Furthering Ascidian Taxonomy Using Molecular Biology, Nicholas Gulnick
Furthering Ascidian Taxonomy Using Molecular Biology, Nicholas Gulnick
Electronic Theses and Dissertations
Ascidians are our closest invertebrate relatives and comprise nearly 3,000 species separated into three orders: Aplousobranchia (most speciose), Stolidobranchia, and Phlebobranchia (least speciose). Ascidians can be classified as either solitary or colonial organisms. Species delimitation using morphological characters alone has had varied results. Well known, widely distributed, morphological species have turned out to be catch all species comprised of several cryptic species. Molecular markers can help mitigate some of the issues presented by strictly using morphological observations, including resolving the status of cryptic species, and accessing the expert knowledge required to identify a species. By incorporating molecular markers and pairing …
Variability In Nutrient Concentrations Across Regions Of The Winyah Bay River Plume, Anne Mackenzie Gossman
Variability In Nutrient Concentrations Across Regions Of The Winyah Bay River Plume, Anne Mackenzie Gossman
Electronic Theses and Dissertations
River plumes are one of the few large-scale features responsible for transporting particulates and terrigenous materials to coastal oceans. Macro-nutrients, such as dissolved inorganic nitrogen (DIN) and phosphorus (PO43-), are basic requirements for marine food webs. Due to the plumes’ ability to transport large quantities of these constituents, these systems contribute heavily to coastal productivity. This study focuses on the dynamics of the Winyah Bay river plume located in Georgetown, South Carolina. River plumes, including the Winyah Bay plume, are dynamic systems whose structures are comprised of a multitude of regions and features including the source, lift off, near-field, mid-field, …
Genetically-Encoded Optical Barcodes For Single-Cell Analysis, Daniel Pritko
Genetically-Encoded Optical Barcodes For Single-Cell Analysis, Daniel Pritko
All Theses
Cell barcodes are capable of being used to answer many different biological questions. They have been used to track the lineage of cells to identifying the function of a gene. While there are multiple different methods to creating cell barcodes, they are limited in their scalability and application. In a previous publication we propose and computationally prove an optical single-cell barcoding method that bridges fast and scalable readouts with the benefits of genetic encoding. In this approach fluorescent proteins (fps) are combined to form fluorescent barcodes that can then be analyzed using a spectral flow cytometer. Here, we test the …
Improving Pedestrian Conspicuity At Night: Testing The Efficacy Of A Video-Based Educational Intervention, Morgan Mccree
Improving Pedestrian Conspicuity At Night: Testing The Efficacy Of A Video-Based Educational Intervention, Morgan Mccree
All Theses
The majority of fatal crashes involving pedestrians occur at night. Insufficient conspicuity has been implicated as a causal factor in these crashes, and there is a critical need for innovative strategies to increase the nighttime conspicuity of pedestrians. This experiment investigated the efficacy of an 11-minute educational video aimed to teach road users about critical and relevant visual perception concepts (e.g., contrast, retroreflectivity, biological motion) that impact nighttime conspicuity. Fifty-four college students were randomly assigned to either watch the video (intervention; n=27) or not (control; n=27). Each participant provided quantitative judgments regarding the nighttime visibility of a pedestrian on an …

Survey Of Nematodes Associated With Strawberry In South Carolina And Evaluation Of Short-Term Heat Exposure On Nematode Reproduction Biology, Ivan A. Alarcon Mendoza
All Theses
Nematodes are a growing threat to the multi-billion-dollar strawberry industry in the US due to the lack of safe and effective management methods. Limited information on the distribution of nematodes in fumigated plasticulture strawberry fields further complicates their management. Additionally, climatic anomalies like rising soil temperatures and heat waves are occurring more often, and their impact on crop growth and nematode reproduction is not well understood. The current study was conducted with two main objectives: i) understand distribution and population densities of nematodes associated with plasticulture strawberry production in South Carolina, and ii) understand the impact of short-term high heat …

Instantons In Φ4 Theories: Transseries, Virial Theorems, And Numerical Aspects, Ludovico T. Giorgini, Ulrich D. Jentschura, Enrico M. Malatesta, Tommaso Rizzo, Jean Zinn-Justin
Physics Faculty Research & Creative Works
We discuss numerical aspects of instantons in two- and three-dimensional φ4 theories with an internal O(N) symmetry group, the so-called N-vector model. By combining asymptotic transseries expansions for large arguments with convergence acceleration techniques, we obtain high-precision values for certain integrals of the instanton that naturally occur in loop corrections around instanton configurations. Knowledge of these numerical properties is necessary in order to evaluate corrections to the large-order factorial growth of perturbation theory in φ4 theories. The results contribute to the understanding of the mathematical structures underlying the instanton configurations.

As high-strength aluminum alloys present several processability issues with additive manufacturing (AM), Scalmalloy®, an Al-Mg-Sc-Zr-based alloy, has been developed. This alloy is age-hardenable, allowing it to precipitate out a strengthening precipitate phase, Al3(Sc,Zr). The manufacturer recommends a single-stage aging treatment at 325 °C for 4 h; however, the majority of the literature studies utilize a powder bed processing known as selective laser melting (SLM) over powder-fed processing directed energy deposition (DED). This study addresses the lack of information on heat treatments for DED fabrication by exploring the application of artificial aging temperatures of 300–400 °C for 2, 4, and 6 …

Although there is considerable evidence showing that the prevention of mental illnesses and adverse outcomes and mental health promotion can help people lead better and more functional lives, public mental health remains overlooked in the broader contexts of psychiatry and public health. Likewise, in undergraduate and postgraduate medical curricula, prevention and mental health promotion have often been ignored. However, there has been a recent increase in interest in public mental health, including an emphasis on the prevention of psychiatric disorders and improving individual and community wellbeing to support life trajectories, from childhood through to adulthood and into older age. Wheat (Triticum aestivum L.) is an important food crop throughout the world. The seed mycobiome is the primary inoculum affecting the overall health and productivity of wheat plants. Seed-borne fungi can have an impact on the production of wheat and cause health complications upon consumption. Therefore, for durable and sustainable wheat production, it is imperative to characterise the wheat mycobiome. The rationale of this study was to investigate the wheat seed mycobiome of two Egyptian wheat cultivars (Sids 14 and Giza 171) using a culture-independent technique. The fungal community in 6 wheat seed samples was identified by high-throughput amplicon sequencing …

Relationship Between Parenting Emotion Regulation, Parenting Style, And Child Emotion Regulation, Sydney R. Ward
All-Inclusive List of Electronic Theses and Dissertations
This study aimed to contribute to the existing literature by investigating parenting factors that contribute to child emotional regulation (ER). Prior research suggests that while emotion dysregulation is not part of current diagnostic criteria for attention deficit hyperactivity disorder (ADHD), children with ADHD often exhibit greater emotion regulation (ER) difficulties compared to their neurotypical peers. Research focused on parenting has found that parent’s ability to manage their emotions within a parenting context (parenting ER) as well as parenting styles (either positive or negative) are associated with child emotional regulation skills. However, previous research has not considered the influence of both …

Photonic Integrated Circuit Data Acquisition And Tunable Laser Control For Biomedical Point Of Care Systems, Maximilian Thoburn
Theses
In the world of healthcare, it has always been desirable to have a single test that can screen for multiple different patient ailments at once. Toward this end, a silicon photonics platform has been developed that uses photonic integrated circuits in combination with lasers to simultaneously perform various biological assays, i.e., to screen for biological markers in a patient’s sample. This system uses inexpensive passive flow cards with inexpensive photonic integrated circuits to create a disposable test platform that can be used in point of care environments. However, the necessary bench-top equipment to run an assay with these cards is …
The Role Of Elevation In The Migration, Physiology, And Genomic Diversification Of Birds, Jessie L. Williamson
The Role Of Elevation In The Migration, Physiology, And Genomic Diversification Of Birds, Jessie L. Williamson
Biology ETDs
Elevational gradients cause profound eco-climatic variation across short distances. Partial pressure of oxygen (PO2) is a severe challenge that organisms face at high elevations; due partly to specialization on PO2, most Neotropical bird species have narrow elevational ranges. However, a small fraction are elevational generalists that span sea level to high elevations, and a subset undertake extreme migratory journeys from low to high elevations biannually. My dissertation combines diverse approaches across levels of biological organization to understand how elevation impacts the ecology, evolution, physiology, and migration of montane birds. I examined ‘extreme’ elevational …
Value Of Macroalgae As Habitat In A Eutrophic Coastal Bay, Dominika Polchowska
Value Of Macroalgae As Habitat In A Eutrophic Coastal Bay, Dominika Polchowska
Theses, Dissertations and Culminating Projects
Eutrophication is a phenomenon caused by changes in nutrient loading that results in harmful algal blooms, an overgrowth of macroalgae, altered aquatic benthic invertebrate communities, and loss of ecosystem functions. Barnegat Bay, NJ is a highly eutrophic system that has experienced a substantial loss of seagrass beds with subsequent expansion of drift macroalgae. Although considered poor quality, these algal beds represent valuable habitat for many benthic invertebrates. This research assesses the value of macroalgae as a habitat for macrofauna in regions where macroalgae are the dominant “vegetation.” Benthic drift algal samples were collected during June and August 2023 to reflect …

Across the country, the mental health crisis continues to rage. Varying degrees of depression, stress, and anxiety are commonly found in college-age students. The COVID-19 pandemic augmented this crisis, but the pandemic is not the sole culprit for the escalation of mental health disorders in the United States. Students across the globe suffer mental health disorders that include depression, anxiety, and stress. Research has shown that graduate students are six times more likely to develop anxiety and depression than the general population. Student registered nurse anesthetists (SRNAs) are vulnerable to mental health disorders. SRNAs enter labor-intensive doctoral programs fraught with …

College Students' Perceptions Of The Student-Teacher Relationship, Lauren E. Biggs
Electronic Theses and Dissertations
Humans, as social creatures, pursue relationships with others to fulfill various needs. As such, when we enter school, we naturally seek to establish relationships with teachers, who act as significant individuals in our lives. In general, research has shown that students with close, warm relationships with their teachers tend to experience more favorable outcomes related to academic achievement, behavior, and social-emotional factors. Students with more negative relationships with their teachers conversely tend to experience poorer outcomes. Though decades of research have consistently shown this correlation, few studies have examined the relationships between college students and their professors. The current study …

Type 2 diabetes mellitus (T2DM) is common and increasing in prevalence worldwide, with devastating public health consequences. While peripheral insulin resistance is a key feature of most forms of T2DM and has been investigated for over a century, research on brain insulin resistance (BIR) has more recently been developed, including in the context of T2DM and non-diabetes states. Recent data support the presence of BIR in the aging brain, even in non-diabetes states, and found that BIR may be a feature in Alzheimer's disease (AD) and contributes to cognitive impairment. Further, therapies used to treat T2DM are now being investigated …

Avian Communities Monitored Along A Successional Chronosequence In Bottomland Hardwood Forests In The Lower Mississippi Alluvial Valley, Eamonn Alasdair Thurmond
Electronic Theses and Dissertations
Bottomland hardwood forests in the Lower Mississippi Alluvial Valley have been extensively reduced due to agriculture conversion and altered hydrology. Remnant forests have undergone further degradation from sources such as a lack of forest management, invasive species, and reduced regeneration of oaks (Quercus spp.). Recent restoration efforts of bottomland hardwood forests have focused on implementing wildlife forestry practices that prioritize restoring wildlife habitat while retaining profitable timber harvests. In this study I surveyed bird communities in bottomland hardwood forests in the Lower Mississippi Alluvial Valley along a successional chronosequence. Birds were sampled using automated recording units over 10 days …

Paclitaxel, which features low water solubility and permeability, is an efflux pump substrate. The current paclitaxel drugs are given intravenously after resolving the solubility issue. Yet, oral delivery to achieve therapeutic bioavailability is not effective due to low absorption. This study evaluated a natural compound, rubusoside, to improve oral bioavailability in an animal model. Free paclitaxel molecules were processed into nano-micelles formed in water with rubusoside. The particle size of the nano-micelles in water was determined using dynamic light scattering. The oral bioavailability of paclitaxel in nano-micelles was determined against Cremophor/alcohol-solubilized Taxol after oral and intravenous administration to pre-cannulated Sprague …

Dietary supplement use in the United States is widespread and increasing, especially among certain population groups, such as older Americans. The science surrounding dietary supplements has evolved substantially over the last few decades since their formal regulation in 1994. Much has been learned about the mechanisms of action of many dietary supplement ingredients, but the evidence on their health effects is still building. As is true of much nutrition research, there are many studies that point to health effects, but not all are at the level of scientific evidence (e.g., randomized controlled interventions), rigor, or quality needed for definitive statements …

Obesity is a highly prevalent chronic disease that impacts >40% of reproductive-aged females. The pathophysiology of obesity is complex and can be understood simply as a chronic energy imbalance whereby caloric intake exceeds caloric expenditure with an energy surplus stored in adipose tissue. Obesity may be categorized into degrees of severity as well as different phenotypes on the basis of metabolic health and underlying pathophysiology. Obesity and excess adiposity have a significant impact on fertility and reproductive health, with direct effects on the hypothalamic-pituitary-ovarian axis, the ovary and oocyte, and the endometrium. There are significant adverse pregnancy outcomes related to …

The popularity of e-cigarettes (vaping) has soared, creating a public health crisis among teens and young adults. Chronic vaping can induce gut inflammation and reduce intestinal barrier function through the production of the proinflammatory molecule hydrogen sulfide (H2S). This is particularly concerning for people with HIV (PWH) as they already face impaired immune function and are at a higher risk for metabolic dysregulation, diabetes, and chronic liver disease. Furthermore, PWH experience unhealthy behaviors, making it crucial to understand the systemic metabolic dysregulation and pathophysiological mechanisms associated with vaping in this population. Here, we employed liquid chromatography–mass spectrometry (LC-MS)-based metabolomics to …

Background: Bioenergetic remodeling of core energy metabolism is essential to the initiation, survival, and progression of cancer cells through exergonic supply of adenosine triphosphate (ATP) and metabolic intermediates, as well as control of redox homeostasis. Mitochondria are evolutionarily conserved organelles that mediate cell survival by conferring energetic plasticity and adaptive potential. Mitochondrial ATP synthesis is coupled to the oxidation of a variety of substrates generated through diverse metabolic pathways. As such, inhibition of the mitochondrial bioenergetic system by restricting metabolite availability, direct inhibition of the respiratory Complexes, altering organelle structure, or coupling efficiency may restrict carcinogenic potential and cancer progression. …

Objective: This study examines the association between political identity and young adults' fertility desires from 1989 to 2019. Background: Understanding the factors that shape fertility preferences is important because these preferences are the critical bridge between social forces and fertility. Identity is a theorized, but understudied, predictor of fertility desires. The increasing salience of political identity suggests that the association between political identity and fertility desires has strengthened over time. Method: Data come from the 1989–2019 waves of Monitoring the Future, a nationally representative study of 12th graders (N = 67,557). Record high temperatures were documented in the McMurdo Dry Valleys, Antarctica, on 18 March 2022, exceeding average temperatures for that day by nearly 30°C. Satellite imagery and stream gage measurements indicate that surface wetting coincided with this warming more than 2 months after peak summer thaw and likely exceeded thresholds for rehydration and activation of resident organisms that typically survive the cold and dry conditions of the polar fall in a freeze-dried state. This weather event is notable in both the timing and magnitude of the warming and wetting when temperatures exceeded 0°C at a time when biological communities and …

Background: Physical activity and metformin pharmacotherapy are associated with improved clinical outcomes in breast and colorectal cancer survivors. Myokines are cytokines secreted from skeletal muscle that may mediate these associations. Methods: This hypothesis-generating analysis used biospecimens collected from a multi-centre 2 × 2 factorial randomized design of 116 patients with stage I–III breast and colorectal cancer who were randomized to 12 weeks of (1) aerobic exercise (moderate intensity titrated to 220 min/week); (2) metformin (850 mg daily for 2 weeks and then titrated to 850 mg twice per day); (3) aerobic exercise and metformin; or (4) control. Fourteen myokines were …
