Optical trapping of metallic nanoparticles investigates phenomena at the interface of plasmonics and optical micromanipulation. This thesis combines ideas from the optical properties of metals, originating in solid-state physics, with the force mechanisms resulting from optical trapping. I explore the influence of enhanced light-matter interaction due to the plasmon resonances of gold particles on their trapping properties. I present theories for trapping mechanisms of gold metallic particles and then verify these theories through experiments. Hereditary spastic parapareses (HSPs) are clinically heterogeneous motor neuron diseases with variable age of onset and severity. Although variants in dozens of genes are implicated in HSPs, much of the genetic basis for pediatric-onset HSP remains unexplained. Here, we re-analyzed clinical exome-sequencing data from siblings with HSP of unknown genetic etiology and identified an inherited nonsense mutation (c.523C>T [p.Arg175Ter]) in the highly conserved RAB1A. The mutation is predicted to produce a truncated protein with an intact RAB GTPase domain but without two C-terminal cysteine residues required for proper subcellular protein localization. The human mitochondrial genome (mtDNA) is a circular DNA molecule with a length of 16.6 kb, which contains a total of 37 genes. Somatic mtDNA mutations accumulate with age and environmental exposure, and some types of mtDNA variants may play a role in carcinogenesis. Recent studies observed mtDNA variants not only in kidney tumors but also in adjacent kidney tissues, and mtDNA dysfunction results in kidney injury, including chronic kidney disease (CKD). An optimized hepatocellular carcinoma (HCC)-targeted methylation next generation sequencing assay was developed to discover HCC-associated methylation markers directly from urine for HCC screening. Urine cell-free DNA (ucfDNA) isolated from a discovery cohort of 31 non-HCC and 30 HCC was used for biomarker discovery, identifying 29 genes with differentially methylated regions (DMRs). Methylation-specific qPCR (MSqPCR) assays were developed to verify the selected DMRs corresponding to 8 genes (GRASP, CCND2, HOXA9, BMP4, VIM, EMX1, SFRP1, and ECE). Using archived ucfDNA, methylation of GRASP, HOXA9, BMP4, and ECE1, were found to be significantly different (p < 0.05) between HCC and non-HCC patients. The four markers together with previously reported GSTP1 and RASSF1A markers were assessed as a 6-marker panel in an independent training cohort of 87 non-HCC and 78 HCC using logistic regression modeling. AUROC of 0.908 (95% CI, 0.8656-0.9252) was identified for the 6-marker panel with AFP, which was significantly higher than AFP-alone (AUROC 0.841 (95% CI, 0.778-0.904), p = 0.0026). Applying backward selection method, a 4-marker panel was found to exhibit similar performance to the 6-marker panel with AFP having 80% sensitivity compared to 29.5% by AFP-alone at a specificity of 85%. This study supports the potential use of methylated transrenal ucfDNA for HCC screening.

Background Human mesenchymal stem cells (hMSCs) are being researched for cell-based therapies due to a host of unique properties, however, genetic modification of hMSCs, accomplished through nonviral gene delivery, could greatly advance their therapeutic potential. Furthermore, expression of multiple transgenes in hMSCs could greatly advance their clinical significance for treatment of multifaceted diseases, as individual transgenes could be expressed that target separate pathogenic drivers of complex diseases. This work is a unified study of stable and unstable steady states of 2D active nematic channel flow using the framework of Exact Coherent Structures (ECSs). ECSs are stationary, periodic, quasiperiodic, or traveling wave solutions of the governing equations that, together with their invariant manifolds, organize the dynamics of nonlinear continuum systems. We extend our earlier work on ECSs in the preturbulent regime by performing a comprehensive study of stable and unstable ECSs for a wide range of activity values spanning the preturbulent and turbulent regimes. While best practices in criminal justice reform have been studied and identified, the point at which reform legislation is decided upon and factors related to its passage is a traditionally understudied. My research will explore the factors that influence the political will of legislators who pass criminal justice reforms. Using a constructivist view of grounded theory, my research uses qualitative data to determine patterns of words and generalities implicit in decision-making for legislators in the state of Louisiana by way of semi-structured interviews with current and former state legislators. The theory guiding this study is attachment theory. Thomas Jefferson’s autobiography reveals his continual struggle against slavery and his frustration at the resistance of the “public mind” in Virginia, predominantly composed of slave-owning aristocrats. Despite vocal condemnations of slavery, attempts to translate his anti-slavery stance into formal documents faced significant resistance from the society he aimed to change. Even within the House of Burgess, Jefferson's support for a bill allowing individual slave owners to free their slaves was met with contempt. There is an overrepresentation of African American youths at every stage of the juvenile justice system in America, from initial interaction with criminal justice to sentencing and imprisonment. This quantitative correlational study aimed to study the perceived racial discrimination of incarcerated African American juveniles by correctional officers. This qualitative study aimed to find the recurring themes amidst Facebook audience engagement of the Netflix drama series. The series sampling size was collected from Nielsen Ratings (2023) and Netflix’s (2022) top ten charts for three consecutive months from 11/4/2022-1/29/2023. Netflix was the most-watched streaming platform, with 80% of viewership, with drama being the most prevalent genre. The theoretical frameworks were medium and new medium theories. The theories guiding this study were Ivan Pavlov’s classical conditioning and Adam Morton’s theory of mind in predictive behaviors. Neutralization assays like the plaque reduction neutralization test (PRNT) and focus reduction neutralization test (FRNT) are the most specific ways to measure neutralizing antibodies, which provide a good estimate of protection for RNA viruses. However, these assays are time-consuming, labor-intensive, and low-throughput. Alternative methods that are faster and convenient are necessary to quantify antibody responses in large-scale studies. This bacterium is notorious for its ability to share genetic content within and outside of its species. OBJECTIVES: Primary and post-primary tuberculosis (TB) are distinct entities. The aim of this study was to study the histopathology of primary and post-primary TB by using the unique human autopsy material from the pre-antibiotic era, 1931-1947.

MATERIAL AND METHODS: Autopsy data were collected from the autopsy journals, and the human tissue was collected from the pathology archives at the Department of Pathology, the Gades Institute.

RESULTS: Histological presentations of TB lesions showed great diversity within a single lung. Post-primary TB starts as a pneumonia forming early lesions, characterized by the infiltration of foamy macrophages containing mycobacterial antigens within alveoli, and …
