Background
Biologic TNF-α inhibitors (bTNFIs) can block cerebral TNF-α in Alzheimer’s disease (AD) if these macromolecules can cross the blood–brain barrier (BBB). Thus, a model bTNFI, the extracellular domain of type II TNF-α receptor (TNFR), which can bind to and sequester TNF-α, was fused with a mouse transferrin receptor antibody (TfRMAb) to enable brain delivery via BBB TfR-mediated transcytosis. Previously, we found TfRMAb-TNFR to be protective in a mouse model of amyloidosis (APP/PS1) and tauopathy (PS19), and herein we investigated its effects in mice that combine both amyloidosis and tauopathy (3xTg-AD).

The accuracy and ease of metabolite assignments from a complex mixture are expected to be facilitated by employing a multi-spectral approach. The 2D 1H-13C HSQC and 2D 1H-1H-TOCSY are the experiments commonly used for metabolite assignments. After decades of being either silenced or portrayed in a villainous perspective, LGBTQ+ individuals are beginning to see more representation on television including shows that feature drag performers. While previous shows such as RuPaul’s Drag Race and Dragula have been released for several years, We’re Here offers a new and unique approach to portrayals of drag on television by omitting the competitive aspect of the prior two shows. Analysis reveals that We’re Here highlights drag as a political tool that can both resist hegemonic standards and create community amongst participants and audience members. Firm Performance Feedback And Organizational Impression Management: The Moderating Role Of Ceo Overconfidence, Hazel H. Dadanlar, Manisha M. Vaswani, Marwan Al-Shammari, Soumendra Nath Banerjee

We propose that while positive performance feedback is positively related to firm sentiment, negative performance feedback is negatively associated with the firm sentiment. Additionally, overconfident Chief Executive Officers (CEOs) will improve the positive relationship between positive performance feedback and firm sentiment and reduce the negative relationship between negative performance feedback and firm sentiment. Using 7,182 firm-year observations for the 2004−2017 period, we show that positive performance feedback positively affects firm sentiment, and negative performance feedback negatively influences firm sentiment. We also found that higher levels of CEO overconfidence will minimize the negative impact of negative performance feedback on firm sentiment. …

For the past decade there has been an emerging disease plaguing wild snakes across the Eastern United States and Europe. In 2006, researchers started investigating the decline of Timber rattlesnake populations in New Hampshire. They discovered a fungal infection killing off the young to mid-juvenal snakes, thus know as Snake Fungal Disease or Ophidiomycosis. In 2011, San Deigo State University, identified the pathogen that causes infection, the fungus Ophidiomyces ophiodiicola. O. ophiodiicola has now affected 30 different snakes from six families within at least 20 different states since it’s discovery. This study pertains to determining the prevalence of Snake Fungal …

Zein nanoparticles (ZNP) (189.4 ± 2.0 nm, +25.7 ± 0.9 mV) and lignin nanoparticles (LNP) (173.6 ± 0.9 nm, - 56.5 ± 2.8 mV) with loaded azoxystrobin (AZO) (5.2 ± 0.8 and 5.5 ± 0.7 wt %, respectively) were designed as antifungal delivery systems for seed treatments. Both particles followed pseudo-first-order kinetics for AZO release at 25 °C, with AZO releasing faster from ZNP. AZO-entrapped ZNP treatments produced the greatest yield (41.15 bushels), followed by empty LNP (40.35 bushels) for inoculated samples; these findings were comparable to yields achieved with the commercial AZO formulation, Dynasty. Certain animal species use the Earth’s magnetic field (i.e. magnetoreception) alongside their other sensory modalities to navigate long distances that include continents and oceans. It is hypothesized that several animals use geomagnetic parameters, such as field intensity and inclination, to recognize specific locations or regions, potentially enabling migration without a pre-surveyed map. However, it is unknown how animals use geomagnetic information to generate guidance commands, or where in the world this type of strategy would maximize an animal’s fitness. To evaluate temporal trends in the prevalence of gram-negative bacteria (GNB) with difficult-to-treat resistance (DTR) in the southeastern United States. Secondary objective was to examine the use of novel β-lactams for GNB with DTR by both antimicrobial use (AU) and a novel metric of adjusted AU by microbiological burden (am-AU). Design:

Black soldier fly larvae (Hermetia illucens; BSFL) composting is biotechnology used for organic waste management and an alternative to traditional composting. We designed a two-phase experiment to evaluate the effect of BSFL composting on the emergence of the following six weed species: barnyardgrass (Echinochloa crus-galli), common ragweed (Ambrosia artemisiifolia), giant foxtail (Setaria faberi), ivyleaf morningglory (Ipomoea hederacea), redroot pigweed (Amaranthus retroflexus), and velvetleaf (Abutilon theophrasti). Teachers play a crucial role in developing intercultural competence among students in our globalized, multilingual classrooms. Over the past few decades, there has been a notable increase in the global burden of central nervous system (CNS) diseases. Despite advances in technology and therapeutic options, neurological and neurodegenerative disorders persist as significant challenges in treatment and cure. Recently, there has been a remarkable surge of interest in extracellular vesicles (EVs) as pivotal mediators of intercellular communication. As marine conservation challenges intensify with accelerating anthropogenic change, informing public deliberation about difficult trade-offs requires commitment to epistemological pluralism. Robust integration of social sciences can improve the realism of policy debates by explicating a range of potential social-ecological outcomes. Funders have long incentivized interdisciplinarity, yet progress is insufficient and embedded in a political economy of knowledge production. Failure to substantively address inequities can stymie collaboration. Institutional expectations for promotion and tenure rarely recognize the extent to which deep engagement transforms epistemological norms and scholarly outputs. Several organizations and programs offer relevant experience and resources. Senior scholars can use their …

African spiritual memory is the thread that connects members of the African diaspora, irrespective where we are or what we are doing. However, because of white supremacy, the longevity of African spiritual memory and its duration as a living, thriving, breathing force often becomes distorted under the weight of anti-black prejudice. This dissertation debunks the myth that people of African descent required outside input from non-Africans and Arabs to establish the world’s oldest and most studied civilizations, Ta-Seti and then the Egyptian Empire. Nutrient pollution in stormwater drives the eutrophication of inland and costal waterbodies which leads to sea grass retreat and the proliferation of harmful algal blooms (HAB). These anthropogenic effects destabilize ecosystems, and some HABs can pose direct human health risk. Bioretention, or the storage and controlled discharge of stormwater run-off in an ecologically engineered setting, is a potential solution to this problem. Vestibular schwannoma (VS), benign cranial nerve sheath tumors of the vestibulocochlear nerve, lack efficacious systemic therapies, especially if they develop in a NF2-related schwannomatosis (NF2) background. They cause hearing loss, tinnitus and balance problems. Metastasis associated in colon cancer 1 (MACC1) is a key driver of metastasis. Although MACC1 expression is associated with highly malignant tumors and VS are considered benign, both are attached to the HGF/MET signaling pathway and MACC1 is a candidate gene localized at a hearing loss-related gene locus. INTRODUCTION: Eosinophils contribute to the pathogenesis of allergic diseases, including asthma, allergic rhinitis, and atopic dermatitis. We previously reported that human tissue eosinophils have high CD69 expression compared to blood eosinophils, and its expression is correlated with disease severity and the number of infiltrated eosinophils. However, biological CD69 signaling activity in eosinophils remains unclear.
METHODS: CD69 expression on lung tissue eosinophils obtained from mice with ovalbumin-induced asthma was measured using flow cytometry. CD69 crosslinking was performed on eosinophils purified from the spleen of IL-5 transgenic mice to investigate CD69 signaling and its function in eosinophils. The molecular mechanisms of venetoclax-based therapy failure in patients with acute myeloid leukemia were recently clarified, but the mechanisms by which patients with myelodysplastic syndromes (MDS) acquire secondary resistance to venetoclax after an initial response remain to be elucidated. Here, we show an expansion of MDS hematopoietic stem cells (HSCs) with a granulo-monocytic-biased transcriptional differentiation state in MDS patients who initially responded to venetoclax but eventually relapsed. While MDS HSCs in an undifferentiated cellular state are sensitive to venetoclax treatment, differentiation towards a granulo-monocytic-biased transcriptional state, through the acquisition or expansion of clones with STAG2 or RUNX1 mutations, affects HSCs' …

Background/aims: We describe our findings in patients with locally advanced lacrimal sac and nasolacrimal duct (NLD) carcinoma who received neoadjuvant systemic therapy.
Methods: We identified patients with locally advanced primary lacrimal sac/NLD carcinoma treated with neoadjuvant systemic intravenous therapy at our institution during 2017-2019.
Results: The study included seven patients, four men and three women; the mean age was 60.4 years (range: 43-76). All patients had locally advanced disease with significant orbital soft tissue invasion with or without skull base invasion making eye-sparing surgery not feasible as an initial step. Some eukaryotic pre-tRNAs contain an intron that is removed by a dedicated set of enzymes. Intron-containing pre-tRNAs are cleaved by tRNA splicing endonuclease, followed by ligation of the two exons and release of the intron. Fungi use a "heal and seal" pathway that requires three distinct catalytic domains of the tRNA ligase enzyme, Trl1. In contrast, humans use a "direct ligation" pathway carried out by RTCB, an enzyme completely unrelated to Trl1. Because of these mechanistic differences, Trl1 has been proposed as a promising drug target for fungal infections. To validate Trl1 as a broad-spectrum drug target, we show that …

Collections of human remains in scientific and private institutions have a long tradition, though throughout history there has often been variable regard for the respect and dignity that these tissues demand. Recent public scandals around the use of human remains, coupled with an increasing community awareness around accountability in such instances, forces scholars to confront the ethical and moral concerns associated with these collections. Protein kinase C epsilon (PKCε) regulates behavioural responses to ethanol and plays a role in anxiety-like behaviour, but knowledge is limited on downstream substrates of PKCε that contribute to these behaviours. We recently identified brain-specific serine/threonine-protein kinase 1 (BRSK1) as a substrate of PKCε. Here, we test the hypothesis that BRSK1 mediates responses to ethanol and anxiety-like behaviours that are also PKCε dependent. We used in vitro kinase assays to further validate BRSK1 as a substrate of PKCε and used Brsk1−/− mice to assess the role of BRSK1 in ethanol- and anxiety-related behaviours and in physiological responses to ethanol. Parkinson's disease (PD) is closely linked to α-synuclein (α-syn) misfolding and accumulation in Lewy bodies. The PDZ serine protease HTRA1 degrades fibrillar tau, which is associated with Alzheimer's disease, and inactivating mutations to mitochondrial HTRA2 are implicated in PD. Here, we report that HTRA1 inhibits aggregation of α-syn as well as FUS and TDP-43, which are implicated in amyotrophic lateral sclerosis (ALS) and frontotemporal dementia. The protease domain of HTRA1 is necessary and sufficient for inhibiting aggregation, yet this activity is proteolytically-independent. Further, HTRA1 disaggregates preformed α-syn fibrils, rendering them incapable of seeding aggregation of endogenous α-syn, while reducing HTRA1 …

The study is a pre-test, post-test control group randomized quasi–experimental research design. The population comprised all SSII Biology Students in Lagos Education District II, with two intact classes of 60 students from two selected public secondary schools. Four instruments were used namely; Mastery Learning Strategy Instructional Aid, Conventional Teaching Strategy Instructional Aid, Biology Achievement Test (r = 0.77) and Biology Attitude Scale (r = 0.82). Six null hypotheses were tested using Analysis of Covariance (ANCOVA). After the many global tragedies of the early 20th century, an emerging sense of social and moral urgency catalyzed the nations of the world to attempt to develop and promulgate ethical standards for clinical research. Seminal documents such as The Nuremberg Code, The Declaration of Helsinki, and The Belmont Report attempted to bridge the chasm that allowed such tragedies. Soil pH is a foundational element of agronomy, profoundly influencing biological, chemical and physical soil processes. Adjusting soil pH is a paramount factor for enhancing soil health and productivity with far-reaching environmental benefits. Over time soil naturally tends towards acidity, a process expedited by intensive agriculture practices. To determine the extent of necessary pH corrections, precise soil pH testing is imperative. Various methods including buffer systems, titrations, lime incubations and algorithms considering soil texture are used to assess a soil's lime requirement and each method carries distinct advantages and disadvantages.
