Traumatic brain injury (TBI) is a worldwide health burden that causes death and disability. Clinical management of TBI involves guideline-based protocols to limit secondary brain injury but there remains an unmet need for therapeutics to target neuroprotection and to improve cognitive outcomes in patients. This dissertation concerns the examination of RNA binding motif 5 (RBM5) as a potential therapeutic gene target to promote neuroprotection in models of TBI. RBM5 is a potent pro-death tumor suppressor gene in cancer. We hypothesized that it also promotes cell death in neurons in the brain and therefore its inhibition would decrease neuropathology following CNS …

Prunella vulgaris L. (PV) is a widely distributed plant species, known for its versatile applications in both traditional and contemporary medicine, as well as in functional food development. Despite its broad-spectrum antimicrobial utility, the specific mechanism of antibacterial action remains elusive. To fill this knowledge gap, the present study investigated the antibacterial properties of PV extracts against methicillin-resistant Staphylococcus aureus (MRSA) and assessed their mechanistic impact on bacterial cells and cellular functions. The aqueous extract of PV demonstrated greater anti-MRSA activity compared to the ethanolic and methanolic extracts. UPLC-ESI-MS/MS tentatively identified 28 phytochemical components in the aqueous extract of PV. …

Somatostatin Receptor Subtype-2 Targeting System For Specific Delivery Of Temozolomide, Solmaz Aghaamiri, Sukhen C Ghosh, Servando Hernandez Vargas, Daniel M Halperin, Ali Azhdarinia

Temozolomide (TMZ) is a DNA alkylating agent that produces objective responses in patients with neuroendocrine tumors (NETs) when the DNA repair enzyme O6-methylguanine-DNA methyltransferase (MGMT) is inactivated. At high doses, TMZ therapy exhausts MGMT activity but also produces dose-limiting toxicities. To reduce off-target effects, we converted the clinically approved radiotracer 68Ga-DOTA-TOC into a peptide-drug conjugate (PDC) for targeted delivery of TMZ to somatostatin receptor subtype-2 (SSTR2)-positive tumor cells. We used an integrated radiolabeling strategy for direct quantitative assessment of receptor binding, pharmacokinetics, and tissue biodistribution. Translocation renal cell carcinoma (tRCC) most commonly involves an ASPSCR1-TFE3 fusion, but molecular mechanisms remain elusive and animal models are lacking. Here, we show that human ASPSCR1-TFE3 driven by Pax8-Cre (a credentialed clear cell RCC driver) disrupted nephrogenesis and glomerular development, causing neonatal death, while the clear cell RCC failed driver, Sglt2-Cre, induced aggressive tRCC (as well as alveolar soft part sarcoma) with complete penetrance and short latency. However, in both contexts, ASPSCR1-TFE3 led to characteristic morphological cellular changes, loss of epithelial markers, and an epithelial-mesenchymal transition. Undergraduate research experiences are an instrumental component of student development, increasing conceptual understanding, promoting inquiry-based learning, and guiding potential career aspirations. Moving one step further, as research continues to become more interdisciplinary, there exists potential to accelerate student growth by granting additional perspectives through collaborative research. This study demonstrates the utilization of a model collaborative research project, specifically investigating the development of sorbent technologies for efficient CO2 capture, which is an important research area for improving environmental sustainability. Autosomal dominant polycystic kidney disease (ADPKD) is a monogenic disorder accounting for approximately 5% of patients with renal failure, yet therapeutics for the treatment of ADPKD remain limited. ADPKD tissues display abnormalities in the biogenesis of the centrosome, a defect that can cause genome instability, aberrant ciliary signaling, and secretion of pro-inflammatory factors. Cystic cells form excess centrosomes via a process termed centrosome amplification (CA), which causes abnormal multipolar spindle configurations, mitotic catastrophe, and reduced cell viability. However, cells with CA can suppress multipolarity via "centrosome clustering," a key mechanism by which cells circumvent apoptosis. Here, we demonstrate that inhibiting …

Cave guano deposits are increasingly being recognized as valuable repositories of paleo-climatic and paleo-environmental information. However, that value is constrained by rates of guano decomposition, and these rates have not been previously well-studied. Here we show that field and laboratory studies of deep insectivorous bat guano sequences in the caves of Borneo demonstrate the extreme dependence of decomposition rate on guano water content. Under tropical conditions, moist guanos exhibit decomposition coefficients (k) values of ~ 0.01, implying the loss of most of the organic content on decadal timescales. Silk has been used for centuries as a natural fiber, but has declined in popularity due to synthetic alternatives. In this pop-science article style paper, the history, present, and possible futures of silk are discussed at a generally accessible level. Anthropology, Biology, Sociology, and Environmental Science are brought together to present a case for a renewed interest in silk as a sustainable fiber.

Mutualistic relationships between endophytic fungi and grasses have shown to improve the hardiness of the host. This relationship is common in grasses, including native cool-season grasses that are important in both forage and grassland ecosystems. Elymus genus members, such as Canada wildrye (CAWR), commonly host the endophytic fungi, Epichloё, while southeastern wildrye (SEWR) may not. In this study, seed of ten Elymus accessions and seed and leaves from local SEWR were assessed for endophyte infection. Virtual fencing (VF) offers promising future prospects for improved grazing management as it has the potential to simplify fencing. VF lines are easily drawn and shifted via GPS coordinates. A VF collar emits an acoustic signal when the animal approaches the VF line. The signal stops immediately when the animal turns around. If the animal continues to move towards the VF line, a short electric pulse is emitted. A teaching and an operating mode are provided by the VF collars. The animals automatically change mode when they respond correctly to 20 consecutive acoustic signals without receiving an electric pulse. A …

High labour requirements for fencing and animal monitoring appear to be general obstacles for the wider use of pastureland for grazing livestock. Virtual fencing (VF) enables a less laborious pasture management. Fence lines can be easily drawn and moved using GPS data. The advantages of VF for reducing the labour inherent to controlled grazing management are obvious. Potential additional animal monitoring opportunities arising from such a VF system that uses real-time GPS data have not yet been studied. Lying is seen as an indicator for assessing comfort or restlessness of cattle. Therefore, we focus on lying behavior in this evaluation. …

This invited talk provides a perspective on what is required to excel in the role as an Extension Specialist. In the USA, most such Extension Specialists are tenure-track faculty, and have state-wide or even multi-state responsibilities. Advice is given on how to balance the high expectations of such a faculty appointment while providing appropriate recommendations to farmers/ranchers and service providers in the forage and grazinglands industry. Virtual fencing is a novel technology which uses a combination of audio and electrical stimuli to contain grazing livestock within a GPS boundary. There are however some concerns around the use of such a technology and its potential effects on the behaviour and welfare of animals. To investigate this, 64 dairy-origin calves were assigned to one of two treatments in a randomised complete block design, with 4 groups of 8 calves per treatment. Today, sustainable management of grazing livestock requires high efforts in management and fencing. Nowadays, several developments in digital technologies for herding grazing animals are arising. We conducted a systematic review on current developments in digital technologies for managing grazing animals within the landscape. We mainly focused on cattle (Bos taurus) and sheep (Ovis aries). We highlight the most promising developments of virtual fencing used in recent research to evaluate effectiveness, animal behaviour and welfare. Megathyrsus maximus is a forage species used over 30 million hectares in Brazil and in the surrounding countries for intensification of the cattle production systems. Due to the large area and distinct biomes in which it is used, there is a need to continuously develop more productive and adapted cultivars to each biome and use. Three apomictic accessions were crossed with five sexual plants at Embrapa Beef Cattle in Campo Grande, MS, Brazil in 2014.
