Secondary Electrons And Compton X-Ray Generation In Solid And Heated Biological Sample In The Presence Of Radiologic Contrast Media, Vanessa Robinson, Daler Djuraev, Fatima Ahmed
Publications and Research
Our goal is identification of secondary electrons and Compton photons when externally infused radiological contrast from iodine and gadolinium interferes with calcium and iron in tissues. This could help develop next generation of x-ray methods as well as mechanistic understanding of toxic effects of radiology contrast materials on living tissue and agriculture. Changes in tissue minerals over time, aging and heat stress are manipulated in our experiments by infusing imaging contrast agents that consist of large atoms like Gadolinium and Iodine. These have chelation properties to alter electron distribution of proteins and carbohydrates of biological matrix. Our experiments try to …

BLUF: Limit your aspartame consumption!
Studies have shown that consecutive aspartame consumption at as little as 7% of the FDA recommended daily intake can cause anxiety symptoms in ~8 days. Not only that, but after 12 weeks of consumption, it was proven that the anxiety symptoms are passed to the offspring two generations later. That means if you have roughly one to two 32 oz Diet Cokes per day, your children and grandchildren will show symptoms of anxiety even if they never consume aspartame themselves. It is important to note that aspartame is in many foods and products labeled as …

Metabolic dysfunction-associated fatty liver disease (MAFLD) is the most common chronic liver disease worldwide. Circadian disruptors, such as chronic jet lag (CJ), may be new risk factors for MAFLD development. However, the roles of CJ on MAFLD are insufficiently understood, with mechanisms remaining elusive. Studies suggest a link between gut microbiome dysbiosis and MAFLD, but most of the studies are mainly focused on gut bacteria, ignoring other components of gut microbes, such as gut fungi (mycobiome), and few studies have addressed the rhythm of the gut fungi. This study explored the effects of CJ on MAFLD and its related microbiotic …

Liquid Biopsy: From Concept To Clinical Application, Catherine Alix-Panabières, Dario Marchetti, Julie E. Lang
Pathology Research and Scholarship
The diagnosis and treatment of cancer presents a physical and mental burden to the patient, often involving diagnostic biopsies and surgeries or chemotherapeutic approaches with severe side-effects. Advances which enable early detection of cancer and close monitoring of the disease course without invasive procedures, and which can underpin a tailored approach to treatment, can therefore make a big difference to the quality of life of patients. Liquid biopsies can be used to access tumor cells and tumor DNA circulating in the blood. Monitoring these species can provide a minimally invasive and repeatable means to detect cancer, or gain information about …
Leucine Lock: An Amplified Rapid Antigen Detection System, Matthew David Goff
Leucine Lock: An Amplified Rapid Antigen Detection System, Matthew David Goff
Theses and Dissertations
Rapid diagnostic tests for COVID-19, influenza, and blood analytes currently have many drawbacks. They vary greatly in accuracy, are expensive to manufacture, and can be insufficiently sensitive to small antigen loads. Leucine lock is designed to rectify these issues that exist in current rapid diagnostics by creating a rapid antigen test with minimal background activity, tunable sensitivity, and low manufacturing cost. Leucine lock consists of a novel leucine zipper containing an intramolecular hairpin, an ScFv antigen binding domain, and a fluorophore reporter in a FRET combination. By altering the number of hydrophobic and electrostatic mismatches in the intramolecular hairpin, as …
Drop Retention And Departure In Shear Flow On Structured Superhydrophobic Surfaces, Blake Morgan Lyons
Drop Retention And Departure In Shear Flow On Structured Superhydrophobic Surfaces, Blake Morgan Lyons
Theses and Dissertations
Water is often found adhering to surfaces in droplet form. Droplets can develop as a result of precipitation, condensation, or chemical reactions. Drops are retained or held on a surface due to a retention force exerted on the drop by the surface. This retention force is a function of the surface tension of the liquid, drop geometry, and the contact angle between the drop and the surface. When external or body forces exceed this retention force the drop begins to move. This thesis seeks to understand and quantify the forces present at the point of droplet departure once it begins …
The Effect Of Percussive Massage On The Maintenance Of Skeletal Muscle Health During Limb Unloading, Mohadeseh Ahmadi
The Effect Of Percussive Massage On The Maintenance Of Skeletal Muscle Health During Limb Unloading, Mohadeseh Ahmadi
Theses and Dissertations
A period of skeletal muscle disuse leads to muscle mass and strength loss and compromised metabolic health. Several studies suggest that passive mechanical stimulation may protect against the detrimental effects of disuse in skeletal muscle. This study assessed the effect of the twice-daily application of percussive massage in mitigating the adverse effects of muscle disuse during ten days of leg unloading. Seventeen participants (7 women and 10 men; 18-35 yrs.) were randomly assigned to either control or massage groups, both undergoing leg unloading. The data revealed that both groups similarly experienced significant reductions in muscle strength (-14%±5.9; p<.0001), quadriceps volume (-4.2%±2.8; p<.0001), myofiber cross-sectional area (-8.2%±1.8; p<.0001), and maximal mitochondrial respiration (-20%±9.1; p<.0001). Fatty acid-supported respiration decreased in the control group (-30.4±18.5%; p= 0.0008) and remained unchanged in the massage group. Hydrogen peroxide (H2O2) flux normalized to oxygen consumption exhibited a significant interaction between group and time, particularly in the absence of ADP, where H2O2 flux increased only in the control group (control: 29.9±26.5%, massage: -13±22.6%; p=0.01). There was no change in the H2O2 flux after the addition of ADP. In conclusion, ten days of limb unloading resulted in diminished muscle size, strength, and mitochondrial respiration. Percussive massage was insufficient to prevent the loss of muscle mass, force production, or carbohydrate-supported respiration. However, it exhibited potential protective effects against the decline in mitochondrial fatty acid-supported respiration.

Structural Features Of Hiv-1 Integrase In The Late Stage Of Viral Replication, Tolga Catmakas
Structural Features Of Hiv-1 Integrase In The Late Stage Of Viral Replication, Tolga Catmakas
Dissertations
HIV-1 integrase (IN) is an essential enzyme for the viral life cycle as it integrates viral genomic material into the host cell genome. In addition to its’ essential catalytic function, IN has another important non-catalytic role where it binds the viral RNA to facilitate successful particle maturation during the viral life cycle. A significant number of IN mutants, addressed as class II IN mutants, show various defects, affecting different steps of the viral life cycle. Class II IN mutants exhibit aberrant morphology where viral ribonucleoprotein particles are mislocalized outside of the capsid core. This phenomenon can be observed in the …

Genomic And Proteomic Analysis Of Five Vi01-Like Phages Reveals Wide Host Range And Multiple Tail Spike Proteins, Evan Harris
Genomic And Proteomic Analysis Of Five Vi01-Like Phages Reveals Wide Host Range And Multiple Tail Spike Proteins, Evan Harris
Theses and Dissertations
The number of illnesses attributable to antibiotic resistant bacterial infections are on the rise. According to the CDC, by 2050, complications due to antibiotic resistant infections are set to become the most prevalent cause of death worldwide, overtaking both cancer and heart disease in number. We are required to look for supplements to aid antibiotics in the fight against bacteria because it's becoming increasingly clear that antibiotics alone will fail in the face of multidrug-resistant strains. Bacteriophages are natural killers of bacteria and can thus be weaponized to treat multidrug-resistant infections. When a phage is discovered, our databases need to …

The persistence of reefs relies on mature corals spawning synchronously to maximize fertilization and produce larvae to replenish local populations. Corals synchronize the release of gametes by responding to temperature, sun, and moon light cycles; however, abnormalities in these patterns can disrupt synchrony. This study is the first to describe regional asynchronous spawning of Acropora cervicornis by quantifying gamete development and spawning times among two reefs, an in situ nursery off Fort Lauderdale, and an in situ nursery in the Florida Keys. While A. cervicornis in the Florida Keys synchronously spawned within the predicted window of 2-5 days after the …

Biophysical Factors Affecting Habitat Suitability For Crassostrea Virginica, Jason D. Tilley
Biophysical Factors Affecting Habitat Suitability For Crassostrea Virginica, Jason D. Tilley
Dissertations
Oyster reefs provide a variety of important ecosystem services. However, the mortality rate of eastern oyster, Crassostrea virginica, the dominant species that produces oyster reefs in the northern Gulf of Mexico, is increasing at an alarming rate due to a variety of abiotic and biological factors. I examined how biophysical factors, including the less-studied fatty acid profiles of the suspended particulate matter on which oysters feed, influenced morphometric condition of C. virginica.
I sampled suspended particulate matter (SPM) and oysters in-situ in the western Mississippi Sound, which historically supported the majority of oyster production in Mississippi waters. Sampling …

Characterizing Stress Granule Regulation By Pas Kinase, Ataxin-2 And Ptc6 And Investigating The Lifespan Of Covid-19 Virus On Currency, Colleen R. Newey
Theses and Dissertations
The protein Ataxin-2 is a known positive regulator of stress granules in humans, mice and yeast (known as yeast PBP1). Due to the role that stress granules play in diseases including Amyotrophic Lateral Sclerosis (ALS) and cancer, this thesis investigates the role of Ataxin-2 and its protein binding partners in stress granule development and its effects on various metabolic phenotypes of the cell. PAS kinase is a sensory protein kinase, conserved from yeast to man, which regulates respiration and lipid biosynthesis. Our lab discovered that PAS kinase phosphorylates and activates Ataxin-2 in yeast, and that PAS kinase overexpression enhances localization …

Interfacial Properties Of Hybrid Lipid-Polymer Bilayers: Applications In Drug Delivery And Biosensors, Keith L. Willes
Theses and Dissertations
Amphiphilic block copolymers are unique macro-molecules capable of self-assembling into bilayers analogous to naturally occurring lipid membranes. When combined with lipids, these copolymers form hybrid membranes with unique and sometimes unpredictable properties, including increased chemical and mechanical stability. These synthetically enhanced biological structures represent a versatile platform suitable for a wide range of applications, from advanced biosensing devices to drug delivery systems. The realization of these advancements necessitates a deep understanding of material properties, including the ability to predict and control interfacial behaviors. It has been shown that in the case of pure lipid membranes, interfacial behaviors are dominated by …
