Zinc And Diabetes: A Connection Between Micronutrient And Metabolism, Rahnuma Ahmad, Ronald A. Shaju, Azeddine Atfi, Mohammed S. Razzaque

School of Medicine Publications

Diabetes mellitus is a global health problem and a major contributor to mortality and morbidity. The management of this condition typically involves using oral antidiabetic medication, insulin, and appropriate dietary modifications, with a focus on macronutrient intake. However, several human studies have indicated that a deficiency in micronutrients, such as zinc, can be associated with insulin resistance as well as greater glucose intolerance. Zinc serves as a chemical messenger, acts as a cofactor to increase enzyme activity, and is involved in insulin formation, release, and storage. These diverse functions make zinc an important trace element for the regulation of blood …

Development Of A Selective Novel Fluorescent Substrate For Sodium-Dependent Transporters, Richard S. Agnes, Raymond F. Muzic Jr.

Faculty Scholarship

Aim: To synthesize, characterize, and validate 6FGA, a fluorescent glucose modified with a Cyanine5.5 at carbon-6 position, for probing the function of sodium-dependent glucose transporters, SGLT1 and SGLT2. Main methods: The synthesis of fluorescent glucose analogue was achieved through “click chemistry” of Cyanine5.5-alkyne and 6-azido-6-deoxy-D-glucose. Cell system studies were conducted to characterize the in vivo transport properties. Key findings: Optical analyses revealed that 6FGA displayed similar spectral profiles to Cyanine5.5 in DMSO, allowing for concentration determination, thus supporting its utility in quantitative kinetic studies within biological assays. Uptake studies in cell system SGLT models, LLC-PK1 and HEK293 cells, exhibited concentration …

Electric Field-Induced Clustering In Nanocomposite Films Of Highly Polarizable Inclusions, Elshad Allakhyarov Aug 2024

Electric Field-Induced Clustering In Nanocomposite Films Of Highly Polarizable Inclusions, Elshad Allakhyarov

Researchers, Instructors, & Staff Scholarship

A nanocomposite film containing highly polarizable inclusions in a fluid background is explored when an external electric field is applied perpendicular to the planar film. For small electric fields, the induced dipole moments of the inclusions are all polarized in field direction, resulting in a mutual repulsion between the inclusions. Here we show that this becomes qualitatively different for high fields: the total system self-organizes into a state which contains both polarizations, parallel and antiparallel to the external field such that a fraction of the inclusions is counter-polarized to the electric field direction. We attribute this unexpected counter-polarization to the …

Deep Learning For Source Localization In A Laboratory Tank, Corey Emerson Dobbs Aug 2024

Deep Learning For Source Localization In A Laboratory Tank, Corey Emerson Dobbs

Theses and Dissertations

Deep learning has been applied to underwater acoustics problems in many forms. One difficulty in applying deep learning techniques to ocean acoustics is the spatially and temporally varying environmental properties. Another challenge is the lack of labeled data for training large networks. The overall goal of this work is to develop deep learning approaches for source localization that can be adaptable to different conditions. In this work, a convolutional neural network was trained on acoustic data measured in a water tank, while the water was at room temperature, to predict source-receiver range. Tests were done to ascertain ideal quantities of …

Hematopoietic Growth Factors Regulate The Entry Of Monocytes Into The Adult Brain Via Chemokine Receptor Ccr5, Xuefang Sophie Ren, Junchi He, Songruo Li, Heng Hu, Michele Kyle, Shinichi Kohsaka, Li-Ru Zhao

Faculty, Staff and Student Publications

Monocytes are circulating macrophage precursors generated from bone marrow hematopoietic stem cells. In adults, monocytes continuously replenish cerebral border-associated macrophages under physiological conditions. Monocytes also rapidly infiltrate the brain in pathological settings. The mechanisms of recruiting monocyte-derived macrophages into the brain under pathological conditions have been extensively studied. However, it remains unclear how monocytes enter the brain to renew border-associated macrophages under physiological conditions. Using both in vitro and in vivo approaches, this study reveals that a combination of two hematopoietic growth factors, stem cell factor (SCF) and granulocyte colony-stimulating factor (G-CSF), complementarily and synergistically enhances the adhesion of monocytes …

Driving Green Investments To Address Climate Change, Baptiste Gibrat Aug 2024

Driving Green Investments To Address Climate Change, Baptiste Gibrat

Dartmouth College Master’s Theses

The climate crisis is no longer news, and society needs to accelerate the transition to net zero emissions. Otherwise, the consequences of climate change will become increasingly catastrophic.

This thesis develops a framework to drive green investments, which reduce society’s dependance on fossil fuels. Initially, the research provides a comprehensive examination of these investments, followed by an exploration of three strategies to enhance their adoption: leveraging financial instruments and capital markets, implementing regulatory frameworks, and harnessing the power of technologies. To integrate these elements within a comprehensive framework, the study employs market research, reviews relevant literature, and includes interviews with …

Linking Geospatial Trends And Geochemical Fingerprints To Anthropogenic Phosphorus Dust Deposition In Subalpine Soils, Austen Michael Lambert Aug 2024

Linking Geospatial Trends And Geochemical Fingerprints To Anthropogenic Phosphorus Dust Deposition In Subalpine Soils, Austen Michael Lambert

Theses and Dissertations

This study shows that fugitive emissions associated with the surface mining of phosphate rock are a significant phosphorus (P) source to surface soils in northeastern Utah. Phosphorus is a nonrenewable resource essential for all life. Anthropogenic alterations to the P-cycle have led to widespread P pollution, and the unsustainable management of P has led to the threat of global depletion of P resources. Thus, accounting for both natural and anthropogenic flow paths of P is essential for the conservation and pollution reduction of P. One such source of human alteration to the P-cycle is the mining of phosphate rock. Mining, …

The rise of cutting-edge precision cancer treatments has led to a growing significance of the optimal biological dose (OBD) in modern oncology trials. These trials now prioritize the consideration of both toxicity and efficacy simultaneously when determining the most desirable dosage for treatment. Traditional approaches in early-phase oncology trials have conventionally relied on the assumption of a monotone relationship between treatment efficacy and dosage. However, this assumption may not hold valid for novel oncology therapies. In reality, the dose-efficacy curve of such treatments may reach a plateau at a specific dose, posing challenges for conventional methods in accurately identifying the …

LCS-1, a putative selective inhibitor of SOD1, is a substituted pyridazinone with rudimentary similarity to quinones and naphthoquinones. As quinones catalytically oxidize H2S to biologically active reactive sulfur species (RSS), we hypothesized LCS-1 might have similar attributes. Here, we examine LCS-1 reactions with H2S and SOD1 using thiol-specific fluorophores, liquid chromatography–mass spectrometry, electron paramagnetic resonance (EPR), UV–vis spectrometry, and oxygen consumption. We show that LCS-1 catalytically oxidizes H2S in buffer solutions to form RSS, namely per- and polyhydrosulfides (H2Sn, n = 2–6). These reactions consume oxygen and produce hydrogen peroxide, but they do not have an EPR signature, nor do …

In response to ongoing coastal urbanization, it is critical to develop effective methods to improve the biodiversity and ecological sustainability of artificial shorelines. Enhancing the topographic complexity of coastal infrastructure through the mimicry of natural substrata may facilitate the establishment of ecosystem engineering species and associated biogenic habitat formation. However, interactions between ecosystem engineers and their substratum are likely determined by organismal size and resource needs, thus making responses to topography highly scale-dependent. Here, we assessed the topographic properties (rugosity, surface area, micro-surface orientations) that underpin the abundance and distribution of two ecosystem engineers (fucoids, limpets) across six spatial scales …

Entropy measures are increasingly being used to analyze the structure of neural activity observed by functional magnetic resonance imaging (fMRI), with resting-state networks (RSNs) being of interest for their reproducible descriptions of the brain's functional architecture. Temporal correlations have shown a dichotomy among these networks: those that engage with the environment, known as extrinsic, which include the visual and sensorimotor networks; and those associated with executive control and self-referencing, known as intrinsic, which include the default mode network and the frontoparietal control network. While these inter-voxel temporal correlations enable the assessment of synchrony among the components of individual networks, entropic …

Targeting Aspergillus Fumigatus Hypoxia Response Pathways For Novel Antifungal Drug Development., Cecilia Gutierrez Perez

Dartmouth College Ph.D Dissertations

Invasive fungal disease present are difficult to diagnose and treat and present a high mortality rate across the world with approximately 1.5 million people a year succumbing to fungal infections worldwide. Currently, there are limited antifungal classes used therapeutically, and antifungal drug resistance on the rise further exacerbates the dire need for novel therapeutics with innovative mode of actions. The filamentous fungi, Aspergillus fumigatus is the causative agent of invasive pulmonary aspergillosis (IPA). It is estimated that contemporary antifungals fail in about 50% of patients with invasive filamentous fungal infections, likely due to the biofilm mode of growth exhibited in …

Effective interplay between the uterus and the embryo is essential for pregnancy establishment; however, convenient methods to screen embryo implantation success and maternal uterine response in experimental mouse models are currently lacking. Here, we report 3DMOUSEneST, a groundbreaking method for analyzing mouse implantation sites based on label-free higher harmonic generation microscopy, providing unprecedented insights into the embryo-uterine dynamics during early pregnancy. The 3DMOUSEneST method incorporates second-harmonic generation microscopy to image the three-dimensional structure formed by decidual fibrillar collagen, named 'decidual nest', and third-harmonic generation microscopy to evaluate early conceptus (defined as the embryo and extra-embryonic tissues) growth. We demonstrate that …

Enhancing Accountability In Cyberspace Through A Three-Tiered International Governance Regime, Dan Efrony Aug 2024

Enhancing Accountability In Cyberspace Through A Three-Tiered International Governance Regime, Dan Efrony

International Law Studies

The Great Power Competition perpetuates the inability to reach a universal consensus on how to resolve normative ambiguity on the application of international law to cyberspace. Adhering to a strategy of ambiguity, the United States and its closest allies have been holding their rival States accountable for “irresponsible State behavior” in cyberspace, based on flawed legitimacy, as reflected in a weakened normative layer, and the national U.S. attribution process. Embracing collective attribution has not cured the flaws. The upshot is a poor framework for holding States accountable and an enduring vicious cycle. This article calls on the United States and …
