Atrial and brain natriuretic peptides (ANP and BNP) bind to guanylyl cyclase A/natriuretic peptide receptor A (GC-A/NPRA), stimulating natriuresis and diuresis and reducing blood pressure (BP), but the role of ANP/NPRA signaling in podocytes (highly specialized epithelial cells covering the outer surfaces of renal glomerular capillaries) remains unclear. This study aimed to determine the effect of conditional deletion of podocyte-specific Npr1 (encoding NPRA) gene knockout (KO) in male and female mice. Tamoxifen-treated wild-type control (PD Npr1 f/f; WT), heterozygous (PD-Cre-Npr1 f/+; HT), and KO (PD-Cre-Npr1 f/-) mice were fed a normal-, low-, or high-salt diet for 4 wk. Podocytes isolated …

Affect Regulation And Allostatic Load Over Time, Amanda E. Ng, Tara Gruenewald, Robert-Paul Juster, Claudia Trudel-Fitzgerald

Psychology Faculty Articles and Research

Objective

Emerging work suggests that affect regulation strategies (e.g., active coping, anger expression) predict disease and mortality risk, with sometimes divergent estimates by sex or education levels. However, few studies have examined potential underlying biological mechanisms. This study assessed the longitudinal association of affect regulation with future allostatic load.

Method

In 2004–2006, 574 participants from the Midlife in the United States study completed validated scales assessing use of nine general and emotion-specific regulatory strategies (e.g., denial, anger expression). As a proxy for how flexibly participants regulate their affect, variability in the use of regulatory strategies was operationalized using a standard …

The time from conception through the first year of life is the most dynamic period in human development. This time period is particularly important for infants born very preterm (< 30 weeks gestation; VPT), as they experience a significant disruption in the normal developmental trajectories and are at heightened risk of experiencing developmental impairments and delays. Variations in the epigenetic landscape during this period may reflect this disruption and shed light on the interrelationships between aging, maturation, and the epigenome. We evaluated how gestational age (GA) and age since conception in neonates [post-menstrual age (PMA)], were related to DNA methylation in buccal cells collected at NICU discharge from VPT infants (n = 538). After adjusting for confounders and applying Bonferroni correction, we identified 2,366 individual CpGs associated with GA and 14,979 individual CpGs associated with PMA, as well as multiple differentially methylated regions. Pathway enrichment analysis identified pathways involved in axonogenesis and regulation of neuron projection development, among many other growth and developmental pathways (FDR q < 0.001). Our findings align with prior work, and also identify numerous novel associations, suggesting that genes important in growth and development, particularly neurodevelopment, are subject to substantial epigenetic changes during early development among children born VPT.

Nucleic acid-sensing Toll-like receptors (TLR) 3, 7/8, and 9 are key innate immune sensors whose activities must be tightly regulated to prevent systemic autoimmune or autoinflammatory disease or virus-associated immunopathology. Here, we report a systematic scanning-alanine mutagenesis screen of all cytosolic and luminal residues of the TLR chaperone protein UNC93B1, which identified both negative and positive regulatory regions affecting TLR3, TLR7, and TLR9 responses. We subsequently identified two families harboring heterozygous coding mutations in UNC93B1, UNC93B1+/T93I and UNC93B1+/R336C, both in key negative regulatory regions identified in our screen. These patients presented with cutaneous tumid lupus and juvenile idiopathic arthritis plus …

Pediatric low-grade glioma (pLGG) is the most common childhood brain tumor group. The natural history, when curative resection is not possible, is one of a chronic disease with periods of tumor stability and episodes of tumor progression. While there is a high overall survival rate, many patients experience significant and potentially lifelong morbidities. The majority of pLGGs have an underlying activation of the RAS/MAPK pathway due to mutational events, leading to the use of molecularly targeted therapies in clinical trials, with recent regulatory approval for the combination of BRAF and MEK inhibition for BRAFV600E mutated pLGG. Despite encouraging activity, tumor …

Sustainability In The Specialty Mushroom Industry And The Fight Against Single-Use Plastic: A Case Study, Amanda Newman

University Honors Theses

Specialty mushroom cultivator, Bridgetown Mushrooms, generates approximately 3 metric tons of polypropylene plastic waste a year. This is just a fraction of the 1-2 million metric tons created by the specialty mushroom industry annually. With only 10% of plastic waste being used as secondary materials, the majority of this waste ends up in landfills or in the environment. Through collaboration with employees within Bridgetown’s operation and government agency mentorship, it was determined that the most viable and ecological solution to reduce on-farm plastic waste within Bridgetown’s operation was to develop a reusable growing apparatus as a direct replacement for polypropylene …

Germinal centers (GC) are microanatomical lymphoid structures where affinity-matured memory B cells and long-lived bone marrow plasma cells are primarily generated. It is unclear how the maturation of B cells within the GC impacts the breadth and durability of B cell responses to influenza vaccination in humans. We used fine needle aspiration of draining lymph nodes to longitudinally track antigen-specific GC B cell responses to seasonal influenza vaccination. Antigen-specific GC B cells persisted for at least 13 wk after vaccination in two out of seven individuals. Monoclonal antibodies (mAbs) derived from persisting GC B cell clones exhibit enhanced binding affinity …

Middle West And The Pastoral Ideal In The American Artistic Landscape, Rebecca Umland

Mythcon

The complex and sometimes contradictory origin and use of the term Middle West—an evolving concept especially during the later American Expansionism period (primarily 1880 and after)—impacted the imagination of its writers. Their works view the Middle West as an idyllic place that occupies a central part of their personal and cultural pasts, and as a mediator between East (aged, urban and jaded) and West (wild, youthful, untamed). Middle West rests on yeomanry, an Old-World concept which Jefferson especially admired, embodying the virtues of maturity, wisdom, and kindness: “The pastoral ideal, a haven midway between the corruptions of urban civilization and …

Modelling Climate Effects On Site Productivity And Developing Site Index Conversion Equations For Jack Pine And Trembling Aspen Mixed Stands, Mahadev Sharma

Aspen Bibliography

Forest site productivity estimates are crucial for making informed forest resource management decisions. These estimates are valuable both for the tree species currently growing in the stands and for those being considered for future stands. Current models are generally designed for pure stands and do not account for the influence of climate on tree growth. Consequently, site index (SI) conversion equations were developed specifically for jack pine (Pinus banksiana Lamb.) and trembling aspen (Populus tremuloides Michx.) trees grown in naturally originated mixed stands. This work involved sampling 186 trees (93 of each species) from 31 even-aged mixed stands (3 …

Tumour-host immune interactions lead to complex changes in the tumour microenvironment (TME), impacting progression, metastasis and response to therapy. While it is clear that cancer cells can have the capacity to alter immune landscapes, our understanding of this process is incomplete. Herein we show that endocytic trafficking at the plasma membrane, mediated by the small GTPase ARF6, enables melanoma cells to impose an immunosuppressive TME that accelerates tumour development. This ARF6-dependent TME is vulnerable to immune checkpoint blockade therapy (ICB) but in murine melanoma, loss of Arf6 causes resistance to ICB. Likewise, downregulation of ARF6 in patient tumours correlates with …

Tolkien’S Orphaned Heroes: Kullervo, Húrin And The Limits Of Fostering, Deidre Dawson

Journal of Tolkien Research

Tolkien’s legendarium features many characters who have lost one or both parents through death or abandonment, and who are consequently fostered or adopted. While there are many positive depictions of fostering in the legendarium, the cases of Kullervo and especially Túrin Turambar stand out because of their similar tragic outcomes involving sibling incest followed by suicide. The contrast between the depiction of fostering in these stories is stark: The Story of Kullervo depicts a case of poor fostering involving violence and neglect, whereas The Children of Húrin describes a loving and nurturing foster environment for its hero, Túrin. Both tales …

Harnessing Cubesat Technology For Precision Pesticide Management In Egyptian Agriculture, Motaz El Sayed, Salem Abdou, Abdelhamid Sherif

Small Satellite Conference

This paper presents the development of an autonomous spraying robot designed to optimize agricultural practices through precise pesticide application, facilitated by the integration of real-time satellite data. The robot utilizes multispectral imaging from satellites, including NDVI and thermal data, to assess crop health and identify specific needs for intervention. The system aims to significantly reduce the volume of chemicals used in farming by targeting only the areas that require treatment, thus minimizing environmental impact while maintaining or enhancing crop yield. By employing advanced algorithms for data processing and machine operation, the robot autonomously navigates through fields, delivering pesticides in an …
