Trichotillomania (TTM) is associated with impairments in response inhibition and cognitive flexibility, but it is unclear how such impairments relate to treatment outcome. The present study examined pre-treatment response inhibition and cognitive flexibility as predictors of treatment outcome, change in these domains from pre- to post-treatment, and associations with TTM severity. Participants were drawn from a randomized controlled trial comparing acceptance-enhanced behavior therapy (AEBT) to psychoeducation and supportive therapy (PST) for TTM. Adults completed assessments at pre-treatment (n=88) and following 12 weeks of treatment (n=68). Response inhibition and cognitive flexibility were assessed using the Stop Signal …

Can Beggars Be Choosers? Adaptive Variation In Reproductive Investment Strategies Shapes Family Conflict And Cooperation In Wild Songbirds, Kelly Lynn O'Neil
Electronic Theses and Dissertations Archive
In this dissertation, I investigate the adaptive function of behavioral plasticity related to reproductive strategies in wild songbirds. Specifically, I uncover how various social, physiological, and environmental factors shape dynamics of conflict and cooperation between parents and offspring and among siblings. I performed 2 multi-year field experiments in which I cross-fostered nestlings between broods of wild songbirds, manipulating nestling size asymmetries (in Carolina wrens) and brood size (in prothonotary warblers) while including relevant controls. I observed nests during two developmental stages and used an intricate scoring protocol to quantify nestling begging dynamics, parental feeding decisions, and the distribution of food …

- The advent of 3-Dimensional (3D) printing has provided orthopaedic surgeons the ability to design custom, patient-specific implants for a variety of different complex problems notably, severe bone loss.
- Rheumatoid arthritis (RA) is a debilitating, inflammatory, systemic disease affecting approximately 1.3 million adults in the United States (US).1 Orthopaedic manifestations are plentiful. Issues unique to the shoulder are: central glenoid wear, periarticular erosions and osteopenia, and subchondral cysts. Disease modifying antirheumatic drugs can be preventative but do not restore the erosive articular damage already done.
- Glenosphere fixation is critical for reverse total shoulder arthroplasty (rTSA) implant stability and longevity. Diminished bone …
An Exploration Of The Potential Impacts Of Philosophical Idealism On Psychotherapeutic Practice, Ian Richard
An Exploration Of The Potential Impacts Of Philosophical Idealism On Psychotherapeutic Practice, Ian Richard
Educational Specialist, 2020-current
This study presents a review of the modern ontologies available regarding the nature of the universe and identifies Bernardo Kastrup’s formulation of idealism as a leading model. Idealism theorizes that consciousness is the basis of reality, as opposed to physical objects. The implications of Kastrup’s idealism on psychotherapeutic practice are considered, including conflicts with the medical model, synergies with the contextual model, and suggestions for approaches that align with the findings of idealism.
Counselor Educator Attitudes And Beliefs Toward Psychedelics And Pat, Kimberly Hughes
Counselor Educator Attitudes And Beliefs Toward Psychedelics And Pat, Kimberly Hughes
Educational Specialist, 2020-current
The psychedelic renaissance of the past two decades has introduced a new era of acceptability of and curiosity in historically maligned psychedelic compounds. With increasing positive evidence-based support, psychedelics and psychedelic-assisted therapies (PAT) are being embraced as legitimate psychotherapeutic mechanisms. The uptick in interest, acceptability, and use means that encountering psychedelics in the counseling room is not a question of if but when. As leaders within the field and educators of future generations of clinicians, counselor educators are uniquely poised to influence the field’s relationship with psychedelics. While no current studies have explicitly examined the attitudes and beliefs of counselor …

Losing It: Anti-Fat Bias And Implications For Counselors, Kate Bailey
Educational Specialist, 2020-current
Anti-fat bias and stigma can be pervasive in the United States and can impact almost every aspect of culture in the U.S. Counselors are likely not exempt from holding anti-fat bias. Yet, anti-fat bias is often overlooked as an issue worthy of attention in a profession dedicated to advocacy. Anti-fat bias can negatively affect larger-bodied clients in the counselor/client relationship when counselors are unaware of the anti-fat bias that they hold. Using a framework of liberatory consciousness (awareness, analysis, action, and accountability) counselors may reduce the harm caused in the counselor/client relationship regarding anti-fat bias.

Let’S Get Creative: Using Creative Arts Interventions To Address Trauma Experiences, Ashley Oginz-Wilson
Educational Specialist, 2020-current
Trauma as a theme in counseling work is something that mental health clinicians are commonly exposed to. While there are standard practices and therapeutic modalities to address trauma in counseling, it is important to maintain flexibility as a clinician to best meet the individual needs of the client. This literature review explores ways to embrace creative counseling interventions in support of clients with trauma experiences. Creative counseling interventions could include art, music, dance and movement, nature, humor, drama, and play. The premise of such interventions is that they offer clients a new way to express their feelings, memories, stories, etc. …

Quantitative tracking of rapidly moving micron-scale objects remains an elusive challenge in microscopy due to low signal-to-noise. This paper describes a novel method for tracking micron-sized motile organisms in off-axis Digital Holographic Microscope (DHM) raw holograms and/or reconstructions. We begin by processing the microscopic images with the previously reported Holographic Examination for Life-like Motility (HELM) software, which provides a variety of tracking outputs including motion history images (MHIs). MHIs are stills of videos where the frame-to-frame changes are indicated with color time-coding. This exposes tracks of objects that are difficult to identify in individual frames at a low signal-to-noise ratio. …

Although research has been conducted on the lived experiences of Black student affairs professionals and academic advisors at Historically White Institutions (HWIs), the literature is scant in its representation of the larger body of Black Professional Staff (BPS) and their experiences. BPS are undervalued and disregarded and are a vital part of the fabric of HWIs. Drawing on Critical Race Theory in Education, anti-Black racism, and plantation politics, the purpose of this critical race methodology study was to describe how Black Professional Staff experienced and navigated racism at an HWI in the context of the Black Lives Matter movement (BLM). …

Electronic Theses and Dissertations Archive
This study examines the conversion of carotenoids to ketocarotenoids, essential for bright plumage in birds, a trait crucial in sexual selection. We used DF-1 avian cells genetically engineered to express BDH1L, CYP2J19, and TTC39B, key enzymes in this conversion. The genetic modifications and enzyme activity were verified via PCR genotyping and RT-qPCR. These cells were treated with yellow carotenoids (zeaxanthin, β-carotene, lutein) and their conversion to red ketocarotenoids (astaxanthin, canthaxanthin, alpha-doradexanthin) was analyzed using High- Performance Liquid Chromatography (HPLC). The study also assessed the impact of antioxidant treatments and mitochondrial uncouplers on this process. Results indicated that the inclusion of …
Methods For The Analysis Of Diffusible Signaling Factors For The Determination Of Elution Activity, Structural Stability And Biological Activity, Brian Hoffman
Electronic Theses and Dissertations Archive
Biofilms pose a threat to public health. The National Institutes of Health estimate that biofilms are linked to 65-80% of hospital-related infections. Biofilms are a complex network of bacterial cells contained within an extracellular matrix and are 10-1000 times more resistant to common antibiotic treatments. Bacteria within a biofilm utilize a form of intercellular communication known as quorum sensing to regulate many cellular functions including the secretion of the extracellular matrix and dispersal of planktonic cells. Compounds known as diffusible signaling factors (DSF) regulate the formation/degradation of the extracellular matrix. One family of DSF consist of medium chain fatty acid …
Vascular Electrophysiology And Pathogenic Consequences Of Cardiovascular Katp Channel Mutations, Alex Hanson
Vascular Electrophysiology And Pathogenic Consequences Of Cardiovascular Katp Channel Mutations, Alex Hanson
Arts & Sciences Graduate Student Theses and Dissertations
The complex cardiovascular disorder Cantú Syndrome (CS) arises from gain-of-function (GoF) mutations in either KCNJ8 or ABCC9, the genes encoding the Kir6.1 and SUR2 subunits of cardiovascular ATP-sensitive potassium (KATP) channels, respectively. CS involves an array of cardiovascular pathologies, including cardiac hypertrophy and hypercontractility, low systemic vascular resistance, and excessively compliant, dilated, and tortuous vessels. Together, the latter features exemplify the hypomyotonic and hyperelastic components of CS vasculopathy. It was recently established that CS vasculopathy drives the associated cardiac pathologies, which are observed even in the absence of cardiac KATP GoF. In this thesis, I carried out experiments to determine …

Inducible Crispr-Targeted ‘Knockdown’ Of Human Gut Bacteroides In Mice Reveals Glycan Utilization Strategies, Zachary Beller
Inducible Crispr-Targeted ‘Knockdown’ Of Human Gut Bacteroides In Mice Reveals Glycan Utilization Strategies, Zachary Beller
Arts & Sciences Graduate Student Theses and Dissertations
Learning how members of the human gut microbiota compete and cooperate for dietary nutrients, such as fiber-associated glycans, should help facilitate development of more precise nutritional recommendations for improving health. To model how bacteria prioritize the utilization of different fiber glycans in the mammalian gut, germ-free mice were colonized with a 13-member consortium of cultured, genome-sequenced human bacterial strains, including seven Bacteroides species. Animals were fed a Western diet supplemented with pea fiber. Once the consortium had time to assemble, an inducible CRISPR-based system was used to deplete the absolute abundance of Bacteroides thetaiotaomicron or B. cellulosilyticus by 10- to …
Impact Of Parent Reported Adverse Experiences And Other Family Stressors On Child Development And Home Visitation Participation Among Immigrant Families, Fithi Andom
Brown School Graduate Student Theses and Dissertations
This dissertation examines variations in early childhood developmental and socioemotional health as well as level of participation in home visitation among recent immigrant families according to reports of adverse experiences (including poverty, intimate partner violence (IPV), and prior history of child protective services (CPS) involvement), maternal depression, and parent–child relationship measures. The period of early childhood is dependent on positive parenting to support optimum development, but parenting may be particularly sensitive to the detrimental influences of adverse experiences. Less is known about how the relationship between parenting, adverse experiences, and child development may differ for immigrant families. Home visitation has …

Αb-Crystallin Phosphorylation Induces A Condensatopathy To Worsen Post-Myocardial Infarction Cardiomyopathy, Md Moydul Islam
Arts & Sciences Graduate Student Theses and Dissertations
Rare genetic mutations in proteins expressed in cardiomyocytes lead to proteotoxic cardiomyopathies which are characterized by widespread protein-aggregate pathology. However, the mechanistic link between proteotoxicity and protein-aggregates in the more common ischemic cardiomyopathy remains unclear. Our study revealed the mis-localization of desmin, actin, and α-actinin to protein-aggregates in the myocardium of patients with ischemic cardiomyopathy and in mouse hearts undergoing post-myocardial infarction ventricular remodeling. These findings mimic observations in autosomal-dominant cardiomyopathy resulting from the R120G mutation in the chaperone protein CRYAB where desmin is mis-localized into the aggregates. Importantly, we observed increased phosphorylation of CRYAB at serine-59 (pS59-CRYAB) and its …

Environmental air irritants including nanosized carbon black (nCB) can drive systemic inflammation, promoting chronic obstructive pulmonary disease (COPD) and emphysema development. The let-7 microRNA (Mirlet7 miRNA) family is associated with IL-17-driven T cell inflammation, a canonical signature of lung inflammation. Recent evidence suggests the Mirlet7 family is downregulated in patients with COPD, however, whether this repression conveys a functional consequence on emphysema pathology has not been elucidated. Here, we show that overall expression of the Mirlet7 clusters, Mirlet7b/Mirlet7c2 and Mirlet7a1/Mirlet7f1/Mirlet7d, are reduced in the lungs and T cells of smokers with emphysema as well as in mice with …

BACKGROUND: Pain and dysfunction persist for most patients following hip-related pain treatment. Additionally, individuals with hip-related pain are typically less physically active than individuals without hip pain, despite evidence that regular physical activity reduces chronic musculoskeletal pain. Poor psychological health is common in patients with hip-related pain and further reinforces low physical activity. Mind-body interventions can improve psychological health and activity levels but have yet to be integrated to provide comprehensive, psychologically informed care for patients with hip-related pain. Thus, we are using the NCCIH intervention development framework to develop Helping Improve PSychological Health (HIPS), a novel, multimodal mind-body intervention …

Relative Influence Of Sea State And Mean, Turbulent, And Heterogenous Refractivity On X-Band Propagation, Douglas M. Pastore, Erin E. Hackett
Marine Science
In predicting electromagnetic wave propagation within the marine atmospheric surface layer, it is common to assume steady homogenous conditions. However, discrepancies between predicted and measured propagation remain, which could be due in-part to turbulent fluctuations of the refractive index, spatially heterogenous evaporative ducting environments, and mischaracterization of the rough ocean bottom boundary. To better understand the relative importance of these contributors, this study explores the sensitivity of X-band propagation to parameters describing sea-state conditions and refractive environments, including turbulent fluctuations and spatially heterogenous conditions. This study employs the extended Fourier Amplitude Sensitivity Test to compute sensitivity indices that evaluate the …

The human gut mycobiome (the fungus that lives in our gut) has been correlated with overall health and disease in humans. There are several factors that affect the human gut mycobiome including delivery method during birth (vaginal vs. c-section) and the presence or absence of antibiotics at birth. The mycobiome of 140 infants aged newborn to 18 months were tested using fecal samples from 3 different time periods (meconium, 6 months, 18 months). A total of 434 fecal samples were collected. A midwife or nurse collected the meconium samples while the child’s parents collected the samples at the 6-month and …

Newborn mammalian cardiomyocytes quickly transition from a fetal to an adult phenotype that utilizes mitochondrial oxidative phosphorylation but loses mitotic capacity. We tested whether forced reversal of adult cardiomyocytes back to a fetal glycolytic phenotype would restore proliferative capacity. We deleted Uqcrfs1 (mitochondrial Rieske iron-sulfur protein, RISP) in hearts of adult mice. As RISP protein decreased, heart mitochondrial function declined, and glucose utilization increased. Simultaneously, the hearts underwent hyperplastic remodeling during which cardiomyocyte number doubled without cellular hypertrophy. Cellular energy supply was preserved, AMPK activation was absent, and mTOR activation was evident. In ischemic hearts with RISP deletion, new cardiomyocytes …

Background: Tuberculosis (TB), caused by Mycobacterium tuberculosis (Mtb), continues to be a major public health problem worldwide. The human immunodeficiency virus (HIV) is another equally important life-threatening pathogen. HIV infection decreases CD4+ T cell levels markedly increasing Mtb co-infections. An appropriate animal model for HIV/Mtb co-infection that can recapitulate the diversity of the immune response in humans during co-infection would facilitate basic and translational research in HIV/Mtb infections. Herein, we describe a novel humanized mouse model. Methods: The irradiated NSG-SGM3 mice were transplanted with human CD34+ hematopoietic stem cells, and the humanization was monitored by staining various immune cell markers …

Group 3 innate lymphoid cells (ILC3s) are key players in intestinal homeostasis. ER stress is linked to inflammatory bowel disease (IBD). Here, we used cell culture, mouse models, and human specimens to determine whether ER stress in ILC3s affects IBD pathophysiology. We show that mouse intestinal ILC3s exhibited a 24-hour rhythmic expression pattern of the master ER stress response regulator inositol-requiring kinase 1α/X-box-binding protein 1 (IRE1α/XBP1). Proinflammatory cytokine IL-23 selectively stimulated IRE1α/XBP1 in mouse ILC3s through mitochondrial ROS (mtROS). IRE1α/XBP1 was activated in ILC3s from mice exposed to experimental colitis and in inflamed human IBD specimens. Mice with Ire1α deletion …

Immune checkpoint inhibitors (ICI) are increasingly used in combination. To understand the effects of different ICI categories, we characterized changes in circulating autoantibodies in patients enrolled in the E4412 trial (NCT01896999) of brentuximab vedotin (BV) plus ipilimumab, BV plus nivolumab, or BV plus ipilimumab-nivolumab for Hodgkin Lymphoma. Cycle 2 Day 1 (C2D1) autoantibody levels were compared to pre-treatment baseline. Across 112 autoantibodies tested, we generally observed increases in ipilimumab-containing regimens, with decreases noted in the nivolumab arm. Among 15 autoantibodies with significant changes at C2D1, all nivolumab cases exhibited decreases, with more than 90% of ipilimumab-exposed cases showing increases. Autoantibody …

The Effects Of Acute Exercise On Retrieval-Induced Forgetting, Anna Katherine Pitts
Honors Theses
This research investigates the relationship between acute exercise on retrieval-induced forgetting. Retrieval-induced forgetting (RIF) is a type of active forgetting that occurs when recalling an item from memory leads to the concurrent forgetting of related items connected to the same memory. Inhibition has been theorized as a potential mechanism behind RIF. Additionally, previous studies have shown that acute exercise can contribute to and enhance cognitive inhibitory processes. Therefore, an exercise-RIF relationship is plausible because the mechanisms that influence RIF, inhibition, are also influenced by exercise. Due to this understanding, the present thesis aims to investigate that when acute vigorous-intensity exercise …
