Myotonic dystrophy type 1 (DM1) is a slowly progressive, multisystem disorder caused by a CTG repeat expansion in the DMPK 3’UTR that leads to global dysregulation of alternative splicing. The resulting decline in physical function is slow, and no reliable biomarkers exist for predicting disease progression; however, an RNA mis-splicing biomarker associated with weakness may have utility in predicting functional outcomes. Here we validate the Splice Index (SI) as a potential biomarker of DM1-associated strength and function. However, labeled activity recognition datasets are costly to collect and highly varied, which challenges machine learning techniques that rely on large datasets. Furthermore, activity recognition in practice needs to account for user trust - models are motivated to enable interpretability, usability, and information privacy. As Murry and colleagues (2018) note, racism serves as a family-level stressor, impacting both Black adolescents and their parents. As Black teens continue to experience frequent and varied racism experiences (English et al., 2020), their Black parents are facing a double challenge: bearing the weight of their own experiences with racism, while also parenting a Black adolescent through the same. Electrospun polymeric biodegradable scaffolds are essential in tissue engineering, particularly for Engineered Tissue Vascular Grafts (ETVGs), which promise advancements in treating coronary artery disease, peripheral arterial disease, congenital cardiovascular defects, and renal disease. These scaffolds present a solution to issues with autologous graft availability and durability. While large-diameter grafts in low-pressure environments have seen success, small-diameter grafts in high-flow scenarios remain challenging. Understanding polymeric scaffold degradation and behavior during incubation, especially under dynamic mechanical loading, is vital for clinical translation of small-caliber ETVGs.

In this study, we assessed the quality of de novo genome assemblies for three species of parasitic nematodes (Brugia malayi, Trichuris trichiura, and Ancylostoma caninum) generated using only Oxford Nanopore Technologies MinION data. Assemblies were compared to current reference genomes and against additional assemblies that were supplemented with short-read Illumina data through polishing or hybrid assembly approaches. For each species, assemblies generated using only MinION data had similar or superior measures of contiguity, completeness, and gene content. In terms of gene composition, depending on the species, between 88.9 and 97.6% of complete coding sequences predicted in MinION data only assemblies …

BACKGROUND: Genetic variants and gene expression predict risk of chronic obstructive pulmonary disease (COPD), but their effect on COPD heterogeneity is unclear. We aimed to define high-risk COPD subtypes using genetics (polygenic risk score, PRS) and blood gene expression (transcriptional risk score, TRS) and assess differences in clinical and molecular characteristics.

METHODS: We defined high-risk groups based on PRS and TRS quantiles by maximising differences in protein biomarkers in a COPDGene training set and identified these groups in COPDGene and ECLIPSE test sets. In 2017, a criminal court judge in rural Tennessee made this offer to people incarcerated through sentences issued by his court. When litigators sought monetary damages and to enjoin the practice, and the local media exposed the scheme, the judge begrudgingly rescinded the order while expressing surprise that it was controversial. The Board of Judicial Conduct issued a letter of reprimand to the judge. Meanwhile, the United States District Court for the Middle District of Tennessee summarily dismissed the constitutional tort claims against the judge based on absolute …

Formally, judicial analysis of a challenged statute's validity should be consistent, regardless of the challenge's pre- or post-enforcement posture. A post-enforcement posture arises when an aggrieved party defensively challenges a purportedly unconstitutional statute being enforced against them. Alternatively, a pre-enforcement posture arises when an impacted party strikes first, attacking the statute by asserting a credible threat that the law will be enforced against them in the future. Either way, judicial evaluation of a statute's validity should turn on its content and effect-not on whether it was challenged before or after enforcement.

Background: Pregnant and postpartum women are at high risk of depression due to hormonal and biological changes. Antenatal depression is understudied compared to postpartum depression and its predictors remain highly controversial.

Aim: To estimate the prevalence of depressive symptoms during pregnancy and investigate factors associated with this condition including vitamin D, folate and Vitamin B₁₂ among participants in the Kuwait Birth Study.

The prevalence of Non-alcoholic Fatty Liver Disease (NAFLD) is increasing, particularly in individuals who consume minimal to no alcohol. Currently, NAFLD affects at least 32% of the global population. Although not all but approximately 5-10% cases of NAFLD progress to non-alcoholic steatohepatitis (NASH), a condition arising from lipotoxicity. Patients with NASH may further develop liver fibrosis, cirrhosis, and hepatocellular carcinoma (HCC). However, it is important to note that progression from NAFLD to NASH and HCC is not inevitable for all patients. Article 103 of the United Nations Charter establishes the primacy of states’ obligations under the Charter over their other international obligations. The effect of Article 103 on states’ obligations has been discussed in the jurisprudence and literature but only in the context of obligations conflicting with binding Security Council decisions. Open data initiatives and knowledge graphs, in synergy, have contributed to an increasing volume of disaster-related data in the Semantic Web. Synthesizing and enriching these data is critical to support all aspects of data-driven disaster risk reduction and management. A standard template that coherently defines, maps, and classifies the wide range of hazards to which communities are exposed is a key input for this task. The UNDRR-ISC Hazard Information Profiles (HIPs) provide evidence-informed standardization of hazard nomenclature and definitions and a “science-backed” classification.
