DNA combing and DNA spreading are two central approaches for studying DNA replication fork dynamics genome-wide at single-molecule resolution by distributing labeled genomic DNA on coverslips or slides for immunodetection. Perturbations in DNA replication fork dynamics can differentially affect either leading or lagging strand synthesis, for example, in instances where replication is blocked by a lesion or obstacle on only one of the two strands. Thus, we sought to investigate whether the DNA combing and/or spreading approaches are suitable for resolving adjacent sister chromatids during DNA replication, thereby enabling the detection of DNA replication dynamics within individual nascent strands. To …

Altered apolipoprotein kinetics play a critical role in promoting dyslipidemia and atherogenesis. Human apolipoprotein kinetics have been extensively evaluated, but similar studies in mice are hampered by the lack of robust methods suitable for the small amounts of blood that can be collected at sequential time points from individual mice. We describe a targeted liquid chromatography tandem mass spectrometry method for simultaneously quantifying the stable isotope enrichment of several apolipoproteins represented by multiple peptides in serial blood samples (15 μl each) obtained after retro-orbital injection of

Senior Theses, Projects, and Awards
Silver nanoparticles (AgNPs) are widely used within commercial products due to their distinct antibacterial and antimicrobial properties. As their utilization continues to increase, their inevitable improper disposal could present lasting environmental ramifications due to their nonspecific biotoxicity. One microorganism of interest is Caulobacter crescentus, a common freshwater bacterium found in locations where AgNPs end after their disposal. C. crescentus has two distinct cell types—a stalked and a swarmer form—each with their own unique function and metabolic outputs. Approximately 15% of the C. crescentus metabolome varies with cell cycle, thus making this an interesting and environmentally relevant model organism for …

The Blurry Line Between Corporation And Cult: A Retrospective Autoethnographic Study, Ernst Graamans
The Blurry Line Between Corporation And Cult: A Retrospective Autoethnographic Study, Ernst Graamans
The Qualitative Report
In popular management literature corporations are sometimes loosely compared to cults. The comparison is a severe allegation as it implies the transgression of subordinate employees’ integrity. This paper explores to what extent such comparisons with cults are warranted as well as the implications this has for the practice of corporate culture management. On grounds of the author’s unique, first-hand experience in both corporate and cultic environments a retrospective autoethnographic (RAE) approach was chosen to further explore the supposed resemblance. The comparison is structured along Lifton’s eight criteria of thought reform and reveals that although akin to cults in all aspects …

A number of large facilities intended for the permanent sequestration of carbon dioxide are being developed in the United States. Several of them will be located in Texas and Louisiana on or near the coast of the Gulf of Mexico, making them easily accessible to ships. At the same time, there is substantial interest in Europe in installing equipment to capture carbon dioxide from certain industrial operations before it is emitted into the atmosphere, but currently there are inadequate facilities existing in Europe to sequester much of this carbon dioxide. Therefore, there is interest in the possibility of using ships …

For years, social media companies have sparred with lawmakers over how much independent access to platform data they should provide researchers. Sharing data with researchers allows the public to better understand the risks and harms associated with social media, including areas such as misinformation, child safety, and political polarization. Yet researcher access is controversial. Privacy advocates and companies raise the potential privacy threats of researchers using such data irresponsibly. In addition, social media companies raise concerns over trade secrecy: the data these companies hold and the algorithms powered by that data are secretive sources of competitive advantage. This Article shows …
Time Series Modeling To Ascertain Age In Fisheries Management, Kathleen Sue Kirch
Time Series Modeling To Ascertain Age In Fisheries Management, Kathleen Sue Kirch
OES Theses and Dissertations
The ability to assign accurate ages of fish is important to fisheries management. Accurate ageing allows for the most reliable age-based models to be used to support sustainability and maximize economic benefit. Structures used to age include bones, scales, and most commonly ear bones (otoliths). Assigning age relies on validating putative annual marks by evaluating accretional material laid down in patterns in fish otoliths, typically by marginal increment analysis. These patterns often take the shape of a sawtooth wave with an abrupt drop in accretion yearly to form an annual band and are typically validated qualitatively. Researchers have shown keen …

As one of the most famous fermented drinks in the world, beer is an especially relatable topic for microbiology courses. Here, we describe a short and easily adaptable module based on the antibacterial properties of hops used in brewing. By the 15th century, beer recipes included hops (the flower of the Humulus lupulus plant) as a bittering agent and antimicrobial. By the 19th century, the highly hopped Indian Pale Ale (IPA) became popular, and a modern myth has emerged that IPAs were invented to survive long ocean voyages such as from Britain to India. With that myth in mind, we …

Throughout history, the pursuit of diagnosing and predicting crop yields has evidenced genetics, environment, and management practices intertwined in achieving food security. However, the sensitivity of crop phenotypes and genetic responses to climate still hampers the identification of the underlying abilities of plants to adapt to climate change. We hypothesize that the PiAnosi and WagNer (PAWN) global sensitivity analysis (GSA) coupled with a genetic by environment (GxE) model built of environmental covariance and genetic markers structures, can evidence the contributions of climate on the predictability of maize yields in the U.S. and Ontario, Canada. The GSA-GxE framework estimates the relative …
