Analyzing Functional Interactions Of Designed Peptides By Nmr Spectroscopy, Wonsuk Choi
Analyzing Functional Interactions Of Designed Peptides By Nmr Spectroscopy, Wonsuk Choi
Pharmaceutical Sciences (MS) Theses
The development of small peptide-based therapeutics can be accelerated by the knowledge of relationships between the peptide structure and its functional interactions. Here, we report the analysis of two groups of synthetic peptides designed for two applications – broad bactericidal action and inhibition of protein-protein interactions in human cells. Novel amphiphilic peptides designed for antibacterial application incorporated arginine as cationic amino acids and non-natural amino acids that have aromatic side chains with similar hydrophobic properties as tryptophan. The interaction of lead cyclic peptides and their linear analogs with a phospholipid bilayer mimicking a bacterial membrane was studied using nuclear magnetic …

Vascularization is a key pre-requisite to engineered anatomical scale three dimensional (3-D) constructs to ensure their nutrient and oxygen supply upon implantation. Presently, engineered pre-vascularized 3-D tissues are limited to only micro-scale hydrogels, which meet neither the anatomical scale needs nor the complexity of natural extracellular matrix (ECM) environments. Anatomical scale perfusable constructs are critically needed for translational applications. To overcome this challenge, we previously developed pre-vascularized ECM sheets with long and oriented dense microvascular networks. The present study further evaluated the patency, perfusability and innate immune response toward these pre-vascularized constructs. Macrophage-co-cultured pre-vascularized constructs were evaluated in vitro to …
+Protective Roles For Interferon-Gamma In Astrocytes During Chronic Autoimmunity, Brandon C. Smith
+Protective Roles For Interferon-Gamma In Astrocytes During Chronic Autoimmunity, Brandon C. Smith
ETD Archive
Multiple sclerosis (MS) is an inflammatory and neurodegenerative disease of the central nervous system (CNS). Infiltrating inflammatory immune cells perpetuate demyelination and axonal damage in the CNS and significantly contribute to pathology and clinical deficits. Interferon (IFN)γ is classically described as deleterious in acute CNS autoimmunity, we and others have shown astrocytic IFNγ signaling also has a neuroprotective role. We found IFNγ signaling on astrocytes upregulates subunits of the immunoproteasome (iP) and programmed death ligand 1 (PD-L1), both of which serve protective roles during autoimmunity. Astrocyte-specific knockouts of Ifngr1 resulted in increased clinical severity in the MS murine model, experimental …
Exploration Of Secondary Structure Directing Agents On The Formation Of Hierarchical And 2d Zeolites, Suleiman Yahaya
Exploration Of Secondary Structure Directing Agents On The Formation Of Hierarchical And 2d Zeolites, Suleiman Yahaya
ETD Archive
Hierarchical and 2D zeolites address the diffusion limitation of conventional zeolites by enhancing diffusion in the mesopores. So far, the surfactant compounds, which successfully produce hierarchical zeolites, have complex molecular structures making the zeolite synthesis expensive and uncompetitive for commercialization, making cost-effective synthesis of hierarchical and 2D zeolite highly desirable. However, the development of effective synthesis protocols relies on a good understanding of the SDA's influence on crystal morphology development, which is largely unknown. Here, we explore inexpensive and readily available compounds via a dual-SDA strategy and use MFI zeolite as the model system for the research. TPAOH served as …

Protective Responses Of The Intestinal Epithelial Cell Line Ht-29 Cells Exposed To Dephosphorylated Salmonella Flagellin [Research Note], Heyeon Baik, Jaiesoon Cho
The Philippine Agricultural Scientist
This study aimed to describe the effects of Salmonella Typhimurium flagellin (SFL) dephosphorylated by sweet potato purple acid phosphatase (PAP) on the protective responses of the intestinal epithelial cell line HT-29 cells. The enzyme was reported to display a broad substrate specificity for various organic phosphorylated conjugates and phosphoproteins. Dephosphorylation of SFL by sweet potato PAP decreased to 35% in the presence of 0.05 mM vanadate as compared with the negative control (p < 0.05). Intact SFL and the SFL treated with sweet potato PAP did not remarkably induce the activation of caspase-3 in HT-29 cells at all the tested levels of the substrate. Intact SFL maximally induced the release of IL (interleukin)-8 in HT-29 cells at 1000 ng/mL (p < 0.05). However, the SFL treated with the enzyme inhibited the release of IL-8 at over 100 ng/mL of the substrate as compared with intact SFL, resulting in an approximately 8-fold decrease even at 1000 ng/mL (p < 0.05). The SFL treated with the enzyme decreased the activation of the total ERK1/2 in the cells to 1.9 and 1.7–fold at 10 and 1000 ng/mL of the substrate, respectively, as compared with intact SFL (p < 0.05). In conclusion, sweet potato PAP could be a promising tool for controlling excessive inflammation during Salmonella infection in animal husbandry, and the enzyme could be a safe alternative that can overcome the drawbacks of chemotherapy.

The Perception Of Natural Resource Management In Nebraska: Efforts For Cross-Boundary Collaborative Management, Daniel Morales
School of Natural Resources: Dissertations, Theses, and Student Research
Nebraska’s agricultural landscapes are rapidly changing, affecting natural resources and their successful management. I utilized two surveys and scenario planning (Chapters 1: statewide survey, 2: local survey, and 3: scenario-planning workshop) to investigate attitudes and perceptions of natural resource management and cross-boundary collaboration. My first objective focused on determining what prevents Nebraskans from addressing natural resources challenges, considering demographics amongst generations and the type of areas they live in (rural versus urban). The second objective focused on whether landowners engaged with their community in managing natural resources. The third objective was to develop alternative future scenarios for the Denton Hills …

Light addressable electrochemical (LAE) sensors have seen great utility in the past several years because they enable multiple localized electrochemical measurements to be performed on a single macroscopic electrode, opening up applications in imaging, biosensing, surface patterning, and multiplexing. In this study, we investigated the effects of electrodeposition on the formation of LAE sensors formed between n-Si and electrodeposited Pt. We prepared sensors by electrodepositing Pt onto freshly-etched n-Si under a variety of conditions, varying the Pt precursor concentration, electrodeposition time, supporting electrolyte, and potential waveform. We characterized the sensors using a combination of atomic force microscopy, electrochemical impedance spectroscopy, …

The dissolution of anthropogenic carbon dioxide (CO2) in seawater has altered its carbonate chemistry in the process of ocean acidification (OA). OA affects the viability of marine species. In particular, calcifying organisms and their early planktonic larval stages are considered vulnerable. These organisms often utilize energy reserves for metabolism rather than growth and calcification as supported by bulk RNA-sequencing (RNA-seq) experiments. Yet, transcriptomic profiling of a bulk sample reflects the average gene expression of the population, neglecting the variations between individuals, which forms the basis for natural selection. Here, we used single-embryo RNA-seq on larval sea urchin Heliocidaris crassispina, which …

The Aegean region makes a prominent contribution to Turkish viticulture. The region, which includes important grape culture centers such as Manisa, Denizli, and İzmir, has extremely favorable conditions for the cultivation of seedless grapes as well as table and wine grape varieties. In this region, which possesses rich grapevine genetic diversity together with its area and production potential, it is seen that there is variety confusion caused by different nomenclature and intravariety variations. In addition, some old grape varieties that are not used in cultivation are at risk of disappearing. In order to address these problems, the viticulture potentials of …

Climate change is the major factor that affects crop yield and productivity in the long term. The changing climate scenario is now impacting farmers’ lands and their outcomes in both developed and developing countries. The intercropping of crop species or varieties that have high arable capability is a promising technique for increasing crop production without increasing agricultural inputs. In intercropping, the selection of varieties, optimum sowing time, method of plant arrangement, and sowing proportion are required to make the design suitable for an intercropping system. Intercropping is one of the most highlighted agronomic practices helping to improve soil health (structure, …

The strawberry tree (Arbutus andrachne L.) is a wild edible fruit that grows naturally in the Mediterranean climate zone, including Türkiye. In this study, the genetic morphological, biochemical, and genetic diversity of 10 superior strawberry tree genotypes growing naturally in Antalya Province in Türkiye were investigated. Morphological and biochemical characterization were performed based on some important fruit characteristics. In addition, genetic variation was detected at the molecular level using simple sequence repeat (SSR) markers. The genotypes exhibited fruit weight, width, length, and shape index values of 1.22–2.12 g, 12.07–15.28 mm, 10.15–14.24 mm, and 0.99–2.22, respectively. The biochemical parameters soluble solid …

A plum pox virus (PPV) susceptible traditional cultivar (Besztercei Bt.2) and a clone of a PPV resistant autochthonous landrace, Nemtudom (Nábrád) of the hexaploid European plum (Prunus domestica) were used to identify and characterize an isoform of the eukaryotic translation initiation factor, eIF(iso)4E, a host susceptibility factor required for PPV infection in plum. The cDNA and intron sequences of the eIF(iso)4E gene and the modeled 3D structure of the encoded proteins revealed sequence alterations that are not expected to critically influence the structure and function of the protein. The introns shared the canonical GT–AG splice sites, although they showed base …

Heat stress (HS) is expected to become a significant abiotic stressor with the continued rise in global temperatures, severely limiting the development and production of bell pepper (Capsicum annuum L.), especially in arid and semiarid regions. Thus, this study looked into the impact of HS on the antioxidant activity, as well as the biochemical responses of bell pepper seedlings under varying temperatures (40, 32, and 25 °C) for 5 days. The results showed that exposure to higher temperatures led to an increase in enzymatic and antioxidant processes. At 40 and 32 °C, HS led to a decrease in chlorophyll a …

Mango is a fruit consumed worldwide, but generates wastes, which have potential to be valorised, because they possess compounds with demonstrated positive implications for the consumer’s health. For this reason, in the present study, biscuits with mango pulp and powders developed from its wastes were developed. The chemical composition was determined for the mango peel, pulp, and seed according to AOAC procedures, but also for the mango biscuit and a reference. The total phenolic content (TPC) was assessed with the Folin–Ciocâlteu method, while the antioxidant potential was tested using the 2,2-diphenyl-1-picrylhydrazyl (DPPH) method. Titratable acidity and alkalinity were also determined. …
