Investigation Of The Prevalence Of Fentanyl Analogs In A Drug Testing Population Using Liquid Chromatography Tandem Mass Spectrometry (Lc-Ms-Ms), Ivan Cordova

Student Theses

This study serves to determine the prevalence of novel fentanyl analogs that are present within a drug testing population in New York State. Samples were analyzed using liquid chromatography tandem mass spectrometry (LC-MS/MS) to qualitatively determine the presence of various possible fentanyl analogs. The sample preparation of this assay consisted of a simple dilute and shoot. This method of sample preparation can be quicker, eliminating sample preparation times as well as being less financially strenuous to clinical laboratories as it eliminates the need for various solvents and solid/liquid phase cartridges. The assay was aimed to see if analytes can be …

Degradable Polymers and Networks Based on Furan-Maleimide Diels-Alder Reactions of Silyl Ether Derivatives and Maleimides. I.

I. Part 1. Two groups of silyl ether monomers, A and B, were synthesized, each containing four different monomers. The monomers of group A were obtained by reacting furfuryl alcohol with four dichlorosilanes with varying siloxane spacers. The monomers of group B were obtained by reacting n-(2-hydroxyethyl)maleimide (HEMI-1c) with the same four dichlorosilanes. Each monomer from group A was individually reacted with each monomer from group B via a Diels-Alder reaction to produce a series of sixteen degradable linear polymers with a variety …

Immunological Feedback Loops Generate Parasite Persistence Thresholds That Explain Variation In Infection Duration, Clayton E. Cressler, Daniel C.G. Metz, David A. Chang Van Oordt, Andrea L. Graham Aug 2024

Immunological Feedback Loops Generate Parasite Persistence Thresholds That Explain Variation In Infection Duration, Clayton E. Cressler, Daniel C.G. Metz, David A. Chang Van Oordt, Andrea L. Graham

School of Biological Sciences: Faculty Publications

Infection duration affects individual host fitness and between-host transmission. Whether an infection is cleared or becomes chronic depends on the complex interaction between host immune responses and parasite growth. Empirical and theoretical studies have suggested that there are critical thresholds of parasite dose that can determine clearance versus chronicity, driven by the ability of the parasite to manipulate host immunity. However, the mammalian immune response is characterized by strong positive and negative feedback loops that could generate duration thresholds even in the absence of direct immunomodulation. Here, we derive and analyse a simple model for the interaction between T-cell subpopulations …

Stanniocalcin 2 Governs Cancer Cell Adaptation To Nutrient Insufficiency Through Alleviation Of Oxidative Stress, Shuo Qie, Haijuan Xiong, Yaqi Liu, Chenhui Yan, Yalei Wang, Lifeng Tian, Chenguang Wang, Nianli Sang

Department of Pharmacology, Physiology, and Cancer Biology Faculty Papers

Solid tumours often endure nutrient insufficiency during progression. How tumour cells adapt to temporal and spatial nutrient insufficiency remains unclear. We previously identified STC2 as one of the most upregulated genes in cells exposed to nutrient insufficiency by transcriptome screening, indicating the potential of STC2 in cellular adaptation to nutrient insufficiency. However, the molecular mechanisms underlying STC2 induction by nutrient insufficiency and subsequent adaptation remain elusive. Here, we report that STC2 protein is dramatically increased and secreted into the culture media by Gln-/Glc- deprivation. STC2 promoter contains cis-elements that are activated by ATF4 and p65/RelA, two transcription factors activated by …

Impacts Of Diets And Weight On Cognitive Decline Diseases Among The Middle-Aged Populations In Louisiana, Danielle Beamon

Walden Dissertations and Doctoral Studies

Cognitive decline is a public health concern since people are living longer, but little is known regarding how to prevent the progression of this condition; furthermore, current treatment options are not always effective after diagnosis. Previous research indicated obesity was associated with diminishing cognitive health, but most studies were conducted on individuals over the age of 65. The purpose of the study was to determine if there is a statistical correlation between cognitive decline and body mass index (BMI) among those between the ages of 40 and 64 years. Data from the 2015-2020 Behavioral Risk Factor Surveillance System Alzheimer’s Disease …

Using Phylogenomics And Herbarium Specimens To Evaluate Relationships In The Plant Genus Aneilema, Family Commelinaceae., Allan Sampao Loinyio

Masters Theses

The genus Aneilema, with approximately 65 species in seven sections, is one of the largest genera in the Commelinaceae. It is mostly distributed across Australia, Madagascar, and Africa, but one species occurs in South America. Early molecular phylogenetic analyses based on a few chloroplast genes and a small number of species have supported the monophyly of the genus with the inclusion of Rhopalephora, which was nested within Aneilema. This study used the Angiosperms353 gene set to investigate the evolutionary and taxonomic relationships within the genus and its close relatives. Through molecular phylogenetic analyses of 35 Aneilema species …

  • ‘Early detection and rapid response’ (EDRR) is the most successful framework for preventative invasive species management, but prioritizing localized EDRR actions with limited resources is challenging. An approach that ranks individual locations, such as waterbodies, for EDRR by combining an invasive species' establishment risk with the practicality of managing it could help set reasonable priorities.
  • Here, we worked with regional practitioners in Arkansas, USA, and the broader Southeastern USA to co-produce a workflow for preventative aquatic invasive species management that (1) estimates establishment risk under current and future climates with a species distribution model, (2) scores waterbodies according to difficulty …

A ‘Nuclear Bomb’ Or Just ‘A Joke’? Groundwater Models May Help Communicate Nuanced Risks To The Great Salt Lake, Matthew D. Laplante, Piyush Dahal, Shih-Yu Simon Wang, Kirsti Hakala, Avik Mukherjee

Plants, Soils, and Climate Faculty Publications

The Great Salt Lake entered the zeitgeist of environmental concern in 2022 when a coalition of scientists and activists warned in a highly publicized report that the lake might be just five years away from complete desiccation, a possibility one state official warned was tantamount to an “environmental nuclear bomb”. Shortly thereafter, an unpredicted and unprecedented pluvial winter resulted in an increase in inflow, temporarily halting the lake’s decline and prompting Utah’s governor to mock the dire prediction as “a joke”, an outcome that speaks to the tension between agenda-setting and trust-building that researchers face when sharing worst-case warnings, particularly …

An Agent-Based Modeling Of Permafrost Melting And Virus Exposure Due To Climate Change, Nishat Ara Mozid

Theses and Dissertations

Climate change has affected human life and natural system significantly. Particularly, there is specific evidence of change in the Arctic region for climate warming. One of the consequences of climate change is the process of permafrost degradation and thawing rate of active layer. Permafrost is defined as frozen grounds that exists at a temperature of or below 0℃ for at least two consecutive years, consists of ice and novel pathogens which are frozen for a long time. Anthrax may cause a serious threat for local environment and livestock due to permafrost melting. The study introduces an agent-based modeling of permafrost …

Can Simultaneous Integrated Boost Provide Better Dosimetric Coverage Compared To Sequential Boost In Prone Breast Treatment Of Large Breast Volumes?, Kinze Kraft

Masters Projects

Introduction: This study investigates the dosimetric differences between sequential boost (SeB) and simultaneous integrated boost (SIB) techniques in prone breast radiotherapy for patients with large breast volumes, utilizing a 3D-CRT planning technique. Prone positioning in radiotherapy has shown to be effective in reducing heart and lung doses, yet limited research exists to compare overall plan quality between SeB and SIB using this technique. This analysis aims to evaluate whether SIB provides better dosimetric coverage and reduced dose to organs at risk (OARs) compared to SeB in prone breast radiotherapy.

Methods: A retrospective analysis was conducted on data from ten patients …

Under climatic warming, glaciers are becoming a secondary source of atmospheric contaminants originally released into the environment decades ago. This phenomenon has been well-documented for glaciers near emission sources. However, less is known about polar ice sheets and ice caps. Radionuclides are one of the contaminants that can be remobilised through ice melting and accumulate in cryoconite material on the surface of glaciers. To understand the cycling of radionuclides in polar glacial contexts, we evaluate the radioactivity of cryoconite samples from Flade Isblink, a High Arctic ice cap in northeast Greenland. The measured radioactivity is among the highest reported across …

A key challenge in aging research is to extend lifespan in tandem with slowing down functional decline so that the life with good health (healthspan) can be extended. Here, we show that monthly clearance of a small number of cells, which highly express p21Cip1 (p21high), starting from 20 months improves cardiac and metabolic function, and extends both median and maximum lifespan in mice. Importantly, by assessing health and physical function of these mice monthly until death, we show that clearance of p21high cells improves physical function at all remaining stages of life, suggesting healthspan extension. Mechanistically, …

Somatic mutations in non-malignant tissues are selected for because they confer increased clonal fitness. However, it is uncertain whether these clones can benefit organ health. Here, ultra-deep targeted sequencing of 150 liver samples from 30 chronic liver disease patients revealed recurrent somatic mutations. PKD1 mutations were observed in 30% of patients, whereas they were only detected in 1.3% of hepatocellular carcinomas (HCCs). To interrogate tumor suppressor functionality, we perturbed PKD1 in two HCC cell lines and six in vivo models, in some cases showing that PKD1 loss protected against HCC, but in most cases showing no impact. However, Pkd1 haploinsufficiency …

Application Of Machine Learning For Multi-Omics Data Integration, Dhoha Abid Aug 2024

Application Of Machine Learning For Multi-Omics Data Integration, Dhoha Abid

McKelvey School of Engineering Graduate Student Theses & Dissertations

Traditionally, machine learning (ML) is used to train a model to predict scores for instances that were not seen by the model during training. In this conventional use of ML, the model is trained to learn general patterns that relate features to labels, so it can predict accurate scores for unseen data. Here, we use ML, unconventionally, to integrate different types of noisy data. Specifically, for biological investigations, in which there is no mean to measure a ground truth. We propose to train a ML model to predict scores on the same instances that were used in its training. In …

In the wild, C. elegans are emersed in environments teeming with a veritable menagerie of microorganisms. The C. elegans cuticular surface serves as a barrier and first point of contact with their microbial environments. In this study, we identify microbes from C. elegans natural habitats that associate with its cuticle, constituting a simple “skin microbiome.” We rear our animals on a modified CeMbio, mCeMbio, a consortium of ecologically relevant microbes. We first combine standard microbiological methods with an adapted micro skin-swabbing tool to describe the skin-resident bacteria on the C. elegans surface. Furthermore, we conduct 16S rRNA gene sequencing studies …

Sequence-based genetic testing identifies causative variants in ~ 50% of individuals with developmental and epileptic encephalopathies (DEEs). Aberrant changes in DNA methylation are implicated in various neurodevelopmental disorders but remain unstudied in DEEs. We interrogate the diagnostic utility of genome-wide DNA methylation array analysis on peripheral blood samples from 582 individuals with genetically unsolved DEEs. We identify rare differentially methylated regions (DMRs) and explanatory episignatures to uncover causative and candidate genetic etiologies in 12 individuals. Using long-read sequencing, we identify DNA variants underlying rare DMRs, including one balanced translocation, three CG-rich repeat expansions, and four copy number variants. We also …

Social network analysis is becoming more relevant with the rise of Large Language Models and other AI frameworks, particularly as online content creation surges. However, analyzing social network data from different platforms to study online opinions and influence campaigns remains challenging. Even more complex is the concept of multi-source social network analysis. Knowledge graphs, an essential part of Web 4.0 and crucial in information modeling and encoding, offer a unique solution. They allow for a more concise encoding of relationships between entities and related discussions. However, there is limited investigation into using multi-source data for examining online information campaigns and …

Potential Contributions Of Xenobiotics To P-Glycoprotein Clearance Of Amyloid Β-Peptide From The Brain, Joseph Jr Asante

Theses and Dissertations

Alzheimer’s disease (AD) is an example of a neurodegenerative disease, mostly age related and characterized by cognitive decline. Currently there is no effective treatment that can prevent or slow progression of the disease. Roughly 95% of cases are sporadic, with unknown etiology. A distinguishing characteristic of the Alzheimer’s brain is the presence of amyloid plaques, with their constituent amyloid-β (Aβ) peptides implicated in causing synaptic dysfunction, memory impairment, and neurotoxicity. Although extensive research has explored the role of Aβ peptides in AD, understanding their metabolism in health versus disease remains insufficient. P-glycoprotein (P-gp), a member of the ATP-binding cassette transporter …
