The evolutionary outcomes we observe in modern organisms, particularly associations between phenotypes and environments, have been and remain an invaluable tool in interpreting the biological phenomena that shape our world. In the following chapters, I leverage a comprehensive phylogenetic and morphological dataset for 351 species comprising a diverse group of arboreal lizards, the Anolis clade, to explore how communities and morphologies evolve. First, I characterized patterns among coexisting lineages of Anolis to reveal a general pattern of phylogenetic relatedness among lineages comprising assemblages and identify differences attributable to a biogeographic variable. The COVID-19 pandemic significantly altered the caregiving landscape, presenting distinct challenges for parents. While existing literature highlights the considerable increase in parenting stress and burnout during this period (Griffith et al., 2022), it also foregrounds the indispensable and often invisible labor of mothering¾emphasizing the disproportionate burdens placed on mothers as they bridged gaps in societal infrastructure (O’Reilly & Green, 2021). Grounded in matricentric feminist theory, this qualitative study delves into the dominant ideologies and social constructs shaping normative motherhood as a framework for understanding maternal stress and resilience during a global health crisis. Bacterial infections are a major public health concern. Bacterial infection can occur in any part of the body and can also cause the body to respond to its presence. Bacteria can be transferred to humans through water, food, air, and living vectors. The modes of transmission for bacterial infections are vectors, airborne, physical contact, and droplets. Current preventative measures have been put in place to combat this issue. The purpose of this quantitative research was to study the relationship between avoidant attachment, coping styles, and body image in adults with trichophagia and onychophagia disorders. The repeated failed attempts of patients to reduce or stop their compulsive behavior persist as a significant health concern for clinicians and researchers alike. Previous literature indicated that a lack of recognition of the underlying mental illness in these conditions exists and may be due to the absence of individualized treatment. Xanthorhiza simplicissima is a plant that is steeped in medical history. It has been used for hundreds of years by the Native Americans, and later by the Europeans, of Appalachia for its medicinal properties. One of the most useful medical applications of this plant are its antibiotic properties, as typified by its use as a tea that was consumed to treat mouth sores. These antibiotic properties can be attributed to some of its secondary alkaloids such as berberine, liriodenine, oxyacanthine, and magnoflorine. Acute myeloid leukemia (AML) is a type of blood cancer of the myeloid cell lineage. Obesity is characterized by an increase in body weight that results in excessive fat accumulation. Obesity has been associated with an increased incidence of many cancers, including blood cancers. This study evaluated the role obesity in AML progression in a novel transgenic mouse model developed by crossing Flt3ITD mice with Lepob/ob mice. Leukemia burden was augmented in obese AML mice. In addition, it was determined that obesity upregulated the ceramide-mediated and ceramide-1-phosphate-mediated NADPH oxidase 2 (NOX2). One of the fundamental requirements for a patent application is a disclosure of the invention via an accurate written description with sufficient detail to enable the recreation of the invention. The U.S. patent system has historically reviewed patent applications from various industries with a uniform set of requirements and standards. Volatile organic compounds (VOCs) and ozone (O3) are harmful pollutants present in indoor air. Indoor concentrations of VOCs are typically higher than outdoors, due to the presence of indoor sources like building materials and ozone-surface reactions. The study aims to identify and quantify the ozone reactivity and primary and secondary emissions of different indoor coatings. The first study explored secondary physical education teachers’ acceptance and use of a supplemental software application, PLT4M (“Plat-form”). The study was an exploratory cross-sectional mixed methods study guided by the Unified Theory of Acceptance and Use of Technology. Quantitative results showed that performance expectancy, attitude toward use, and price value significantly predicted PE teachers’ behavior intention to use PLT4M. Qualitative data revealed the following themes regarding teachers’ use: (1) differentiation and modifications; (2) accessibility; (3) limitations and challenges. The mechanical forces experienced during movement and the time constants of muscle activation are important determinants of the durations of behaviours, which may both be affected by size-dependent scaling. The mechanics of slow movements in small animals are dominated by elastic forces and are thus quasistatic (i.e. always near mechanical equilibrium). Muscular forces producing movement and elastic forces resisting movement should scale identically (proportional to mass2/3), leaving the scaling of the time constant of muscle activation to play a critical role in determining behavioural duration. PURPOSE: Identifying molecular and immune features to guide immune checkpoint inhibitor (ICI)-based regimens remains an unmet clinical need.
EXPERIMENTAL DESIGN: Tissue and longitudinal blood specimens from phase III trial S1400I in patients with metastatic squamous non-small cell carcinoma (SqNSCLC) treated with nivolumab monotherapy (nivo) or nivolumab plus ipilimumab (nivo+ipi) were subjected to multi-omics analyses including multiplex immunofluorescence (mIF), nCounter PanCancer Immune Profiling Panel, whole-exome sequencing, and Olink.
RESULTS: Higher immune scores from immune gene expression profiling or immune cell infiltration by mIF were associated with response to ICIs and improved survival, except regulatory T cells, which were associated with worse …

Papillary thyroid carcinoma (PTC) is the most frequent form of thyroid cancer. PTC commonly presents with mutations of the serine/threonine kinase BRAF (BRAFV600E), which drive ERK1/2 pathway activation to support growth and suppress apoptosis. PTC patients often undergo surgical resection; however, since the average age of PTC patients is under 50, adverse effects associated with prolonged maintenance therapy following total thyroidectomy are a concern. The development of mutant-selective BRAF inhibitors (BRAFi), like vemurafenib, has been efficacious in patients with metastatic melanoma, but the response rate is low for mutant BRAF PTC patients. This essay provides a metaphysical and epistemological account of gender. The metaphysics involves a ‘non-reductionist view of gender’, which is to say that gender cannot be reduced to a single factor. On this view gender begins with one’s innate gender inclinations (in the vein of Julia Serano). These inclinations pick out a specific range of gender possibilities from the gender spectrum that apply to a person. At the same time we live in communities and cultures with certain gender norms. A rare earth metal, gadolinium has unique magnetic properties and the largest neutron absorption cross section making it highly sought after in emerging technology. However, unlike most metals, it is difficult to work with and lacks a damascene process. When incorporated into alloys, gadolinium’s properties are muted and the film thickness must increase to account for the low concentration. Gadolinium metal films have been deposited successfully through sputter coating, but these films have density deficiencies and are limited to line-of-sight substrates. Attention-deficit/hyperactivity disorder (ADHD) is a common condition that affects a significant number of the world’s population, especially young adults (CDC, 2022). ADHD has its roots in behaviors of hyperactivity, inattention, and impulsiveness, and while it is new to mental health, these patterns have been observed throughout history. ADHD is a stigmatized diagnosis, and with this stigma can come low self-esteem (Mueller et al., 2012). In this study, a longitudinal regression model for covariance matrix outcomes is introduced. The proposal considers a multilevel generalized linear model for regressing covariance matrices on (time-varying) predictors. This model simultaneously identifies covariate-associated components from covariance matrices, estimates regression coefficients, and captures the within-subject variation in the covariance matrices. Optimal estimators are proposed for both low-dimensional and high-dimensional cases by maximizing the (approximated) hierarchical-likelihood function. These estimators are proved to be asymptotically consistent, where the proposed covariance matrix estimator is the most efficient under the low-dimensional case and achieves the uniformly minimum quadratic loss among all linear combinations of the …

BACKGROUND: Icariin (ICA), a natural flavonoid compound monomer, has multiple pharmacological activities. However, its effect on bone defect in the context of type 1 diabetes mellitus (T1DM) has not yet been examined.
AIM: to explore the role and potential mechanism of ICA on bone defect in the context of T1DM.
METHODS: The effects of ICA on osteogenesis and angiogenesis were evaluated by alkaline phosphatase staining, alizarin red S staining, quantitative real-time polymerase chain reaction, Western blot, and immunofluorescence. N6-Methyladenosine (m6A), a prevalent posttranscriptional modification, plays an important role in cancer progression. Clear cell renal cell carcinoma (ccRCC) is chiefly associated with the loss of the von Hippel-Lindau (VHL) gene, encoding a component of the E3 ubiquitin ligase complex. In this issue of the JCI, Zhang and colleagues unveiled a function of VHL beyond its canonical role as an E3 ubiquitin ligase in regulating hypoxia-inducible factors (HIFs). It also governed m6A modification by orchestrating the assembly of m6A writer proteins METTL3 and METTL14, thereby stabilizing PIK3R3 mRNA. Vitamin A deficiency (VAD) induced TGF-β hyperactivation and reduced expression of cell adhesion proteins in the lung, suggesting that the disruption of retinoic acid (RA) signaling leads to epithelial-mesenchymal transition (EMT). To elucidate the role of lung vitamin A status in EMT, several EMT markers and the expression of the proprotein convertase furin, which activates TGF-β, were analyzed in two experimental models. Our in vivo model included control rats, VAD rats, and both control rats and VAD rats, treated with RA. For the in vitro studies, human bronchoalveolar epithelial cells treated with RA were used. Whole-brain connectome data characterize the connections among distributed neural populations as a set of edges in a large network, and neuroscience research aims to systematically investigate associations between brain connectome and clinical or experimental conditions as covariates. A covariate is often related to a number of edges connecting multiple brain areas in an organized structure. However, in practice, neither the covariate-related edges nor the structure is known. Therefore, the understanding of underlying neural mechanisms relies on statistical methods that are capable of simultaneously identifying covariate-related connections and recognizing their network topological structures. The nuclear receptor, farnesoid X receptor (FXR/NR1H4), is increasingly recognized as a promising drug target for metabolic diseases, including nonalcoholic steatohepatitis (NASH). Protein-coding genes regulated by FXR are well known, but whether FXR also acts through regulation of long non-coding RNAs (lncRNAs), which vastly outnumber protein-coding genes, remains unknown. Utilizing RNA-seq and global run-on sequencing (GRO-seq) analyses in mouse liver, we found that FXR activation affects the expression of many RNA transcripts from chromatin regions bearing enhancer features.
