How Monitoring Food Is Protecting People, Grace Sugden May 2024

How Monitoring Food Is Protecting People, Grace Sugden

Capstone Projects and Master's Theses

While the issue of food safety has always been a concern for humanity, what that looks like has changed drastically over time. Instead of trying to judge if a stream has safe drinking water, consumers are now faced with deciding which preservatives and pesticides they expose themselves to. With the multitude of United States agencies tasked with protecting the public, standards and quality control processes should be transparent and efficient. Since the creation of the Bureau of Chemistry, numerous acts have been passed that regulate ingredients, processing procedures, and minimize contamination risks. When a spoiled product enters the food web, …

A literature review and surveys with parents and teachers indicated that the lack of parental involvement in schools contributed to students’ behaviors, lack of …

Whispers Of Thyme: Echoes Of Herbal Wisdom, Gianna Constantine May 2024

Whispers Of Thyme: Echoes Of Herbal Wisdom, Gianna Constantine

Capstone Projects and Master's Theses

Traditional herbal medicines used for centuries are now highly uncommon compared to the man made pharmaceuticals doctors have prescribed us with. Yes, they help and do what they are meant to but how come we go to these first? Why instead of peppermint or ginger tea, do we take pepto bismol? Without a thought we immediately turn to the chemically man made medicines marketed to us rather than trying something natural that has the same purpose first. We pollute the Earth that has done nothing but given us the tools to help ourselves and our discomforts, while relying on our …

Increasing Preschool Children’S Scientific Inquiry, Dianna Leon May 2024

Increasing Preschool Children’S Scientific Inquiry, Dianna Leon

Capstone Projects and Master's Theses

This capstone project focused on increasing scientific inquiry in young children. The problem that exists is that there is not enough focus on scientific learning in preschool settings compared to school settings with older age groups. According to Dejarnette (2018), young children can learn about science and are naturally curious, making them natural scientists. Science curriculum has many benefits for children, such as the development of observation, documentation, investigation, communication, and literacy skills. Science curriculum can create curiosity and enjoyment in young children while they learn about the world around them, creating a lifelong interest in learning (Larimore, 2020). Exciting …

Cholesterol is essential for both normal cell viability and cancer cell proliferation. Aberrant activity of squalene monooxygenase (SM, also known as squalene epoxidase), the rate-limiting enzyme of the committed cholesterol synthesis pathway, is accordingly implicated in a growing list of cancers. We previously reported that hypoxia triggers the truncation of SM to a constitutively active form, thus preserving sterol synthesis during oxygen shortfalls. Here, we show SM truncation is upregulated and correlates with the magnitude of hypoxia in endometrial cancer tissues, supporting the in vivo relevance of our earlier work. To further investigate the pathophysiological consequences of SM truncation, we …

Faculty, Staff and Student Publications

B-cell acute lymphoblastic leukemia (B-ALL) remains a hard-to-treat disease with a poor prognosis in adults. Mucosa-associated lymphoid tissue lymphoma translocation protein 1 (MALT1) is a para-caspase required for B-cell receptor (BCR)-mediated NF-κB activation. Inhibition of MALT1 in preclinical models has proven efficacious in many B-cell malignancies including chronic lymphocytic leukemia, mantle cell lymphoma and diffuse large B-cell lymphoma. We sought to examine the role of MALT1 in B-ALL and determine the biological consequences of its inhibition. Targeting MALT1 with both Z-VRPR-fmk and MI-2 efficiently kills B-ALL cells independent of the cell-of-origin (pro, pre, mature) or the presence of the Philadelphia …

Dlk-Mapk Signaling Coupled With Dna Damage Promotes Intrinsic Neurotoxicity Associated With Non-Mutated Tau, Sanming Li, Ethan R Roy, Yanyu Wang, Trent Watkins, Wei Cao

Faculty, Staff and Student Publications

Alzheimer's disease (AD) is the most prevalent form of neurodegeneration. Despite the well-established link between tau aggregation and clinical progression, the major pathways driven by this protein to intrinsically damage neurons are incompletely understood. To model AD-relevant neurodegeneration driven by tau, we overexpressed non-mutated human tau in primary mouse neurons and observed substantial axonal degeneration and cell death, a process accompanied by activated caspase 3. Mechanistically, we detected deformation of the nuclear envelope and increased DNA damage response in tau-expressing neurons. Gene profiling analysis further revealed significant alterations in the mitogen-activated protein kinase (MAPK) pathway; moreover, inhibitors of dual leucine …

Phosphorylation of proteins on tyrosine (Tyr) residues evolved in metazoan organisms as a mechanism of coordinating tissue growth1. Multicellular eukaryotes typically have more than 50 distinct protein Tyr kinases that catalyse the phosphorylation of thousands of Tyr residues throughout the proteome1-3. How a given Tyr kinase can phosphorylate a specific subset of proteins at unique Tyr sites is only partially understood4-7. Here we used combinatorial peptide arrays to profile the substrate sequence specificity of all human Tyr kinases. Globally, the Tyr kinases demonstrate considerable diversity in optimal patterns of residues surrounding the site of phosphorylation, revealing the functional organization of …

Cancer immunotherapy is an attractive strategy because it stimulates immune cells to target malignant cells by regulating the intrinsic activity of the immune system. However, due to lacking many immunologic markers, it remains difficult to treat glioma, a representative "cold" tumor. Herein, to wake the "hot" tumor immunity of glioma, Porphyromonas gingivalis (Pg) is customized with a coating to create an immunogenic tumor microenvironment and further prove the effect in combination with the immune checkpoint agent anti-PD-1, exhibiting elevated therapeutic efficacy. This is accomplished not by enhancing the delivery of PD-1 blockade to enhance the effect of immunotherapy, but by …

Heart disease involves irreversible myocardial injury that leads to high morbidity and mortality rates. Numerous cell-based cardiac in vitro models have been proposed as complementary approaches to non-clinical animal research. However, most of these approaches struggle to accurately replicate adult human heart conditions, such as myocardial infarction and ventricular remodeling pathology. The intricate interplay between various cell types within the adult heart, including cardiomyocytes, fibroblasts, and endothelial cells, contributes to the complexity of most heart diseases. Consequently, the mechanisms behind heart disease induction cannot be attributed to a single-cell type. Thus, the use of multi-cellular models becomes essential for creating …

Complications occurring after lymphodepleting chemotherapy (LDC) may delay chimeric antigen receptor (CAR) T-cell infusion. The effect of these delays on clinical outcomes is unclear. We performed a retrospective analysis of 240 patients with relapsed/refractory large B-cell lymphoma treated with standard-of-care axicabtagene ciloleucel (axi-cel) and identified 40 patients (16.7%) who had delay in axi-cel infusion. Of these, 85% had delay due to infection. At time of LDC initiation, patients with delayed infusion had lower absolute neutrophil count (P=0.006), lower platelets (P=0.004), lower hemoglobin (P5 days (4.6 vs. 8.2 months; P=0.036), but not 1 day (5.7 vs. 8.2 months; P=0.238). Following propensity …

Plant Model Of Α-Synucleinopathy: Expression Of Α-Synuclein A53t Variant In Hairy Root Cultures Leads To Proteostatic Stress And Dysregulation Of Iron Metabolism, Jasmina Kurepa, Kristen A. Bruce, Greg A. Gerhardt, Jan A. Smalle May 2024

Smalle

Neurology Faculty Publications

Synucleinopathies, typified by Parkinson’s disease (PD), entail the accumulation of α- synuclein (αSyn) aggregates in nerve cells. Various αSyn mutants, including the αSyn A53T variant linked to early-onset PD, increase the propensity for αSyn aggregate formation. In addition to disrupting protein homeostasis and inducing proteostatic stress, the aggregation of αSyn in PD is associated with an imbalance in iron metabolism, which increases the generation of reactive oxygen species and causes oxidative stress. This study explored the impact of αSyn A53T expression in transgenic hairy roots of four medicinal plants (Lobelia cardinalis, Artemisia annua, Salvia miltiorrhiza, and Polygonum multiflorum). In all …

Mems Optical Cavities For Low Power Communication., Jacob Schopp May 2024

Mems Optical Cavities For Low Power Communication., Jacob Schopp

Electronic Theses and Dissertations

This thesis discusses the use of MEMS (microelectromechanical systems) optical cavities for use in low power communication. MEMS optical cavities are thin air gaps between an actuated MEMS mirror and a fixed glass encapsulation. By moving the MEMS mirror the reflectivity of the MEMS optical cavity can be changed. This change in reflectivity can be used to transmit information passively by modulating reflected light intensity instead of generating new light using a light source. These MEMS optical cavities are demonstrated using optical fibers for illumination delivery and reflected light collection. In addition to being reflected for communication, the illuminating light …

Electronic Theses and Dissertations

The continuous pursuit of effective methods for gene transfection in primary human T cells is crucial for advancing cell-based therapies. Viral vectors, currently used in cell-based therapies, involve a lengthy and costly process. This thesis explores the efficacy of coupling acoustofluidic and sonoporation techniques as an alternative non-viral gene transfection approach with primary T cells. Sonoporation, in conjunction with microbubbles, has been demonstrated to induce cavitation, resulting in the formation of pores in the cell membrane through which molecules can enter. Using flow cytometry, we compared the loading efficiency of primary T cells with that of Jurkat T cells to …

An Investigation Of Protein Arginine Methyltransferase 1 Substrate Recognition Through Ribonucleoprotein Modifications, Isabella Beverly Lonardo

Undergraduate Honors Capstone Projects

Just as humans need to communicate with each other, our cells have a series of processes that allows them to communicate from one to another. Protein Arginine Methyltransferase I is an enzyme that helps mediate cellular communication by selectively binding to target proteins and performing chemistry that has broad-reaching downstream effects. However, the mechanisms that underly PRMT1 selectivity have yet to be elucidated. This study uses alterations to a series of known targets, called ribonucleoproteins, to identify some of the characteristics that govern recognition by the PRMT1 enzyme. I have made significant progress in the creation, expression, and purification of …
