Background: Globally, approximately 50 million couples experience one form of infertility, and 10 million cases of subfertility have been reported in sub-Saharan Africa. Infertility is characterized by a lack of clinical conception among couples who live together for more than one year with regular coitus, without the use of contraception. Factors related to fertility vary by sex and geographical region. These factors include age, lifestyle, infectious diseases, and genetic disorders. In African culture, children are considered a simple inheritance and a measure of masculinity, so efforts are needed to address the growing problem of male infertility in this context. …

The information-containing nature of the DNA molecule has been long known and observed. One technique for quantifying the relationships existing within the information contained in DNA sequences is an entity from information theory known as the average mutual information (AMI) profile. This investigation sought to use principally the AMI profile along with a few other metrics to explore the structure of the information contained in DNA sequences.
Treating DNA sequences as an information source, several computational methods were employed to model their information structure. Maximum likelihood and maximum a posteriori estimators were used to predict missing bases in DNA sequences. …

The stigmatisation of queer identities is a constant in cinematic history, especially so in horrorgenre movies. A transgressive and confronting medium, horror cinema presents numerous opportunities to align queer identities with cultural symbols of degeneracy; perpetuating associations that contribute to negative cultural attitudes towards queerness. While public discourse on such topics has advanced to entertain ideas of equity and authenticity in these representations, negative values and attitudes are yet evident in contemporary cultural texts. Existing network analysis methods struggle to optimize observer placements in dynamic environments with limited visibility. This dissertation introduces the novel ROBUST (Ranged Observer Bipartite-Unipartite SpatioTemporal) framework, offering a significant advancement in modeling, analyzing, and optimizing observer networks within complex spatiotemporal domains. ROBUST leverages a unique bipartite-unipartite approach, distinguishing between observer and observable entities while incorporating spatial constraints and temporal dynamics.

Bayou St. John is a degraded waterway located within the City of New Orleans. The Bayou, which is disconnected from Lake Pontchartrain by floodgates, has undergone restoration efforts to improve connectivity through frequent floodgate openings and marsh habitat restoration outside of the floodgates. To assess possible responses of local fish assemblages to these restoration efforts, I analyzed long-term fish assemblage data from three sites located inside of the floodgates and one site located at the restored marsh outside of the floodgates. This paper describes the creation and development of an implementation of the NeuroEvolution of Augmenting Topologies (NEAT) architecture to train an agent to play Super Mario Brothers. Building off of a basic implementation of NEAT, this thesis project shows the process of refining the fitness calculation that ranks the networks in the population and also defines the creation and application of a dataset to train the agent. This study investigates the influence of specific frameworks in performance and vocal pedagogy on artists' capacity to enact evidence-based learning outcomes, with emphasis on vocal physiology and educator-performer relationships. Employing an experimental design, five philosophies that support capacity-building were tested across two trial settings, varying in intensity. The participant utilized narrative reasoning, diversified methods, and mentorship from specialized educators to apply frameworks. Findings indicate that, in practicing evidence-based learning and maximizing anatomic foundations of the voice, substantial developments in artistic capacity ensued. Educator-performer relationships mediate this process through reciprocal collaborations that foster capacity-building. Performers display optimal artistic capacity by effectively …

This thesis describes the design, implementation, and testing of a novel procedural narrative system called the Procedurally Adaptive Webbed Narrative (PAWN) system. PAWN procedurally generates characters and, responding to choices made by the player, produces more responsive characters and relationships involving the player and these narrative agents. Initially, this thesis discusses other interactive narrative types that exist, such as emergent or event-driven narratives, along with their strengths and weaknesses. It then examines each aspect of PAWN, starting with initial actor generation, then moving to the capturing of game events and translating them into logical objects called Occurrences. The community of Slidell, Louisiana, sits across the lake from New Orleans on the north shore of Lake Pontchartrain. Slidell quietly shrouds a multitude of fascinating historic narratives with roots that extend back to before the 1718 founding of New Orleans. The first documented European presence in the area near Slidell occurred in 1699 when Iberville camped at Goose Point a few miles west of present-day Slidell. This area would play a vital role in the survival, growth, and development of the early New Orleans settlement.

In this thesis, petroleum-contaminated groundwater from three spill-affected Minnesota sites—two contaminated with crude oil; one with diesel—is investigated to elucidate whether differences in degradation products from different parent oils exist. After spill events, parent molecules from the leaked oil seep underground and undergo microbial degradation over time, transforming into hydrocarbon oxidation products (HOPs), water-soluble contaminants that infiltrate groundwater aquifers, flowing into nearby environments and towns while exhibiting environmental persistence, toxicity to aquatic biota, and potential risks to humans that become exposed. Human studies of early brain development have been limited by extant neuroimaging methods. MRI scanners present logistical challenges for imaging young children, while alternative modalities like functional near-infrared spectroscopy have traditionally been limited by image quality due to sparse sampling. In addition, conventional tasks for brain mapping elicit low task engagement, high head motion, and considerable participant attrition in pediatric populations. BACKGROUND: Influenza, respiratory syncytial virus (RSV), and human metapneumovirus (hMPV) are common respiratory viruses causing similar symptoms. Optimal tools to assess illness severity for these viruses have not been defined. Using the Hospitalized Acute Respiratory Tract Infection (HARTI) study data, we report symptom severity by clinician-rated clinical severity scores (CSS) in adults with influenza, RSV, or hMPV and correlations between CSS and patient-reported outcomes (PROs).

Inorganic mercury (Hg) is converted to potential neurotoxic methylmercury (MeHg) by a natural process called Hg methylation. MeHg can be biomagnified in the food chain, thus the consumption of Hg-contaminated fish contributes to harmful human health issues. Selenium (Se) inhibits the Hg bioavailability to the methylating bacteria by forming mercuric selenide (HgSe). Pseudodesulfovibrio mercurii, a type of sulfate-reducing bacteria (SRB) cultures were grown in anaerobic environments using an estuarine sulfate lactate growth medium to evaluate the effects of Hg concentrations, bacterial growth phase, and sodium selenate in the Hg methylation process. Exploration of the effects of maternal Adverse Childhood Experiences (ACEs) on child externalizing behaviors were conducted in the proposed study. Subtypes of ACEs, such as household dysfunction and maltreatment, and their effects on child externalizing behaviors were also examined. Maternal ACEs have shown to have negative effects on their children through use of more negative parenting practices (NPP). However, the role of maternal difficulty in emotion regulation in the link between maternal ACEs, NPP, and child externalizing behaviors has not been previously shown in the literature. This project focuses on the importance of the relationship between Fe-Ti oxide abundance and interconnectivity, and crystal-plastic deformation in Atlantis Bank, Southwest Indian Ridge by utilizing Micro-CT scanning. This was done through comparing the Micro-CT scans of 15 samples from three different Ocean Drilling Program and International Ocean Discovery Program drill holes using the Nikon NIS Advanced Research 3-D visualization program and producing plots and statistics from MATLAB. Results indicate a wide range of textures across a range of crystal-plastic fabric intensity and Fe-Ti oxide abundance. Saudi Arabia was heavily dependent on oil, which caused an economic crisis due to the collapse of global oil prices. In 2016, the Saudi government launched Vision 2030 to diversify the Saudi economy and avoid dependence on oil revenues. One aim of Vision 2030 is to initiate numerous tourism initiatives to boost the tourism industry's economic impact, elevating its contribution from 3% of GDP to 10% by 2030.

The role of polyunsaturated fatty acid (PUFA) biosynthesis in acute myeloid leukemia (AML) remains largely undefined. A comparative expression analysis of 35 genes encoding fatty acid biosynthesis enzymes showed that fatty acid desaturase 1 (FADS1) was highly expressed across multiple AML subtypes relative to healthy controls and that elevated FADS1 expression correlates with worse overall AML patient survival. Functionally, shRNA-mediated inhibition of FADS1 reduced AML cell growth in vitro and significantly delayed leukemia onset in an AML mouse model. Today, more than 90% of people with cystic fibrosis (pwCF) are eligible for the highly effective cystic fibrosis transmembrane conductance regulator (CFTR) modulator therapy called elexacaftor/tezacaftor/ivacaftor (ETI) and its use is widespread. Given the drastic respiratory symptom improvement experienced by many post-ETI, clinical studies are already underway to reduce the number of respiratory therapies, including antibiotic regimens, that pwCF historically relied on to combat lung disease progression. Early studies suggest that bacterial burden in the lungs is reduced post-ETI, yet it is unknown how chronic Pseudomonas aeruginosa populations are impacted by ETI. IMPORTANCE: Diabetes is associated with poorer prognosis of patients with breast cancer. The association between diabetes and adjuvant therapies for breast cancer remains uncertain.
OBJECTIVE: To comprehensively examine the associations of preexisting diabetes with radiotherapy, chemotherapy, and endocrine therapy in low-income women with breast cancer.
DESIGN, SETTING, AND PARTICIPANTS: This population-based cohort study included women younger than 65 years diagnosed with nonmetastatic breast cancer from 2007 through 2015, followed up through 2016, continuously enrolled in Medicaid, and identified from the linked Missouri Cancer Registry and Medicaid claims data set. IMPORTANCE: Early-onset colorectal cancer (EOCRC), defined as a diagnosis at younger than age 50 years, is increasing, and so-called red flag signs and symptoms among these individuals are often missed, leading to diagnostic delays. Improved recognition of presenting signs and symptoms associated with EOCRC could facilitate more timely diagnosis and impact clinical outcomes.
OBJECTIVE: To report the frequency of presenting red flag signs and symptoms among individuals with EOCRC, to examine their association with EOCRC risk, and to measure variation in time to diagnosis from sign or symptom presentation.

Careful regulation of the complement system is critical for enabling complement proteins to titrate immune defense while also preventing collateral tissue damage from poorly controlled inflammation. In the eye, this balance between complement activity and inhibition is crucial, as a low level of basal complement activity is necessary to support ocular immune privilege, a prerequisite for maintaining vision. Dysregulated complement activation contributes to parainflammation, a low level of inflammation triggered by cellular damage that functions to reestablish homeostasis, or outright inflammation that disrupts the visual axis. Astrocytes are a ubiquitous and enigmatic type of non-neuronal cell and are found in the brain of all vertebrates. While traditionally viewed as being supportive of neurons, it is increasingly recognized that astrocytes play a more direct and active role in brain function and neural computation. BACKGROUND: Neuroimaging studies often quantify tau burden in standardized brain regions to assess Alzheimer disease (AD) progression. However, this method ignores another key biological process in which tau spreads to additional brain regions. Understanding the genetic basis of cortical bone traits can allow for the discovery of novel genes or biological pathways regulating bone health. Mice are the most widely used mammalian model for skeletal biology and allow for the quantification of traits that cannot easily be evaluated in humans, such as osteocyte lacunar morphology. PURPOSE: Histone 3 (H3) K27M-mutant diffuse midline glioma (DMG) has a dismal prognosis with no established effective therapy beyond radiation. This integrated analysis evaluated single-agent ONC201 (dordaviprone), a first-in-class imipridone, in recurrent H3 K27M-mutant DMG.
METHODS: Fifty patients (pediatric, n = 4; adult, n = 46) with recurrent H3 K27M-mutant DMG who received oral ONC201 monotherapy in four clinical trials or one expanded access protocol were included.
