In recent years, the demand for high-performance micro and nanodevices has surged, necessitating the exploration of novel dielectric materials to replace conventional silicon dioxide. Following the continuation of the Moorse law, as device dimensions reduce to nanoscale levels, the properties of silicon dioxide can degrade, leading to issues such as increased leakage current and reduced gate control. Early human embryogenesis is difficult to study due to practical and ethical limitations, but the successful culture of “naïve” human pluripotent stem cells (hPSCs) has expanded our ability to model cells akin to the pre-implantation epiblast. Naïve hPSCs are readily obtained through in vitro resetting of “primed” hPSCs, cells which model the post-implantation epiblast. Naïve hPSCs offer the advantages of modeling the pre-implantation transcriptome and epigenome, the ability to form embryonic and extraembryonic tissues, and the capacity to self-organize into integrated human embryo models. However, a persistent drawback of naïve hPSCs is the loss of parent-specific epigenetic marks, called imprints. …

Chimeric Antigen Receptor T-cell (CAR-T) therapy has transformed the treatment landscape for hematological malignancies, showing high efficacy in patients with relapsed or refractory (R/R) disease and otherwise poor prognosis in the pre-CAR-T era. These therapies have been usually administered in the inpatient setting due to the risk of cytokine release syndrome (CRS) and immune effector cell-associated neurotoxicity syndrome (ICANS). This dissertation interrogates the use of poetic fragmentation as a tool of ecological representation and coins the term “ecotonal poetics.” Ecotonal poesis, this dissertation explains, is the stretching of lyric across the field of the page, using space, sound, language, and parataxis in its attempt to not only represent but create ecotonal ecologies through text, sound, and visual cues. This dissertation puts forth four case studies of contemporary American women poets turning the space of the page into an ecotone where text, white space, author, reader, body, and environment meet. The following paper provides insight into the Flint, Michigan water crisis of 2014 through the lens of environmental racism. The Flint, Michigan water crisis saw the contamination of the municipal water supply of the town of Flint as a result of the decision to switch the main water source from Lake Huron to the Flint River as a temporary measure as the town of Flint switched to the Karegnondi Water Authority contract to supply their municipal water. Autism Spectrum Disorders (ASDs) are neurodevelopmental disorders (NDDs) in which children display differences in social interaction/communication and repetitive stereotyped behaviors along with variable associated features. Cul3, a gene linked to ASD, encodes CUL3 (CULLIN-3), a protein that serves as a key component of a ubiquitin ligase complex with unclear function in neurons. Cul3 homozygous deletion in mice is embryonic lethal; thus, we examine the role of Cul3 deletion in early synapse development and neuronal morphology in hippocampal primary neuronal cultures. Mutations in the LMNA gene encoding lamins A/C cause an array of tissue-selective diseases, with the heart being the most commonly affected organ. Despite progress in understanding the perturbations emanating from LMNA mutations, an integrative understanding of the pathogenesis underlying cardiac dysfunction remains elusive. Using a novel conditional deletion model capable of translatome profiling, we observed that cardiomyocyte-specific Lmna deletion in adult mice led to rapid cardiomyopathy with pathological remodeling. Before cardiac dysfunction, Lmna-deleted cardiomyocytes displayed nuclear abnormalities, Golgi dilation/fragmentation, and CREB3-mediated stress activation. Translatome profiling identified MED25 activation, a transcriptional cofactor that regulates Golgi stress. Autophagy is disrupted …

Hydrocephalus is characterized by the abnormal accumulation of cerebrospinal fluid (CSF) within the brain ventricles. In post-infectious hydrocephalus (PIH) cases, the condition presents challenges in understanding the immune response. PIH is a complex condition, often persisting after the initial infection is treated and thus requiring a deeper understanding of the immune mechanisms involved in its development. Most research on the probability of winning a tennis match is based on the assumption that the points are independent and identically distributed, treating each point as a Bernoulli trial with fixed probability of success. This assumption, however, seems to contradict experience. Players' performance appears to fluctuate as the match progresses due to the psychological effect of past performance. To test this counterintuitive yet central assumption, previous research has attempted to test the independence hypothesis. The use of wetlands as a treatment approach for nitrogen in runoff is a common practice in agroecosystems. However, nitrate is not the sole constituent present in agricultural runoff and other biologically active contaminants have the potential to affect nitrate removal efficiency. In this study, the impacts of the combined effects of four common veterinary antibiotics (chlortetracycline, sulfamethazine, lincomycin, monensin) on nitrate-N treatment efficiency in saturated sediments and wetlands were evaluated in a coupled microcosm/mesocosm scale experiment. Background:

Pathogenesis of seborrheic dermatitis involves lipid secretion by sebaceous glands, Malassezia colonization, and an inflammatory response with skin barrier disruption. Each of these pathways could be modulated by diet, obesity, and nutritional supplements. Current treatment options provide only temporary control of the condition; thus, it is essential to recognize modifiable lifestyle factors that may play a role in determining disease severity.

Objective:

This study aimed to summarize published evidence on diet, nutritional supplements, alcohol, obesity, and micronutrients in patients with seborrheic dermatitis and to provide useful insights into areas of further research.

The inability of adult mammalian cardiomyocytes (CMs) to undergo cell renewal following ischemic injury stands as a major impediment to heart function recovery and contributes to heart failure (HF). Current therapeutic approaches do not offer an effective approach of reactivating CM cell division to prevent HF in humans. Fungal resistance to commonly used medicines is a growing public health threat, and there is a dire need to develop new classes of antifungals. We previously described a peptide produced by Enterococcus faecalis, EntV, that restricts Candida albicans to a benign form rather than having direct fungicidal activity. Moreover, we showed that one 12-amino acid (aa) alpha helix of this peptide retained full activity, with partial activity down to the 10aa alpha helix. In this thesis, we improved an existing structured light methodology to more accurately estimate tissue optical properties imaged in a single snapshot. The developed approach is a major enhancement of the traditional three-phase spatial frequency domain imaging system: It greatly simplifies the operational procedure while also maintaining high accuracy in optical property measurements .

Leveraging this improved approach, we have developed a low-cost, compact, and radial-view endoscope prototype. Metal (oxyhydr)oxide surfaces regulate the accessibility of trace metals in soils and aquatic systems. Up to half of the trace metals in urban environments, river systems, and contaminated soils are associated with mineral surfaces yet this pool often remains bioaccessible. Past studies of adsorption-desorption suggest that macroscopic rates of adsorption at mineral surfaces are likely mass transfer-limited and show biphasic rates, with a fast-adsorbing fraction equilibrating in seconds to minutes and a slower-adsorbing pool binding over a timescale of hours to days. In addition, adsorption-desorption hysteresis has been observed for some metals after aging at metal (oxyhydr)oxide surfaces, indicating that …

Classical dendritic cells (cDCs) are specialized antigen presenting cells that comprise distinct subsets distinguished by transcriptional developmental differences and in the types of immune responses they initiate. Classical type 1 dendritic cells (cDC1) are thought to perform antigen cross-presentation, which is required to prime CD8 T cells, whereas cDC2 have been suggested to be specialized for priming CD4 T cells. Some of the earliest studies of glycans were performed on mammalian corneas and lenses with many of the key concepts we currently recognize as being fundamental to our understanding of basic cell biology arising from these studies. Proteoglycans and their GAG side chains are essential components of the ECM of the lens capsule. They also are present in the anterior corneal epithelial basement membrane and the posterior (Decemet's) basement membrane, and they organize collagen fiber diameters and spacing in the corneal stroma to maintain stromal clarity. PURPOSE: This study explored employee health behavior changes and health care utilization after workplace genetic testing (wGT). Wellness-program-associated wGT seeks to improve employee health, but the related health implications are unknown.

METHODS: Employees of a large US health care system offering wGT (cancer, heart disease, and pharmacogenomics [PGx]) were sent electronic surveys. Self-reported data from those who received test results were analyzed. Descriptive statistics characterized responses, whereas logistic regression analyses explored correlates of responses to wGT.

RESULTS: 53.9% (n = 418/776) of respondents (88.3% female, mean age = 44 years) reported receiving wGT results. 12.0% (n = 48/399) received results …

Human T-cell leukemia virus type-1 (HTLV-1) is an oncogenic retrovirus that causes multiple disorders, including adult T-cell leukemia and HTLV-1-associated myelopathy/tropical spastic paraparesis. HTLV-1 retroviral integrase binds to the regulatory B’56γ subunit of the host cell Protein Phosphatase 2A (PP2A). Integrase contains a highly conserved LxxIxE motif that is essential for binding, which increases integration efficiency and facilitates HTLV-1 hijack of host cell machinery. We aim to understand how mutations introduced in the highly conserved binding site can affect viral particle production and infectivity. Diabetic foot ulcers (DFUs) significantly affect the lives of patients and increase the risk of hospital stays and amputation. We suggest a remote monitoring platform for better DFU care. This system uses digital health metrics (scaled from 0 to 10, where higher scores indicate a greater risk of slow healing) to provide a comprehensive overview through a visual interface. The platform features smart offloading devices that capture behavioral metrics such as offloading adherence, daily steps, and cadence. Childhood-onset essential hypertension (COEH) is an uncommon form of hypertension that manifests in childhood or adolescence and, in the United States, disproportionately affects children of African ancestry. The etiology of COEH is unknown, but its childhood onset, low prevalence, high heritability, and skewed ancestral demography suggest the potential to identify rare genetic variation segregating in a Mendelian manner among affected individuals and thereby implicate genes important to disease pathogenesis. However, no COEH genes have been reported to date. This thesis explores the fluid and often elusive concept of the body as mediated through technology and art, questioning the boundaries between the physical and virtual. By investigating the interactions of cultural ideals, technological mediation, and material experimentation, the research delves into how contemporary art practices can challenge and expand our understanding of embodiment.

Central to this exploration is the use of varied mediums such as sculpture, digital imagery, and installation art to create what I term "virtual bodies"—conceptual entities that exist at the intersection of imagination and material reality.
