This dissertation addresses critical challenges in urban heat management, sustainable energy resource utilization, and water quality communication through three studies. Study 1 investigates the impact of roof color on urban heat islands in Tucson, Arizona where approximately 70% of roofs display high albedo (i.e., light) colors. Energy consumption simulations conducted indicate that converting dark- to light-colored roofs could save Tucson approximately $1,400,000 annually in energy costs, highlighting the potential of cool roofs for energy savings and improved thermal comfort. Produced waters (PW) are a major byproduct of the oil and gas industrial hydraulic fracturing processes that create a major waste disposal issue with a need for disposal, treatment, and reuse method developments. The current primary disposal method is subsurface injection, which can lead to high pressures in the subsurface. PWs are often highly saline from mineral leaching due to long residence in subsurface reservoirs. According to the USGS National Minerals Information Center, bromine, calcium chloride, iodine, lithium, magnesium, and sodium chloride have been found in and extracted from these domestic subsurface brines, suggesting that there may be other economically …

Due to the ecological and physical responses of arctic tundra to warming, organic carbon (C) stored in frozen permafrost soils is increasingly at risk of mineralization and export to aquatic systems, the coast, and atmosphere. Although many coastal aquatic systems in the Arctic are likely net sources of CO2 to the atmosphere, little is known about the drivers of inorganic C flux and how these systems contribute to regional C budgets. Neural networks are used in many real-world applications, ranging from classification tasks to medical diagnostics. For each task, a neural network is typically able to make predictions due to its ability to extract meaningful patterns from processing large amounts of data. Thus, given the increases in available data in recent decades, the performance of neural networks in making accurate predictions has greatly increased. However, this data often comes with ingrained uncertainties due to measurement errors or the inherent variability of individual data points. There is a paradigm shift towards data-centric AI, where model efficacy relies on quality, unified data. The common research analytics and data lifecycle environment (CRADLE™) is an infrastructure and framework that supports a data-centric paradigm and materials data science at scale through heterogeneous data management, elastic scaling, and accessible interfaces. We demonstrate CRADLE’s capabilities through five materials science studies: phase identification in X-ray diffraction, defect segmentation in X-ray computed tomography, polymer crystallization analysis in atomic force microscopy, feature extraction from additive manufacturing, and geospatial data fusion. CRADLE catalyzes scalable, reproducible insights to transform how data is captured, stored, and analyzed. …

The emergence of large, general-purpose foundation models has sparked significant interest in the broader machine learning community. Among the many models being released, the Segment Anything Model (SAM) has demonstrated exceptional capabilities for object segmentation in various settings. Given its expansive training data, SAM has been used for image segmentation across various downstream tasks ranging from tumour segmentation to aerial object detection. However, the majority of pretraining data used by SAM consists of naturally-occurring images, which have significantly different characteristics from images of tumours or images taken by drones. Recent declines in bee populations and ranges have been cause for concern due to the valuable pollination service that they provide. Several factors have been proposed to contribute to these declines, including habitat loss, pathogen spread, and pesticide usage, so many pollinator conservation schemes have involved the addition of pollinator-friendly habitat through wildflower plantings and artificial nesting sites. Because of this, many efforts have been made to enhance bee populations across different landscape types, including natural, agricultural, urban, and industrial areas. Osteoarthritis (OA) is a degenerative joint disease characterised by the breakdown of cartilage, causing pain, stiffness, and limited movement. Early diagnosis is crucial for effective management but remains challenging due to non-specific early symptoms. This study explores the application of Discriminant Function Analysis (DFA) to classify OA patients and healthy volunteers based on biomarker concentrations of Interleukin-6 (IL-6), Tumour necrosis factor-alpha (TNF-α), and Myeloperoxidase (MPO). DFA was employed to analyse biomarker data from 86 participants (58 patients, 28 volunteers) to evaluate the discriminatory power of these biomarkers in predicting OA. CD4+ T cells can either enhance or inhibit tumour immunity. Although regulatory T cells have long been known to impede antitumour responses1-5, other CD4+ T cells have recently been implicated in inhibiting this response6,7. Yet, the nature and function of the latter remain unclear. Here, using vaccines containing MHC class I (MHC-I) neoantigens (neoAgs) and different doses of tumour-derived MHC-II neoAgs, we discovered that whereas the inclusion of vaccines with low doses of MHC-II-restricted peptides (LDVax) promoted tumour rejection, vaccines containing high doses of the same MHC-II neoAgs (HDVax) inhibited rejection. Recent advancements in artificial intelligence (AI) have accelerated the prediction of unknown protein structures. However, accurately predicting the three-dimensional (3D) structures of fusion proteins remains a difficult task because the current AI-based protein structure predictions are focused on the WT proteins rather than on the newly fused proteins in nature. Following the central dogma of biology, fusion proteins are translated from fusion transcripts, which are made by transcribing the fusion genes between two different loci through the chromosomal rearrangements in cancer. Neutrophil extracellular traps (NETs) function to control infectious agents as well as to propagate inflammatory response in a variety of disease conditions. DNA damage associated with chromatin decondensation and NACHT domain-leucine-rich repeat-and pyrin domain-containing protein 3 (NLRP3) inflammasome activation have emerged as crucial events in NET formation, but the link between the two processes is unknown. In this study, we demonstrate that poly(ADP-ribose) polymerase-1 (PARP-1), a key DNA repair enzyme, regulates NET formation triggered by NLRP3 inflammasome activation in neutrophils. BACKGROUND: GP-2250, a novel analog of taurultam (TRLT), has emerged as a potent anti-neoplastic drug; however, the mechanisms underlying its effects are not well understood. Here, we investigated the mechanism of action and the biological effects of GP-2250 using in vitro and in vivo models.
METHODS: We carried out a series of in vitro (MTT assay, Annexin V/PI assay, colony formation assay, reverse-phase protein array [RPPA], and HRLC/IC analysis) to determine the biological activity of GP-2250 and investigate the mechanism of action. OBJECTIVES: Cachexia is a metabolic disorder and comorbidity with cancer and heart failure. The syndrome impacts more than thirty million people worldwide, accounting for 20% of all cancer deaths. In acute myeloid leukemia, somatic mutations of the metabolic enzyme isocitrate dehydrogenase 1 and 2 cause the production of the oncometabolite D2-hydroxyglutarate (D2-HG). Increased production of D2-HG is associated with heart and skeletal muscle atrophy, but the mechanistic links between metabolic and proteomic remodeling remain poorly understood. Therefore, we assessed how oncometabolic stress by D2-HG activates autophagy and drives skeletal muscle loss.

The RNA exosome is an evolutionarily conserved complex required for both precise RNA processing and decay. Pathogenic variants in EXOSC genes, which encode structural subunits of this complex, are linked to several autosomal recessive disorders. Here, we describe a missense allele of the EXOSC4 gene that causes a collection of clinical features in two affected siblings. This missense variant (NM_019037.3: exon3:c.560T>C) changes a leucine residue within a conserved region of EXOSC4 to proline (p.Leu187Pro). The two affected individuals show prenatal growth restriction, failure to thrive, global developmental delay, intracerebral and basal ganglia calcifications, and kidney failure. Neurogenetic disorders, such as neurofibromatosis type 1 (NF1), can cause cognitive and motor impairments, traditionally attributed to intrinsic neuronal defects such as disruption of synaptic function. Activity-regulated oligodendroglial plasticity also contributes to cognitive and motor functions by tuning neural circuit dynamics. However, the relevance of oligodendroglial plasticity to neurological dysfunction in NF1 is unclear. Here we explore the contribution of oligodendrocyte progenitor cells (OPCs) to pathological features of the NF1 syndrome in mice. INTRODUCTION: Vascular contributions to cognitive impairment and dementia (VCID) represent a major factor in cognitive decline in older adults. The present study examined the relationship between cerebrovascular reactivity (CVR) measured by magnetic resonance imaging (MRI) and cognitive function in a multi-site study, using a predefined hypothesis.
METHODS: We conducted the study in a total of three analysis sites and 263 subjects. Each site performed an identical CVR MRI procedure using 5% carbon dioxide inhalation. A global cognitive measure of Montreal Cognitive Assessment (MoCA) and an executive function measure of item response theory (IRT) score were used as outcomes.

In this study, we investigate the influence of cold-plasma-induced enhanced performance and efficiency of SAPO-34 membranes in the separation of CO2 and CH4 mixtures. Placing the herein presented research in a broader context, we aim to address the question of whether cold plasma can significantly impact the membrane performance. We subjected SAPO-34 membranes to plasma mild disturbances and analyzed their performance in separating CO2 and CH4. Our findings reveal a notable enhancement in membrane efficiency and sustained performance when exposed to cold plasma. In this study, contour fields of mean age and turbulence kinetic energy (TKE) in the post-stenotic region of model arteries were analyzed to explore their correlation with FFR. While both high and low FFR cases exhibited similarities with an inner low-age region and an outer high-age region, low FFR cases displayed asymmetry in their low-age regions due to eccentric stenosis. Moreover, gradients of age across the artery diameter were more pronounced for low FFR cases. Additionally, low FFR cases demonstrated significantly higher TKE in the post-stenotic blood jet stream compared to high FFR cases that are reflected in low-age boundary …

Objective: To assess the efficacy of irrigation solution containing reuterin in inhibiting gene expression of E. faecalis, F. nucleatum, and P. gingivalis during biofilm formation in ex vivo root canal model. Material and Methods: Reuterin was isolated from an Indonesian strain of L. reuteri. Seventy-two premolar root samples were subjected to inoculation with E. faecalis, F. nucleation, and P. gingivalis for 24 hours. Rapid population growth and development in Western states are exerting strain on the region’s limited water resources. Urbanization exacerbates this issue by increasing impervious surfaces, limiting infiltration of precipitation during storm events and snowmelt, which results in changes to hydrologic conditions with higher runoff volumes and higher peak flows. Stormwater transports pollutants as it flows across impervious surfaces, discharging high volumes of runoff and elevated loads of urban contaminants into receiving waters. Suspended sediments can have benefits and drawbacks on aquatic ecosystems depending on the concentrations. The need to quantitatively monitor water bodies and watersheds is key to reduce excessive suspended concentrations. This study aims to integrate remote sensing technology with numerical watershed and hydrodynamic models and apply them on the Tennessee River and the surrounding tributaries to better assess the fate and transport of suspended sediment concentrations (SSC) there. The obtained results indicate that using Google Earth Engine (GEE), Landsat satellite can be used to estimate historical turbidity in the Tennessee River. New topological and geometrical methods in knot theory provide a way to rigorously measure the entanglement of open curves in 3-space for the first time. These methods enable the topological analysis of physical systems of filaments, such as those formed by proteins. In this dissertation we employ tools from knot theory in combination with available experimental data and molecular simulations to analyze the topological complexity of tau proteins. We present a comprehensive framework that amalgamates inputs from multiple sensors. Prior research has established a role for both social isolation and exposure to high fat Western diets in altering a range of behaviors from reduced memory performance to increased depression-like behaviors. The present study scrutinizes the interplay among these variables during the peri-adolescent developmental phase, utilizing Long-Evans rats as the experimental model. Our overarching hypothesis is that rats exposed to either social isolation, a high-fat diet, or both will result in heightened pain sensitivity, diminished cognitive flexibility, and increased neuroinflammatory responses within brain regions implicated in sociability, cognition, memory, and pain processing. Secure attachment with parents predicts less emotional distress and greater subjective well-being across the lifespan. One mechanism by which parental attachment supports mental health outcomes is through psychological needs satisfaction (PNS). In addition to the attachment to parents, relationships with fictive kin caregivers are prevalent and important among Black families. The present study investigated the association between parental attachment, fictive kin bonds, psychological needs satisfaction and mental health outcomes (i.e., emotional distress and subjective well-being) in a sample of Black American adults.
