38), we demonstrated that CXCR2 blockade significantly reduced RT-induced neutrophilic infiltration. CXCR2 blockade showed the same effects on RT-induced tumor inhibition and host survival as direct neutrophil depletion. Background: Electronic nicotine delivery systems (ENDSs) are rapidly increasing in the U.S., however, information about their long-term risks and benefits remains limited. This study examined the relationship between ENDS use and periodontal health among U.S. adults. (2) Methods: Data came from 33,822 adults who participated in the 2016–2018 wave of the Population Assessment of Tobacco and Health (PATH) study. Inclusion criteria were adults without a history of cigarette smoking or diabetes. Logistic regression analysis was performed to estimate the associations between ENDS use and a history of periodontal disease, with multivariable logistic regression adjusting for factors associated with poor oral …

Steel monopile support structures for offshore wind turbines require protection from corrosion on their external and internal surfaces. Cathodic protection (CP) works effectively to protect the external surfaces of monopiles, but internally, byproducts from aluminum sacrificial anode CP (SACP) and impressed current CP (ICCP) induce acidification that accelerates steel corrosion. This project investigated the effects of perforations on the performance of aluminum SACP and ICCP when used inside of steel pipes. Additionally, marine growth on the external and internal surfaces of the pipes was assessed to determine if CP byproducts affect marine organism development.

As a result of the exacerbation of the problem of water pollution, research was directed towards studying the treatment using ceramic membranes, which proved to be highly effective in treating all water sources. The research aims to study the possibility of preparing a new type of ceramic membranes from Syrian zeolite that was not previously used in this field. The aim of this investigation is to determine how different weight percentages of alumina nanoparticles, including 0.02, 0.04, and 0.06 percent wt, affect the physical characteristics of Poly Acrylamide (PAAM). Using a hot plate magnetic stirrer, 10 g of poly acrylamide powder was dissolved in 90 g of di-ionized distillate water for 4 hours to produce PAAM with a concentration of 0.11 g/ml. Sweet pepper (Capsicum annuum L.) is an economically important vegetable crop. Wilt disease caused by Fusarium oxysporum f. sp. capsici is a specific pathogen that affects the pepper. Four isolates of F. oxysporum f. sp. capsici Fo3, Fo6, Fo7 and Fo8 were obtained from diseased pepper plants that were collected from different pepper fields in Baghdad. Fo6 isolate that has high pathogenicity to pepper seeds, Trichoderma harzianum (Th) was tested in vitro against F. oxysporum f. sp. capsici showed a high inhibition rate for the isolate Fo6, the concentration of chelated iron Fe-EDDHA 0.5% reduced the radial growth of Fo6 …

In this paper, previous studies about Fuzzy regression had been presented. The fuzzy regression is a generalization of the traditional regression model that formulates a fuzzy environment's relationship to independent and dependent variables. All this can be introduced by non-parametric model, as well as a semi-parametric model. Moreover, results obtained from the previous studies and their conclusions were put forward in this context. So, we suggest a novel method of estimation via new weights instead of the old weights and introduce another suggestion based on artificial neural networks.

Diazotization reaction between quinolin-2-ol and (2-chloro-1-(4-(N-(5-methylisoxazol-3-yl)sulfamoyl)phenyl)-2l4-diazyn-1-ium was carried out resulting in ligand-HL, this in turn reacted with the next metal ions (Ni2+, Pt4+, Pd2+, and Mn2+) forming stable complexes with unique geometries such as (tetrahedral for both Ni2+ and Mn2+, octahedral for Pt4+ and square planer for Pd2+ ). The creation of such complexes was detected by employing spectroscopic means involving ultraviolet-visible which proved the obtained geometries, fourier transfer proved the formation of azo group and the coordination with metal ion through it. Topic Modelling is an established research area where the quality of a given topic is measured using coherence metrics. Often, we infer topics from Neural Topic Models (NTM) by interpreting their decoder weights, consisting of top-activated words projected from individual neurons. Transformer-based Language Models (TLM) similarly consist of decoder weights. However, due to its hypothesised superposition properties, the final logits originating from the residual path are considered uninterpretable. Therefore, we posit that we can interpret TLM as superposed NTM by proposing a novel weight-based, model-agnostic and corpus-agnostic approach to search and disentangle decoder-only TLM, potentially mapping individual neurons to multiple …

Spatial reasoning in text plays a crucial role in various real-world applications. Existing approaches for spatial reasoning typically infer spatial relations from pure text, which overlook the gap between natural language and symbolic structures. Graph neural networks (GNNs) have showcased exceptional proficiency in inducing and aggregating symbolic structures. However, classical GNNs face challenges in handling multi-hop spatial reasoning due to the over-smoothing issue, i.e., the performance decreases substantially as the number of graph layers increases. Neurodevelopmental disorders have their roots in multifaceted origins that manifest during the earliest stages of embryonic growth, often linked to sophisticated gene regulation dynamics. A perturbed metabolic environment, specifically owing to maternal obesity, can lead to dyslipidemia, subsequently threatening the optimal development of the embryonic central nervous system. Growing evidence suggests that such metabolic irregularities during pregnancy can alter neurogenesis pathways, thus predisposing the developing fetus to neurodevelopmental disorders. The efforts of this thesis are directed towards understanding the thermodynamics and kinetics underlying the dimerization reaction of a chloride/proton antiporter membrane protein, CLC-ec1, in lipid membranes. Due to the inherent challenges of working with membrane proteins, thermodynamic and kinetic information for the protein association reactions in membranes has been historically limited. The research presented in this thesis demonstrates the technological advancements for these measurements in a model membrane protein system, thereby setting an important milestone for the field. The genetic potentials of rice cultivars will need to be expressed to their fullest if global rice production is to be expanded enough by 2050 to meet the increased demand of the expanding population while the availability of land and water per capita dwindles. Across watershed science, two key variables emerge–streamflow and solute concentration–which serve as the basis for efforts ranging from basic watershed biogeochemistry research to policy decisions surrounding watershed management. However, we rarely account for how error in discharge (Q) impacts estimates of downstream nutrient loading. Here, we examined the impact of uncertainty in streamflow measurements on estimates of downstream nitrate export using publicly available data from the U.S. Geological Survey (USGS). We characterized how uncertainty in stage-discharge relationships impacts annual flux estimates across 70 USGS gages. Critical advances in radionuclide therapy have led to encouraging new options for cancer treatment through the pairing of clinically useful radiation-emitting radionuclides and innovative pharmaceutical discovery. Of the various subatomic particles used in therapeutic radiopharmaceuticals, alpha (α) particles show great promise owing to their relatively large size, delivered energy, finite pathlength, and resulting ionization density. This review discusses the therapeutic benefits of α-emitting radiopharmaceuticals and their pairing with appropriate diagnostics, resulting in innovative "theranostic" platforms. Fused Deposition Modeling (FDM) has gained widespread popularity as an affordable, versatile, and user-friendly additive manufacturing technique. However, ensuring consistent and high-quality prints remains a significant challenge. This study investigated the potential use of Nondestructive Evaluation (NDE) in the form of Acoustic Emission (AE) to optimize the FDM 3D printing process, including filament defects and the selection of print parameters. AE monitoring involves the detection and analysis of acoustic waves generated during the printing post-process, providing valuable insights into the dynamic behavior of a specimen with respect to the selected print parameters and integrity of the system. The gut is a major source of bacteria and antigens that contribute to neuroinflammation after brain injury. Colonic epithelial cells (ECs) are responsible for secreting major cellular components of the innate defense system, including antimicrobial proteins (AMP) and mucins. These cells serve as a critical regulator of gut barrier function and maintain host-microbe homeostasis. In this study, we determined post-stroke host defense responses at the colonic epithelial surface in mice. Females demonstrate greater fatigue resistance compared to males in tasks ranging from single-limb contractions to whole-body exercise, including running. Many of the studies investigating sex differences in fatigability following running, however, occur after long duration, low-intensity tasks and it is unknown whether there is a sex difference in fatigability following high-intensity running. This study compared fatigability and recovery following a 5 km running time trial in young males and females. Sixteen recreationally active participants (8 males, 8 females, age: 23 ± 4 years) completed a familiarisation and experimental trial. The apolipoprotein-E (APOE) ε4 allele is the greatest genetic risk factor for late-onset Alzheimer's disease (AD), but alone it is not sufficiently predictive. Because neuropathological changes associated with AD begin decades before cognitive symptoms, neuroimaging of healthy, cognitively intact ε4 carriers (ε4+) may enable early characterization of patterns associated with risk for future decline. Research in the cerebral cortex highlights a period of compensatory recruitment in elders and ε4+, which serves to maintain cognitive functioning. Yet, AD-related changes may occur even earlier in the cerebellum. Advances in electroencephalography (EEG) source localization now allow effective modeling of cerebellar activity. Background
Recent advances in data-driven computational approaches have been helpful in devising tools to objectively diagnose psychiatric disorders. However, current machine learning studies limited to small homogeneous samples, different methodologies, and different imaging collection protocols, limit the ability to directly compare and generalize their results. Here we aimed to classify individuals with PTSD versus controls and assess the generalizability using a large heterogeneous brain datasets from the ENIGMA-PGC PTSD Working group.
Methods

There is more human-generated noise and light today than ever before, a trend tied to ongoing urbanization and with reverberating impacts on a wide range of organisms. While research on these impacts is mounting and particularly plentiful when it comes to birds, there is still much to explore within its effects on avian behavior, fitness, and evolution. In chapter 1, we investigated how fledgling movement is influenced by noise and light pollution. While adult birds have well-documented species-specific responses to these stimuli, fledglings are sorely understudied in this context. Lichens are an underrepresented group of organisms when it comes to both research and conservation efforts. At the same time, lichens face increasing threats from anthropogenic sources including wildfires, climate change and urbanization. With this thesis, I seek to ascertain and publish ecological data on a critically endangered lichen to inform future conservation designations and efforts to preserve and protect the species in perpetuity.
