In this thesis I looked at how legend tripping is mediated through the affordances of new social technologies and apps, specifically TikTok. Short video formats allowed for folk cinematography to spread legend narratives and at the same time create more visual online landscapes for people to interact with. I defined folk cinematography as the way in which people make videos based on a vernacular level. Isotopic analysis has been a common practice used in paleontology to study ancient ecosystems, and the lives of animals therein. Oxygen isotopic analysis is of particular importance not only because of its use in studying paleoecology, diet, and migration, but also its use in studying mountain elevation, and its impact on ancient ecosystems. I use stable oxygen isotopic analysis to study the habits and ecology of brontotheres from intermountain basins surrounding the Uinta Mountains of Utah and Wyoming, USA. Fiddler crabs live on the edge of their thermal tolerance limits, balancing the need to thermoregulate with the need to mate. Previous research suggests that increased temperature reduces fiddler crab surface activity time, thus decreasing mating opportunities, throughout the range species with greatest impacts to low latitude populations. The less studied missed mating opportunities associated with behavioral compensation for thermal stress are crucial to understanding species vulnerability to climate change. Oyster larval microbiomes are shaped by their aquatic microbiomes. Antioxidant therapies are of interest in the prevention and management of ocular disorders such as cataracts. Although an active area of interest, topical therapy with antioxidants for the treatment of cataracts is complicated by multiple ocular anatomical barriers, product stability, and solubility. Depression is a highly prevalent mental disorder in older adults, and among its highest risk factors is loneliness. Although this relationship is commonly evident, the mechanisms underlying it, such as sense of mattering and psychological adjustment, lack exploration. The current research aims to examine the relationship between loneliness and geriatric depression and explore the mediating roles of sense of mattering and psychological adjustment.

Purpose: Oropharyngeal squamous cell cancers (OPSCCs) are traditionally managed with surgery and, if indicated, adjuvant radiation therapy (RT) with or without chemotherapy. NCCN recommends keeping the time from surgery to the start of RT (TSRT) within 6 weeks to avoid possibly compromising patient outcomes. HPV+ OPSCCs behave more favorably than HPV- OPSCCs. We hypothesized that TSRT beyond 6 weeks may not portend poorer outcomes for the former. Methods: We identified nonmetastatic, high-risk HPV+ OPSCCs treated with multimodal therapy at 2 institutions. This research investigates the determinants of breastfeeding initiation and continuation among African American women in the United States and evaluates the effectiveness of an intervention using WordPress to disseminate culturally relevant breastfeeding information to African American women. First, secondary data from the Future of Families and Child Wellbeing Study (FFCWS) was used to test factors influencing breastfeeding initiation and continuation among White and Black women. The results showed that the foreign-born (vs. US-born) status, residing (vs. non-residing) with grandparents, and strong (vs. weak) cultural identity were positively associated with breastfeeding initiation and continuation across racial groups. Variegated leaves with disjointed, patterned coloration of the leaf blade are often hypothesized to have decreased photosynthetic capacity in comparison to uniformly colored leaves. Yet, many forest understory plants in tropical, subtropical, and temperate climates possess leaf variegation. Moreover, mixed findings from previous research on the physiological impacts of leaf variegation suggest variegation is adaptative under certain environmental conditions despite decreased photosynthetic tissue area. Background: Patients with major depressive disorder (MDD) can present with altered brain structure and deficits in cognitive function similar to those seen in aging. However, the interaction between age-related brain changes and brain development in MDD remains understudied. In a cohort of adolescents and adults with and without MDD, we assessed brain aging differences and associations through a newly developed tool that quantifies normative neurodevelopmental trajectories. To prevent forward contamination from microbes aboard spacecraft intended for exploration of solar system bodies there is a need for effective sterilization methods. However, current techniques are both time-consuming and expensive. For example, dry heat sterilization requires removal from the assembly site and several days of treatment. Furthermore, some components such as optics and electronics are not compatible with current sterilization techniques. In this thesis, a novel femtosecond laser surface processing technique for the rapid sterilization of spacecraft hardware is reported. This study investigated the factors influencing patient adoption of telemedicine, including patient experiences and economic considerations. Paclitaxel (PTX), a chemotherapeutic that alters microtubule dynamics, is known to induce nociceptive hypersensitivity and sensory neuron damage in human patients and in Drosophila larvae. We used a modified feeding protocol to establish a genetically tractable Drosophila model of PTX-induced hypersensitivity. Larvae fed PTX exhibited hypersensitivity to thermal and mechanical stimuli. Thermal hypersensitivity was observed at low concentrations of PTX (even below 1 μM per ml of food), begins within 8 hours of PTX feeding, and did not completely resolve at the larval stage. Live imaging of peripheral thermal nociceptors showed that this hypersensitivity precedes observable neuronal damage. At low …

Cantú syndrome is a multisystem disorder caused by gain-of-function (GOF) mutations in KCNJ8 and ABCC9, the genes encoding the pore-forming inward rectifier Kir6.1 and regulatory sulfonylurea receptor SUR2B subunits, respectively, of vascular ATP-sensitive K+ (KATP) channels. In this study, we investigated changes in the vascular endothelium in mice in which Cantú syndrome-associated Kcnj8 or Abcc9 mutations were knocked in to the endogenous loci. We found that endothelium-dependent dilation was impaired in small mesenteric arteries from Cantú mice. Loss of endothelium-dependent vasodilation led to increased vasoconstriction in response to intraluminal pressure or treatment with the adrenergic receptor agonist phenylephrine. We also …

PURPOSE: Dysregulated cholesterol metabolism is critical in the pathogenesis of AMD. Cellular senescence contributes to the development of numerous age-associated diseases. In this study, we investigated the link between cholesterol burden and the cellular senescence of photoreceptors.

METHODS: Retinas from rod-specific ATP binding cassette subfamily A member 1 (Abca1) and G member 1 (Abcg1) (Abca1/g1-rod/-rod) knockout mice fed with a high-fat diet were analyzed for the signs of cellular senescence. Brachytherapy is a critical component of locally advanced cervical cancer treatment, and patients ineligible for brachytherapy historically have poor outcomes. Delivery of boost with stereotactic body radiation therapy (SBRT) has been studied, though toxicity is a concern. Recent case reports have explored adaptive radiation boost, which can adjust plans for inter-fraction motion using magnetic resonance guidance. Herein, we report the first patient with locally advanced cervical cancer ineligible for brachytherapy who was treated with a cone-beam computed tomography (CBCT)-guided adaptive boost following completion of chemoradiation. Age-related vision loss caused by retinal neurodegenerative pathologies is becoming more prevalent in our ageing society. To understand the physiological and molecular impact of ageing on retinal homeostasis, we used the short-lived African turquoise killifish, a model known to naturally develop central nervous system (CNS) ageing hallmarks and vision loss. Bulk and single-cell RNA-sequencing (scRNAseq) of three age groups (6-, 12-, and 18-week-old) identified transcriptional ageing fingerprints in the killifish retina, unveiling pathways also identified in the aged brain, including oxidative stress, gliosis, and inflammageing. This study explores the bioaffinity of aptamer-target interactions through a comprehensive analysis. Aptamers, oligonucleotides that can bind to target molecules, have emerged as powerful tools in various fields, including diagnostics and therapeutics. The primary objective of this research was to assess the binding affinity and specificity of selected aptamers to their respective targets using a combination of experimental techniques and computational models. The binding affinity of 10 different complexes were measured through surface plasmon resonance (SPR) followed by molecular simulations. Both the federal government and photojournalism have played a substantial role in the “othering” of Appalachia while simultaneously attempting to save it. Examining both institutions demonstrates the exploitative processes that alter the perception of rural communities and their surrounding environment. Stacey Kranitz’s archival exhibit, As It Was Give(n) To Me, internalizes this precarious role of the camera in a region heavily journalized. For the marine industry biofouling has a negative reputation. On ship hulls, the accumulation of these unwanted plants and animals can lead to increased drag, fuel consumption, and greenhouse gas emissions. Offshore platforms are also subject to biofouling which can result in corrosion and hydrodynamic loading, thus shortening their lifespan. While the harmful impacts of biofouling are commonly reported throughout the literature, biofouling can also benefit both aquatic and human populations. Emotions are biological responses to stimuli that allow individuals to derive meaning, appraise experiences, and prepare to respond. However, individuals perceive emotions differently based on emotion socialization which not only dictates the way emotions are viewed and managed but also has been directly linked with mental health outcomes. Furthermore, research shows emotion socialization is informed by demographic variables such as gender such that the expectations of emotionality differ; where women are taught to express emotions, men are taught to conceal.
